Through employing analytical chemistry within the realm of lipidomics, researchers analyze traditional Chinese medicine (TCM). OBJECTIVE To establish the fingerprint of Bushen ningshen ointment， determine the contents of its major constituents， and investigate its in vitro antioxidant activity. METHODS High performance liquid chromatography （HPLC） fingerprints of 10 batches of Bushen ningshen ointment were established. Similarity evaluation and identification of common peaks were subsequently performed. The contents of 10 components such as salidroside were determined using the same HPLC method. Using the scavenging rates against 2，2′-azino bis（3-ethylbenzothiazoline-6-sulfonic acid） （ABTS） and 1，1-diphenyl-2- picrylhydrazyl （DPPH） radicals， as well as ferric ion reducing antioxidant power （FRAP） as indicators， the anti-oxidant activity of the ointment was evaluated； grey relational analysis and partial least squares regression were conducted using SIMCA 14.1 software to establish the spectrum-effect relationship. RESULTS The fingerprint chromatogram of 10 batches of Bushen ningshen ointment contained 24 common peaks， with similarity values all exceeding 0.96. Eleven peaks were identified as adenosine （peak 1）， salidroside （peak 4）， morroniside （peak 6）， catechin （peak 7）， paeoniflorin （peak 10）， spinosin （peak 11）， ferulic acid （peak 12）， isoquercitrin （peak 13）， E-mail：wli1743@163.com verbascoside （peak 14）， paeonol （peak 23）， and emodin （peak 24）. Content determination results showed that the average contents of salidroside， morroniside， catechin， paeoniflorin， spinosin， ferulic acid， isoquercitrin， verbascoside， paeonol， and emodin were 0.725， 1.962， 0.214， 3.395， 0.124， 0.107， 0.286， 0.019， 0.034 and 0.067 mg/g， respectively. The antioxidant potency composite index （APC） for the 10 batches ranged from 85.08% to 96.35%. Spectrum-effect relationship analysis indicated that all 24 common peaks were positively correlated with the antioxidant capacity. Seventeen peaks had variable importance in projection values ＞1， specitically peaks 2， 5， 6， 7， 9， 10， 13- 21， 23， and 24. CONCLUSIONS This study successfully established the HPLC fingerprint and content determination method for Bushen ningshen ointment. The compounds represented by the 17 common peaks such as morroniside may be the active components contributing to its antioxidant effects.

Objective To analyze the trend of global cellulitis disease burden from 1990 to 2019, and to provide a theoretical basis for the prevention and control of cellulitis disease. Methods The Global Burden of Disease 2021 (GBD2021) data were collected, and data on the incidence, mortality, and disability-adjusted life year (DALY) of cellulitis were analyzed for each country worldwide. The estimated annual percentage change (EAPC) and age-standardized rate (ASR) were used to estimate the trend change of cellulitis from 1990 to 2021. Results The global burden of cellulitis increased significantly in 2021, with 55.96 million cases, 28.9 million deaths and 876.1 million DALYs, respectively. Incidence and mortality rates were generally higher in males than in females. The incidence and DALYs were higher in high SDI regions, with the highest burden observed in South Asia. In contrast, East Asia exhibited the lowest burden and demonstrated a declining trend. There were significant differences between countries, with India having the highest prevalence, the United States having the highest incidence, and Bahrain having the fastest growing rate.In 2021, China had the lowest age-standardised incidence of cellulitis in the world and the fastest declining age-standardised incidence and age-standardised DALYs. Conclusion The global disease burden of cellulitis is increasing from 1990-2021, and cellulitis remains an an important global public health problem. Targeted preventive meausres should be taken in areas with different economical levels. Men, middle-aged and elderly people, and newborns are the key groups in need of attention and health education.

Objective:To investigate the cortical activation characteristics of patients with post-stroke foot drop during task-oriented ankle rehabilitation robot training using functional near-infrared spectroscopy(fNIRS).Method:Twenty patients with post-stroke foot drop were recruited.A 51-channel fNIRS system were used to measure changes in oxygenated hemoglobin(HbO2)in the cortical regions during task-oriented ankle rehabilita-tion robot training.The regions of interest(ROIs)included the bilateral dorsolateral prefrontal cortex(DLP-FC),primary motor cortex(M1),primary somatosensory cortex(S1),premotor area and supplementary mo-tor area(pSMA),and inferior frontal gyrus(IFG).Data processing and statistical analysis were conducted us-ing MATLAB(R2021b)and SPSS 25.0 software.Result:Significant cortical activation was observed in channels 19,26,28,29,35,36,39,42,and 48(P＜0.05)during task-oriented ankle rehabilitation robot training in patients with post-stroke foot drop.Based on the channel-to-brain region registration,significant activation was noted in the bilateral pSMA,bilateral DLPFC,unaffected side S1,and unaffected side IFG's area.Conclusion:Task-oriented ankle rehabilitation robot training can lead to increased cortical activation in pa-tients with post-stroke foot drop,which may represent a potential neural mechanism underlying its therapeutic effects.

Objective To investigate the predictive value of inflammatory markers including:fibrinogen-to-albumin ratio(FARI)and lymphocytes × albumin(LA)product before treatment in patients with hypopharyngeal carcinoma for patient's prognosis by creating and internally evaluating columnar graphs based on inflammatory markers.Methods Hypopharyngeal cancer patients who atten-ded the Affiliated Hospital of Xuzhou Medical University from 2009 to 2019 were collected,and the patients who met the criteria were di-vided into the training group and the validation group according to the nativity conditions.The optimal cutoff value was determined by sub-ject operating characteristic(ROC)curve analysis,and the survival curve was analyzed by Kaplan-Meier method.The factors affecting the prognosis were analyzed univariately and multivariately using COX risk regression analysis,and after determining the independent prog-nostic indexes related to overall survival(OS),the prognostic column line graph was constructed and internally validated.Results The relevant data of 134hypopharyngeal cancer patients were retrospectively analyzed,and they were randomly assigned to the training group(93 cases)and the validation group(41 cases).Univariate and multivariate COX regression analyses were performed to obtain the follow-ing factors affecting the survival of patients with hypopharyngeal cancer:body mass index(BMI),lymph node stage,whether or not post-operative adjuvant radiotherapy was performed,and pre-treatment FARI,LA,PLR,and NLR.A prognostic column chart was drawn based on the independent prognostic factors obtained.The C-index of the column-line diagrams for the training and validation groups were0.81(95％CI:0.76-0.87)and0.71(95％CI:0.60-0.82),respectively.In addition,our column-line diagrams demon-strated higher accuracy in predicting patient prognosis compared with conventional TNM staging.Conclusion Our findings confirm the prognostic significance of pretreatment inflammatory markers for patients with hypopharyngeal cancer,with high FARI(＞0.083)and low LA(≤77.30)associated with poorer OS in patients with hypopharyngeal cancer.We can predict individual prognosis more accurately and effectively based on the prognostic column chart.

The adipose tissue of mammals represents an important energy-storing and endocrine organ, and its dysfunction is relevant to the onset of several health problems, including non-alcoholic fatty liver disease (NAFLD). However, whether treatments targeting adipose dysfunction could alleviate NAFLD has not been well-studied. Adrenomedullin 2 (ADM2), belonging to the CGRP superfamily, is a protective peptide that has been shown to inhibit adipose dysfunction. To investigate the adipose tissue-specific effects of ADM2 on NAFLD, adipose-specific ADM2-overexpressing transgenic (aADM2-tg) mice were developed. When fed a high-fat diet, aADM2-tg mice displayed decreased hepatic triglyceride accumulation compared to wild-type mice, which was attributable to the inhibition of hepatic de novo lipogenesis. Results from lipidomics studies showed that ADM2 decreased ceramide levels in adipocytes through the upregulation of ACER2, which catalyzes ceramide catabolism. Mechanically, activation of adipocyte HIF2α was required for ADM2 to promote ACER2-dependent adipose ceramide catabolism as well as to decrease hepatic lipid accumulation. This study highlights the role of ADM2 and adipose-derived ceramide in NAFLD and suggests that its therapeutic targeting could alleviate disease symptoms.
