1.Efficacy evaluation of repeated transcranial magnetic stimulation combined with bipolar depression triple therapy in reducing female adolescents' non-suicidal self-injury.

In order to explore effective ways to reduce non-suicidal self-injury (NSSI) among female adolescents, a total of 45 female adolescent patients with NSSI in West China Hospital of Sichuan University and Guizhou Second Provincial People's Hospital from June 2021 to June 2024 were selected randomly that divided into groups A, B and C, with 15 cases in each group. Group A was treated with repeated transcranial magnetic stimulation (rTMS) and bipolar depression triple therapy, and group B was treated with bipolar depression triple therapy to compare the effectiveness and safety. Group C received bipolar depression triple therapy combined with sham stimulation which only produced stimulating sounds but no stimulating magnetic field as a control in the study. After treatment, the Hamilton Anxiety Score (HAMA), Hamilton Depression Score (HAMD) and Nurses' Global Assessment of Suicide Risk (NGASR) in group A were significantly lower than those in group B and C ( P < 0.01). rTMS combined with bipolar depression triple therapy has a definite effect on reducing NSSI in female adolescents, which can reduce the incidence rate of short-term NSSI behavior in patients.

2.Analysis of the Results of Thalassemia Gene Screening in 9 334 Cases in Guiyang Region.

OBJECTIVE:
To investigate the common genotypes and distribution characteristics of thalassemia in Guiyang region, and preliminarily analyze the rare mutations of globin genes in this area.
METHODS:
A total of 9 334 individuals who came to our hospital for thalassemia screening from June 2016 to February 2023 were included in this study. They were examined for common thalassemia mutations using PCR-based flow-through hybridization technology. Meanwhile, rare and unknown mutations were detected by Sanger sequencing.
RESULTS:
Among the 9 334 cases, 895 positive cases of common thalassemia were detected, with a positive rate of 9.59%. Among the positive samples, 565 cases (63.13%) were confirmed to be α thalassemia, of which the most common genotypes were αα/-α3.7 (46.37%), followed by αα/--SEA(26.55%) and αα/-α4.2(10.62%); 310 cases (34.64%) were diagnosed as β thalassemia, with βCD17/βN (39.35%) being the most frequent genotype, followed by βCD41-42 /βN (31.29%) and β IVS-II-654/ βN (12.90%). There were 20 cases (2.23%) of αβ complex thalassemia, mainly being αα/-α3.7 combined with βCD17 /βN . Additionally, 8 cases of rare globin gene mutations were found by Sanger sequencing, including 7 mutation types. Among them, HBB: c. -137C> T (-87 C>T) was reported for the first time in Guizhou; HBA1 : c.*29C>T and HBB : c. 93-50C>T (IVS I-81C>T) were new mutations that had not been recorded in either the HbVar or IthaGenes database.
CONCLUSION
Guiyang region has a high incidence of thalassemia mutations, and these mutations are diverse and complex. Analyzing gene mutation types of thalassemia in this area can contribute to the prevention of the birth of children with severe thalassemia.

3.Pseudomonas monteilii ZMU-T06 produces 2-substituted quinolines by oxidative dehydroaromatization.

2-substituted quinolines are the building blocks for the synthesis of natural products and pharmaceuticals. In comparison with classical methods, dehydroaromatization of 2-substituted-1,2,3,4-tetrahydroquinolines has emerged in recent years as an efficient and straightforward method to synthesize quinolines due to its high atom economy and sustainability. However, existing chemical methods need transition metal catalysts and harsh reaction conditions. Biocatalysis with high efficiency, high selectivity, and mild reaction conditions has become an important method of organic synthesis. We mined a strain Pseudomonas monteilii ZMU-T06 capable of producing monoamine oxidase for the dehydroaromatization of 2-substituted-1,2,3,4-tetrahydroquinolines to synthesize 2-substituted quinolines (8 substrates, yields of 45.7%-48.4%) and then hypothesized the catalytic mechanism, providing a new method for green synthesis of 2-substituted quinolines.

4.Content determination and chemometric analysis of 8 components in Smilax glabra

OBJECTIVE To provide reference for Smilax glabra quality control. METHODS UPLC method was established to simultaneously determine the contents of 5-O-caffeoylshikimic acid, neoastilbin, astilbin, neoisoastilbin, isoastilbin, engelitin, resveratrol and isoengelitin in 20 batches of S. glabra from different areas (No. S1-S20). The quality evaluation of 20 batches of samples was performed by chemometrics; the differential biomarkers that affected the quality of S. glabra were screened. RESULTS The measured 8 components had good linear relationship within the range of measured concentration (r≥0.999 6). RSDs of precision, repeatability and stability tests (24 h) were all lower than 2.00% (n=6). The average recoveries varied between 97.60% and 106.40% (RSDs were all lower than 2.00%, n=6). Cluster analysis showed that the samples produced in Zhejiang (S1-S5) and Jiangxi (S6-S10) were clustered into one category; the samples produced in Hunan (S11-S15) were clustered into one category; the samples produced in Yunnan (S16-S20) were clustered into one category. Principal component analysis showed that the first two principal components could represent 85.60% information of 8 components in S. glabra. Partial least squares-discriminant analysis showed that variable importance projection values of 5-O-caffeoylshikimic acid, astilbin, isoastilbin and neoastilbin were all greater than 1. CONCLUSIONS There are differences in the contents of the above 8 components in S. glabra from different origins; 5-O-caffeoylshikimic acid, astilbin, isoastilbin and neoastilbin may be differential markers affecting the quality of S. glabra.
5.Meta-analysis of the efficacy and safety of PD-1/PD-L1 inhibitors combined with bevacizumab in the treatment of advanced non-small cell lung cancer

OBJECTIVE To evaluate the efficacy and safety of programmed death-1/programmed death-ligand 1 (PD-1/PD-L1) inhibitors combined with bevacizumab in the treatment of advanced non-small cell lung cancer (NSCLC) based on platinum- containing dual therapy. METHODS Retrieved from CNKI, Wanfang, VIP, Web of Science, PubMed and other Chinese and English databases, cohort studies or randomized controlled trial studies on the treatment of advanced NSCLC with platinum- containing double agents in combination with PD-1/PD-L1 inhibitors and bevacizumab (trial group) versus platinum-containing double agents with or without PD-1/PD-L1 inhibitor or bevacizumab (control group) were collected from the inception to April 25, 2024. After screening literature, extracting data and evaluating quality, meta-analysis and sensitivity analysis were performed by using RevMan 5.4.1 software. RESULTS A total of 15 pieces of literature were included, involving 13 clinical studies with a total of 3 282 patients. Compared with the control group, partial response rate [RR=0.75,95%CI(0.68,0.82),P<0.000 01], complete response rate [RR=0.47,95%CI(0.29,0.76),P=0.002], progressive disease rate [RR=1.23,95%CI(1.11,1.37),P<0.000 1], objective response rate (ORR) [RR=0.72,95%CI(0.67,0.79),P<0.000 01] and disease control rate (DCR) [RR=0.85, 95%CI (0.77,0.95),P=0.003] were higher in the trial group. There was no statistically significant difference in the stable disease rate [RR=1.25, 95%CI (0.86, 1.83), P=0.25] or overall adverse drug reaction incidence rate [RR=0.95, 95%CI (0.90, 1.00), P= 0.07] between the two groups of patients. Sensitivity analyses showed robust and reliable results for all outcome indicators. CONCLUSIONS PD-1/PD-L1 inhibitors combined with bevacizumab based on platinum-containing dual therapy in the treatment of advanced NSCLC can improve patients’ clinical benefits, such as ORR and DCR, without increasing the risk of adverse drug reaction.

7.Effects and mechanisms of wogonin on airway inflammation in rats with chronic obstructive pulmonary disease

OBJECTIVE To study the effects and potential mechanism of wogonin (Wog) on airway inflammation in rats with chronic obstructive pulmonary disease (COPD). METHODS Eighty-four rats were randomly divided into control group, model group, Wog low-dose and high-dose groups (intragastric administration of 50, 100 mg/kg), aminophylline group (positive control, intragastric administration of 2.3 mg/kg), recombinant rat receptor-interacting protein kinase 1 [rRIPK1, receptor-interacting protein kinase 1 (RIPK1) activator] group (tail vein injection of 8 µg/kg), and Wog high-dose+rRIPK1 group (intragastric administration of Wog 100 mg/kg+tail vein injection of rRIPK 8 µg/kg), with 12 rats in each group. Except for control group, COPD model of other groups was induced by smoking combined with tracheal injection of lipopolysaccharide. Twenty-four hours after successful modeling, the rats were administered once a day for 4 weeks. The changes of peak inspiratory flow (PIF), peak expiratory flow (PEF) and minute ventilation (MV),forced expiratory volume in one second(FEV1)/forced vital capacity(FVC) were measured after the last medication; the serum levels of interleukin 1β(IL-1β), IL-6 and tumor necrosis factor-α (TNF-α) were measured by ELISA; the pathological changes of lung tissue in rats were observed; the apoptotic rate of pulmonary epithelial cells was detected. mRNA expressions of RIPK1, RIPK3 and mixed lineage kinase domain-like protein (MLKL), and protein expressions of RIPK1, RIPK3 and p-MLKL were all detected in lung tissue of rats. RESULTS Compared with control group, PIF, PEF, MV and FEV1/FVC of model group were decreased significantly (P<0.05), while the levels of IL-1β, IL-6 and TNF- α were increased significantly (P<0.05); there was a large number of inflammatory cells infiltration in the lung tissue and bronchialwall thickening in model group; the apoptotic rate of pulmonary epithelial cells,mRNA expressions of RIPK1, RIPK3 and MLKL, protein expressions of RIPK1, RIPK3 and p-MLKL were increased significantly (P<0.05). Compared with model group, above indexes of rats were improved significantly in Wog low-dose and high-dose groups (P<0.05), and pathological injuries were alleviated significantly. The corresponding indexes of rats were worsened in rRIPK1 group (P<0.05), and pathological damage had further worsened. rRIPK1 significantly attenuated the inhibitory effect of high-dose Wog on airway inflammation and RIPK1/RIPK3/ MLKL pathway in COPD rats (P<0.05). CONCLUSIONS Wog may improve airway inflammation in COPD rats by inhibiting RIPK1/RIPK3/MLKL signal pathway.
8.Expression and significance of eIF2B1 in hepatocellular carcinoma

Objective:
To investigate the expression of eukaryotic translation initiation factor 2B1(eIF2B1) in HCC and its correlation with clinical prognosis.
Methods:
Through the follow⁃up of the clinical data of 743 hepatocellular carcinoma patients in the specimen bank ,91 HBV⁃related liver cancer patients with complete follow⁃up information were screened out , and their postoperative tumor tissues were selected. Meanwhile , the adjacent tumor tissues with a
distance of more than 2 cm from the tumor margin were collected to make tissue chips. Western blot and immunohistochemical experiments were performed. Image J was used to analyze the proportion of positive cells stained by tissue chip and the gray value of Western blot results. Clinical data and follow⁃up data of included patients were statistically analyzed by R4. 0. 5 software.
Results:
Immunohistochemical results suggested that eIF2B1 was more strongly expressed in HCC than in para⁃cancerous tissues. Western blot results showed that eIF2B1 was more strongly expressed in HepG2. 2. 15 cells than in HepG2 cells. The expression of eIF2B1 in HCC tissues was correlated with
tumor number (P < 0. 05) . Cox multivariate regression analysis showed that the expression of eIF2B1 was an independent risk factor for the overall clinical prognosis of HBV⁃DNA positive patients (HR = 4. 28 ,P = 0. 018) .
Conclusion
The expression of eIF2B1 is significantly enhanced in HBV⁃associated HCC tissues and is significantly
associated with overall survival in HBV DNA positive patients.
9.Experimental study of PD⁃1/PD⁃L1 signal pathway probe targeting osteosarcoma

Objective :
To explore a non⁃invasive assay for screening osteosarcoma patients with positive programed death receptor⁃1 ( PD⁃1) /programmed death receptor ligand⁃1 ( PD⁃L1) signaling pathway expression
Methods :
The subcutaneous tumor bearing mouse model of human Osteosarcoma cells ( OS⁃732) was established by the method of tumor formation , and toxicity test was performed to verify the toxicity of PD⁃L1 antibody to mouse organs . 124 I ⁃anti⁃PD⁃L1 monoclonal antibody molecular probe was further synthesized , and OS⁃732 rats were injected with 18 . 5 MBq 124 I ⁃anti⁃PD⁃L1 probe in tail vein to start OS⁃732 osteosarcoma Micro Positron E mission Tomography/ Computed Tomography (Micro⁃PET/CT) imaging.
Results :
OS⁃732 osteosarcoma model was successfully constructed , immunohistochemistry confirmed the presence of PD⁃L1 expression in OS⁃732 osteosarcoma , and Micro⁃PET/CT imaging was successfully performed at different time points (2 , 24 and 48 h after probe injection) to achieve non⁃invasive real⁃time observation of PD⁃L1 in OS⁃732 osteosarcoma .
Conclusion
In this study , the 124 I ⁃anti⁃PD⁃L1 monoclonal antibody molecular probe was constructed in vitro , and Micro PET /CT imaging verified that the probe successfully targeted the PD⁃L1 receptor of OS⁃732 Osteosarcoma , and showed clear immune imaging , indicating that it is hopeful to achieve non⁃invasive screening of Osteosarcoma patients with PD⁃L1 positive expression in clinical practice , and this technology might benefit the majority of Osteosarcoma patients .
10.Accurate Reconstruction of Traffic Accident Based on Multiple Optimization Algorithms and Evaluation of Craniocerebral Injury Risk

Objective To investigate the effect of different optimization algorithms on accurate reconstruction of traffic accidents. Methods Non-dominated sorting genetic algorithm-II ( NSGA-II), neighborhood cultivation genetic algorithm (NCGA) and multi-objective particle swarm optimization (MOPSO) were used to optimize the multi-rigid body dynamic reconstruction of a real case. The effects of different optimization algorithms on convergence speed and optimal approximate solution were studied. The optimal initial impact parameters were simulated as boundary conditions of finite element method, and the simulated results were compared with the actual injuries. Results NCGA had a faster convergence speed and a better result in optimization process. The kinematic response of pedestrian vehicle collision reconstructed by the optimal approximate solution was consistent with the surveillance video. The prediction of craniocerebral injury was basically consistent with the cadaver examination. Conclusions The combination of optimization algorithm, rigid multibody and finite element method can complete the accurate reconstruction of traffic accidents and reduce the influence of human factors.
