1.Association of insulin-like growth factor binding protein-3 gene polymorphism at the rs10225396 locus with susceptibility to hepatocellular carcinoma

ObjectiveTo investigate the association of the single nucleotide polymorphism (SNP) of the insulin-like growth factor binding protein-3 (IGFBP3) gene at rs10225396 (A>G) locus with the susceptibility to hepatocellular carcinoma, as well as its potential molecular regulatory mechanisms, and to provide novel genetic biomarkers and a theoretical basis for early screening and precise targeted therapy for hepatocellular carcinoma. MethodsA total of 192 patients with hepatocellular carcinoma who were admitted to The Affiliated Hospital of Yanbian University and Yanbian Cancer Hospital from January 2009 to August 2016 were enrolled as experimental group, and 190 healthy individuals who underwent physical examination in Physical Examination Center of Yanbian Hospital during the same period of time were enrolled as control group. Peripheral blood samples were collected from all subjects, and after DNA was extracted from whole blood, the DNA samples meeting quality standards were sent to Beijing Genomics Institute Research Center Co., Ltd., for Mass ARRAY mass spectrometry, while genotyping was completed. The independent-samples t test was used for comparison of continuous data between two groups, and the chi-square test was used for comparison of categorical data between two groups. A binary Logistic regression model was used to analyze the association of the SNP of the IGFBP3 gene at rs10225396 locus with the susceptibility to hepatocellular carcinoma, and odds ratio (OR) and 95% confidence interval (CI) were calculated to assess the risk of developing hepatocellular carcinoma in individuals carrying different genotypes. ResultsThere were two alleles (G and A) at the rs10225396 locus of the IGFBP3 gene, yielding the three genotypes of AA, AG, and GG, and its genotype distribution was consistent with the Hardy-Weinberg equilibrium (χ²=0.072, P=0.789). The stratified genetic analysis showed that carriers of the IGFBP3 rs10225396 AA genotype had a significant increased risk of hepatocellular carcinoma among individuals of age <63 years, male sex, smoking history, drinking history, and the Chinese Korean population (all P<0.001). The binary Logistic regression analysis showed that after adjustment for related risk factors in the codominant model, the population with genotype AG and GG had a significant reduction in the risk of hepatocellular carcinoma compared with the population with genotype AA (P<0.001), and in the dominant model, the population with genotype AG+GG had a significant reduction in the risk of hepatocellular carcinoma compared with the population with genotype AA (P<0.001). ConclusionThrough a genotyping analysis of hepatocellular carcinoma patients and healthy individuals in Yanbian Korean Autonomous Prefecture of Jilin Province, China, this study shows that genotype AA at rs10225396 locus of the IGFBP3 gene is significantly associated with the susceptibility to hepatocellular carcinoma, and this genotype can significantly increase the risk of developing hepatocellular carcinoma with the presence of specific risk factors, which provides a potential genetic marker for early screening and precise treatment of hepatocellular carcinoma.
2.Relationship between HLA⁃A rs3132682 and susceptibility to hepatocellular carcinoma in Yanbian area

Objective:
To study the relationship between the single nucleotide polymorphism of HLA⁃A rs3132682 and the risk of liver cancer.
Methods:
The author selected 291 cases of liver cancer patients in Yanbian area as the experimental group and 272 healthy people as the control group. The genotypes and allele frequencies of the two loci were detected by MassARRAY SNPS mass spectrometry. Odds ratio (OR) and 95% confidence interval (CI) were calculated by unconditional logistic regression to evaluate the risk of liver cancer in patients with different genotypes.
Results:
There were G and C alleles in HLA⁃A rs3132682 locus , GG , GC and CC genotype. After adjusting for confounding factors in HLA⁃A rs3132682 locus , there was correlation between CC genotype and the risk of liver cancer compared with GG genotype (P < 0. 05) . Stratification analysis showed that compared with the population with CG + GG genotype , the Korean population with CC genotype increased the risk of liver cancer by 3. 331 times.
Conclusion
The single nucleotide polymorphism of HLA⁃A rs3132682 is correlated with the risk of liver cancer in Yanbian area.
3.Application of transcatheter aortic valve replacement in elderly patients with native pure aortic regurgitation

Objective:This study sought to investigate the feasibility, anatomical indications and operating points of transcatheter aortic valve replacement (TAVR) procedure in the treatment of pure aortic regurgitation (AR).Methods:The medical records of 4 elderly patients with pure AR who were treated in the cardiology department, Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology from March 2020 to March 2021 were retrospectively analyzed. All patients were implanted with self-expandable valve stents via peripheral artery approach for TAVR treatment. The feasibility, anatomical indications and key points of TAVR were analyzed.Results:The 4 patients with pure AR who were carefully screened had an average age of 66 years, and all achieved TAVR treatment success without serious perioperative complications and death. Postoperative examination and follow-up data showed that cardiac functions and cardiac remodeling indexes were significantly improved.Conclusions:This exploratory study shows that TAVR is technically feasible and effective treatment option for selected elderly patients with native pure AR, which is worthy of further study.
4.Serum levels of interleukin(IL-13) and prostaglandin(PGE1) and their clinical significance in patients with vital hepatitis B

Objective:To explore the relationship between type I hypersensitivity reaction and the mechanism of the viral hepatitis B through the research of interleukin(IL-13 ) and prostaglandin(PGE1) levels in serum of patients with viral hepatitis B, and therefore, to provide new theory for the perfection of the immunological mechanism of hepatitis B, especially acute hepatitis B.Methods:Serum IL-13 and PGE1 levels were measured in 50 patients with viral hepatitis B and 35 normal cases using double antibody sandwich ELISA method. AST and ALT levels were studied using omni automatic biochemistry analyzer and those correlation with IL-13 and PGE1 were observed.Results:①The IL-13 and PGE1 levels of 10 acute hepatitis B were significantly higher than those of the normal cases; ②The IL-13 and PGE1 levels of 35 chronic hepatitis B were also significantly higher than those of the normal cases; ③The levels of IL-13 and PGE1 were significantly positively correlated with serum ALT and AST.Conclusion:IL-13 and PGE1 levels in patients with acute hepatitis B are most remarkably increased, and correlate with the damage of liver.
