1.Dendritic cell subpopulations in the tumor microenvironment: biological behaviors and targeted therapeutic strategies

[摘 要] 树突状细胞(DC)作为抗肿瘤免疫应答的核心启动者与调控者,已成为肿瘤免疫治疗的重要靶点。DC疫苗等靶向DC的免疫治疗策略在抗肿瘤治疗中展现出独特优势,但仍存在抗原提呈效率不足、免疫抑制微环境抵抗、功能特异性调控困难等瓶颈问题。肿瘤微环境(TME)中的DC存在高度异质性,不同DC亚群在分化发育、免疫调控及效应转归方面呈现复杂的多样性。解析DC亚群的精确表型与功能机制对于开发新型DC靶向性免疫治疗策略具有重要意义。TME中募集的经典DC、浆细胞样DC、单核细胞来源DC能与肿瘤浸润免疫细胞和微环境中的非免疫细胞(包括肿瘤细胞、成纤维细胞、内皮细胞等)相互作用激活抗肿瘤免疫应答,而TME还会通过转录调控、表观遗传调控、代谢重塑等多种方式抑制DC招募和抗原提呈能力,甚至诱导其向耐受性DC转化。值得注意的是,新近发现的富含免疫调节分子的成熟DC具有双向免疫调控作用,其起源路径和免疫调控网络仍有待深入研究。随着单细胞分析与空间组学分析等技术的发展,有望在单细胞分辨率下系统解析DC亚群的功能多样性及其与TME的时空相互作用网络,为开发下一代精准免疫治疗策略提供新靶点和新思路。

Experimental teaching of Medical Immunology is an important way to achieve the teaching goal of cultivating"prac-tical,applied,and innovative high-quality medical talents".Applying the ATR teaching mode to Medical Immunology experimental teaching,fully utilizing the advantages of artificial intelligence teaching assistants,can not only stimulate students'learning enthusi-asm,achieve simultaneous progress of"knowledge learning"and"thinking exercise",promote deep learning,and improve teaching quality,but also enhance students'teamwork ability,achieve a dual approach of"value thinking"and"practical exercise",integrate ideological and political education into the curriculum,and achieve moral education.
3.Progress of cytotoxic T lymphocyte-associated antigen-4 inhibitors in treatment of melanoma

Melanoma is characterized with high heterogeneity and invasiveness. The advent of cytotoxic T lymphocyte-associated antigen-4 (CTLA-4) inhibitors have shown the obvious therapeutic efficacy in the treatment of melanoma patients, which promotes the progression of melanoma immunotherapy. However, therapy resistance caused by immune escape of melanoma and immune-related adverse events caused by treatment have limited the therapeutic effects of CTLA-4 inhibitors, which have become a clinical challenge. This article reviews the current role of CTLA-4 inhibitors in melanoma therapy, aiming to provide references for clinical immunotherapy strategies for melanoma.
4.Design of integrated device for cleaning,sterilizing and drying glass graduated pipette

Objective To design an integrated device for cleaning,sterilizing and drying the glass graduated pipette with considerations on its characteristics.Methods The integrated device had its shell made of 304 stainless steel and hardware part composed of three modules for control,cleaning and drying.The control module consisted of a human machine interface(HMI)touch screen,a programmable logic controller(PLC),solenoid valves and etc;the cleaning module was made up of an ultrasonic release disk,a heat preservation cylinder,a bracket and etc;the drying module comprised a blower,a heating plate,a heat preservation cylinder and connected pipes.The HMI was developed with the programming language of EasyBuilder,and the PLC control logic was written with SCL high-level programming language.Results The device developed could be used for cleaning,sterilizing and drying the glass graduated pipette,and the pipette could be stored in the device after drying.Conclusion The integrated device developed eliminates manual operation and enhances the efficiency of the cleaning,sterilization and drying of the glass graduated pipette.[Chinese Medical Equipment Journal,2025,46(4):108-112]

8.The Influence of Family Adversities on Longitudinal Changes in Physical Inactivity Among Korean Adolescents During the COVID-19 Pandemic

Objectives:
Lack of physical activity has a critical effect on the physical and mental health of adolescents. This study examined the influence of family adversities on the longitudinal changes in physical inactivity among adolescents during the coronavirus disease 2019 (COVID-19) pandemic.
Methods:
The study used multi-wave data from the Korean Children and Youth Panel Survey, including 2590 Korean adolescents aged 12-14 years. The longitudinal trajectory of physical inactivity among adolescents and the effects of related factors were estimated using a latent growth modeling method.
Results:
Our results revealed a significant increase in physical inactivity among adolescents over time. At the onset of the pandemic, approximately one-seventh of Korean middle schoolers reported a lack of physical activity. However, 3 years later, during the quarantine, nearly one-fifth of these adolescents reported a significant increase in their physical inactivity. Initially, low level parental education was predictive of adolescents’ physical inactivity, but this effect diminished over time, becoming statistically insignificant by the end of the 3-year period. Moreover, the increase in physical inactivity over the 3 years was significantly influenced by parental rejection.
Conclusions
These findings suggest that adolescents who experience parental rejection are more likely to report an increase in sedentary behaviors in contexts such as the COVID-19 pandemic.

Lithocarpus litseifolius is rich in the chalcones phloridzin and trilobatin, the biosynthesis pathways of which have not been fully demonstrated. Chalcone synthase(CHS) is the first key rate-limiting enzyme in the biosynthesis of flavonoids in plants. To explore the functions of CHS gene family in chalcone synthesis of L. litseifolius, this study screened out two CHS genes(LlCHS1 and LlCHS2) from the transcriptome data of this plant, and then bioinformatics analysis and functional characterization were performed for the two genes. The bioinformatics analysis showed that LlCHS1 and LlCHS2 were acidic hydrophilic stable proteins with no transmembrane domain, composed of 395 and 390 amino acid residues, respectively. Both of them contained the characteristic amino acid sequence "WGVLFGFGPGL" and highly conserved active sites(Cys-164, Phe-215, His-303, and Asn-336) of the CHS family. The phylogenetic tree showed that LlCHS1 shared the same clade with similar genes in Aquilaria sinensis, and LlCHS2 was closely related to similar genes in Malus domestica. Under exogenous addition of phloretic acid, co-expression of LlCHS1 or LlCHS2 with Aa4CL from Aromatoleum aromaticum in Escherichia coli catalyzed the production of phloretin from phloretic acid. This study laid a theoretical foundation for revealing the functions of CHS in plants and provided new enzymatic modules for producing phloretin by synthetic biology.

10.Artificial Light at Night and Type 2 Diabetes Mellitus

The widespread and pervasive use of artificial light at night (ALAN) in our modern 24-hour society has emerged as a substantial disruptor of natural circadian rhythms, potentially leading to a rise in unhealthy lifestyle-related behaviors (e.g., poor sleep; shift work). This phenomenon has been associated with an increased risk of type 2 diabetes mellitus (T2DM), which is a pressing global public health concern. However, to date, reviews summarizing associations between ALAN and T2DM have primarily focused on the limited characteristics of exposure (e.g., intensity) to ALAN. This literature review extends beyond prior reviews by consolidating recent studies from 2000 to 2024 regarding associations between both indoor and outdoor ALAN exposure and the incidence or prevalence of T2DM. We also described potential biological mechanisms through which ALAN modulates glucose metabolism. Furthermore, we outlined knowledge gaps and investigated how various ALAN characteristics beyond only light intensity (including light type, timing, duration, wavelength, and individual sensitivity) influence T2DM risk. Recognizing the detrimental impact of ALAN on sleep health and the behavioral correlates of physical activity and dietary patterns, we additionally summarized studies investigating the potential mediating role of each component in the relationship between ALAN and glucose metabolism. Lastly, we proposed implications of chronotherapies and chrononutrition for diabetes management in the context of ALAN exposure.
