Objective:To compare the impact of the AlignRT-based optical guidance method with the traditional marker line method on the accuracy of positioning, and explore the effect of patient′s different thoracic characteristic parameters on the precision of optical guidance positioning. Methods:A total of sixty breast cancer patients who received post breast-conserving radiotherapy at Anhui Cancer Hospital from July 2022 to September 2023 were retrospectively selected. Then these patients were equally divided into traditional cross hairs set-up (marker line group) and surface-guided set-up (SGRT group). The cone-beam CT scanning results were used as the gold standard, the three-dimensional set-up errors and the CTV-PTV target area external radiotherapy boundaries of two groups were studied comparatively. Multiple patient′s thoracic characteristic parameters were defined and the correction between each parameter and set-up error of SGRT was analyzed. Results:The mean value of three-dimensional set-up errors in the SGRT group and the marker line group was within 0.2 cm and 0.5 cm, respectively. The differences in three-dimensional set-up errors and total displacements between two groups were statistically significant ( z=-2.93, -3.21, -2.59, -4.76, P<0.05). The SGRT group reduced the CTV-PTV boundary from 0.5 cm of conventional marker line group to 0.3 cm. The thoracic aspect ratio H/W, the affected side pinch angle α1, and the healthy side pinch angle α2 were positively correlated with the x-direction posing error ( r=0.49, 0.59, 0.71, P<0.05); whereas, the affected side and the healthy side mammary gland heights D1 and D2 were negatively correlated with the z-direction posing error ( r=-0.46, -0.49, P<0.05). Conclusions:For breast-conserving postoperative radiotherapy patients, SGRT can obtain a more accurate radiotherapy set-up than the traditional marker line method, and can effectively reduce the target area externally expanded range. Meanwhile, the monitoring accuracy of SGRT is affected by the patient′s thoracic characteristic parameters, and clinical attention should be paid to breast-conserving radiotherapy patients with thick body shape, narrow body width, and small breast glands.

As the need for antibody production rises, there is an urgent need to lower the costs and enhance the efficiency of the separation process. Currently, the chromatographic media used for antibody separation and purification often focus on individual properties of antibodies, such as affinity, hydrophobicity, and charge, leading to issues like low purification efficiency or inadequate adsorption capacity. To address this, an electrostatically coupled polypeptide affinity medium (FD7-3, 5-diaminobenzoic acid n-sepharose, FD7-DA-Sepharose) was developed for rapid purification of antibodies from cell culture supernatant. This medium utilized 3, 5-diaminobenzoic acid as a spacer to attach the heptapeptide-affinity ligand (FYEILHD, FD7) to agarose microspheres. Antibodies could be adsorbed through charge interactions with the carboxyl functional group of the FD7-DA-Sepharose spacer, while FD7 enhanced electrostatic coupling and affinity adsorption through synergistic effects, significantly increasing the adsorption capacity while maintaining the affinity and specificity. The influences of pH and ionic strength on adsorption capacity were investigated with human immunoglobulin as a model protein. The static adsorption capacity (Qm) of FD7-DA-Sepharose in the solution of pH 6.0 reached 67.73 mg/mL, representing a 52.68% increase compared with that (44.36 mg/mL) of the commercial Protein A affinity medium. Furthermore, the elution conditions for FD7-DA- Sepharose were mild (20 mmol/L PB, 0.5 mol/L NaCl, pH 6.0), in contrast to the harsh acidic elution (pH 2.7-3.6) typically associated with Protein A, which can damage antibody integrity. The FD7-DA-Sepharose medium was then employed to purify antibodies from cell culture supernatant, achieving the yield of 94.8% and the purity of 98.4%. The secondary structure of the purified antibody was determined by circular dichroism spectroscopy. The results demonstrated that FD7-DA-Sepharose enabled efficient purification of antibodies from cell culture supernatant, which provided a cost-effective solution (approximately one-third the price of commercial Protein A affinity medium) with gentle elution conditions that preserve the natural conformation of antibodies. This approach paves a novel, economical, and efficient way for the separation and purification of antibodies from cell culture supernatant.

To perform the phylogenetic characterization of an isolated porcine rotavirus(PoRV)and investigate its pathogenicity in suckling mice and piglets.A G4P[23]genotype PoRV strain JSJR2023 was successfully isolated from the diarrheic piglet feces through propagation in MA104 cells. The viral proliferation kinetics were analyzed using TCID50 assays, followed by complete genome sequencing through Sanger sequencing platforms.Comprehensive genotyping and phylogenetic reconstruction were conducted using MEGA7.0 with maximum likelihood algorithms.Pathogenicity was assessed in the following animal models:5-day-old C57BL/6 mice and 3-day-old piglets.Multidimensional evaluation included clinical monitoring(diarrhea scoring, growth parameters),virological detection, and histopathological analysis of intestinal tissues. The virus strain JSJR2023 could replicate efficiently in MA104 cells, achieving peak titers of 107.5 TCID50/mL.Whole genome genotype analysis showed that the strain belonged to G4-P[23]-I5-R1-C1-M1-A8-N1-T1-E1-H1.Phylogenetic analysis indicated that the VP3 and NSP4 genes of JSJR2023 strain were most closedrelated to human species rotaviruses, suggesting genetic reassortment between human and porcine RV strains. The animal experiments in suckling mice showed that the JSJR2023 strain infection caused diarrhea symptoms, intestinal edema and congestion, and shedding of intestinal villus epithelial cells. The pathogenicity experiments in piglets showed that compared with the control group, the challenged group of pig-lets had severe diarrhea symptoms, accompanied by reduced appetite and listlessness.Post-mortem examination revealed that the intes-tines were significantly thinner, congested, and filled with yellow watery contents. The challenged piglets showed typical pathological changes such as thinning of the intestinal wall and shortening and shedding of intestinal villi. In conclusion, this study successfully iso-lated a human-porcine recombinant G4P[23]PoRV strain and established the infection models in suckling mice and piglets, providing important tools for investigating the pathogenic mechanism of PoRV,evaluating vaccines and developing antiviral drug.

Objective:To analyze the current situation of dosimetry audits for linear accelerators in China comprehensively based on the literature, including the range of output dose deviation and the qualified rate of output dose deviation. Methods:Literature search including non-full-text was carried out in CNKI database for literature on output dose verification using dosimeter comparison and quality control test, respectively. Data extraction and analysis were carried out for each kind of eligible literature. The data on the number of tested linear accelerators, the number of qualified linear accelerators, the qualified rate of output dose deviation and the range of output dose deviation were extracted, and the total qualified rate of output dose deviation was calculated based on the total number of tested linear accelerators and the total number of qualified linear accelerators. The data on qualified rate of output dose deviation and output dose deviation in past years were sorted out and their trend was analyzed. Results:A total of 11 literatures using dosimeter comparison method for output dose verification were retrieved, and the total qualified rate of the output dose deviation reported in these literatures was 90.7%. The qualified rate of the output dose deviation in the early years was either as low as 75% or as high as 100%, and remained around 90% in recent years. A total of 19 literatures using quality control test method for output dose verification were retrieved, and the total qualified rate of X-ray and electron beam output dose deviation reported in these literatures was 86.5% and 78.4%, respectively. Of the 12 data on X-ray output dose qualified rate in 2014-2019, 8 were below 90.0% and 4 were above 90.0%. Of the 7 data on X-ray output dose qualified rate in 2020-2023, 2 were below 90.0% and 5 were greater than or equal to 90.0%. The qualified rate of electron beam output dose was 46.2%-80.0% before 2021 and 100% from 2021 to 2023.Conclusions:The qualified rate of the output dose deviation of linear accelerators in China reported in the literature in recent years is on the rise, but the total qualified rate of the output dose deviation of linear accelerators in China is still lower than in developed countries. In order to ensure the therapeutic effect of the patients undergoing radiotherapy, measures should be taken to reduce the output dose deviation of linear accelerators and improve the level of qualified rate of output dose verification.
