A matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS)method was developed for simultaneous analysis of 10 kinds of aphrodisiac drugs,including sildenafil,acetildenafil,nor-acetildenafil,homosildenafil,lodenafil carbonate,sildlenafil dimer impurity,vardenafil dimer,hydroxyacetildenafil,N-desmethylsildenafil and udenafil.By using different matrices(α-cyano-4-hydroxycinnamic acid(HCCA)and sinapic acid(SA)),the effects of solvents,amount of matrices,laser intensity and spotting methods on the peak strength and signal intensity of MALDI-TOF MS for the aphrodisiac drugs were investigated.As a result,SA was chosen as the matrix,and then dispersed in methanol-water(50∶50,V/V),and spotted by matrix firstly and sample secondly approach with the reflect linear positive mode and laser power of 60%.Under optimal conditions,the proposed MALDI-TOF MS method could obtain stable signal,high intensity and well-repeated mass spectrometric results.The results of method validation showed a linear range from 10 to 100 ng/mL,and the regression coefficients(r)were all above 0.985.The limits of detection(LOD)were 0.1-1.0 ng/mL,and the recoveries were in the range of 72.9%-109.9%for health wine,soft capsule and instant coffee samples,while the relative standard deviations(RSDs)ranged from 0.7%to 11.3%(n=3).The test results of 10 kinds of aphrodisiac drugs by MALDI-TOF MS were in accordance with the results of high-performance liquid chromatography-tandem mass spectrometry.This method exhibited high sensitivity,high accuracy,good anti-interference ability,low chemical solvents consumption and environmentally friendliness,and could be used to detect drugs without standards.The developed MALDI-TOF MS method could realize the non-target screening and detection of 10 kinds of illegally added aphrodisiac drugs in foods.
4.Preparation and Characterization of Supramolecular Solvent and Its Extraction of Sulfonamide Antibiotics from Milk

As a cheap and effective antibiotic,sulfonamides are often used in animal husbandry.However,their residues in animal-derived foodstuffs will threaten human health.Consequently,a high-performance liquid chromatography(HPLC)method integrated with supramolecular solvent microextraction was successfully established for simultaneous quantification of sulfonamide residues sulfachlorpyridazine,sulfamethoxazole,sulfamethoxypyridazine and sulfadimethoxine in milk matrices.This approach exhibited prominent characteristics of operational simplicity,environmental sustainability,and high extraction efficiency.The supramolecular solvents prepared by tributyl octylphosphine tetrafluoroborate and tetrahydrofuran were employed as extraction solvents.The analytes underwent isolation and concentration via dispersive liquid-liquid microextraction(DLLME)prior to quantitative determination using high-performance liquid chromatography(HPLC).The composition and microscopic morphology of the supramolecular solvent were characterized through a series of analytical techniques,including phase diagram,Fourier transform infrared spectroscopy,scanning electron microscopy,and inverted fluorescence microscopy and so on.The density and pH value of supramolecular solvents were determined.The extraction conditions were optimized through the one-factor experiments.The experimental results demonstrated that under the optimal extraction conditions,the four kinds of sulfonamide antibiotics exhibited excellent linearity within respective detection range(R2 ≥ 0.9998)and the limits of detection(LOD)were 0.67-1.45 μg/L.Compared with literature methods,this approach offered some advantages such as simplicity of operation and less reagent consumption,and could be used for analysis and detection of sulfonamide antibiotic residues in milk samples.The present method provided technical support for food safety regulation and paved a new way for the application of supramolecular solvents in the field of extraction and separation.
5.Exploration on effects of Bushen Antai Mixture on placental oxidative damage in mice with RSA investigation based on Nrf2/HO-1/NF-κB signaling pathways

Objective:To observe the effects of Bushen Antai Mixture on nuclear transcription factor E2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1)/nuclear transcription factor-κB (NF-κB) in placental tissue of recurrent spontaneous abortion (RSA) mice.Methods:The CBA/J mice (female) and DBA/2 (male), CBA/J mice (female) and BALB/c (male) were caged at a ratio of 2 : 1 to establish RSA and normal pregnant mice, respectively. The next morning, the vaginal plug was found or the sperm was seen under the microscope as the successful modeling, which was counted as the first day of pregnancy. CBA/J×BALB/c mice were set as normal group, and the RSA model pregnant mice were divided into model group, progesterone group, and Bushen Antai Mixture low-, medium-, and high-dosage groups using a random number table method, with 6 mice in each group. The normal group and the model group were given distilled water by gavage. Bushen Antai Mixture low-, medium-, and high-dosage groups were given 3.9 g/kg, 11.7 g/kg and 23.4 g/kg Bushen Antai Mixture by gavage. The progesterone group was given progesterone capsule aqueous solution 26 mg/kg by gavage for 14 consecutive days. The embryo loss rate of mice was calculated. The pathological changes of placental tissues were observed by HE staining. The expressions of Nrf2, HO-1 and NF-κB protein and mRNA in placental tissues were detected by Western blot and RT-qPCR.Results:Compared with the model group, the embryo loss rate of mice in the Bushen Antai Mixture low-, medium-, and high-dosage groups and the progesterone group significantly decreased ( P<0.05, P<0.01); the pathological morphology of placental tissue improved; the expressions of Nrf2, HO-1 protein and mRNA in placental tissue significantly increased ( P<0.05, P<0.01), and the expression of NF-κB protein and mRNA significantly decreased ( P<0.05, P<0.01). Conclusion:Bushen Antai Mixture may may treat RSA by activating the Nrf2/HO-1 pathway, inhibiting NF - κB expression, and improving oxidative stress and inflammatory response in the placental tissue of RSA pregnant mice.

Objective To establish a stable,reliable,and clinically relevant porcine model of endotoxin-induced acute respiratory distress syndrome(ARDS).Methods Ten 8-month-old male Bama minipigs were deeply sedated,followed by invasive mechanical ventilation and electrocardiographic monitoring.Lipopolysaccharide(LPS)was intravenously pumped at 600 μg/(kg·h)for 3 hours,then maintained at 15 μg/(kg·h)thereafter.Dynamic monitoring was performed at five time points after LPS injection(LPS 0,1,3,5,and 8 h),including arterial blood gas analysis and chest computed tomography(CT)scans.Pathological examination of lung tissues obtained via bronchoscopic biopsy(HE staining and transmission electron microscopy)was conducted.These indicators were comprehensively used to evaluate the success of the animal model.Results At 5 hours after LPS administration,8 minipigs developed symptoms such as skin cyanosis,elevated body temperature,and respiratory distress.The oxygenation index decreased to<300 mmHg.Chest CT scans showed diffuse pulmonary infiltrates.Histopathology revealed alveolar edema and hyaline membrane formation.Transmission electron microscopy demonstrated disruption of pulmonary blood-air barrier,depletion of lamellar bodies in type Ⅱ pneumocytes,inflammatory cell infiltration,and exudation of plasma proteins and fibrin.Compared with LPS 0 h,at LPS 8 h,the oxygenation index and arterial blood pH were significantly decreased(P<0.001),while blood lactic acid and serum potassium were significantly increased(P<0.05);serum calcium and base excess were significantly decreased(P<0.05),and the lung injury score based on HE-stained lung sections was significantly increased(P<0.01).Conclusion The porcine ARDS model established by continuous LPS injection can dynamically simulate the pathophysiological characteristics and typical pathological manifestations of clinical septic ARDS,making it an effective tool to study the pathogenesis,prevention,and treatment strategies of septic ARDS.
7.IDENTIFICATION AND COMPOSITION OF MOSQUITO SPECIES IN EMEIFENG NATURE RESERVE,FUJIAN PROVINCE,CHINA

Objective In this preliminary survey,we sought to determine the composition of mosquito species inhabiting the Emeifeng Nature Reserve,Fujian Province,China.Methods Mosquito larvae were collected by straw and spoon trapping,and adult mosquitoes were collected by lamp trapping at selected breeding sites in the reserve.The specimens were initially identified based on morphology,with subsequent verification using molecular biology methods.Results A total of 34 mosquito species in 13 genera were collected,among which,there were 4 species of Anopheles(Genus Anopheles Meigen,1818),2 species of Lutzia(Genus Lutzia Theobald,1903),15 species of Culex(Genus Culex Linnaeus,1758),4 species of Stegomyia(Genus Stegomyia Theobald,1901),and single species of Hulecoeteomyia(Genus Hulecoeteomyia Theobald,1904),Luius(Genus Luius Reinert,Harbach et Kitching,2008),Aedes(Genus Aedes Meigen,1818),Downsiomyia(Genus Downsiomyia Vargas,1950),Collessius(Genus Collessius Reinert,Harbach et kitching,2006),Uranotaenia(Genus Uranotaenia Lynch 1891),Armigeres(Genus Armigeres Theobald,1901),Toxorhynchites(Genus Toxorhynchites Theobald,1901),and pestle mosquito(Genus Tripteroides Giles,1904).Conclusions The species composition of mosquitoes sampled in the Emeifeng Nature Reserve will provide a basis for further research on mosquito vectors and contribute to measures for local mosquito control.
8.Construction and validation of a predictive model for early acute kidney injury in patients with cardiac arrest after cardiopulmonary resuscitation

Objective:To construct a nomogram model for predicting the occurrence of acute kidney injury (AKI) in patients with cardiac arrest (CA) after cardiopulmonary resuscitation (CPR), and to verify its validity for early prediction.Methods:The study retrospectively included patients aged 18 years and older who received CPR for CA and were admitted to the emergency room of Tianjin Medical University General Hospital from February 2016 to September 2023. The general information, underlying diseases, resuscitation related indicators, and first laboratory test results of patients were collected. The patients were randomly divided into training and validation groups at a ratio of 7:3. AKI diagnosis was based on the diagnostic criteria of the Kidney Disease Improving Global Outcomes. Univariate and multivariate logistic regression models were used to identify independent risk factors for AKI in patients with cardiac arrest, and a nomogram was constructed on the basis of the independent risk factors. The predictive performance was evaluated by the area under the curve (AUC) of the receiver operating characteristic. The calibration curve, decision curve and clinical impact curve were used to evaluate the model. Bootstrap and cross validation methods were used for internal validation.Results:A total of 527 patients with cardiac arrest were included in the study, 230 patients developed AKI, with an AKI incidence of 43.6%. There was no statistically significant difference in clinical baseline data between the training and validation groups (all P>0.05), indicating comparability between the two groups of data. Multivariate logistic analysis revealed that age ( OR=1.346, 95% CI: 1.197-1.543, P<0.001), CA to CPR time ( OR=2.214, 95% CI: 1.512-3.409, P=0.016), adrenaline dosage ( OR=1.921, 95% CI: 1.383-2.783, P=0.004), APACHE-Ⅱ score ( OR=1.531, 95% CI: 1.316-1.820, P<0.001), baseline creatinine ( OR=1.137, 95% CI: 1.090-1.196, P<0.001), and lactate ( OR=2.558, 95% CI: 1.680-4.167, P<0.001) were the independent risk factors for AKI in patients with cardiac arrest. Initial defibrillable rhythm ( OR=0.214, 95% CI: 0.051-0.759, P=0.023) was a protective factor for AKI in patients with cardiac arrest. A nomogram prediction model was constructed based on the above variables. The AUC of the training group was 0.943 (95% CI: 0.921-0.965) and that of the validation group was 0.917 (95% CI: 0.874-0.960). This prediction model demonstrated good discrimination, calibration and clinical applicability. Conclusions:A nomogram predictive model was constructed on the basis of age, CA to CPR time, initial defibrillable rhythm, adrenaline dosage, the APACHE-Ⅱ score, and baseline creatinine and lactate levels. This nomogram has good predictive value for the early occurrence of AKI in patients with cardiac arrest after cardiopulmonary resuscitation, which can provide new strategies for the early identification of AKI and precise intervention.
9.Interactive network dynamic nomogram for predicting poor neurological outcomes of post-cardiac arrest brain injury patients

Objective:To develop and validate an interactive network dynamic nomogram for early prediction of poor neurological prognosis in patients with post-cardiac arrest brain injury (PCABI).Methods:A retrospective study was conducted on hospitalized patients who achieved return of spontaneous circulation after cardiac arrest at Tianjin Medical University General Hospital between January 2020 and April 2024. Patients were classified into favorable and poor prognosis groups based on the Glasgow-Pittsburgh Cerebral Performance Category at discharge. Eligible patients were randomly assigned to a training cohort and an internal validation cohort in a 7:3 ratio. Univariate and multivariate logistic regression analyses were performed to identify independent predictors of poor neurological outcomes in PCABI, which were subsequently used to develop a nomogram prediction model. The predictive performance of the nomogram was evaluated by comparing its area under the curve (AUC) of receiver operating characteristic with those of individual predictors using the DeLong test. Model calibration and clinical utility were assessed using calibration curves and decision curve analysis, respectively. Internal validation was conducted, and an interactive dynamic nomogram was developed using web-based visualization techniques.Results:A total of 276 PCABI patients were enrolled (training set: 196; validation set: 80), with 82 cases (29.7%) classified as poor prognosis. Multivariate logistic regression analysis identified age ( OR=1.071, 95% CI: 1.021-1.124, P=0.005), APACHEⅡ score ( OR=1.746, 95% CI: 1.393-2.190, P<0.001), initial shockable rhythm ( OR=0.142, 95% CI: 0.025-0.819, P=0.029), defibrillation ( OR=0.228, 95% CI: 0.060-0.869, P=0.030), cardiopulmonary resuscitation duration ( OR=2.116, 95% CI: 1.487-3.010, P<0.001), and lactate level ( OR=1.392, 95% CI: 1.005-1.927, P=0.047) as independent predictors of poor neurological outcomes in PCABI. A nomogram prediction model was developed based on these factors, achieving an AUC of 0.965 (95% CI: 0.939-0.989) in the training cohort and 0.987 (95% CI: 0.967-1.000) in the internal validation cohort. The nomogram demonstrated significantly superior predictive performance compared to individual predictors ( P<0.001) and exhibited excellent discrimination, calibration, and clinical net benefit. The interactive dynamic nomogram, developed through web-based visualization, further enhanced its applicability in clinical practice. Conclusions:The interactive network dynamic nomogram, developed based on age, APACHEⅡ score, initial shockable rhythm, defibrillation, cardiopulmonary resuscitation duration, and lactate level, demonstrated favorable predictive value for poor neurological outcomes in PCABI. This tool facilitates clinical application and offers a novel strategy for early identification and targeted interventions in high-risk patients.
10.Prediction of early in-hospital major adverse events by D-dimer level in patients with acute type A aortic dissection: A retrospective cohort study

Objective To investigate the association between D-dimer levels within 2 hours of admission and in-hospital major adverse events (MAEs) in patients with acute type A aortic dissection (ATAAD) who underwent total arch replacement combined with frozen elephant trunk (FET) implantation. Methods This retrospective study included patients with ATAAD who underwent total arch replacement with FET implantation at Fuwai Yunnan Cardiovascular Hospital from September 2017 to December 2022. Patients were divided into two groups based on the occurrence of in-hospital MAEs: a MAEs group and a non-MAEs group (control). Perioperative data were compared between the two groups. Univariate and multivariate logistic regression analyses were performed to identify risk factors for in-hospital MAEs, which included in-hospital death, gastrointestinal bleeding, paraplegia, acute kidney injury, low cardiac output syndrome, stroke, respiratory failure, multiple organ dysfunction syndrome, and severe infection. The predictive value of D-dimer was evaluated using the area under the receiver operating characteristic curve (AUC). Results A total of 218 patients were included (157 males, 61 females), with a mean age of (51.54±9.79) years. There were 152 patients in the non-MAEs group and 66 in the MAEs group. The overall incidence of in-hospital MAEs was 30.3%, and the in-hospital mortality rate was 2.8% (6/218). Compared to the non-MAEs group, the MAEs group had significantly higher levels of D-dimer and lactate, as well as longer cardiopulmonary bypass time, aortic cross-clamp time, and ICU length of stay (all P<0.05). Multivariate logistic regression analysis identified D-dimer as an independent risk factor for in-hospital MAEs [OR=1.077, 95%CI (1.020, 1.137), P=0.013]. The AUC for the D-dimer level within 2 hours of admission to predict in-hospital MAEs was 0.83 [95%CI (0.736, 0.870), P<0.001]. The optimal cutoff value was 2.2 μg/mL, with a sensitivity of 84.8% and a specificity of 73.0%. Conclusion The serum D-dimer level is an independent risk factor for in-hospital MAEs in patients with ATAAD following total arch replacement with FET implantation. D-dimer levels on admission can help clinicians optimize risk stratification and perioperative management, potentially reducing the incidence of early adverse events.
