1.Accurate evaluation model of burning mouth syndrome based on inflammatory factors and psychological scores: a clinical study

Objective:To explore the correlation between anxiety, depression, and inflammatory markers in the body fluids of patients with burning mouth syndrome (BMS), creating a preliminary assessment model based on clinical data.Methods:Forty-one BMS patients were recruited according to the predefined inclusion criteria and exclusion criteria from the Department of Oral Medicine, Shanghai Ninth People′s Hospital, Shanghai Jiao Tong University School of Medicine between December 2021 and September 2023, along with 12 healthy controls. The pain intensity of the 41 patients was assessed using the visual analog scale (VAS). Meanwhile, anxiety and depression were assessed using the Zung self-rating anxiety scale (SAS) and the Zung self-rating depression scale (SDS). The concentrations of brain-derived neurotrophic factor (BDNF), interferon-γ (IFN-γ), interleukin (IL)-1β, IL-6, IL-8, and tumor necrosis factor-alpha (TNF-α) in saliva and serum were measured using the Luminex 200 TM system and enzyme-linked immunosorbent assay. Statistical analyses were performed using the Chi-square test, independent samples t-test, ANOVA, Spearman′s correlation analysis, and multiple linear regression, respectively. Results:The VAS score for the 41 BMS patients was (3.56±1.90), with 48.8% (20/41) and 41.5% (17/41) of patients experiencing mild and moderate pain, respectively. Only 7.3% (3/41) of patients had severe pain. Among the 41 BMS patients, 61.0% (25/41) exhibited anxiety and/or depression, in whom 39.0% (16/41) had both anxiety and depression, 9.8% (4/41) experienced anxiety without depression, and 12.2% (5/41) had depression without anxiety. The concentrations of IFN-γ and TNF-α in the serum and saliva of BMS patients were significantly higher than those in healthy controls (all P<0.05). In contrast, the BDNF concentration in serum was significantly lower in BMS patients than in healthy participants ( P<0.01), but was significantly higher in saliva ( P<0.001). Serum TNF-α was positively correlated with IFN-γ, IL-1β, and IL-6 (β=0.803, β=0.812, β=0.592; all P<0.01), while saliva TNF-α was negatively correlated with both anxiety and depression (β=-0.325, β=-0.321; all P<0.05). SAS scores were linearly correlated with saliva TNF-α concentrations (SAS=51.374-1.154×saliva TNF-α); saliva TNF-α concentrations were linearly correlated with saliva IFN-γ and IL-1β (saliva TNF-α=2.408+0.281×saliva IFN-γ+0.002×saliva IL-1β). Conclusions:This study provides a preliminary exploration of a clinical assessment model for BMS based on inflammatory markers and psychological scores, offering an exploratory framework for further research and optimization of the model.
2.Design of performance testing platfrom for proportional solenoid valves in ventilators

Objective To design a performance testing platform to evaluate the working status and performance characteristics of the ventilator proportional solenoid valve.Methods The performance testing platform had its hardware including a high-pressure gas source,a pressure regulating valve,sensors and etc,and its software designed based on PyQt5 and composed of several modules for data acquisition,parameter setting,image display,indicator computation,result output and etc.Two kinds of proportional solenoid valves(Valve 1、Valve2)were selected for static and dynamic tests to verify the performance of the platform.Results The platform developed facilitated the proportional solenoid valve to carry out accurate computation of static and dynamic indicators at real time and time domain and waveform feature extraction of sensor data by precision control and data acquisition for the proportional solenoid valve.Static tests showed that Valve 1 gained advantages over Valve 2 in static flow characteristics involving in lowered repeatability,return error and offset while enhanced stability;dynamic tests indicated Valve 2 had rapid flow variations and significant flow fluctuation impacts,Valve 1 showed smooth dynamic response changes,and Valve 2 behaved better than Valve 1 in dynamic performance.Conclusion The testing platform developed comprehensively demonstrates the performance characteristics and working performance of the ventilator proportional solenoid valve,which is of great significance to enhance the reliability and safety of the ventilator.[Chinese Medical Equipment Journal,2025,46(1):13-19]

Allergen-specific immunotherapy(AIT) remains the only therapeutic approach with the potential to modify the natural course of allergic rhinitis(AR). Nevertheless, considerable inter-individual variability exists in patients'responses to AIT. To facilitate more reliable assessment of treatment efficacy, the China Rhinopathy Research Cooperation Group(CRRCG) convened young and middle-aged nasal experts in China to formulate the present consensus. The recommended subjective outcome measures for AIT comprise symptom scores, medication scores, combined symptom and medication scores, quality-of-life assessments, evaluation of disease control, and assessment of comorbidities. Objective indicators may supplement these measures. Currently available objective approaches include skin prick testing, nasal provocation testing, and allergen exposure chambers. However, these methods remain constrained by practical limitations and are not yet appropriate for routine implementation in clinical efficacy evaluation. In addition, several biomarkers, including sIgE and the sIgE/tIgE ratio, sIgG4, serum IgE-blocking activity, IgA, cytokines and chemokines, as well as immune cell surface molecules and their functional activity, have been shown to have associations with AIT outcomes. While these biomarkers may complement subjective assessments, they are subject to significant limitations. Consequently, large-scale multicenter trials and real-world evidence are required to strengthen the evidence base. The present consensus underscores the necessity of integrating patients'subjective experiences with objective testing throughout the treatment process, thereby providing a more comprehensive and accurate framework for efficacy evaluation. Looking forward, future investigations should prioritize the incorporation of multi-omics data and artificial intelligence methodologies, which hold promise for overcoming current limitations in assessment strategies and for advancing both the standardization and personalization of AIT.

4.Tasquinimod promotes the sensitivity of ovarian cancer cells to cisplatin by down-regulating the HDAC4/p21 pathway

To investigate whether Tasquinimod can influence cisplatin resistance in drug-resistant ovarian cancer (OC) cell lines by regulating histone deacetylase 4 (HDAC4) or p21, we explored its effects on the cell cycle, and associated mechanisms.RT-PCR and Western blot analyses, flow cytometry, CCK8 assay, and immunofluorescence were utilized to investigate the effects of Tasquinimod on gene expression, cell cycle, apoptosis, viability, and protein levels in OC cells. The results showed that Tasquinimod inhibited cell viability and promoted apoptosis in SKOV3/DDP (cisplatin) and A2780/DDP cells more effectively than DDP alone. In combination with cisplatin, Tasquinimod further enhanced cell apoptosis and reduced cell viability in these cell lines, an effect that could be reversed following HDAC4 overexpression. Tasquinimod treatment down-regulated HDAC4, Bcl-2, and cyclin D1, and CDK4 expression and up-regulated the cleaved-Caspase-3, and p21 expression in SKOV3/DDP and A2780/ DDP cells. Additionally, Tasquinimod inhibited DDP resistance in OC/DDP cells. These effects were similarly observed in OC mouse models treated with Tasquinimod. In conclusion, Tasquinimod can improve OC cells' sensitivity to DDP by down-regulating the HDAC4/p21 axis, offering insights into potential strategies for overcoming cisplatin resistance in OC.

Background:
The genetic basis for hyperglycaemia in pregnancy remain unclear. This study aimed to uncover the genetic determinants of gestational diabetes mellitus (GDM) and investigate their applications.
Methods:
We performed a meta-analysis of genome-wide association studies (GWAS) for GDM in Chinese women (464 cases and 1,217 controls), followed by de novo replications in an independent Chinese cohort (564 cases and 572 controls) and in silico replication in European (12,332 cases and 131,109 controls) and multi-ethnic populations (5,485 cases and 347,856 controls). A polygenic risk score (PRS) was derived based on the identified variants.
Results:
Using the genome-wide scan and candidate gene approaches, we identified four susceptibility loci for GDM. These included three previously reported loci for GDM and type 2 diabetes mellitus (T2DM) at MTNR1B (rs7945617, odds ratio [OR], 1.64; 95% confidence interval [CI],1.38 to 1.96]), CDKAL1 (rs7754840, OR, 1.33; 95% CI, 1.13 to 1.58), and INS-IGF2-KCNQ1 (rs2237897, OR, 1.48; 95% CI, 1.23 to 1.79), as well as a novel genome-wide significant locus near TBR1-SLC4A10 (rs117781972, OR, 2.05; 95% CI, 1.61 to 2.62; Pmeta=7.6×10-9), which has not been previously reported in GWAS for T2DM or glycaemic traits. Moreover, we found that women with a high PRS (top quintile) had over threefold (95% CI, 2.30 to 4.09; Pmeta=3.1×10-14) and 71% (95% CI, 1.08 to 2.71; P=0.0220) higher risk for GDM and abnormal glucose tolerance post-pregnancy, respectively, compared to other individuals.
Conclusion
Our results indicate that the genetic architecture of glucose metabolism exhibits both similarities and differences between the pregnant and non-pregnant states. Integrating genetic information can facilitate identification of pregnant women at a higher risk of developing GDM or later diabetes.
7.Diagnostic value of CD4+T cells combined with interleukin-8 in human immunodeficiency virus-associated cytomegalovirus retinitis

AIM: To investigate the diagnostic value of CD4+T cells combined with interleukin-8(IL-8)in human immunodeficiency virus(HIV)-associated cytomegalovirus retinitis(CMVR).METHODS: A retrospective study was conducted on 80 HIV-infected patients who visited the Second Hospital of Nanjing from June 2021 to December 2024, including 51 males and 29 females, aged from 22 to 56(44.25±6.31)years, and they were divided into CMVR group(49 cases)and non-CMVR group(31 cases)based on whether the patients had CMVR. The clinical data of the two groups were compared. Restricted cubic spline analysis was performed to analyze the dose-response relationship between CD4+ T cells, IL-8 and the risk of CMVR in HIV-infected patients. Receiver operating characteristic(ROC)curves were used to analyze the diagnostic value of CD4+T cells and IL-8 alone or in combination for HIV-associated CMVR.RESULTS: The IL-8 level in the CMVR group was significantly higher than that in the non-CMVR group, while the levels of neutrophils, neutrophil to lymphocyte ratio(NLR), and CD4+T cells were significantly lower than the non-CMVR group(all P<0.05). The results of restrictive cubic spline analysis showed that there was a significant nonlinear relationship between IL-8, CD4+T cells and the risk of CMVR(Chi-Square=13.625, 5.406, P=0.001, 0.045). The areas under the ROC curve for IL-8 and CD4+ alone and in combination were 0.777, 0.743 and 0.836, respectively.CONCLUSION: Both IL-8 and CD4+T cells showed good diagnostic value for HIV-related CMVR, and their combination further enhances diagnostic efficiency.
