Objective To explore the effect of CB-103 on pyroptosis in oral squamous cell carcinoma(OSCC)cells via the reactive oxygen species(ROS)/caspase-3/gasdermin-E(GSDME)signaling pathway.Methods The OSCC cell line SCC-7 was treated with varying concentrations of CB-103(0,20,40,60,80,and 100 μg/mL).The proliferation activity of CB-103 cells was measured using cell counting kit-8(CCK-8)assay.The cell migration was assessed using a wound-healing assay.Morphological changes in the cells were observed under an inverted microscope.Western blotting was performed to evaluate the protein expression of GSDME,GSDME-N ter-minal,and cleaved caspase-3.ATP release was measured using an ATP detection kit.The levels of intracellular high-mobility group box 1(HMGB1)and ROS were quantified by immunofluorescence.Results Compared to the control group,CB-103 inhibited the proliferation and migration of SCC-7 cells.Additionally,CB-103 increased ROS levels and upregulated the expression of cleaved caspase-3 protein.Furthermore,CB-103 promoted the cleavage of GSDME and increased the number of GSDME-N terminal fragments(all P＜0.05),thereby inducing pyroptosis in SCC-7 cells.Conclusion CB-103 promotes pyroptosis in OSCC cells by activating the ROS/caspase-3/GSDME pathway.

OBJECTIVE：To provide evidence-ba sed evidence for clinical treatment and decision by evaluating efficacy ，safety and cost-effectiveness of denosumab in the treatment of giant cell tumor of bone （GCTB）. METHODS ：Retrieved from PubMed ， the Cochrane Library ，ScienceDirect，CNKI，Wanfang database and VIP as well as health technology assessment （HTA）organi- zation websites ，HTA reports ，randomized controlled trials （RCTs），single-arm trials and retrospective studies were included about denosumab in the treatment of GCTB in the adults and adolescents with mature bone ，and their qualities were evaluated. HTA reports were analyzed with descriptive analysis qualitatively ；Meta-analysis was conducted for single-arm clinical studies and retrospective studies by using R version 3.6.0 software. RESULTS ：Among 49 screened literatures ，there were 6 HTA reports ，5 single-arm trials and 3 retrospective studies ．No eligible RCTs were retrieved. HTA reports of various countries generally believed that denosumab possessed good efficacy and safety ；HTA reports of France ，Austrila and other contries showed that denosumab possessed economics. For the GCTB patients who was unresectable ，denosumab would bring the clinical benefits to about 81％ [95％CI（77％，86％）] of patients. The complete response rate and partial response rate was around 14％[95％CI（10％，19％）] and 51％[95％CI（32％，70％）]，respectively. For the GCTB patients was resectable ，denosumab would prevent some patients from receiving surgery （35％）[95％CI（21％，49％）] or bring surgical down staging to them （40％）[95％CI（36％，45％）]，the postoperative recurrence rate after experiencing the denosumab therapy was about 19％[95％CI（7％，35％）]，and median relapse time was approximately 6.73 months [ 95％CI（3.92，9.55）] after receiving surgery. Main grade 3-4 or high frequercy ADR requiring treatment was back pain ，limbs pain ，hypophosphatemia and jaw osteonecrosis. CONCLUSIONS ：Based on the currently available evidence，among the studies and regions covered in this study ，denosumab has favorable efficacy ，safety and cost-effectiveness in the treatment of TCTB.

OBJECTIVE：To provide reference for scientific decision-making of evaluation and application of clinical comprehensive value of drugs from the perspective of the public decision-making in China. METHODS ：Based on relevant documents and literatures ，drug clinical comprehensive value in other countries and areas ，the core indicators of drug clinical comprehensive value evaluation in China were confirmed. The concept and evaluation methods of each indicator were evaluated ， and relevant suggestions were put forward. RESULTS & CONCLUSIONS ：It’s suggested that the core indicators of China ’s drug clinical comprehensive value evaluation are safety and efficacy ，economy and affordability ，accessibility and fairness adherence ， suitability. The concepts of safety ，efficacy，economy，affordability and adherence indexes are clearly defined ，evaluation methods are systematic ，data sources are sufficient ，and international consensus exists in research design. There is no uniform definition of accessibility which composed of availability ，deliverability，availability，affordability and timeliness. The concept of equity is clearly defined and the evaluation method is systematic ，but the data source is insufficient. Appropriateness is evaluated by drug suitability index but there is no clear definition. Taking efficacy as an example ，efficacy includes intermediate indicators ，outcome indicators and quality of life indicators ，which are often confirmed by epidemiological research ，systematic evaluation and expert consensus. It ’s suggested that the Real World Study data should be used as much as possible when selecting or formulating the clinical comprehensive value indicators of drugs. With the help of multi-criteria decision-making analysis ，technical support should be provided by professional evaluation institutions ，and with the support of experts and researchers ，the above indicators should be empowered to form the clinical comprehensive value judgment of a drug by different public decision-making departments ，so as to decide whether to purchase ，use clinically ，or incorporate medical insurance reimbursement.

OBJECTIVETo explore the prevalence and the different risk factors for asthma in children between urban and rural areas in Fuzhou, Fujian province.

METHODThe epidemiological survey of asthma in 0-14 years old children was conducted from October 2009 to October 2010 between Fuzhou urban and rural areas in Fujian province. The investigation subjects were selected in urban and rural areas by phased stratified random cluster sampling. The 2010 third national epidemiological survey questionnaire of children with asthma was used for screening for possible patients. Diagnosis of asthma was confirmed by physical examination. The children with asthma were designated as the positive cases, while non asthmatic children who were age, gender, ethnic, and living environment matched with asthmatic patients were designated as negative control. Comparison of the prevalence of asthma in children between Fuzhou urban and rural areas was performed. The influencing factors of asthma were analyzed and screened by the regression equation model of two element Logistic regression.

RESULTTotally 12 235 questionnaires of children with asthma and allergic disease screening were issued and 11 738 questionnaire were sent back (6 221 were male and 5 517 were female). The return rate was 95.9% in urban Fuzhou; 648 children were diagnosed as asthma. The prevalence of asthma in male was 6.48% and female children was 4.44% (comparison of the prevalence of gender χ(2) = 23.267, P < 0.001) in urban areas . A total of 6 000 questionnaires of children with asthma and allergic disease screening were sent out and 5 860 were responded (male children 3 228, female children 2 632). The recovery rate was 97.7% in rural Fuzhou; 135 children with asthma was diagnosed. The prevalence of asthma in male was 2.73%and female children and was 1.79%. Adding protein supplement before 6 months (OR = 1.908, 95%CI:1.233-2.959), the use of antibiotics in the treatment of asthma (OR = 14.541, 95%CI:8.920-23.705), furniture materials (non wood) (OR = 2.432, 95%CI:1.563-3.785) were the main risk factors of children with asthma in urban. Adding protein supplement before 6 months(OR = 3.021, 95%CI:1.357-6.711), the use of antibiotics in the treatment of asthma(OR = 14.784, 95%CI:3.842-56.885), the use of coal as fuel (OR = 63.339, 95% CI: 7.993-501.943), domesticated livestock (OR = 13.659, 95% CI:1.342-139.068), the family smoking before and after birth (OR = 6.226, 95%CI:2.674-14.495) and chemical fiber pillow (OR = 3.638, 95%CI:1.241-10.666) were the main risk factors of children with asthma in rural areas.

CONCLUSIONThe prevalence of children with asthma in urban areas was higher than that in rural areas. The prevalence of asthma in male children was higher than in female children. Adding protein food supplement before 6 months, the use of antibiotics and non solid wood furniture material were the main risk factors in children with asthma in urban areas. Adding protein supplement before 6 months, the use of antibiotics, domesticated livestock, the use of coal as fuel and the family smoking before and after birth were the main risk factor of asthma in children in rural areas.

Objective To investigate the feasibility and efficiency of unilateral pedicle screw combined with contralateral translaminar facet screw fixation by percutaneous and interbody fusion to treat low lumbar vertebra diseases. Methods Thirty patients with low lumbar vertebra diseases were entered into the study, including 8 males and 22 females with an average age of 53.7 years. All patients underwent discectomy, spinal canal decompression, cage implantation and lumbar fixation by unilateral pedicle screw combined with contralateral translaminar facet screw under gunsight guiding by percutaneous. Clinical outcomes were assed by JOA questionnaires before and after operation. Operative time, blood loss, and postoperative draiming were recorded. Radiological examination was obtained to assess position of translaminar facet screw.Results Mean operation time was 89 min with a blood loss of 285 ml. Position of translaminar facet screw grade Ⅰ were 24 cases, and grade 11 were 6. Mean follow-up was 22.5 months. 29 cases got bony fusion, and the fusion rate was 96.7%. There were no instability and evidence instrument failure during follow-up. The JOA grades improved from 13.0 preoperation to 25.2 at final follow-up, with the excellent and good rate of 72.5 %. Conclusion Unilateral pedicle screw combined with contralateral translaminar facet screw fixation by percutaneous and interbody fusion provide simple procedure, little trauma, forceful fixation, high fusion rate, and less complication, etc. Therefore, the surgical maneuver is a good choice for partial low lumbar vertebra diseases.

Objective; To investigate the cytotoxicity of monocytes/macrophages against Tca8113 cells in hypoxic microenvironment in vitro. Methods; Peripheral blood mononuclear cells were extracted from healthy person^ blood and cultivated to transform into monocytes/macrophages. The monocytes/macrophages were cultured with Tca8113 cells in hypoxic microenvironment (O_2 < 1%) and in normal microenvironment (20% O_2). The cytotoxicity of monocytes/macrophages against Tca8113 cell was examined by MTT assay. Results;The cytotoxicity of monocyles/macrophages against Tca8113 cells in hypoxic microenvironment was lower than that in normal environment(P<0.05). Conclusion; In hypoxic microenvironment, the cytotoxicity of monocytes/macrophages against Tca8113 cells is decreased. Bui the mechanism of monocytes/macrophages' function in tumors need further research.
