1.Screening of diagnostic markers for endothelial cell Senescence in mice with radiation-induced heart disease and analysis of immune infiltration

BACKGROUND:Radiotherapy significantly improves survival rates in patients with various malignant tumors.However,with prolonged post-treatment survival,many patients face the risk of radiation-related cardiac toxicity.This is especially true after chest radiotherapy,where the risk of radiation-induced heart disease significantly increases,becoming one of the most severe complications affecting prognosis survival.OBJECTIVE:To identify diagnostic markers of endothelial cellular senescence in radiation-induced heart disease through systematic transcriptomic analysis.METHODS:Firstly,genes associated with cellular senescence were screened from the CellAge database and intersected with the transcriptomic training dataset of a mouse model of radiation-induced heart disease to identify differentially expressed senescence-related genes.Secondly,weighted gene co-expression network analysis and machine learning were used to identify key hub genes that play critical roles in radiation-induced heart disease.The expression of these genes was validated using a dataset of radiation-induced endothelial injury.Additionally,the quanTlseq method was employed to assess the immune infiltration status related to radiation-induced heart disease.The expression levels of key genes and their association with survival in esophageal squamous cell carcinoma patients receiving chest radiotherapy were explored through the analysis of The Cancer Genome Atlas database.RESULTS AND CONCLUSION:(1)Systematic transcriptomic analysis identified CCND1 as the core gene of endothelial cellular senescence in radiation-induced heart disease,and this finding was validated in the mouse model of radiation-induced heart disease.(2)The diagnostic model constructed from these data indicated that CCND1 had high specificity and sensitivity for diagnosing radiation-induced heart disease.(3)Immune infiltration analysis revealed significant immune response dysregulation in the mouse model of radiation-induced heart disease,and CCND1 was closely related to various immune cells.(4)Kaplan-Meier survival analysis showed that CCND1 was associated with poorer disease-specific survival in esophageal squamous cell carcinoma patients receiving chest radiotherapy.This study systematically uncovers,for the first time,the pivotal role of CCND1 in endothelial cell senescence associated with radiation-induced heart disease.CCND1,a gene integral to cell cycle regulation,can induce cellular senescence when abnormally expressed.Furthermore,the findings highlight its potential as an early diagnostic marker.
2.Pharmaceutical care for a rare case of diffuse pulmonary microabscesses complicated with organizing pneumonia

OBJECTIVE To investigate the role of clinical pharmacists in the individualized treatment of a rare case of diffuse pulmonary microabscesses caused by Streptococcus constellatus complicated with organizing pneumonia (OP) in a non-immunocompromised patient. METHODS For a middle-aged, non-immunocompromised patient with a one-year disease course, whose imaging findings showed the coexistence of diffuse microabscesses and OP caused by S. constellatus pulmonary abscess, the clinical pharmacist, based on pharmacokinetics/pharmacodynamics characteristics, assisted the physician in optimizing the previously ineffective anti-infective regimen to Ceftriaxone sodium for injection combined with Metronidazole and sodium chloride injection, so as to enhance coverage of mixed anaerobes and lesion penetration. After the patient’s hemoptysis ceased, the clinical pharmacist recommended timely initiation of low-dose Methylprednisolone sodium succinate for injection to manage OP. Upon discharge, sequential oral therapy with Linezolid tablets, Metronidazole tablets, Methylprednisolone tablets and Omeprazole enteric-coated tablets was prescribed. RESULTS After implementation of the individualized treatment regimen, the patient’s hemoptysis resolved, pulmonary lesions significantly regressed, and the patient was discharged in a stable condition. At the three-month follow-up, the patient remained stable. CONCLUSIONS For chronic persistent pulmonary infection complicated with OP in a non-immunocompromised host, clinical pharmacists assist clinicians in optimizing anti-infective regimens and recommend the timely initiation of anti-inflammatory therapy after infection control, thereby contributing to favorable clinical outcomes. Individualized treatment of such complex cases requires comprehensive consideration of pathogen coverage, pathological barriers, and the timing of anti-inflammatory intervention.

4.Involvement of piriform cortex in 5-choice serial reaction time task(5-CSRTT)attention behavior in mice

Objective To investigate the role of the piriformcortex in regulating attentional behavior in mice.Methods Adult male C57BL/6J mice were subjected to a 5-choice serial reaction time task(5-CSRTT).Immunofluorescence staining was performed to detect expressions of cellular FBJ murine osteosarcoma viral oncogene homolog protein(c-Fos)in the piriform cortex.The changes of attentional behavior in the 5-CSRTT test were explored following either stereotactic injection of diphtheria toxin A virus to specifically damage piriform cortex neurons or chemogenetical inhibition of the neuronal activity inthe piriform cortex.Results It was found that the expression of c-Fos in the 5-CSRTT-tested mice was significantly increased compared to the control.Both lesion and chemogenetic inhibition of piriform cortex neuronsreduced the accuracy of attention,but omission rates and premature responses remained unaffected in the 5-CSRTT test.Conclusion Piriform cortex neuronsmay play a critical role in modulating attentional processes in mice.
5.The effectiveness and safety of a percutaneous controllable curved plasma radiofrequency ablation device of nucleus pulposus

Objective To verify the safety and effectiveness of a new percutaneous controllable curved plasma radiofrequency instrument for nucleus pulposus ablation. Methods A new percutaneous controllable curved plasma radiofrequency instrument were designed (controllable curved group), and its ablation effect was compared with the currently used straight head non-bendable plasma ablation instrument (non-bendable group) on gross specimens. The ablation instrument was placed through the right intervertebral foramen, and continuous ablation on the same intervertebral disc was conducted for three times. The ablation range and trajectory were recorded, and the temperature changes in the front, back, left, and right of the ablation center during and 15 seconds after ablation were monitored by the inserted temperature probe. Results There were no difference in temperature changes in the front, back, right regions of the ablation center during and 15 seconds after ablation between the two groups. The temperature changes in the left region of the ablation center both during and 15 seconds after 3rd ablation were larger than those in the non-bendable group (P<0.01). Compared with the non-bendable group, the controllable curved group achieved angle control and larger single ablation area (2.282 5 mm² vs 1.135 8 mm², P<0.000 1). Conclusions This new percutaneous controllable curved plasma ablation instrument can achieve angle control and ablation on the side opposite to the puncture site, increase ablation volume, and is safe.
6.Epidemiological characteristics of scrub typhus in Huai'an, Jiangsu Province in 2006 - 2024

Objective To describe and analyze the epidemiological characteristics of scrub typhus in Huai'an, Jiangsu Province from 2006 to 2024 and explore the long-term incidence trend and distribution of high-risk areas, and to formulate targeted prevention and control strategies. Methods The scrub typhus case report data of Huai'an from 2006 to 2024 in the Chinese Disease Prevention and Control Information System were extracted for descriptive analysis. Results A total of 898 cases of scrub typhus were reported in Huai'an, with an average annual incidence rate of 0.96 per 100 000 from 2006 to 2024. There was a turning point in the incidence trend of scrub typhus in 2011. From 2006 to 2011, the annual percentage change (APC) was 47.09% (95% CI: 7.53 - 859.39), and the upward trend was statistically significant (P < 0.05). From 2012 to 2024, the APC was -2.12% (95% CI: -29.09 - 3.75), and the downward trend was not statistically significant. October and November were the high-incidence months, and the total concentration from 2006 to 2024 was 0.93, indicating that scrub typhus had strict seasonality. The circular distribution method estimated that the peak period of the epidemic was from October 11th to November 25th, and the peak day of incidence was November 3rd. Jinhu County was a high-incidence area. The ratio of male to female cases was 1.03. The age group with the highest reported incidence was 40 to < 80 years old. The occupation with the highest proportion was farmers, accounting for 78.03%. Conclusion From 2006 to 2024, scrub typhus in Huai'an shows a peak every 3 - 4 years. Middle-aged and elderly farmers are the key population at risk, and Jinhu County is a key area. In the future, targeted health education should be carried out to effectively control the prevalence of scrub typhus.
7.Construction of A Chiral Separation Method Using Polystyrene-cyclodextrin Metal-organic Framework Coating for Open-tubular Capillary Electrochromatography

By using a strategy of leveraging the ability of metal-organic frameworks(MOFs)materials to precisely regulate the spatial orientation of cyclodextrins(CD),a polystyrene-modified γ-cyclodextrin metal-organic frameworks(PS-CD-MOFs)capillary coating was established and applied to the chiral separation of amino acids in open-tubular capillary electrochromatography(OT-CEC)based on the excellent film-forming property of polystyrene(PS).Characterization results by Fourier transform infrared spectroscopy(FT-IR),X-ray diffraction(XRD)indicated that PS was successfully grafted onto the surface of CD-MOFs.The modification significantly improved the structural stability and thermal stability of CD-MOFs while maintaining the integrity of the MOFs.The PS-CD-MOFs coated capillary electrophoresis system exhibited excellent performance in separating dansylated amino acid enantiomers(Dns-D,L-AAs).Specifically,under the optimal separation conditions(Sodium dodecyl sulfate/boric acid buffer system,20 cm capillary,and 10 kV effective voltage),good separation was achieved for D,L-methionine(D,L-Met)and D,L-serine(D,L-Ser).Further quantitative analysis showed that Dns-D,L-Met presented a good linear relationship in the concentration range of 10.0-1500.0 μmol/L,with a correlation coefficient close to 0.998,demonstrating high sensitivity and repeatability.Not only did it overcome the problem that traditional CD(used as additives in capillary electrophoresis)could not precisely control the spatial orientation of chiral resolvents,but also it solved the issues of insufficient stability and bonding amount of CD-MOFs coatings by utilizing the excellent film-forming property of polymers on the inner wall of capillaries.This study provided an efficient and controllable new strategy for construction of chiral separation stationary phases.

Forensic medicine has been designated as a first-level discipline,presenting new opportunities and challenges for the development of forensic medicine.Since the 1980s,the establishment of foren-sic medicine discipline and the cultivation of high-level forensic talents have become hot topics in the development of forensic medicine in China.Since the 13th Five-Year Plan,the forensic team of Shanxi Medical University has been aiming at the forefront,proposing the development goals of"Five First-class"and the discipline development path"Six Major Achievements".It has selected benchmark disci-plines,identified gaps in disciplinary development,unified thoughts,formulated completion timelines,concentrated superior resources,assigned tasks to individuals,and created an"Organized Fill-in-the-Blank Format"forensic medicine discipline construction model with the characteristics of the new era.The construction model of forensic medicine has achieved good results in the goals,discipline frame-work,scientific research,talent cultivation,discipline team and platform construction,forming a rela-tively complete discipline construction and management system,and accumulating valuable experience for the construction of first-level discipline and high-level talent cultivation of forensic medicine.
9.Fluorescent Probe Development for Rapid Detection of Tiletamine Based on Cop-per Nanozyme and Molecular Imprinting Technology

Objective To develop a rapid detection method for tiletamine that is easy to operate and low-cost under the premise of ensuring sensitivity and accuracy,to assist in carrying out rapid screening and drug control work on-site.Methods This study integrates dual-ligand copper nanozymes with mo-lecular imprinting technology.Initially,copper nanozymes were synthesized using readily available raw materials at 120℃.Subsequently,specific cavities were imprinted on their surface at room temperature using a sol-gel method to construct a novel fluorescent sensing probe.This probe was characterized and methodologically validated,and then applied to the detection of actual samples.Results The developed probe exhibited stable fluorescence properties,strong anti-interference capability,and excellent specificity and sensitivity,with a detection limit of 5 ng/mL and a quantitative concentration range from 15 to 500 ng/mL.It enabled the rapid detection of tiletamine in real samples such as blood and e-cigarette oil.Conclusion This fluorescent probe can be used for rapid detection and on-site preliminary screening of tiletamine in various types of samples.It significantly improves the detection efficiency and reduces analysis costs,showing high research value and broad application prospects.
10.The Effect of Tai Chi Practice on Balance Control Ability in Single-Leg Stance for the Individuals with Functional Ankle Instability:An Intervention Study

Objective To determine the effect of Tai Chi practice on balance control ability of the individuals with functional ankle instability(FAI)in single-leg stance(SLS),and further observe the augmented effect from attention focus on ankle.Methods A total of 46 individuals with FAI were randomly divided into ankle focus group,free focus group and control group.The ankle focus group and free focus group performed Tai Chi practice for 10 weeks;the ankle focus group focused their attention on the ankle joint,while the free focus group didn't require attention focus;the control group maintained the origin lifestyle and didn't engage in special physical activities.The three-dimensional force plate and Y balance test(YBT)were used to evaluate the static(with eyes open and closed)and dynamic(instant and continuous)balance ability of individuals with FAI.Results For ankle focus group and free focus group,the anterior-posterior(AP)and medial-lateral(ML)sway distances of center of pressure(COP)in SLS with eyes open and closed were significantly reduced,and the reach distance in YBT in three directions were increased.For the ankle focus group,the ML stability index and the dynamic postural stability index were significantly reduced during single-leg drop landing.The ankle focus group and free focus group had a significantly smaller ML COP sway distance than control group in SLS with eyes open;the ankle focus group had a significantly smaller ML COP sway distance than free focus group and control group in SLS with eyes closed;and the reach distance of ankle focus group in posterior-medial&posterior-lateral direction in YBT was longer than that of free focus group and control group.Conclusions Tai Chi practice can comprehensively improve static and dynamic balance control ability of the individuals with FAI in SLS,and ankle attention-focused Tai Chi practice is more effective,especially for the enhancement of their ML control ability in static state with eyes closed and in continuous dynamic SLS.
