Objective To study the key quality markers of Shugan Liqi Mixture in the treatment of liver depression and qi stagnation and menstrual irregularities;To provide a scientific basis for the selection of index components in the process of its quality standardization study.Methods The characteristic chromatogram of Shugan Liqi Mixture was established by HPLC;the serum components of Shugan Liqi Mixture were analyzed by LC-MS;the results of the characteristic chromatogram and serum chemistries were integrated,and the network pharmacology was used to construct a component-target network of Shugan Liqi Mixture,so that important components related to liver depression and qi stagnation and menstrual dysfunction could be obtained.The key quality markers of Shugan Liqi Mixture were screened out by combining with relevant literature reports.Results A total of 12 common peaks were obtained from the characteristic chromatogram of 15 batches of Shugan Liqi Mixture,and 7 components were identified,including paeoniflorin,hyperoside,naringin,hesperidin,neohesperidin,baicalin and saikosaponin b2.Totally 26 prototype blood components,including isorhamnetin,nobiletin and tetrahydropalmatine,were identified by serum pharmacochemistry study.According to the"five principles"of quality markers,and combining the characteristic chromatogram and serum medicinal chemistry results,the five components of baicalin,naringin,tetrahydropalmatine,paeoniflorin and saikosaponin a were finally identified as the key quality markers of Shugan Liqi Mixture combined with the data from network pharmacology and the literature.Conclusion This study preliminarily identifies five key quality markers of Shugan Liqi Mixture,providing index components in the process of its quality standard research.

Objective To screen the anti-Helicobacter pylori gastritis active components of the ethyl acetate extract of Alpinia officinarum Hance.Methods The"knock-out"strategy combined with high-performance liquid chromatography(HPLC)detection was developed to separate the components of the ethyl acetate extract of A.officinarum while obtaining the negative samples without the components.A human gastric epithelial cell(GES-1)model of H.pylori gastritis was established,and the levels of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),interleukin-8(IL-8)and interleukin-1β(IL-1β)in the supernatant of the cells were determined by enzyme-linked immunosorbent assay(ELISA).Results The total flavonoid fraction,the negative fraction without total diphenylheptanoids,the negative fraction without 5-hydroxy-7-(4-hydroxy-3-methoxyphenyl)-1-phenyl-3-heptanone(DHPA),and galangin significantly reduced IL-6 levels in the supernatant of H.pylori infected GES-1 cells at a concentration of 8 μg·mL-1 with 24 h incubation.The total flavonoid fraction strongly inhibited the release of IL-6,TNF-α,IL-8,and IL-1β from H.pylori gastritis GES-1 cells at a concentration of 16 μg·mL-1.Conclusions The total flavonoid fraction is the major anti-H.pylori gastritis active component of the ethyl acetate extract of A.officinarum.The results lay the foundation for further elucidation of the material basis of A.officnarum against H.pylori gastritis.

Objective:To prospectively investigate the prevalence of heparin-induced thrombocytopenia (HIT) in critically ill children during extracorporeal membrane oxygenation(ECMO) and explore the clinical characteristics and prognosis of HIT during ECMO.Methods:A total of 22 critically ill children, who had received ECMO support for more than 96 hours in the Intensive Care Unit at the Children′s Hospital of Fudan University from March 2019 to December 2020, were enrolled.According to the 4T score and the heparin/platelet factor 4(PF4) antibody, children whose 4T scores were not less than 6 and heparin/ PF4 antibodies were positive, were classified into HIT group ( n=6), and non-HIT group ( n=16). The clinical outcomes and the incidence of thrombotic events were compared between two groups. Results:The incidence of HIT during ECMO in critically ill children was 27% in this study.The incidence of thrombosis in the ECMO circuit in the HIT group was higher (100% vs.63%, P=0.133), and the average time to the first thrombosis in ECMO circuit in the HIT group was shorter than that in the non-HIT group (3.70 d vs.5.44 d, P=0.06). During the first 14 days of ECMO, the proportion of children with thrombotic events no less than twice was higher in the HIT group (67% vs.19%, P=0.054). There was no significant difference regarding the survival rate at 28 days after ECMO withdrawal between two groups (33% vs.50%, P=0.664). Conclusion:The prevalence of HIT during ECMO in critically ill children is high.Thrombosis events tend to occur earlier and more extensively in children with HIT during ECMO.No significant effect of HIT on the survival rate of children during ECMO is found.Whether HIT has effect on the survival rate of children with ECMO requires a prospective and large clinical study.

Objective:To investigate the correlation and consistency between the parameters of the thromboelastography(TEG) and of routine coagulation tests and platelet count(PLT) in critically ill children in the pediatric intensive care unit.Methods:We conducted a retrospective analysis on the results of the simultaneous TEGs, coagulation tests and PLT of 231 children in pediatric intensive care unit at Children′s Hospital Affiliated to Fudan University from March 2019 to July 2019, including the correlation analysis, the chi-square test and Kappa consistency analysis.Results:The reaction time(R) was linearly dependent on activated partial thromboplastin time(APTT), prothrombin time(PT) and thrombin time(TT), respectively.The maximal amplitude(MA), clot time formation(K) and alpha-angle(α) were linearly dependent on fibrinogen.The MA, K and α were linearly dependent on PLT, too.And the correlation between MA and PLT was the best( R=0.656). There were significant differences( P<0.001) and poor consistency(Kappa value: 0.112, 0.047, 0.124) between R and APTT, PT, TT.There was no significant difference( P>0.05) and there was moderate consistency(Kappa value: 0528, 0.518, 0.408) between Ma, K, α and PLT.There were significant differences( P<0.001) and the consistency was poor or fair(Kappa value: 0.324, 0.188, 0.170) between Ma, K, α and fibrinogen. Conclusion:TEG is significantly correlated with PLT in evaluating platelet function and the consistency is good.The TEG parameters are linearly correlated with the relevant results of routine coagulation tests in evaluating the coagulation factor and fibrinogen function, but consistency is weak.Therefore, it is concluded that these two methods are irreplaceable.

Objective:To develop an HPLC method for determining matrine , oxymatrine and oxysophocarpine in wine-processed Sophora flavescens.Methods:An HPLC method was used with a Dalian Elite HYP-ODS column (250 mm ×4.6 mm, 5 μm).The mobile phase consisted of methanol-phosphate buffer (12:88) with the flow rate of 1.0 ml· min-1 .The detection wavelength was 205 nm, the column temperature was 25℃and the injection volume was 20 μl.Results:The calibration curve was linear within the range of 0.105-0.420 μg for matrine,0.760-3.040μg for oxysophocarpine and 1.615-6.460μg for oxymatrine.The average recovery of ma-trine, oxysophocarpine and oxymatrine was 99.70％(RSD=1.92％), 99.41％( RSD=1.93％) and 98.60％(RSD=1.79％)(n=6), respectively.Conclusion:The method is accurate and reliable with good reproducibility , promising specificity and high sensitivi-ty, which can be used for the quality control of wine-processed Sophora flavescens.

Objective:To provide scentific evidence for determining the harvest period of Cortex Abizziae by studying the dynamic variation of the content of lignan glycoside I in Cortex Abizziae.Methods:Chromatography was performed on a Boston Green ODS C18 (250 mm×4.6 mm,5 μm) column,the mobile phase was acetonitrile-0.04% phosphate (18∶82),the flow rate was 1.0 ml·min-1,the detection wavelength was 204 nm,and the column temperature was at 25℃.Results:There were differences in the contents of lignan glycoside I in Cortex Abizziae at different harvest periods.The content of lignan glycoside I reached the highest level in January,decreased quickly from January to March,and gradually increased from April to December.Conclusion:The content of lignan glycoside I in Cortex Abizziae at different harvest periods is different,and the optimum harvest time of Cortex Abizziae is determined from November to January of the following year.

Farnesoid X receptor(FXR), a member of the nuclear receptor superfamily, plays an important role on the regulating bile acid, lipid and glucose homeostasis. FXR antagonist can stimulate the conversion of total cholesterol into bile acid by improving the activity of CYP7A1. It can also reduce the content of triglyceride, low-density lipoprotein and increase that of high-density lipoprotein by regulating the relevant target genes. Considering the above facts, FXR antagonist is expected to become a potential drug to treat hyperlipidemia. In recent years, great progress has been made in the research on FXR antagonists with their various structural types having been obtained. This article introduces natural and synthetic FXR antagonists reported in recent years.

Objective To establish the quality standard of Zhimiling Suppository (Cortex Phellodendri, Radix Sophorae Flavescentis, Catechu, Borneolum Syntheticum, Alumen). Methods Catechu, Borneolum Syntheticum, Alumen in the preparation were identified by physics and chemstry method. Berberine Hydrochloride was determined by HPLC. Results The standard curve for Berberine Hydrochloride was linear in the range of 0.080 2～0.802 ?g, the average recovery was 100.4%, RSD was 0.85% (n=6). Conclusion The methods are simple, accurate and reproducible.

Objective To establish the method of the GC fingerprints for Ligusticum chuanxiong essential oil and its quality control.Methods The temperature at both feed inlet and detector was 250℃,the carrier gas was nitrogen,and the flow rate was 2 mL/min.GC of the essential oil in ten batches of L.chuanxiong was analyzed by adopting temprature programmer and the fingerprint peaks were identified by GC-MS.Results To set up the GC fingerprints for L.chuanxiong essential oil,13 peaks were marked out altogether and the GC fingerprints were analyzed by similarity analysis.Conclusion The method is of high degree of presion,reproducibility,and stability.The resolving of each component in the essential oil is good.The established fingerprints could be used as one of the quality control index for L.chuanxiong essential oil.
