1.Biomarkers Screening and Mechanisms Analysis of the Restraint Stress-Induced Myocardial Injury in Hyperlipidemia ApoE-/-Mice

Objective To explore the biomarkers and potential mechanisms of chronic restraint stress-induced myocardial injury in hyperlipidemia ApoE-/-mice.Methods The hyperlipidemia combined with the chronic stress model was established by restraining the ApoE-/-mice.Proteomics and bioinformatics techniques were used to describe the characteristic molecular changes and related regulatory mechanisms of chronic stress-induced myocardial injury in hyperlipidemia mice and to explore potential diagnostic biomarkers.Results Proteomic analysis showed that there were 43 significantly up-regulated and 58 sig-nificantly down-regulated differentially expressed proteins in hyperlipidemia combined with the restraint stress group compared with the hyperlipidemia group.Among them,GBP2,TAOK3,TFR1 and UCP1 were biomarkers with great diagnostic potential.KEGG pathway enrichment analysis indicated that fer-roptosis was a significant pathway that accelerated the myocardial injury in hyperlipidemia combined with restraint stress-induced model.The mmu_circ_0001567/miR-7a/Tfr-1 and mmu_circ_0001042/miR-7a/Tfr-1 might be important circRNA-miRNA-mRNA regulatory networks related to ferroptosis in this model.Conclusion Chronic restraint stress may aggravate myocardial injury in hyperlipidemia mice via ferrop-tosis.Four potential biomarkers are selected for myocardial injury diagnosis,providing a new direction for sudden cardiac death(SCD)caused by hyperlipidemia combined with the restraint stress.
2.Clinical application of Chaihu Jia Longgu Muli Decoction based on modern pathophysiology mechanism.

Chaihu Jia Longgu Muli Decoction was firstly recorded in Treatise on Cold Damage(ZHANG Zhong-jing, Eastern Han dynasty). According to this medical classic, it is originally used in the treatment of the Shaoyang and Yangming syndrome. Based on the modern pathophysiological mechanism, this study interpreted the classic provisions of Chaihu Jia Longgu Muli Decoction. Original records of "chest fullness" "annoyance" "shock" "difficult urination" "delirium" "heavy body and failing to turn over" all have profound pathophysiological basis, involving disorders in cardiovascular, respiratory, nervous, and mental systems. This formula is widely used, which can be applied to treat epilepsy, cerebral arteriosclerosis, cerebral infarction, and other cerebrovascular diseases, hypertension, arrhythmia, and other cardiovascular diseases, insomnia, constipation, anxiety, depression, cardiac neurosis and other acute and chronic diseases as well as diseases in psychosomatic medicine. The clinical indications include Bupleuri Radix-targeted syndrome such as fullness and discomfort in chest and hypochondrium, bitter taste mouth, dry throat, and dizziness, the insomnia, anxiety, depression, susceptibility to fright, upset, dreamfulness and other psychiatric symptoms, red tongue, thick and yellow tongue coating, and wiry hard and powerful pulse. This formula was found to be used in combination with other formulas, such as Gualou Xiebai Decoction, Wendan Decoction, Zhizhu Pills, Juzhijiang Decoction, Suanzaoren Decoction, and Banxia Baizhu Tianma Decoction.

3.Value of computed diffusion-weighted imaging in diagnosis of benign and malignant lesions of gallbladder

Objective To explore the value of computed diffusion-weighted imaging(cDWI)for diagnosing benign and mali-gnant lesions of the gallbladder.Methods Totally 54 gallbladder disease patients confirmed pathologically from February 2018 to February 2022 in some hospital were selected retrospectively,including 27 patients with malignant tumors(malignant group)and 27 patients with benign lesions(benign group).All the patients in the 2 groups were examined using MR equipment and apparent diffusion coefficient(ADC)images were generated,then the generated ADC images were transferred to Ziostation and cDWI images with b-values of 1 500,2 000,3 000 and 4 000 s/mm2 were formed according to standard mono-exponential fitting.The ADC values of the two groups were compared and the diagnostic efficacy of ADC was analyzed using ROC curves;comparison analyses were carried out on the diagnostic efficacy of cDWI and cDWI combined with ADC for benign and mali-gnant lesions of the gallbladder in case of different high b values,the diagnostic efficacy of common diffusion weighted imaging(mDWI),cDWI,mDWI combined with ADC and cDWI combined with AUC in case b had the value of 1 000 s/mm2 or 1 500 s/mm2.SPSS 21.0 was used for statistical analysis.Results The mean ADC value of the malignant group was 1.697× 10-3 mm2/s,which was significantly lower than that of the benign group(2.203×10-3 mm2/s),and the difference was statistically significant;the AUC was the largest when the ADC value was 1.796×10-3 mm2/s;the AUC value peaked at 0.858 at an ADC value of 1.796×10-3 mm2/s,with the sensitivity and specificity being 81.3%and 90.9%respectively;when b had the value of 1 500 s/mm2,cDWI combined with ADC showed the highest diagnostic efficacy and the sensitivity,specificity and accuracy were 92.59%,85.19%and 88.89%respectively,with the accuracy and specificity significantly higher than those of mDWI and mDWI combined AUC at b=1 000 s/mm2 and those of cDWI at b=1 500 s/mm2(P<0.05)while the sensitivity differences not statistically significant(P>0.05).Conclusion In case of b value of 1 500 s/mm2 cDWI combined with ADC gains advan-tages over mDWI in differentiating benign and malignant diseases of the gallbladder.[Chinese Medical Equipment Jour-nal,2023,44(11):66-70]
4.Effect of Shenshuai Xiezhuo Decoction and Its Deficiency Tonifying and Pathogen Eliminating Components on Renal Interstitial Fibrosis in UUO rats

Objective:To study the effect of Shenshuai Xiezhuo decoction and its deficiency tonifying and pathogen eliminating components on renal interstitial fibrosis in UUO rats. Method:A rat model of unilateral ureteral obstruction (UUO) was established through ligation of a unilateral ureter. The rats were divided into six groups: sham operation group, model group, benazepril group, Shenshuai Xiezhuo decoction group, Buxufang group, and phlegm group, with 24 rats in each group. On the third day after operation, the rats in the Shenshuai Xiezhuo decoction group, Buxufang group, and phlegm group were given Shenshuai Xiezhuo decoction concentrating agent at a dose of 8.0 g·kg-1·d-1, the rats in the benazepril group were given benazepril 1.5 mg·kg-1·d-1, and the rats in the sham operation group and the model group were given the same volume of saline. On the 7th, 14th and 21st days after operation, the expressions of peripheral cells and relevant signal pathway markers in renal tissue were detected by Western blot and Real-time fluorescent quantitative polymerase chain reaction (Real-time PCR) respectively. Result:The stromal damage score and the interstitial collagen accumulation on the 14th and 21st days after UUO were significantly lower in the Shenshuai Xiezhuo decoction group, Buxu prescription group and Qixie prescription group than those in the model group (
5.Evaluation of isolation and field effect of ginseng disease resistance and growth-promoting bacteria.

This study was aimed to isolate the strains with both disease resistance and growth-promoting, and clarify the field application effects of the strain for laying the further application foundation. The strains with good antagonistic effect were isolated from the 298 strains in Panax ginseng and the soil by plate confrontation method. The nitrogen fixation potential was verified by Ashby medium. The Salkowski method was used to determine the ability of producing IAA. Silicate medium screening and flame spectrophotometry was used to determine the ability of dissolving potassium. CAS method was applied to detect the ability of producing siderophores to determine its growth characteristics. The morphological, physiological and biochemical and 16S rRNA sequences were used to identify the species. The method of root irrigation was used to determine the effects of its disease control and growth-promoting on ginseng. A strain TY15 with broad spectrum of antimicrobial effect, nitrogen fixation, potassium-dissolving and the capacity of producing IAA and siderophores was obtained by screening. And the strain TY15 was identified as Pantoea agglomerans. The control effect of TY15 on the disease of ginseng in the field was 68.02%, which was equivalent to 68.94% of 30 billion per gram of beneficial microecological bacterium agent. The fresh weight of P. ginseng treated with TY15 strain was increased by 22.73% compared with the control group treated with water. And finally a strain TY15 with good application prospects was obtained.
6.Transcutaneous electrical acupoint stimulation for asthenozoospermia.

Objective:
To study the effect of transcutaneous electrical acupoint stimulation (TEAS) in the treatment of asthenozoospermia.
METHODS:
We randomly divided 72 asthenozoospermia patients into a 2 Hz TEAS (n = 29), a 100 Hz TEAS (n = 20), and a blank control group (n = 23), those in the former two groups treated by 30 minutes of TEAS at 2 Hz and 100 Hz respectively, applied to the acupoints of bilateral Shenshu, left Zusanli, and Guanyuan, once a day for 60 days, while those in the blank control group left untreated. Using computer-assisted sperm analysis (CASA), we examined sperm concentration and motility as well as the percentages of grade a and grade a+b sperm in different groups of the patients.
RESULTS:
Compared with the baseline, 2 Hz TEAS significantly increased sperm motility ([12.76 ± 1.39] vs [18.89 ± 2.46]%, P<0.05) and the percentage of grade a+b sperm ( [10.68 ± 1.22] vs [16.32 ± 2.10]%, P<0.05) in the asthenozoospermic patients, while 100 Hz TEAS improved not only sperm motility ([12.32 ± 2.21] vs [23.81 ± 3.42]%, P<0.01) and the percentage of grade a+b sperm ([10.45 ± 1.98] vs [20.25 ± 2.82 ]%, P<0.01), but also the percentage of grade a sperm ([6.44 ± 1.16] vs [13.31 ± 2.30]%, P<0.05). Moreover, in comparison with the blank control group, 2 Hz TEAS also remarkably increased sperm motility ([9.57 ± 1.60] vs [18.89 ± 2.46]%, P<0.05) and the percentage of grade a+b sperm ([7.81 ± 1.31] vs [16.32 ± 2.10]%, P<0.05) in the asthenozoosperma patients, while 100 Hz TEAS improved not only sperm motility ([9.57 ± 1.60] vs [23.81 ± 3.42]%, P<0.01) and the percentage of grade a+b sperm ([7.81 ± 1.31] vs [20.25 ± 2.82]%, P<0.01) but also the percentage of grade a sperm ([4.87 ± 1.01] vs [13.31 ± 2.30]%, P<0.01). Meanwhile, the rate of clinical effectiveness was significantly higher in the 100 Hz TEASthan in the blank control group either in intention-to-treat (ITT) analysis (100% vs 18.18%) orper-protocol (PP) analysis (90% vs 0%), and so was it than in the 2 Hz TEAS group based on the data of ITT (100% vs 33.33%).
CONCLUSIONS
Both 2 Hz and 100 Hz TEAS are effective for the treatment of asthenozoospermia by improving sperm motility and vitality.
