1.Protective effect of overexpression of miR-140-5p mediated by lentivirus on lipopolysaccharide-induced liver injury in rats

Objective:To explore the protective effect of lentivirus-mediated miR-140-5p overexpression on lipopolysaccha-ride-induced liver injury in rats.Methods:A total of 40 rats were randomly divided into control group,model group,miR-NC group and miR-140-5p-mimic group,with 10 rats in each group.Rats in miR-NC group and miR-140-5p-mimic group received tail vein injection of lentivirus empty vector miR-NC and recombinant miR-140-5p respectively,while rats in control group and model group received tail vein injection of normal saline.Three days later,rats in model group,miR-NC group and miR-140-5p-mimic group received intraperi-toneal injection of 10 mg/kg lipopolysaccharide,while rats in control group received intraperitoneal injection of normal saline.All rats were killed 12 hours after injection,and the serum levels of ALT,AST,endotoxin,TNF-α,IL-6,IL-1β,SOD,MDA and CAT were detected.Pathological morphology and apoptosis of liver tissue were observed by HE and TUNEL staining.TargetScanHuman was used to predicted binding site of miR-140-5p and TLR4,and double luciferase reporter gene assay was used to analyse the targeted regula-tion relationship between miR-140-5p and TLR4.Level of miR-140-5p in liver tissue was detected by qRT-PCR,mRNA and protein expressions of TLR4,MyD88 and NF-κB in liver tissue were detected by qRT-PCR and Western blot,and expression of p-NF-κB was detected by Western blot.Results:In control group,the hepatic lobule was clearly visible,and there was no hepatocyte degeneration,necrosis and inflammatory cell infiltration.Obvious hepatocyte necrosis,vacuolar degeneration,hyperemia and infiltration of inflam-matory cells into hepatic sinusoids were observed in model group and miR-NC group.Hepatocyte hyperemia,necrosis,vacuolar degen-eration and inflammatory cell infiltration in miR-140-5p-mimic group were significantly less than those in model group and miR-NC group.miR-140-5p can directly target TLR4.Compared with control group,levels of ALT,AST,endotoxin,apoptotic cells,TNF-α,IL-6,IL-1β,MDA,TLR4,MyD88 mRNA and protein,NF-κB mRNA,p-NF-κB in model group,miR-NC group and miR-140-5p-mimic group were increased(P<0.05),while SOD and CAT were decreased(P<0.05).Compared with model group and miR-NC group,levels of ALT,AST,endotoxin,apoptotic cells,TNF-α,IL-6,IL-1β,MDA,TLR4,MyD88 mRNA and protein,NF-κB mRNA and p-NF-κB in miR-140-5p-mimic group were decreased(P<0.05),while levels of SOD and CAT were increased(P<0.05).Conclusion:Overexpression of miR-140-5p can reduce the level of inflammatory reaction and oxidative stress in rats with lipopolysac-charide-induced liver injury,protect hepatocytes and reduce liver pathological injury,and its mechanism may be related to TLR4/MyD88/NF-κB signal pathway.

3.Anti-mGluR5 encephalitis with mental disorders as the initial symptom: a case report

This article reported the clinical features of a rare patient with anti-metabotropic glutamate receptor 5 (mGluR5) encephalitis with mental disorders as the initial symptom, so as to provide references for clinical diagnosis and treatment. The patient was a 38-year-old male, developed pharyngeal pain as prodromal symptoms, and the main clinical manifestations included rapidly progressive memory loss, anxiety and depression, and psychomotor excitement symptoms including irritability and impulsive behaviors. The disease had a progressive deterioration. In the most severe state, the patient became unconscious in a shallow coma, with further cognitive decline, hallucinations and delusions, and lack of self-awareness. Both cerebrospinal fluid and serum anti-mGluR5 antibody were strongly positive (1∶100). After two sessions of hormone shock therapy, the patient showed significantly improvement in consciousness, cognitive, emotional and psychiatric dimensions.
4.Expression of monocarboxylate transporter 8 mRNA in the brain tissue of rats with cerebral ischemia.

OBJECTIVETo investigate the mRNA expression of monocarboxylate transporter 8 (MCT8), a thyroid hormone transport protein, in the lateral ventricle of rats with cerebral ischemia.
METHODSImmunofluorescence staining was used to observe the expression of MCT8 in the lateral ventricle of 5 normal SD rats. Another 20 adult male SD rats were randomized into 4 groups and subject to permanent ligation of both the common carotid arteries (2-vessel occlusion, 2VO) for 3 days, 2 weeks, or 5 weeks, or no ligation (control). At the end of the experiment, the transcriptional level of MCT8 in the brain tissue of the rats were detected using fluorescent quantitative PCR.
RESULTSMCT8 mRNA levels in 3-day and 2-week 2VO groups were comparable with that in the control group (P=0.909; P=0.694), but increased significantly in 5-week 2VO group compared with that in the control and 3-day 2VO groups (P=0.029; P=0.023). No significance was found in MCT8 mRNA between the 2-week and 5-week 2VO groups (P=0.065).
CONCLUSIONProlonged cerebral ischemia causes compensatory increase of MCT8 mRNA expression on the capillary endothelial cell membranes in the lateral ventricle of rats.

5.MRI Diagnosis of Ligament Trauma of Knee

Objective To evaluate the value of magnetic resonance imaging(MRI) in diagnosing ligament injury of the knee.Methods MRI manifestations of ligament injury of the knee confirmed by operation or joint-screen examination in 32 cases were retrospectively analyzed. All cases underwent routine MR scanning. The cases with suspicious anterior cruciate ligament injury undenwent oblique saqittal section fast spin echo-T_2 weight (FSE-T_2WI )and the cases with suspicious medical accessory ligament injury underwent coronal section fat suppressed-fast spin echo-T_2 weight(FS-FSE-T_2W).Results 36 ligaments were found having injury in all cases, including 16 anterior cruciare ligaments, 14 posterior cruciate ligaments, 5 medical accessory ligaments and 1 lateral accessory ligament. The ligaments injuries included tear completely and part.Conclusion MRI is the ideal non-invasive method for studying ligament injury of the knee.
6.Analysis of clinical characteristics and prognosis of young patients with rectal carcinoma

Objective To investigate the clinical characteristics of young rectal carcinomain in order to improve the diagnostic level and therapeutic efficacy at the early stage.Methods Clinical data,including clinical manifestations,misdiagnosis,clinical pathology and prognosis which from 47 cases of young rectal carcinoma were analyzed retrospectively and compared with those from 99 cases of old rectal carcinoma.Results The average course of disease was 6.98 months in young patient group,it was shorter than that in old patient group(9.16 months).The misdiagnosis rate of young group(61.70%) was significantly higher than that in old group(28.28%)(P 0.05).Conclusion Compared with the old patients,advanced stage and poorly differentiated carcinoma are more common in young patients with rectal carcinoma.But the 5-year survival rates of the young and old patients are similar.In addition,T stage and lymph node metastasis are the independent prognostic factors.Therefore,early diagnosis and treatment are crucial to improve the survival rate in young patients with rectal carcinoma.
