Objective:
To investigate the effects of different doses of X ⁃rays irradiation combined with allogeneic lymphocyte infusion on the establishment of aplastic anemia in mice.
Methods:
Forty BALB/c mice were randomly divided into four groups : the 3 Gy group (n = 9) , the 4 Gy group (n = 9) , the 5 Gy group (n = 10) , and the con⁃trol group (n = 12) . In the 3 Gy , 4 Gy , and 5 Gy groups , the experimental mice were exposed to corresponding do⁃ses of X ⁃ray and then intravenously infused with 0. 2 mL mixed suspension of the thymus and spleen cells from DBA/2 mice , at a concentration of 1 × 107 cells/mL , within 4 hours after irradiation. The control group did not un⁃dergo X ⁃ray irradiation and infused with an equivalent volume of physiological saline instead. Blood samples were collected from the orbital venous plexus of BALB/c mice and analyzed using an animal automated hematology analy⁃zer to measure peripheral blood parameters , including red blood cells ( RBC) , white blood cells ( WBC) , and platelets (PLT) . The general condition of mice was monitored daily , and survival rates were recorded for each group. At the experimental endpoint , the tibias were harvested for hematoxylin and eosin (HE) staining , while the femurs were used to prepare bone marrow smears for morphological examination. For the 5 Gy group , T ⁃cell subsets(ELISA) at the endpoint.
Results :
In the 3 Gy group , pancytopenia was observed , but platelet recovery occured rapidly , returning to normal levels by day 17 post⁃modeling. No deaths occurred during the observation period. At myeloid⁃to⁃erythroid (M/E) ratio , and no significant morphological abnormalities were noted in cells at any devel⁃with hematopoietic cells. In the 4 Gy group , pancytopenia persisted throughout the observation period. The survival rate was 90% . Endpoint analysis showed hypocellular marrow by morphological examination. HE staining indicated minimal fatty infiltration in the bone marrow tissue. In the 5 Gy group , pancytopenia was observed , though erythro⁃cyte counts returned to normal levels by day 24. The survival rate during the observation period was 50% . Endoint analysis revealed vacuolization of marrow particles and reduced hematopoietic cells with predominantly non⁃hematopoietic cells in bone marrow morphology. HE staining demonstrated severe fatty infiltration in the bone mar⁃row tissue , with scarcity of immature cells and hematopoietic precursor cells. Flow cytometry analysis showed a de⁃creased proportion of CD4 + T cells (% ) and an increased proportion of CD8 + T cells (% ) . ELISA confirmed elevated secretion of negative hematopoietic regulators : interferon⁃gamma ( IFN⁃γ) and tumor necrosis factor⁃alpha (TNF⁃α ) .
Conclusion
Combined administration of varying radiation doses with allogeneic lymphocyte infusion consistently induced peripheral blood cytopenia in mice , characterized by reductions in RBC , WBC , and PLT counts. Integrated analysis of bone marrow morphology , histopathological assessment via HE staining , and immuno logical parameters confirmed that a mouse model of aplastic anemia can be successfully established using 5 Gy X ⁃ ray irradiation coupled with infusion of 2 × 106 allogeneic lymphocytes.

Integrated metabolomic and lipidomic profiling, utilizing liquid chromatography coupled with high-resolution mass spectrometry(LC-HRMS),has emerged as a pivotal strategy for biomarker discovery. However, the inherent polarity disparity between metabolites and lipids complicates simultaneous analysis. To address this,a dual-stationary phase polarity-extended liquid chromatography(PELC)system was developed, which surpassed conventional one-dimensional LC(1D-LC)by enabling comprehensive coverage of both polar and non-polar compounds within a single injection. This system enhanced chromatographic resolution, peak capacity, and throughput while minimizing analytical variability.Extracellular vesicles(EVs),lipid bilayer-enclosed nanoparticles ubiquitously present in biofluids, had gained prominence as reservoirs of cancer biomarkers due to their cargo stability and pathophysiological relevance.Herein, the application of PELC-HRMS for concurrent metabolome-lipidome profiling in EVs was pioneered.A total of 193 metabolites were identified using this technique coupled with MS-DIAL software and Human Metabolome Database.Subsequently, this technique was employed to explore potential biomarkers for prostate cancer(PCa).Multivariate analysis identified 17 differentially abundant metabolites in PCa,implicating dysregulated pathways including purine metabolism, starch and sucrose metabolism, galactose metabolism, cysteine and methionine metabolism, and biosynthesis of unsaturated fatty acids.Notably,creatine(AUC=0.92)and DG 42:5(AUC=0.80)demonstrated robust diagnostic efficacy, attributable to their broad polarity ranges and EV-specific enrichment. This study established PELC as a high-fidelity platform for multi-omics integration in complex biospecimens, advancing mechanistic insights into metabolic rewiring and disease pathophysiology.

Objective:To explore the feasibility and clinical outcomes of transperineal prostate multimodal image fusion-targeted biopsy under the day surgery model. Methods:Clinical data of 258 patients who underwent transperineal prostate biopsy at the Second Affiliated Hospital of Xi’an Jiaotong University between December 2022 and June 2024 were retrospectively analyzed. Patients were divided into two groups, and the experimental group( n = 141)underwent transperineal prostate multi-modal image-fusion targeted biopsy in the day-surgery mode, with age of(70.0 ± 8.8)years,median prostate-specific antigen(PSA)level of 11.10(7.63?17.06)ng/ml,and median Prostate Imaging Reporting and Data System(PI-RADS)score of 4(3,5). The control group( n = 117)underwent traditional transperineal systematic biopsy, with age of(69.3 ± 7.4)years,median PSA of 25.20(16.18-54.40)ng/ml,and median PI-RADS score of 4(3,5). The experimental group was given the day surgery mode:preoperatively, multiparametric magnetic resonance imaging(mpMRI)of the prostate was multimodally fused with ultrasound images, the location of target lesions in the prostate was manually mapped, and a targeted biopsy plan was developed. Intraoperatively, under ultrasound guidance, precise puncture was performed at the lesion sites, followed by systematic biopsy. After the operation, the patients were observed for 4-6 hours, and could be discharged if there were no obvious abnormalities, with the total hospitalization time within 24 hours. For the control group, the conventional biopsy mode was used. Intraoperatively, under ultrasound guidance, the standard 12-core transperineal systematic biopsy protocol was adopted, and sampling was conducted according to the anatomical regions of the prostate(base, midportion, apex, transition zone, and peripheral zone)to cover the entire gland. Patients in this group required routine hospitalization, with a hospital stay of 3-5 days. Operative time, intraoperative pain Numerical Rating Scale(NRS)score, complications within one week postoperatively, treatment costs, overall prostate cancer detection rate, and clinically significant prostate cancer(csPCa,defined as Gleason score ≥ 7 or pathological stage ≥ T 2b)detection rate were compared between the two groups. Results:All procedures were successfully completed without special incidents. The operative time was(13.49 ± 2.00)min in the experimental group and(13.05 ± 2.89)min in the control group, showing no significant difference( P > 0.05). The intraoperative pain NRS scores were(3.01 ± 1.17)and(3.10 ± 1.25)in the experimental and control groups, respectively, with no significant difference( P > 0.05). Pathological examination revealed that the overall prostate cancer detection rates were 46.8%(66/141)in the experimental group and 42.7%(50/117)in the control group, while the csPCa detection rates were 35.5%(50/141)and 31.6%(37/117),respectively. The differences were not statistically significant( P > 0.05). The complication rate was 6.4%(9/141)in the experimental group(including 2 cases of acute urinary retention,3 hematuria,3 fever, and 1 sepsis)and 6.0%(7/117)in the control group(including 3 acute urinary retention,1 hematuria,2 fever, and 1 sepsis),with no significant difference( P > 0.05). All complications improved after symptomatic treatment. The treatment costs were(4 063.25 ± 67.26)yuan in the experimental group and(5 185.14 ± 469.15)yuan in the control group, demonstrating a statistically significant difference( P < 0.05). Conclusions:Transperineal prostate multimodal image fusion-targeted biopsy can be safely performed under the day surgery model, offering advantages including a relatively high detection rate for csPCa,low complication rate, and better cost-effectiveness.
