1.Improvement effect and mechanism of salidroside on radiation-induced parotid gland injury in rats by VIP-cAMP pathway

OBJECTIVE To explore the improvement effect and mechanism of salidroside on radiation-induced parotid gland injury in rats. METHODS Rats were randomly assigned into normal group, radiation group, salidroside low-dose (salidroside-L, 50 mg/kg) group, salidroside high-dose (salidroside-H, 100 mg/kg) group, and salidroside-H+inhibitor (100 mg/kg salidroside+0.1 µmol/kg H-89) group, with 10 rats in each group. Except for the normal group, rats in the other groups were subjected to radiation exposure to establish a model of radiation-induced parotid gland injury. Rats in each group were intraperitoneally injected with the corresponding drug or normal saline, once a day, for 40 consecutive days. After the last administration, the levels of reactive oxygen species (ROS), cyclic adenosine monophosphate (cAMP), superoxide dismutase (SOD), and amylase in serum were detected; the pathological changes of parotid gland tissue were observed; the apoptosis rate of parotid gland tissue cells, the expression levels of B-cell lymphoma-2 (Bcl-2) and its associated X protein (Bax), mRNA expression levels of interleukin-6 (IL- 6) and tumor necrosis factor-α (TNF-α), the protein expression levels of type Ⅲ collagen (Col Ⅲ), vasoactive intestinal peptide (VIP), and the phosphorylation level of protein kinase A (PKA) in parotid gland tissue were determined. RESULTS Compared with normal group, the levels of ROS, amylase, apoptosis rate, Bax expression level, mRNA expression levels of IL-6 and TNF- α, and protein expression level of Col Ⅲ in the radiation group were significantly increased, while the levels of cAMP, SOD, Bcl-2 expression level, VIP protein expression level and PKA phosphorylation level were significantly decreased (P<0.05). Compared with radiation group, the above indicators in the salidroside-L group and salidroside-H group were significantly improved (P<0.05), and the improvement in the salidroside-H group was more significant (P<0.05); inhibitor H-89 significantly reversed the changes in the above indicators of the salidroside-H group (P<0.05). CONCLUSIONS Salidroside can effectively alleviate radiation-induced parotid gland injury in rats, and its mechanism may be related to the activation of the VIP-cAMP pathway.
2.Effect of fine skin care on the severity and recurrence of skin lesions in patients with psoriasis

Objective:To explore the application effect of fine skin care in patients with psoriasis, and provide evidence-based theoretical basis for the development of skin care in patients with psoriasis.Methods:A randomized controlled trial was conducted. Patients with psoriasis admitted to the People′s Hospital of Wenshan Zhuang and Miao Autonomous Prefecture, Yunnan Province from December 2022 to March 2024 were selected as the research subjects by the convenience sampling method and divided into the control group and the experimental group by the random number table method. The control group was given routine care. The experimental group was given fine skin care on the basis of control group. Before and after the intervention, the itching symptoms, skin lesions, comfort and quality of life were evaluated using 12-item Pruritus Severity Scale (12-PSS), Psoriasis Area and Severity Index (PASI), General Comfort Questionnaire (GCQ), 36-item Short Form (SF-36) and compared between the two groups. The incidence of complications and recurrence rate in the two groups were counted 3 months after intervention.Results:Finally, 96 patients were included in the study, including 48 patients in the experimental group, 28 males and 20 females, aged (59.31 ± 17.31) years old; 48 cases in the control group, 29 males and 19 females, aged (61.54 ± 18.11) years old. Before the intervention, there were no statistically significant differences in the scores of 12-PSS, PASI, GCQ and SF-36 between the two groups (all P>0.05). After the intervention, the scores of 12-PSS and PASI in the experimental group were (3.65 ± 2.96), (5.08 ± 1.15) points respectively, which were lower than (8.29 ± 2.00), (7.37 ± 1.34) points in the control group, the differences were statistically significant ( t=9.00, 8.99, both P<0.05). The scores of GCQ and SF-36 in the experimental group were (41.42 ± 4.01), (95.08 ± 4.47) points respectively, which were higher than (33.94 ± 5.74) and (84.19 ± 8.52) points in the control group, the differences were statistically significant ( t=7.40, 7.84, both P<0.05). The total incidence of complications and recurrence rate in the experimental group were 4.17% (2/48), 2.08% (1/48) respectively, which were lower than 18.75% (9/48), 14.58% (7/48) in the control group, the differences were statistically significant ( χ2=5.03, 4.91, both P<0.05). Conclusions:Fine skin care can improve the itching symptoms of patients with psoriasis and reduce the severity of skin lesions. It can also improve the comfort and quality of life of patients and reduce the incidence of complications and recurrence rate, and the clinical application effect is good.

It can also improve the comfort and quality of life of patients and reduce the incidence of complications and recurrence rate, and the clinical application effect is good.
4.Clinical Performance of Single-incision Laparoscopic Sigmoid Colon Vaginoplasty in Women with Congenital Absence of Vagina

Objective To explore the feasibility of single?incision laparoscopic sigmoid colon vaginoplasty in women with congenital absence of va?gina. Methods Clinical data of thirty?two patients with congenital absence of vagina admitted in Shengjing Hospital of China Medical University be?tween January 2008 and June 2014 was retrospectively analyzed. Nineteen patients underwent laparoscopic assisted sigmoid colon vaginoplasty,and thirteen paitiens underwent single?incision laparoscopic sigmoid colon vaginoplasty. The operation time,intraoperative blood loss,evacuation time and follow up were compared between the two groups. Results In the group of single?incision laparoscopic sigmoid colon vaginoplasty,the opera?tion time was shorter and intraoperative blood loss was less. There was no difference between two groups in evacuation time. There was no infection or leakage of anastomotic stoma of sigmoid colon,and no postoperative stenosis of the neovagina and its length was satisfied. Patients were satisfied with the quality of sexual life after the surgery. Patients underwent single?incision laparoscopic sigmoid colon vaginoplasty were all satisfied with the ap?pearance of abdomen. Conclusion The single?incision laparoscopic sigmoid colon vaginoplasty can simplify the procedure of intestinal anastomo?sis,shorten the operation time,and reduce the intraoperative blood loss. Consequently,the single?incision laparoscopic sigmoid colon vaginoplasty is effective and safe.
5.Acupoint therapy can reduce airway inflammation and control asthma symptoms

Objective To investigate the effects of acupoint therapy on inflammatory factors and its clini-cal efficacy in relieving bronchial asthma. Methods Selected patients with bronchial asthma which was in remis-sion were randomly divided into a treatment group that was treated with acupoint therapy and a control group that was given Seretide. Each group had 30 cases. The treatment period was 4 weeks. Both groups were evaluated in terms of Asthma Control Test ( ACT) scores and the serum content of interleukin-5 ( IL-5) and interleukin-10 ( IL-10) before and at one month ( short-term) , as well as three months after the end of the treatment ( long-term) . The asthma control situation ( fully controlled, partially controlled or uncontrolled) was evaluated. Results Before treatment the average ACT scores of the two groups were not significantly different. After the treatment both the short-term and long-term average ACT scores of the treatment group were significantly higher than those of the con-trol group. The total effectiveness rate of asthma control in the treatment group in the short term ( 93%) was signifi-cantly higher than that in the control group ( 70%) . After the treatment the IL-5 and IL-10 levels in the treatment group were improved to a significantly greater extent than those in the control group. Conclusion Acupoint thera-py can reduce airway inflammation, control bronchial asthma symptoms and show good clinical efficacy, probably by regulating IL-5 and IL-10 levels.
6.Accuracy of endoscopic ultrasound in the preoperative staging and the guidance of transanal endoscopic microsurgery for rectal cancer.

OBJECTIVETo explore the accuracy of endoscopic ultrasound (EUS) in preoperative staging of rectal cancer and to guide the treatment of transanal endoscopic microsurgery (TEM) in early rectal cancer.
METHODSClinical data of 80 patients with rectal cancer receiving EUS examination for preoperative staging in our department between June and December 2012 were retrospectively analyzed. Consistence comparison of EUS preoperative staging and pathological staging was performed to identify the accuracy of EUS preoperative staging. All the patients underwent operation within 1 week after EUS examination. According to preoperative staging, early rectal cancer(Tis or T1N0M0) patients with lesions less 20 cm to anus underwent TEM.
RESULTSThe overall accuracy of EUS for preoperative T stage was 68.8%(55/80), and for T1, T2, T3, T4 was 91.3%(73/80), 83.8%(68/80), 77.5%(62/80), 85.0%(67/80), which had a good consistence with postoperative pathological T staging(Kappa=0.562). The overall accuracy of EUS for preoperative N stage was 52.7%(39/74), and for N0, N1, N2 stage was 64.9%(48/74), 55.4%(41/74), 85.1%(63/74), which had a poor consistence with postoperative pathological N staging(Kappa=0.235). Six patients underwent TEM successfully, with mean operation time 99(65 to 123) min, without intraoperative and postoperative complication, and were discharged 2-3 days after operation. Enteroscope showed good recovery 1 month later. Pathology confirmed that all the lesions were early rectal cancer. During postoperative follow-up of 14.8 (11 to 19) months, there was no local recurrence and distant metastasis.
CONCLUSIONPreoperative EUS has a good accuracy with pathologic T stage, and can guide TEM in early rectal cancer.

7.Construction and application of a new rat-holding device

Objective To provide a practical device and protocol to hold conscious rats for subsequent operations which can overcome the disadvantages of existing methods .Users can complete the experiment more efficiently , with or without prior experience .Methods Using transparent plastic film , plastic sealing machine and sponge to make a simple device for holding rats , by taking advantage of their escaping nature .To compare the performance of the new method and existing methods for holding and injecting rats .Results Compared with existing methods , the new device and method can reduce the time-consuming to hold rats by 44.7%, from 18.13 seconds to 10.03 seconds.For holding and injecting , the new method can reduce the time-consuming by 55.3%, from 139.33 seconds to 52.26 seconds .Conclusions The new device and method is good for holding and injecting rats or drawing blood from the caudal veins .It can shorten the time of operation and reduce the stress reaction in the animals .It’ s especially helpful for inexperienced experimenters such as students in teaching and research tasks .
8.Effect of Pinggan Qianyang recipe on the expression of Tpx II HSP27 and ANXA1 in the hypothalamus of spontaneously hypertensive rats with hyperactivity of liver-YANG syndrome.

OBJECTIVE:
To investigate the effect Pinggan Qianyang recipe on expression of Tpx, HSP27 and ANXA1 in the hypothalamus of spontaneously hypertensive rats (SHRs) with the hyperactivity of liver-YANG syndrome.
METHODS:
A total of 30 SHRs were subjected to administration of Aconiti Praeparatae Decoction to establish the model of SHR with liver-YANG hyperactivity first, then they were randomly divided into three groups: the control group, the model group and the treatment group (n=10 per group). A total of 10 SD rats were served as the normal group. The rats in control group and treatment group were given Enalapril plus Pinggan Qianyang recipe for four weeks. The change of behavior and blood pressure of rats were monitored. RT-PCR and Western-blot were performed to detect the expression of Tpx II, HSP27 and ANXA1 mRNA and protein in the hypothalamus, respectively.
RESULTS:
Compared with the normal SD rats, the heart rate, blood pressure and grade of irritability were significantly increased while rotation endurance time was dramatically reduced in the SHR model with liver-YANG hyperactivity (P<0.01), these changes were reversed by the application of Enalapril plus Pinggan Qianyang recipe. Compared with the normal SD rats, the protein and mRNA expression of Tpx II and ANXA1 in the model group were significantly upregulated (P<0.01) while the HSP27 was significantly downregulated (P<0.01). Compared with the model group, the protein and mRNA expression of Tpx II and ANXA1 in the control group or treatment group were significantly decreased (P<0.05 or P<0.01) while HSP27 was significantly increased (P<0.05 or P<0.01). Compared with the control group, the expression of Tpx II and ANXA1 protein in treatment group were significantly reduced (P<0.05 or P<0.01).
CONCLUSION
Pinggan Qianyang recipe can improve the blood pressure and behavior in SHRs with hyperactivity of Liver-YANG syndrome, which might be related to the regulation of Tpx, HSP27 and ANXA1 expression in hypothalamuses.
