1.Association between daily physical activity patterns and dyslipidemia among people receiving physical examination aged 40-65 years

Objective:To analyze the association between daily physical activity patterns and dyslipidemia among people receiving physical examination aged 40-65 years.Methods:This cross-sectional study consecutively enrolled 864 participants aged 40-65 years and met the inclusion and exclusion criteria who underwent health check-ups at the Physical Examination Center of Fuquan First People′s Hospital from March to November in 2022. The data of general characteristics, physical activity, physical examination findings, and lipid profiles were collected. The daily physical activity patterns were identified using K-means clustering analysis. The unconditional binary logistic regression was employed to explore the associations between these activity patterns and dyslipidemia, followed by subgroup analyses.Results:The physical activity of the 864 study participants (517 males and 347 females) included in the analysis was divided into 4 patterns (G1: low physical activity; G2: active commuting; G3: housework; G4: leisure exercise). Using G1 as a reference, after adjusting for confounders, G4 was negatively associated with low high density lipoprotein cholesterol (HDL-C) ( OR=0.37, 95% CI: 0.14-1.00) ( P=0.05). In the male, G3 was negatively associated with dyslipidemia ( OR=0.44, 95% CI: 0.21-0.93) and low HDL-C ( OR=0.25, 95% CI: 0.10-0.68) (both P<0.05). In the subjects aged 50 years and above, G2 was negatively associated with dyslipidemia ( OR=0.52, 95% CI: 0.30-0.90), hypertriglyceridemia ( OR=0.50, 95% CI: 0.28-0.90) and low HDL-C ( OR=0.47, 95% CI: 0.24-0.91) (all P<0.05). In those who never or occasionally stayed up late, G2 was negatively associated with hypertriglyceridemia ( OR=0.31, 95% CI: 0.13-0.75) ( P<0.05); in those who stayed up late often, G4 was negatively associated with dyslipidemia ( OR=0.33, 95% CI: 0.13-0.85) and low HDL-C ( OR=0.19, 95% CI: 0.04-0.84) (both P<0.05). In the centrally obese population, G2 was negatively associated with dyslipidemia ( OR=0.55, 95% CI: 0.35-0.88) and hypertriglyceridemia ( OR=0.54, 95% CI: 0.33-0.86) (both P<0.05). Conclusions:Association between different physical activity patterns and dyslipidemia varied among adults aged 40-65 years undergoing health check-ups. Leisure-time exercise is associated with a reduced risk of dyslipidemia, while household activities also emerges as a beneficial factor linked to lower dyslipidemia risk particularly in the male population.
2.Mendelian randomization study on cervical cancer and immune cell phenotypes

Objective This study aims to explore the causal relationships between 731 immune cell phenotypes and cervical cancer using a two-sample Mendelian randomization(MR)analysis.Methods Utilizing results from genome-wide association study(GWAS),inverse variance weighted(IVW),MR-Egger regression,weighted median,simple mode and weighted mode were used for MR analysis,and sensitivity analysis was used for quality control of heterogeneity and horizontal pleiotropy.Results Using IVW as the primary analytical method and ensuring consistency in the direction of results across all five MR methods,we identified five immune cell phenotypes potentially causally associated with cervical cancer.Among these,relative count of central memory CD4+T cells in all CD4+T cells(CM CD4+%CD4+)(ORIVW=0.804,95%CI:0.685-0.943,PIVW=0.0074),absolute count of naive CD8 bright T cell(Naive CD8br AC)(ORIVW=0.493,95%CI:0.359-0.676,PIVW=0.000 01)and median fluorescence intensities of CD45 on immature myeloid-derived suppressor cells(CD45 on Im MDSC)(RIVW=0.746,95%CI:0.609-0.914,PIVW=0.0048)were positively associated with cervical cancer risk,while median fluorescence intensities of IgD on IgD+CD38-unswitched memory B cell(IgD on IgD+CD38-unsw mem)(RIVW=1.393,95%CI:1.148-1.690,PIVW=0.000 78)and median fluorescence intensities of CD3 on terminally differentiated CD8 bright T cell(CD3 on TD CD8br)(ORIVW=1.267,95%CI:1.082-1.483,PIVW=0.0033)were negatively associated with cervical cancer risk.Conclusion This study identifies five immune cell phenotypes closely related to cervical cancer risk through genetic analysis,providing new insights for early prevention and the development of novel immunotherapies for cervical cancer.
3.Application of contrast agent dual-phase double-flow method in CMR"one-stop"examination process of patients with old myocardial infarction

Objective:To explore the influence of injection methods of different contrast agents on CMR image quality,and further optimize"one-stop"examination process under the premise of the analysis for high-pressure bolus pressure curve of contrast-enhanced(CE)cardiac magnetic resonance(CMR).Methods:The data of CMR examination of 70 patients with old myocardial infarction who admitted to the department of emergency of Xuanwu Hospital,Capital Medical University from December 2023 to December 2024 were selected as research objects.They were divided into an observation group and a control group by a simple randomization method,with 35 cases in each group.The observation group adopted gadolinium contrast agent injection with double-phase double-flow,with two bolus injections at 4 ml/s and 2 ml/s sequentially.The control group adopted the injection with single-phase double-flow,with only one bolus injection at 4 ml/s.The dynamic pressure time curve was drawn,and the total bolus dose and pressure peak of two kinds of injection methods were compared.The residual liquid samples of the observation group and the control group were collected after injection of contrast agent was conducted.After the residual liquids of 10 ml,20 ml and 30 ml were derived respectively,the T1 mapping sequence was used to collect signal intensity values,and to compared the difference of that between two groups.The signal-to-noise ratio(SNR)of left ventricular blood pool,infarcted myocardium and distal myocardium,and the contrast-to-noise ratios(CNR)between infarcted myocardium and blood pool,and between distal myocardium and blood pool were compared between the observation group and the control group.Results:There was no statistical difference in the total dose of bolus injection between the observation group and the control group(P>0.05).The peak value of the pressure of bolus injection in the observation group and control group were respectively(87.4±11.3)pound force per square inch(PSI)and(104.0±20.1)PSI,and the difference was statistically significant(t=5.81,P<0.05).T1 signal intensity values of 10 ml residual liquid and 20 mL residual liquid were respectively 1984.43±70.26 and 2190.56±195.96 in observation group,and they were respectively 1203.36±184.99 and 2884.64±349.39 in control group,and the differences were significant(t=-3.57,6.03,P<0.05).The T1 signal intensity value of 30mL residual fluid sample was 4371.75±75.16 in observation group,and that was 4261.86±110.68 in control group,and the difference was no significant(P>0.05).The SNR value of blood pool was(4.88±1.01)in observation group,which was lower than(8.25±1.36)in control group,and the difference of that between two groups was significant(t=6.11,P<0.05).The CNR values of infarcted myocardium and blood pool,of remote myocardium and blood pool between two groups were statistically significant(t=-4.79,-5.39,P<0.05).Conclusion:The contrast agent of high-pressure bolus injection with dual-phase dual-flow method can not only explore the time window of the extravasation of contrast agent,but also improve the SNR and the CNR of CMR images,and further optimize the CMR examination process.

8.Pharmacodynamic evaluation of Polygonum capitatum nanofiber functional dressing

OBJECTIVE To evaluate the efficacy of the functional dressing of Polygonum capitatum nanofibers (P-PVP-PCL). METHODS P-PVP-PCL were prepared by electrospinning technology, and the microstructure of P-PVP-PCL was observed. The antibacterial activity and antioxidant activity of P-PVP-PCL and its effects on the survival rate, adhesion and migration rate of mouse fibroblast L929 cells were investigated. The effects of medical gauze dressing, blank nanofiber dressing (PVP-PCL) and P- PVP-PCL on the healing rate of the wound were investigated by establishing the back skin wound model of rats. The pathological changes of the wound tissue and collagen fiber deposition were observed, as well as the number of platelet endothelial cell adhesion molecule-1 (CD31) positive blood vessels and the expression of transforming growth factor-β (TGF-β) protein in wound tissue. RESULTS P-PVP-PCL had a smooth surface and a double-layer structure at the cross-section. The inhibition rates of P-PVP-PCL against Staphylococcus aureus and Escherichia coli were (98.88±0.66)% and (94.75±1.41)% , respectively. The antioxidant activity of P-PVP-PCL was (83.69±1.56)%, and the cell activity of the P-PVP-PCL group was significantly higher than those of the control group and PVP-PCL group (P<0.05). Compared with medical gauze dressings, P-PVP-PCL was more conducive to L929 cell adhesion; at 48 hours, the cell scratches in this group had basically healed. Compared with the medical gauze dressing group, the wound healing rates of the PVP-PCL group and the P-PVP-PCL group were significantly increased (P<0.05). On the 14th day of intervention, the wounds in the P-PVP-PCL group had basically healed, there was no dermal necrosis in the wound tissue, and the collagen fibers were arranged relatively neatly and the density was relatively uniform. The number of CD31 positive blood vessels and the expression of TGF-β protein showed a downward trend compared with the 7th day of intervention, and the number of CD31 positive blood vessels was significantly lower than those of the medical gauze dressing group and PVP-PCL group (P<0.05), but the protein expression of TGF- β was significantly higher than those of the medical gauze dressing group and the PVP-PCL group (P<0.05). CONCLUSIONS P-PVP-PCL has good antibacterial and antioxidant activity in E-mail:444096585@qq.com vitro, and can promote the proliferation, adhesion and migration of L929 cells. It can promote wound healing of rats in vivo.
9.Target delineation and dosimetry analysis of 125I interstitial brachytherapy for recurrent adenoid cystic carcinoma in the parotid gland

Objective:To analyze the target design and dosimetric parameters of patients with recurrent adenoid cystic carcinoma (ACC) in the parotid gland who were treated with 125I interstitial brachytherapy alone. Methods:A retrospective analysis was conducted for 25 patients with recurrent adenoid cystic carcinoma in parotid gland who were histopathologically diagnosed between January 2015 and October 2019. These patients were treated with 125I interstitial brachytherapy alone, with prescribed doses of 100-120 Gy. The target volume was designed according to the pathological characteristics of ACC and recurrence sites. The pre- and post-operation dosimetric parameters (i.e., local control rates) were calculated using the treatment planning system. Results:In this study, the local recurrence sites included the superficial lobe (10/25) and deep lobe (7/25) of the parotid gland primarily and the skull base region (four patients) and mastoid posterior region (four patients) secondarily. The number of 125I seeds ranged from 16 to 111, with a median number of 59. The activity of radioactive particles was 18.5-25.9 MBq. The 3- and 5-year local control rates were 81.5% and 61.5%, respectively. No significant differences were found between pre- and post-operative dosimetric parameters such as D90, V100, and V150. There was no significant difference in local control rates among the four different recurrence sites. Conclusions:125I interstitial brachytherapy is proven to be an effective approach in the treatment of recurrent adenoid cystic carcinoma in the parotid gland. Satisfying local control rates can be achieved through target delineation performed according to recurrence sites and perineural invasion characteristics of ACC.
10.Knockdown the expression of ku70 and lig4 in HEK293T cells by CRISPR/Cas13 system.

Clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated proteins (Cas) system is a hotspot of gene editing and gene expression research, in which CRISPR/Cas13 system provides a new direction for RNA interference and editing. In this study, we designed and synthesized the corresponding gRNAs of CRISPR/Cas13a and CRISPR/Cas13b systems in non-homologous end joining (NHEJ) pathway, such as Ku70 and Lig4, and then detected the expression of ku70 and lig4 in HEK293T cells. The CRISPR/Cas13a system could efficiently knockdown the mRNA expression of ku70 and lig4 more than 50%, and CRISPR/Cas13b system also suppressed ku70 and lig4 about 92% and 76%, respectively. Also, CRISPR/Cas13a, b systems could down-regulate Ku70 and Lig4 proteins level to 68% and 53%, respectively. The study demonstrates that the CRISPR/Cas13 system could effectively knockdown the expression of RNA and protein in HEK293T cells, providing a new strategy for gene function and regulation research.
