Lutetium-177 (177Lu)-prostate specific membrane antigen (PSMA)-617 is a small-molecule radioligand therapy (RLT) drug targeting PSMA. By selectively delivering the β- radiation emitted by 177Lu to PSMA-positive prostate cancer cells, it induces tumor cell death. The agent has been approved in multiple nations and regions for the treatment of PSMA-positive metastatic castration-resistant prostate cancer, thereby expanding therapeutic options for this patient population. This review outlines the development, pharmacological properties, and current clinical applications of 177Lu-PSMA-617, aiming to provide a theoretical basis for the clinical practice of RLT in prostate cancer treatment.

BACKGROUND:Osteoarthritis is pathologically characterized by progressive degeneration of the articular cartilage and abnormal deformation of the subchondral bone.In recent years, with the deepening of medical research,it has been found that the mitogen-activated protein kinases(MAPK)signaling pathway has a regulatory role in inflammatory cell infiltration, inflammatory factor release, and chondrocyte proliferation, which is particularly important for the treatment of osteoarthritis.OBJECTIVE:To briefly review the main research progress in the mechanism of MAPK signaling pathway regulating osteoarthritis in recent years, aiming to provide new ideas for the treatment of osteoarthritis.METHODS:CNKI,WanFang and PubMed databases were searched for relevant literature using the search terms of"mitogen-activated protein kinases, osteoarthritis, extracellular signal-regulated MAP kinases,p38 mitogen-activated protein kinases,JNK mitogen-activated protein kinase"in Chinese and English.Relevant literature published from January 2019 to November 2024 was searched, and 108 articles were finally included for summary analysis.RESULTS AND CONCLUSION:(1)Various stimuli inside and outside the cells activate the MAPK signaling pathway, regulate gene transcription and protein synthesis, and promote the release of inflammatory factors, such as tumor necrosis factor-α,interleukin-1β,and interleukin-6.The release of these inflammatory factors aggravates the progression of osteoarthritis.(2)The active ingredients of traditional Chinese medicine, mainly saponins and flavonoids,as well as Chinese herbal formulas and preparations with the main effects of activating blood circulation and removing blood stasis, tonifying the liver and kidney, can play a therapeutic role in osteoarthritis by inhibiting the MAPK signaling pathway, regulating the release of matrix metalloproteinases, balancing the homeostatic state of osteogenesis and osteoblastogenesis, attenuating the synovial inflammation, decreasing the release of inflammatory factors and inflammatory vesicles, decreasing cellular pyroptosis, promoting autophagy, and ameliorating oxidative stress.(3)Although traditional Chinese medicine has become popular in the treatment of osteoarthritis by virtue of its own advantages of multi-components,multi-targets,multi-pathways,and low side effects, the use of MAPK signaling pathway to guide the treatment of individual osteoarthritis is the difficulty of the technology, which needs to be continuously researched and explored.(4)Therefore,further development of relevant herbal inhibitors that can modulate the MAPK signaling pathway may be a potential drug strategy for the treatment of osteoarthritis in the future.

ObjectiveTo investigate growth hormone secretagogue receptor (GHSR) rs2922126 gene polymorphisms and their association with genetic susceptibility to nonalcoholic fatty liver disease (NAFLD) in the Chinese Han population in Qingdao, China, and to provide a basis for diagnosis and treatment. MethodsA total of 220 patients who were admitted to Qingdao Municipal Hospital from June 2022 to June 2023 and were diagnosed with NAFLD based on radiological examination were enrolled as NAFLD group, and 167 healthy individuals during the same period of time were enrolled as control group. Fasting blood samples were collected from all subjects, and related biochemical parameters were measured. Whole blood DNA was extracted, and polymerase chain reaction and MALDI-TOF mass spectrometer were used for genotyping. The chi-square test was used for comparison of categorical data between groups, and the independent-samples t test or the Mann-Whitney U test was used for comparison of continuous data between groups. The binary logistic regression analysis was used to investigate the risk of NAFLD. ResultsCompared with the control group, the NAFLD group had significantly higher age, body mass index (BMI), fasting plasma glucose, triglyceride, gamma-glutamyl transpeptidase, alkaline phosphatase, alanine aminotransferase, and aspartate aminotransferase, as well as a significantly lower level of high-density lipoprotein (all P0.05). The distribution of GHSR rs2922126 genotypes was consistent with the Hardy-Weinberg equilibrium, suggesting population representativeness in the subjects enrolled (NAFLD group: P=0.106; control group: P=0.849). There was no significant difference in the distribution of AA, TA, and TT genotypes at GHSR rs2922126 locus between the control group and the NAFLD group (P=0.099), and there was also no significant difference in allele frequency between the two groups (P=0.063). In the recessive model of A allele, there was a significant difference in the distribution of AA homozygote and TA+TT genotype between the NAFLD group and the control group (P=0.032). The binary logistic regression analysis showed that in the recessive model of A allele, AA homozygote carriers had an increased risk of NAFLD compared with TA+TT genotype carriers (odds ratio [OR]=1.712, 95% confidence interval [CI]: 1.045 — 2.807, P=0.033). There was still a significant difference after adjustment for sex, age, and BMI (OR=2.156, 95%CI: 1.221 — 3.808, P=0.008). In the NAFLD group, AA genotype carriers had a significantly higher serum level of total cholesterol (TC) than TT+TA carriers (Z=-1.99,P=0.046). ConclusionGHSR rs2922126 AA genotype may be associated with the increased risk of NAFLD in the Chinese Han population in Qingdao, and GHSR rs2922126 AA genotype is associated with the increase in TC in NAFLD patients.

The translocator protein (TSPO) positron emission tomography (PET) can noninvasively detect neuroinflammation associated with epileptogenesis and epilepsy. This study explored the role of the TSPO-targeting radioligand [18F]F-TFQC, an m-trifluoromethyl ER176 analog, in the PET neuroimaging of epileptic rats. Initially, [18F]F-TFQC was synthesized with a radiochemical yield of 8%-10% (EOS), a radiochemical purity of over 99%, and a specific activity of 38.21 ± 1.73 MBq/nmol (EOS). After determining that [18F]F-TFQC exhibited good biochemical properties, [18F]F-TFQC PET neuroimaging was performed in epileptic rats at multiple time points in various stages of disease progression. PET imaging showed specific [18F]F-TFQC uptake in the right hippocampus (KA-injected site, i.e., epileptogenic zone), which was most pronounced at 1 week (T/NT 1.63 ± 0.21) and 1 month (T/NT 1.66 ± 0.20). The PET results were further validated using autoradiography and pathological analysis. Thus, [18F]F-TFQC can reflect the TSPO levels and localize the epileptogenic zone, thereby offering the potential for monitoring neuroinflammation and guiding anti-inflammatory treatment in patients with epilepsy.

Objective:
Previous studies have discovered a correlation between cigarette smoking and cortical thickness and surface area, but the causal relationship remains unclear. The objective of this investigation is to scrutinize the causal association between them.
Methods:
To derive summary statistics from a genome-wide association study (GWAS) on cortical thickness, surface area, and four smoking behaviors: 1) age of initiation of regular smoking (AgeSmk); 2) smoking initiation (SmkInit); 3) smoking cessation (SmkCes); 4) cigarettes per day (CigDay). Linkage disequilibrium score regression (LDSC) was employed to examine genetic association analysis. Furthermore, for traits with significant genetic associations, Mendelian randomization (MR) analyses were conducted.
Results:
The LDSC analysis revealed nominal genetic correlations between AgeSmk and right precentral surface area, left caudal anterior cingulate surface area, left cuneus surface area, left inferior parietal surface area, and right caudal anterior cingulate thickness, as well as between CigDay and left caudal middle frontal surface area, between SmkCes and left entorhinal thickness, and between SmkInit and left rostral anterior cingulate surface area, right rostral anterior cingulate thickness, and right superior frontal thickness (rg=-0.36–0.29, p<0.05). MR analysis showed a unidirectional causal association between left caudal middle frontal surface area and CigDay (βIVW=0.056, pBonferroni=2×10-4).
Conclusion
Left caudal middle frontal surface area has the potential to serve as a significant predictor of smoking behavior.

China Medical Equipment 2025;22(2):104-108
Objective:To explore the relationship between the stability of the pulse perfusion index(PI)value and Pulse oximetry(SpO2)alarms in monitor for neonates,so as to reduce the false alarm rate. Methods:A monitor with dual blood oxygen module(Module 1 and Module 2)was used to simultaneously monitor different parts of neonates. If both modules alarmed simultaneously,it was considered as a valid alarm. If only one module alarmed while the others was normal,it was considered as an invalid alarm. The PI values were classified into A,B and C categories based on the calculated variance of PI values. The distribution of valid alarms in the three categories was analyzed by using statistical method. The eighteen neonates who admitted to the neonatal department of Shanxi Children's Hospital from May11,2023 to May 19,2023 were monitored, with an average monitoring time of 7.3 hours for each case. The false alarm rates of the monitor with dual blood oxygen module were compared among categories A,B and C,as well as the correlations between different PI values and SpO2 alarms. Results:The times of monitored valid alarm of two non-invasive blood oxygen modules(Module 1 and Module 2)were 41 times, and the false alarm rates of categories A and C were significantly higher than that of category B.The times of categories A,B and C of the monitored PI values of Module 1 and Module 2 were respectively(6,34,1)and(8,30,3),which appeared weak correlation with the times of valid alarm of SpO2(r=0.230,0.259,P<0.05).The result of univariate binary logistic regression indicated PI value could not predict SpO2 alarms in SpO2 alarm. Conclusion:PI value can reflect the affected accuracy of SpO2 alarm when the data that are collected by prober are instability. The PI value that is collected by single SpO2 prober can indirectly reflect that the instable data of collection can affect the correctness of SpO2 alarm, while the simultaneous monitor of adopting monitor with dual oxygen module can improve the accuracy of SpO2 alarms in monitoring for neonates.
