1.Bioinformatics analysis and experimental validation of BPS and BPF on signaling pathways of male reproductive system damage

Objective:To investigate the pathways involved in bisphenol S(BPS)and bisphenol F(BPF)induced male reproductive injury by bioinformatics methods and experimental verification.Methods:Bioinformatics analysis was conducted to screen the genes related to male reproductive system diseases associated with BPF and BPS from the Comparative Toxicogenomics Database(CTD).Functional enrichment using Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed to predict potential signaling pathways and key genes.Cell counting kit-8(CCK-8)method was used to assess the cell viability in various groups treated with different concentrations of BPS and BPF(1×10-3,1×10-2,1×10-1,1×100,1×101,and 1×102 μmol·L-1).TM3 cells were divided into control group(0.1%DMSO),different doses of BPS groups,and different doses of BPF groups.The cells were treated with 20,40,and 80 μmol·L-1 of BPS and BPF for 72 h,respectively.Real-time fluorescence quantitative PCR(RT-qPCR)and Western blotting method were used to detect the expression levels of key genes mRNA and proteins in various groups.Results:The bioinformatics analysis results revealed that 507 and 447 male systemic disease genes related to BPS and BPF were screened by CTD,respectively.The GO enrichment analysis results showed that the selected genes were primarily enriched in biological processes(BP)such as reproductive system development and reproductive structure development.The KEGG pathway analysis results indicated that these genes were significantly enriched in signaling pathways such as phosphatidylinositol 3-kinase/protein kinase B(PI3K/AKT),hypoxia-inducible factor-1(HIF-1),and cellular senescence(P<0.001).The CCK-8 method results showed that compared with control group,the cell viabilities in 1× 102 μmol·L-1 BPF and BPS groups were significantly decreased(P<0.05),while the viabilities of TM3 cells in other groups had no significant changes(P>0.05).After BPS treatment,compared with control group,the expression levels of PI3K,AKT,hypoxia-inducible factor-1α(HIF-1α),and CREB-binding protein(CBP)mRNA in low,medium,and high doses of BPS groups were decreased(P<0.05),the expression levels of PI3K protein were decreased(P<0.05),the expression levels of B-cell lymphoma-2-associated X protein(Bax)protein were increased(P<0.05),and the expression levels of serine protease inhibitor clade B,member 10(SERPINB10)mRNA were increased(P<0.01);the expression levels of Bax and intraflagellar transport 80 homolog(IFT80)mRNA in the cells in medium and high doses of BPS groups were increased(P<0.05);the expression levels of B-cell lymphoma-2(Bcl-2)mRNA and protein in low and high doses of BPS groups were decreased(P<0.05);the expression levels of additional sex combs like 2(ASXL2)mRNA in low and medium doses of BPS groups were decreased(P<0.01).After BPF treatment,compared with control group,the expression levels of Bcl-2,HIF-1α,and structural maintenance of chromosomes protein 1B(SMC1B)mRNA in low,medium,and high doses of BPF groups were decreased(P<0.05),and the expression levels of IFT80 mRNA(P<0.01)and Bax protein(P<0.01)were increased;the expression levels of PI3K,AKT,and ring finger protein 130(RNF130)mRNA in low and high doses of BPF groups were decreased(P<0.05);the expression level of CBP mRNA in medium dose of BPF group was decreased(P<0.05),while the expression level of RNF130 mRNA was increased(P<0.05);the expression levels of PI3K and Bcl-2 proteins in high dose of BPF group were decreased(P<0.05).Conclusion:BPF and BPS may cause cell cytotoxicity and impair male reproductive health through PI3K/AKT and HIF-1 signaling pathways.RNF130 and SMC1B may be important targets for their induction of male reproductive toxicity.
2.Development and evaluation on reliability and validity of a sense of gain scale for the elderly based on classical test theory and optimal test assembly

Objective:To develop a scale to measure the sense of gain of the elderly, and to evaluate its reliability and validity.Methods:According to the content of the sense of gain, combined with the literature research method and interview method, the scale entry pool was established.After the expert consistency evaluation and the optimal test assembly method (OTA) analysis, the formal version of the scale was formed.The stratified sampling method was used to select 1 043 community-dwelling elders for the survey, and the reliability and validity of the scale were evaluated by methods based on classical test theory.Results:The scale of elderly sense of gain, which was screened by OTA method, included four dimensions: health status, relationship and communication, social security, and ideal expectation, with a total of 17 items. The Cronbach's α value of the formal version of the scale was 0. 850, and the Cronbach's α coefficients of the four dimensions were 0.721, 0.772, 0.779, and 0.930, respectively. The CR values of the combined reliabilities of each dimension were all above 0.7. In terms of AVE values, except for the health status dimension was acceptable, the other three dimensions were all above 0.36. The correlation coefficient between the sense of health acquisition and life satisfaction of the elderly was 0.531 ( P<0.01). Conclusion:The developed scale for measuring the sense of gain of the elderly has good reliability and validity, which can be used as an effective tool for measuring the sense of gain of the elderly.
3.Quantile regression and random forest analysis on influencing factors of the subjective well-being of the elderly

Objective:To explore the influencing factors of the subjective well-being of elderly individuals and improve the accuracy of well-being predictions.Methods:A multi-stage stratified sampling method was conducted in Guangdong province and Guangxi Zhuang autonomous region from August 2019 to October 2020. The health status and subjective well-being of older adults were assessed using a self-administered multidimensional health questionnaire, while overall well-being was evaluated with a single-item 5-point scale. The statistical analysis was performed using R software (version 4.3.2), with analytical methods including χ2 tests, univariate analysis, quantile regression and random forest model. Results:A total of 2 620 participants were recruited, and 70.9% of whom reported feeling happy.The depression symptom score of the elderly was 3.00(1.00, 6.00), emotional status score was (28.26±4.14), activities of daily living (ADL) score was 69.00(52.00, 81.00), and the body mass index(BMI) was (22.68±3.81)kg/m 2.The quantile regression results indicated that marital status (married)(Q 10: β=0.049; Q 50: β=0.014; Q 90: β=0.005) and depressive symptoms (no depression, depressive tendency)(Q 10: β=1.000; Q 50: β=1.000; Q 90: β=0.025) had varying effects on older adults' subjective well-being across three quantiles (Q 10, Q 50, Q 90)(all P<0.05). In contrast, social activity (no social activity)(Q 10: β=-0.072; Q 50: β=-0.011; Q 90: β=-0.006), ADL (severe disability, moderate disability, mild disability)(Q 10: β=-0.103, Q 50: β=-0.006, Q 90: β=-0.048; Q 10: β=-0.063, Q 50: β=-0.012, Q 90: β=-0.005; Q 10: β=-0.078, Q 50: β=-0.016, Q 90: β=-0.009), and emotional status (poor emotion, fair emotion)(Q 10: β=-0.261, Q 50: β=-0.009, Q 90: β=-0.025; Q 10: β=-0.140, Q 50: β=-0.017, Q 90: β=-0.029) had a negative impact on elderly subjective well-being at the three quantile points(all P<0.05). Random forest results indicated that depressive symptoms, ADL, emotional status, household monthly income per capita, BMI, social activity, multimorbidity status, and marital status were the most important determinants of subjective well-being. Conclusion:Depressive symptoms and emotional status have a significant impact on the subjective well-being of older adults. Additionally, ADL, household income per capita, BMI, and social engagement also play important roles in influencing well-being, while the emotional support provided by marriage should not be overlooked.
4.Comparative analysis of medium-to-long-term effects of Ceramage? polymer ceramic composite resoin onlay and CAD/CAM fabricated all-ceramic onlays in restoring endodontically treated posterior teeth

Objective:To compare the midium-to-long-term clinical outcomes of Ceramage? polymer ceramic composite resoin on-lay and CAD/CAM fabricated all-ceramic onlays in the restoration of endodontically treated posterior teeth.Methods:A total of 75 patients with 92 endodontically treated posterior teeth exhibiting class Ⅱ defects were included and randomly divided into two groups:the Ceramage? composite resin group(48 teeth)and the CAD/CAM fabricated all-ceramic group(44 teeth).The respective restora-tions were performed,and patients were followed up for 24 months.Clinical outcomes of the restorations and periodontal conditions were evaluated using the modified USPHS criteria,and patient satisfaction was assessed.Results:After 24 months of follow-up,the Ceramage? composite resin group showed higher rates of secondary caries,marginal discrepancy,and marginal discoloration,as well as higher plaque index,gingival index,and sulcus bleeding index compared to the CAD/CAM fabricated all-ceramic group.Howev-er,the Ceramage? group exhibited lower rates of restoration wear and fracture(P<0.05).Patient satisfaction was similar between the two groups(P>0.05).Conclusion:Both Ceramage? polymer ceramic composite resoin onlay and CAD/CAM fabricated all-ce-ramic onlays achieved favorable mid-to-long-term clinical outcomes.The choice of restoration method should comprehensively con-sider the patient's oral condition and restorative needs.

8.Clinical eficacy of customized titanium mesh combined with concentrated growth factors for restoration of anterior alveolar ridge defects

Objective:To observe the clinical efficacy of customized titanium mesh combined with concentrated growth factors(CGF)for restoration of anterior alveolar ridge defects.Methods:105 patients with anterior alveolar ridge defects were included and underwent implant restoration.The patients were classified into 2 groups by random number table method,those in control group(n=53)received customized titanium mesh,in study group(n=52)received customized titanium mesh combined with CGF.After 1 year follow-up,the success rate of implantation,alveolar ridge height,alveolar ridge bone augmentation,alveolar bone mineral density,bone thickness,aesthetic results and complications were compared between the 2 groups.Results:In 1-year follow-up,no implant loosening and shed-ding occurred in the 2 groups,and the success rate of implantation was 100%.The height of the alveolar ridge,the horizontal bone aug-mentation,the alveolar bone mineral density and bone thickness of study group were greater than those of the control group(P<0.05).Study group scored higher on pink aesthetic/white aesthetic score(PES/WES)than control group(P<0.05).In study group,there were 2 cases of postoperative swelling and pain,2 cases of periodontal infection,1 case of gingival bleeding and 1 of allergy.In control group,there was 1 case of postoperative swelling,1 of pain,1 of gingival bleeding,1 of allergy and 2 of periodontal infection(P>0.05).Conclusion:Application of customized titanium mesh combined with CGF for restoration of anterior alveolar ridge defects can improve the height of the alveolar ridge and bone metabolism,ensure the aesthetic results.
9.Analysis of Influencing Factors on the Sense of Gain for Elderly Chronic Patients based on Ordered Logistic Regression and Decision Tree Methods

Objective To understand the sense of gain of elderly chronic patients,explore their influencing factors,and provide a basis for the subsequent enhancement of the sense of gain of elderly chronic patients.Methods The data for this study were obtained from a survey of elderly chronic patients in the affiliated hospital of Guangdong Medical University.Convenience sampling was used to survey 380 elderly chronic patients with hypertension,coronary heart disease and benign prostatic hyperplasia treated in the hospital.The questionnaire included sense of gain,demographic factors,physical function,psychological function,and social function,etc.One-way multi-sample rank test,Spearman′s correlation analysis,multifactorial ordered logistic regression model and CHAID categorical decision tree analysis were performed using SPSS 26.0.Results Among the 380 older chronically ill people surveyed,2.9%had no sense of gain at all,40.5%had unclear or moderate sense of gain,and 66.6%had a very strong sense of gain.The results of multifactorial ordered logistic regression showed marital status,education level,social support and spiritual pillars are associated with sense of gain with all above variables being statistically significant(P<0.05).The OR values for unmarried,below junior high school level,high school and specialist,able to interact with others as they did before they got sick,spiritual pillars to make them no/a little feel the meaning of life,and limited moderate activities are 0.318,0.172,0.307,0.500,0.145,0.407,0.545,respectively,indicating relatively low sense of gain.The elderly chronic patients who usually did not feel depressed or sad,and did not have financial difficulties in their families due to their illness and treatment had a higher sense of gain with OR values being 3.042 and 1.988,respectively.The results of categorical decision tree analysis showed that whether or not spiritual support can make feel the meaning of life or the value of life,whether or not interact with other people as did before the disease,whether or not the disease has caused family′s financial difficulties,and whether or not have limitations on moderate activities are the most important factors influencing the sense of gain of the elderly chronic patients.Conclusion It is necessary to pay attention to the mental aspects of the elderly chronic patients and maintain interpersonal communication,and take targeted measures to help them reduce the degree of activity limitation as much as possible,so as to enhance the sense of gain of the elderly chronic patients.
10.O-arm navigation assisted hemivertebra resection and bone grafting and internal fixation for the congenital cervicothoracic hemivertebra

Objective:To investigate the clinical efficacy and precautions of O-arm combined with navigation-assisted steotomy and hemivertebra resection for congenital cervicothoracic hemivertebra.Methods:From February 2016 to October 2020, the clinical data of 12 patients with cervicothoracic hemivertebra admitted in Henan Provincial People's Hospital were retrospectively analyzed, including 5 males and 7 females, aged 9.4±2.6 years (range, 4-15 years). Intraoperative neural monitoring system was used to ensure the safety of surgical correction process and O-arm navigation system assisted the implantation of pedicle screws,hemivertebra resection, and scoliosis deformity correction. Postoperative CT was used to evaluate the accuracy of screw placement, and routine preoperative and postoperative X-ray films of the full-length spine in standing position were taken to measure the coronal and sagittal Cobb angles. The correction rate of scoliosis and kyphosis, internal fixation, shoulder height difference and bone graft fusion were calculated at the final follow-up.Results:A total of 108 pedicle screws were inserted in 12 patients, and the screw placement accuracy rate was 96.3% (104/108). The follow-up time was 37.9±10.2 months (range, 24-61 months). The number of fused segments was 5.4±1.1 (range, 4-7). One week after surgery, the correction rate of Cobb angle was 78.5%±3.2% for scoliosis and 70.1%±5.4% for kyphosis. There were statistically significant differences in side and kyphosis Angle and Scoliosis Research Society (SRS)-22 score between preoperative and 1 week after surgery ( P<0.05). There was no significant difference between the operation and the last follow-up ( P>0.05). At the last follow-up, all the 12 patients achieved gradeⅠ fusion. SI was 2.4±0.8 cm before operation, 1.0±0.6 cm at 1 week after operation, and 0.7±0.5 cm at last follow-up, and the difference was statistically significant ( F=38.30, P<0.001). No pseudojoint formation, significant loss of correction Angle, or rupture of internal fixation relaxant occurred during the operation or during follow-up. Conclusion:O-arm combined with navigation-assisted steotomy and hemivertebra resection for the treatment of congenital cervicothoracic hemivertebra has the advantages of good orthopedic effect, reduced radiation exposure and fewer complications, and accurate pedicle screw implantation and hemivertebra resection.
