1.Study on the Distribution of Traditional Chinese Medicine Syndrome Types in Patients with Lumbar Disc Herniation and Its Correlation with Serum HIF-1α,NLRP3 and IL-1β Levels

Objective To investigate the distribution of traditional Chinese medicine(TCM)syndrome types in patients with lumbar disc herniation(LDH)and its correlation with serum levels of hypoxia-inducible factor 1α(HIF-1α),NOD-like receptor thermal protein domain associated protein 3(NLRP3)and interleukin 1β(IL-1β)of the patients.Methods Data collection was conducted in 160 patients(LDH group)admitted to the LDH outpatient and inpatient department of Wuhan Hankou Hospital from October 2022 to June 2024 and in 20 healthy volunteers(healthy control group)who underwent physical examination in the hospital during the same period.The levels of serum HIF-1α,NLRP3 and IL-1β were compared between the two groups,and then the correlation of serum HIF-1α,NLRP3 and IL-1β levels with Visual Analogue Scale(VAS)score of pain,Japanese Orthopedic Association(JOA)score and Oswestry Dysfunction Index(ODI)score in LDH patients was explored.The distribution of TCM syndrome types in LDH patients was statistically analyzed,and then the correlation between TCM syndromes and serum HIF-1α,NLRP3 and IL-1β levels in LDH patients was investigated.Furthermore,the correlation between serum HIF-1α,NLRP3,IL-1β levels and prognosis of LDH was analyzed.Results(1)Compared with the healthy control group,the serum levels of HIF-1α,NLRP3 and IL-1β in the LDH group were significantly increased,and the differences were statistically significant(P<0.01).(2)Spearman correlation analysis showed that VAS and ODI scores were positively correlated with serum HIF-1α,NLRP3 and IL-1β levels,and JOA score was negatively correlated with serum HIF-1α,NLRP3 and IL-1β levels,the differences being statistically significant(P<0.01).(3)The percentage of TCM syndrome types in 160 patients with LDH in descending order was as follows:blood stasis syndrome(65 cases,40.63%)>kidney deficiency syndrome(45 cases,28.13%)>cold-damp syndrome(30 cases,18.75%)>damp-heat syndrome(20 cases,12.50%).(4)There were statistically significant differences in serum levels of HIF-1α,NLRP3 and IL-1β among LDH patients with different TCM syndromes(P<0.01):the serum levels in patients with cold-damp syndrome were higher than those in patients with kidney deficiency syndrome(P<0.05),serum levels in patients with damp-heat syndrome were higher than those in patients with cold-damp syndrome(P<0.05),and serum levels in patients with blood stasis syndrome were higher than those in patients with damp-heat syndrome(P<0.05).(5)Multivariate logistic regression analysis showed that serum HIF-1α,NLRP3 and IL-1βwere risk factors for the prognosis of LDH patients,and they were negatively correlated with the prognosis of LDH(P<0.05 or P<0.01).Conclusion Blood stasis syndrome is the predominated syndrome type in LDH patients,and then comes kidney deficiency syndrome.The levels of serum HIF-1α,NLRP3 and IL-1β vary in LDH patients with different TCM syndrome types,and are expected to be the potential biological indicators for TCM syndrome differentiation of LDH.Serum HIF-1α,NLRP3 and IL-1β are the risk factors for the prognosis of LDH.
2.Construction of expression vector containing rat rCB1 gene and its influence on the apoptosis in human cervical cancer CaSki cell line

Objective To construct rCB1 gene eukaryotic expression vector, detect its expression in the cell, and explore its influence on apoptosis in human cervical cancer CaSki cells.Methods The total RNA was extracted from rat brains.The rCB1gene was amplified by RT-PCR.The pcDNA3.1(+)-rCB1 was constructed by enzyme digestion, purifi-cation, bind the PCR purification products and pcDNA3.1 (+) DNA.The pcDNA3.1 (+)-rCB1 plasmid was transfect-ed into HEK293 and CaSki cells by liposomes.The expression and localization of rCB1 were detected by Western blot and immunofluorescence combined with confocal laser scanning microscopy.The apoptosis rate of CaSki cells was detected by flow cytometry.The expression of rCB1, Bcl-2, Bax and Bad was detected by Western blot and real-time fluorescence quantitative RT-PCR (qRT-PCR).Results The 5300 bp pcDNA3.1(+) and 1500 bp rCB1 were obtained after diges-ting the pcDNA3.1 ( +)-rCB1.The result of sequencing was in agreement with the sequence of rCB1 gene ( NM_012784.4 ) .The rCB1 expressed in the membrane and cytoplasm when pcDNA3.1 (+)-rCB1 plasmid was transfected into HEK293 cells.The apoptosis rate of rCB1 group was increased compared with the blank group when pcDNA3.1 (+)-rCB1 plasmid was transfected into CaSki cells (P<0.05).Compared with the blank group, rCB1 gene upregulated the expres-sion of Bax and Bad, and suppressed the expression of Bcl-2.The statistical difference was significant ( P <0.05). Conclusions The pCDNA3.1(+)-rCB1 eukaryotic expression vector is constructed successfully.It is found that rCB1 is expressed in membrane and cytoplasm of HEK293 cells.rCB1 can significantly promote the apoptosis in cervical cancer CaSki cells by up-regulating the expression of Bax and Bad, and down-regulating the expression of Bcl-2 as well.
3.Application of reverse dot blotting for detection of Tyzzer’ s organism

Objective To establish a simple, stable, specific and sensitive method for detection of Tyzzer ’ s or-ganism by reverse dot blotting ( RDB) .Methods Primers and specific probes were designed according to the conservative sequence of Tyzzer 16S rDNA.The forward primer was labeled with biotin .The reverse dot blotting method was established followed by PCR amplification .The specificity and sensitivity of this method were determined .Next, 41 mice and 38 rats were examined by RDB , ELISA and IFA .Results The RDB method showed a high specificity , and in the testing of the 79 laboratory animals , its limit of detection was 4.5 ng/μL.Compared the results of ELISA and IFA , its consistence with ELISA was 100%and the positive rate was 7.59%(6/79), the consistence with IFA was 92.4%(73/79), and the posi-tive rate was 0%.Conclusions An accurate, sensitive and specific method in combination with PCR and RDB in detection of Tyzzer’ s organism is established in this study .
4.Clinical Observation on Simple Obesity Treated by Acupuncture plus Ear Point-Sticking Method

Objective: In order to observe the clinical therapeutic effect of acupuncture plus ear point-sticking for simple obesity. Methods: Fifty-two cases of the patients with simple obesity were treated by acupuncture and ear point-sticking method for one course. Results:After the treatments, the results showed remarkable effect in 22 cases, effect in 25 cases and failure in 5 cases, and the total effective rate was 90.4%. There was no obvious change in various indexes of the body in the male and female patients before and after the treatment.Conclusion: Acupuncture plus ear point-sticking for simple obesity is stable in the therapeutic effect, simple and safe in application, without side effects, and cheap in cost and needs to be greatly popularized.
5.Influence of Electroacupuncture on Serum Leptin in Patients with Simple Obesity

To investigate the influence of electroacupuncture on leptin, the serum leptin level was observed by radioimmunoassay before and after electroacupuncture treatment in 30 patients with simple obesity. After stimulation of electroacupuncture, serum leptin level dropped in the patients with simple obesity.
