1.Clinical significance of galectin-3 and HPV16/18-E6 expression in laryngeal carcinoma patients

Objective:To observe the expression and clinical significance of galectin-3 and HPV16/18-E6 proteins in laryngeal carcinoma patients.Methods:Immunohistochemistry and Western blot method were used to observe the expressions of galectin-3 protein and HPV16/18-E6 protein in laryngeal carcinoma tissues of 60 patients. SAS 9.4 software was used to analyze the relationship between the galectin-3 protein and the clinical stage severity, pathological grades and lymph node metastasis in all the cases. Results:The result of immunohistochemical examinations showed that the positive rate of galectin-3 protein expression was 78.3% (47/60), and the Western blot result showed that the galectin-3 protein expression in tumor tissues was significantly higher than that in adjacent normal tissues. Statistical analysis showed that galectin-3 expression was correlated with the severity of clinical staging, differentiation of the tumor and lymph node metastasis of laryngeal cancer. There was no correlation between the expression of galectin-3 protein and the 16/18-E6 protein of human papillomavirus.Conclusions:The expression of galectin-3 protein showed a gradually increasing trend with the aggravation of the clinical severity of laryngeal carcinoma, providing certain experimental data for the early diagnosis and treatment of laryngeal cancer.
2. Expression of galectin-1 in Chinese laryngeal squamous cell carcinoma patients and its clinical significance

Objective:
To observe the expression and clinical significance of galectin-1 in Chinese laryngeal squamous cell carcinoma patients.
Methods:
Immunohistochemical technique was used to test the galecin-1 protein expression in the 50 laryngeal squamous cell carcinoma patients and Western blot method was used to test the galecin-1 protein expression in the 3 of the patients. SAS 9.4 software was used to analyze the relationship between the galectin-1 protein and the clinical degrees, pathological grades and lymph node metastasis in all the cases.
Results:
In the 50 laryngeal squamous cell carcinoma patients, the positive rate of galectin-1 protein was 74% (37/50). The expression of galectin-1 protein was correlated with the severity of clinical staging, pathological differentiation and lymph node metastasis.
Conclusions
High expression of galectin protein related with the occurrence and development of head and neck squamous cell carcinomas.
3.Clinical observation on plum-blossom needle combined with rehabilitation training for hand spasm after stroke.

OBJECTIVETo observe the effect difference between plum-blossom needle combined with rehabilitation training and conventional rehabilitation training for hand spasm after stroke.
METHODSA total of 61 patients were randomly divided into a comprehensive treatment group (30 cases) and a rehabilitation training group (31 cases). In the rehabilitation training group, Bobath occupational therapy, OT training, and hand function training were adopted, once every day; on the basis of treatment in the rehabilitation group, plum-blossom needle was applied at the lung meridian of hand-, heart meridian of hand-, pericardium meridian of hand- in the comprehensive treatment group. The treatment was given once every two days, three weeks as one course in the two gnoups. After 3 courses of treatment, clinical efficacy evaluation was performed, and the modified Ashworth scale and Fugl-Meyer (FMA) motor function scores were assessed before and after treatment.
RESULTSAfter treatment, the grade for Ashworth scale and FMA scores in the comprehensive treatment group and the rehabilitation trainning group were better than those before treatment (all <0.05), and the improvements in hand spasm and hand fuction in the comprehensive treatment group were superior apparently to those in the rehabilitation trainning group (both <0.05) The total effective rate of hand function was 93.3% (28/30) in the comprehensive treatment group, which was better than 74.2% (23/31) in the rehabilitation training group (<0.05).
CONCLUSIONPlum-blossom needle combined with rehabilitation training are more effective than simple rehabilitation training for hand spasm after stroke.
4. Analysis of human papillomavirus infection and p53 expression in patients with hypopharynx squamous cell carcinoma

Objective:
To analyze the infection of human papillomavirus and expression of p53 in patients with hypopharynx squamous cell carcinoma.
Methods:
The pathological samples of 15 clinically diagnosed hypopharynx cancer patients were collected. Lunimex and PCR techniques were used to detect the human papillomavirus (HPV) gene and immunohistochemical method was used to analyze the HPV protein expression in the samples. PCR and direct sequencing were used to screen the mutations in p53 gene and immunohistochemistry was used to evaluate the expressing profiles of p53 proteins.
Results:
In the 15 cases, 6 were positive for HPV16/18 gene and 2 cases were positive for HPV16/18 E6 protein expression. The total positive rate was about 46.7% (7/15). In addition, 9 of 15 (60.0%) cases had sequencing-confirmed p53 mutations and p53 immunohistochemistry (IHC) confirmed 9 (60.0%) cases had positive staining.
Conclusions
The high HPV infection rate and p53 expression in hypopharynx squamous cell carcinoma patients in the study lays a foundation for further elucidating the pathogenesis of this kind of cancer.
5.Influence of intestinal early enteral nutrition therapy on intestinal barrier function and immune response of patients with radiation enteritis

Objective To investigate the influence of early enteral nutrition therapy on the intestinal barrier function and immune response of the patients with radiation enteritis (ER) so as to find a relatively simple and effective method to treat RE.Methods Fifty-six patients with radiation enteritis (RE) diagnosed by colonoscopy,X-rays,and pathology were randomly divided into 2 equal groups:experimental group undergoing enteral inutrition therapy,and control group undergoing conventional therapy only.Peripheral blood samples were collected 1,11,and 21 days after admission.Plasma diamine oxidase (DAO),D-lactic acid,endotoxin,and lactulose/mannitol (L/M) ratio,and levels of IgG,IgM,and IgA,and CD4/CD8 ratio were examined.Five cases from the experimental group and 5 cases from the control group underwent second-time operation because of incomplete intestinal obstruction,intestinal stenosis,or recurrent tumor respectively.The biopsy specimens of the terminal ileum or distal descending colon taken during the first and second operations underwent pathological examination.Peripheral blood samples were collected 1,11,and 21 days after admission.Plasma diamine oxidase (DAO),D-lactic acid,endotoxin,and lactulose/mannitol (L/M) ratio,and levels of IgG,IgM,and IgA,and CD4/CD8 ratio were examined.Results There were no significant differences in the intestinal function and blood immunological indices between these 2 groups.The levels of DAO,D-lactic acid,and endotoxin,and the L/M ratio 11 days after admission of the experiment group were all significantly lower than those of the control group (t =2.568,2.427,2.143,2.443,P < 0.05),and all those indices 21 days after admission of the experiment group were all much more significantly lower in comparison with the control group (t =6.019,12.834,7.837,7.997,P < 0.01).The levels of IgG,IgM,and IgA,and CD4/CD8 ratio 11 days after admission of the experimental group were all significantly higher than those of the control group (t =2.096,2.211,2.182,P< 0.05,and t=2.301,P< 0.05),and the differences became much more significant 21 days after admission (t =2.703,2.679,3.138,P < 0.01,and t =5.107,P < 0.01).The height of the intestinal villa and the thickness of the mucosa of the specimens taken at the second-time operation were both much greater than those at the first-time operation.Conclusion Early enteral support therapy helps effectively maintain the intestinal barrier and immune response function of the RE patients.
6.Protective effects of Dahuangfuzi decoction on the intestine barrier functional of acute necrotizing pancreatitis in rats

Objective To observe the effects of Dahuangfuzi decoction on the intestine barrier functional of acute necrotizing pancreatitis in rats. Methods The 60 rats were randomly divided into sham operation group ( n = 19 ), ANP group ( n = 21 ), and Dahuangfuzi treatment group ( n = 20). The rats of ANP group were induced by injecting 1 ml/kg of 4% sodium taurocholate into the pancreatiobiliary duct, and jejunal fistula was esablished. The rats of treatment group received Dahuangfuzi decoction (2 ml, repeated at 4 and 8 h)through jejunum distal stoma tube 0. 5 h after ANP induction. The other 2 groups received same amount of normal saline. Blood sample was collected through abdominal aorta, 24 h after ANP induction, and the serum amylase, endotoxin, D-lactate, plasma diamine oxidase (DAO) were detected. Pancreas, small intestine tissue was harvested for pathologic examination, index of intestinal epithelial damage was measured and ultrastructural changes in small intestinal mucosa was observed. Results The expression of serum amylase, endotoxin,D-lactate, DAO in sham operation group was ( 152 ± 32 ) U/L, (6.95 ± 2.10) pg/L, ( 3.96 ± 1.08 ) μg/mland ( 14.26 ± 2.67 ) μg/ml, while the corresponding values were ( 1549 ± 93 ) U/L, (40.48 ± 3.41 ) pg/L,( 12.34 ± 1.23 ) μg/ml and ( 80.28 ± 3.54) μg/ml in ANP group, and they were (655 ± 49 ) U/L, ( 19.55 ±2.50) pg/L, (6.75 ± 1.36 ) μg/mland ( 20.69 ± 7.53 ) μg/ml in treatment group. The values in ANP group were significantly higher than those in sham operation group. The values in treatment group were significantly lower than those in ANP group, but significantly higher than those in sham operation group ( P < 0.05 or P <0. 01 ). The thickness and height of intestinal mucosa in ANP group were ( 389.44 ± 29.87 )μm and ( 16.52 ±3.73) μm, which were significantly lower than those in treatment group [(501.95 ± 45.38 )μm, (27.82 ±5.17)] μm, and in sham operation group [( 658.72 ± 57.49 ) μm, ( 35.49 ± 6.43 )μm, Index of intestional epitholial donage in ANP group was 3.72 ± 0.65 which is significently higher than those in theatment (2.12 ±0.37 ) and in sham operation group (0.85 ± 0.24). The intestinal mucosa histological and ultrastructural changes in Dahuangfuzi treatment group were better than those in ANP group. Conclusions Dahuangfuzidecoction can significantly decrease the damage of intestine barrier function in ANP rats.
