1.Enzyme-independent functions of HDAC3 in the adult heart.

The cardioprotective effects of histone deacetylase (HDAC) inhibitors (HDIs) are at odds with the deleterious effects of HDAC depletion. Here, we use HDAC3 as a prototype HDAC to address this contradiction. We show that adult-onset cardiac-specific depletion of HDAC3 in mice causes cardiac hypertrophy and contractile dysfunction on a high-fat diet (HFD), excluding developmental disruption as a major reason for the contradiction. Genetically abolishing HDAC3 enzymatic activity without affecting its protein level does not cause cardiac dysfunction on HFD. HDAC3 depletion causes robust downregulation of lipid oxidation/bioenergetic genes and upregulation of antioxidant/anti-apoptotic genes. In contrast, HDAC3 enzyme activity abolishment causes much milder changes in far fewer genes. The abnormal gene expression is cardiomyocyte-autonomous and can be rescued by an enzyme-dead HDAC3 mutant but not by an HDAC3 mutant (Δ33-70) that lacks interaction with the nuclear-envelope protein lamina-associated polypeptide 2β (LAP2β). Tethering LAP2β to the HDAC3 Δ33-70 mutant restored its ability to rescue gene expression. Finally, HDAC3 depletion, not loss of HDAC3 enzymatic activity, exacerbates cardiac contractile functions upon aortic constriction. These results suggest that the cardiac function of HDAC3 in adults is not attributable to its enzyme activity, which has implications for understanding the cardioprotective effects of HDIs.

Digital technologies have become an integral part of complete denture restoration. With advancement in computer-aided design and computer-aided manufacturing (CAD/CAM), tools such as intraoral scanning, facial scanning, 3D printing, and numerical control machining are reshaping the workflow of complete denture restoration. Unlike conventional methods that rely heavily on clinical experience and manual techniques, digital technologies offer greater precision, predictability, and efficacy. They also streamline the process by reducing the number of patient visits and improving overall comfort. Despite these improvements, the clinical application of digital complete denture restoration still faces challenges that require further standardization. The major issues include appropriate case selection, establishing consistent digital workflows, and evaluating long-term outcomes. To address these challenges and provide clinical guidance for practitioners, this expert consensus outlines the principles, advantages, and limitations of digital complete denture technology. The aim of this review was to offer practical recommendations on indications, clinical procedures and precautions, evaluation metrics, and outcome assessment to support digital restoration of complete denture in clinical practice.

Paraplegia caused by spinal cord injury is a serious neurological complication, for which surgery is currently the main treatment method. Due to different surgical approaches, patients are usually expected to maintain a passive prone position for a long time or switch between the supine and prone positions. Affected by multiple factors such as neurogenic sensory disorders, pathological changes in muscle tone and operative duration, the risk of intraoperative acquired pressure injury (IAPI) is significantly increased. Current clinical prevention strategies for IAPI in these patients predominantly focus on localized pressure relief during positioning, lacking systematic, standardized comprehensive prevention protocols or evidence-based guidelines. To address it, Department of Nursing, Orthopedics Branch, China International Exchange and Promotive Association for Medical and Health Care, Spinal Trauma Professional Committee, Orthopedics Branch, Chinese Medical Doctor Association, Nursing Group of Spine and Spinal Cord Professional Committee of Chinese Association of Rehabilitation Medicine organized experts in relevant fields to formulate Guideline for the prevention of intraoperative acquired pressure injury in paraplegic patients with spinal cord injury ( version 2025), based on evidence-based medical evidence and latest research results and clinical practice at home and abroad. Eleven recommendations were put forward from the aspects of preoperative risk assessment, intraoperative prevention strategies, postoperative handover and monitoring, and supportive mechanisms for IAPI prevention, aiming to standardize the prevention measures and management strategies of IAPI in paraplegic patients with spinal cord injury and accelerate the recovery of patients and improve the therapeutic effect.
4.Epidemiological study on traditional Chinese medicine treatment for inflammatory bowel disease in Jiangsu Province from 2019 to 2023

Objective:To explore patterns of traditional Chinese medicine (TCM) use among patients with inflammatory bowel disease (IBD) in Jiangsu Province, China from 2019 to 2023.Methods:Using data from the IBD health data platform of the National Healthcare Big Data (Eastern) Center, a retrospective cohort study was conducted. We performed descriptive analyses on hospitalised patients diagnosed with IBD between 2019 and 2023, who received TCM treatment.Results:The study included 11 095 case records from 4 760 patients, with TCM diagnoses primarily indicating diarrhoea and abdominal pain. Ulcerative colitis (UC) accounted for 4 782 hospitalizations (3 103 patients), while Crohn's disease (CD) accounted for 6 313 hospitalizations (1 657 patients). Patient demographics showed a trend towards younger age and a higher proportion of males. Treatment utilisation was highest in southern Jiangsu compared with the central and northern regions. In terms of disease burden, all treatment costs showed a downward trend. In terms of external TCM therapies, UC patients tend to prefer plasters and enemas, while CD patients are more inclined to use acupuncture. Regarding herbal medicine, licorice, white atractylodes, and white peony root are commonly used single herbs for IBD patients.Conclusions:The number of IBD patients treated with TCM in Jiangsu Province has steadily increased from 2019 to 2023. It is important to identify effective TCM treatment methods to reduce the burden of patients.
5.Relationship between ultrasound differences of bilateral middle cerebral arteries and prognosis of AACIS after endovascular treatment

Objective To investigate the blood flow difference of transcranial Doppler ultrasound of bilateral middle cerebral arteries in patients with acute anterior circulation ischemic stroke(AACIS)after endovascular treatment,and analyze the influencing factors for prognosis of AACIS patients treated with stent combined with thrombolysis.Methods A retrospective analy-sis was conducted on 168 AACIS patients undergoing mechanical embolectomy in our department from May 2020 to April 2022.According to the treatment regimens,they were divided into simple thrombolysis group(72 patients)and combined thrombolysis group(96 patients).The combined thrombolysis group was assigned into good prognosis subgroup(65 cases)and poor prognosis subgroup(31 cases)according to the score Modified Rankin scale(mRS).Kaplan-Meier curve and multivariate logistic regression analyses were employed to identify the influencing factors for the poor prognosis in AACIS patients.Results Kaplan-Meier survival curve analysis showed that the combined thrombolysis group was associated with a good prognosis(P=0.013).After treatment,the combined thrombolysis group obtained significantly higher average blood flow velocity(Vm)of middle cerebral artery(MCA)and Alberta stroke program early computed tomography score(ASPECTS)than the simple thrombolysis group(P<0.01).The good prognosis subgroup had significantly younger age,larger ratios of blood sugar ≤6.8 mmol/L and ASPECTS>2,higher collateral circulation score in MCA region,and elevated Vm than the poor prognosis subgroup(P<0.05,P<0.01).Multivariate logistic regression analysis showed that blood glucose ≤ 6.8 mmol/L,collateral circulation score in MCA region,ASPECTS>2 and Vm were protective fac-tors for good prognosis of AACIS patients(P<0.05,P<0.01).Conclusion Stent combined with thrombolytic therapy for AACIS has a lower risk for poor prognosis,and blood glucose ≤6.8 mmol/L,higher Tan score,DWI-ASPECTS>2,and higher Vm are protective factors.
6.Evaluation of Ice Hockey Helmet Protective Performance Grade Based on Entropy Weight Technique for Order Preference by Similarity to an Ideal Solution Combined with Rank Sum Ratio Method

Objective The combined entropy weight technique for order preference by similarity to an ideal solution(TOPSIS)and rank sum ratio(RSR)methods were utilized to rate the protective performance of ice hockey helmets,and the effectiveness and influencing factors of the rating system,as well as the relationship between protective performance and purchase price were explored.Methods The linear acceleration data during impact drops of twenty-four ice hockey helmets were collected using a uniaxial accelerometer in a collision test machine after low and ambient temperature treatments.The protective performance of ice hockey helmets was rated using the STAR model combined with the entropy weight TOPSIS and RSR methods.One-way ANOVA or the Kruskal-Wallis H test was employed to assess the differences in protective performance among helmets across different grade groups.The correlation between different indicators was analyzed by Pearson's correlation coefficient.Results The comprehensive protective performance ratings of the CCM TACKS 310,IBX,BAUD,and WARRIORS COVERT RS PRO helmets were classified as poor,whereas the BAUER REAKT 150,HYPERLITE,and REAKT 200 helmets were rated as excellent.The remaining helmets were rated as moderate.There was a moderate to high positive correlation between the ambient temperature STAR(A)and Rowson's STAR values,the comprehensive Ti and low temperature Ti,the comprehensive T,and ambient temperature Ti(P<0.05).The comprehensive Ti satisfied the homogeneity of variance(P>0.05)and exhibited significant differences among groups(P<0.05).Significant differences were found in the low temperature indicators among different groups(P<0.05),and their weight coefficients ranked among the top three.The ambient temperature indicators were not affected by the protective performance grade(P>0.05).A weak positive correlation existed between the comprehensive Ti and purchase price(P<0.05).Conclusions According to the energy absorption test protocol,the combined entropy weight TOPSIS and RSR methods can efficiently rate the comprehensive protective performance of ice hockey helmets.The effectiveness of using low temperature indicators for the comprehensive evaluation and rating of ice hockey helmet protective performance is superior to that of ambient temperature indicators and purchase price.Consumers are advised not to use price as a criterion for evaluating the comprehensive protective performance of ice hockey helmets.
7.Intratumoral and peritumoral CT radiomics for predicting Ki-67 expression level of lung adenocarcinoma with mixed ground glass nodules

Objective To observe the value of intratumoral and peritumoral CT radiomics for predicting Ki-67 expression level of lung adenocarcinoma with mixed ground glass nodules.Methods Totally 284 cases of pathologically confirmed lung adenocarcinoma with mixed ground glass nodules were retrospectively enrolled,among them 197 cases were taken as training set(54 cases with high and 143 cases with low Ki-67 expression)and 87 cases as validation set(27 cases with high and 60 cases with low Ki-67 expression).Intratumoral and peritumoral radiomic features were obtained from non-contrast chest CT,and radiomic models for predicting Ki-67 expression in lung adenocarcinoma with mixed ground glass nodules were established using adaptive boosting,light gradient boosting machine(LightGBM)and multilayer perceptron algorithms based on intratumoral features,peritumoral features,as well as intratumoral+peritumoral features,respectively,and the optimal radiomics signature was selected according to the area under the receiver operating characteristic curve(AUC).Univariate and multivariate logistic regression analysis were performed to identify independent impact factors of Ki-67 expression level,and a clinical model was constructed,and the efficacy of the models were evaluated.Results Among radiomics models,LightGBMintratumoral+peritumoral model had the highest AUC(0.934 in training set and 0.845 in validation set),which were superior to that of clinical model(0.616 in training set and 0.684 in validation set)(both P<0.05)Conclusion Intratumoral and peritumoral CT radiomics had good efficacy for predicting Ki-67 expression level of lung adenocarcinoma with mixed ground glass nodules.
