Objective The aim of this study was to investigate the correlation between the interaction of uric acid and inflammatory factors and hyperuricemia in overweight/obese patients. Methods The personnel with hyperuricemia who underwent physical examination in our hospital from September 2021 to September 2022 were selected as the study subjects, and they were divided into 100 cases of overweight group and 90 cases of obese group according to the BMI index; 120 cases of healthy and non-hyperuricemic personnel were randomly selected as the control group; venous blood of the three groups was collected in 5 mL after 8 h of fasting, and were tested respectively for serum uric acid, lipid indexes and inflammatory factors: IL-6, IL-2, IFN-γ, TNF-α, IL-4, IL-10. Results Glucose, triglycerides, total cholesterol, and LDL were significantly higher in the obese group versus the overweight group (P<0.001), while HDL was significantly lower than the control group (P<0.001), and these changes were more pronounced in the obese group (P<0.001).The Pearson correlation coefficient pointed out that the levels of serum uric acid in patients with hyperuricosuric acid were significantly associated with the pro-inflammatory factors IL- 6, IL-2, IFN-γ, and TNF-α were significantly positively correlated (P<0.001), whereas they were significantly negatively correlated with the anti-inflammatory factors IL-4, IL-10 (P<0.001). Conclusion High uric acid levels in overweight/obese patients can cause enhanced inflammatory responses and reduced expression levels of anti-inflammatory factors, and the interaction between uric acid and pro-inflammatory factors aggravates the condition of patients with hyperuricemia.
2.Research progress in relationship between deubiquitinase OTUB2 and occurrence and development of tumor
Journal of Jilin University(Medicine Edition) 2025;51(6):1763-1770
Deubiquitinating enzymes are important hydrolases in the protein ubiquitin system, which regulate the dynamic balance of protein ubiquitination and deubiquitination by reversing ubiquitination, and play important roles in biological processes such as regulating protein stability, cell signal transduction, and cell apoptosis. As a member of the deubiquitinating enzyme family, ovarian tumor domain-containing protease(OTU)domain ubiquitin aldehyde binding 2(OTUB2)can play a key role in DNA damage, signal transduction, cell metabolism, and tumorigenesis by regulating the ubiquitination levels of various substrates.OTUB2 can be abnormally expressed in various cancers, and as a regulatory factor,it significantly affects the growth, development, proliferation, migration, and invasion of cancer cells, and is closely related to the prognosis of the patients, potentially becoming a novel cancer therapeutic target and prognostic biomarker.Currently, there are many studies on the molecular mechanisms of OTUB2,but there are few review reports focusing on its role in cancer.Based on the research results about OTUB2 at home and abroad, this article reviewed the biological function of OTUB2,related signaling pathways, and its latest progress in cancer research, analyzed the application of OTUB2 in cancer treatment and prognosis evaluation, and provided new ideas for future research on OTUB2.

Objective To explore the prevalence rate and related factors of urban and rural residents with hyperuricemia (HUA). Methods A total of 360 subjects in physical examination department of Sanliusan Hospital from January 2020 to January 2023 were selected and divided into urban residents and rural residents according to their permanent residence addresses, and the demographic information, living habits and underlying diseases were collected. Fasting blood glucose (FBG), serum uric acid (SUA), body mass index (BMI) and triglyceride (TG) were measured. The risk factors of HUA were analyzed by logistics regression analysis. Results The incidence rates of HUA in urban and rural residents were 12.18% and 12.88%. There were statistically significant differences in education level, occupation, BMI, sleep time, alcohol drinking, FBG and TG between urban and rural residents (all P<0.05). Logistics regression analysis showed that male, BMI>24 kg/m2, alcohol drinking and chronic kidney disease were independent risk factors for HUA occurrence among urban residents (all P<0.05). Chronic kidney disease, FBG≥7.0 mmol/L and TG≥2.3 mmol/L were independent risk factors for hyperuricemia occurrence among rural residents (all P<0.05). Conclusion Rural residents should strengthen health education and blood glucose and lipid control, and urban residents should pay more attention to reasonable exercise, control alcohol consumption and reduce HUA occurrence.
4.Research progress on the relationship between leptin and perimenopausal depression
Journal of Apoplexy and Nervous Diseases 2024;41(1):92-96
Perimenopausal period is a period with high incidence of female depression, which shows the symptoms of depressed mood and disturbed mind. The pathogenesis of the disease is not clear and it is urgent to search for efficient treatment. Leptin is a hormone secreted by fat cells and plays a role in both metabolism and neuroprotection. In recent years, leptin has been reported to ameliorate perimenopausal depression, and leptin is closely related to estrogen synthesis and secretion. Therefore, leptin may be used as a potential molecular target for the treatment of perimenopausal depression. In this paper, the research on the relationship between leptin and perimenopausal depression is reviewed.

Objective To investigate the effect of LncRNA HULC on radiosensitivity of osteosarcoma cells. Methods Osteosarcoma cells OS732 was infected by shRNA HULC lentivirus, and the interference effect was determined by qRT-PCR. Osteosarcoma cells infected with shRNA HULC lentivirus were irradiated with 8 Gy X-rays. MTT, PI monochrome staining and Annexin V-FITC/PI double staining were used to detect cell proliferation, cell cycle and apoptosis, respectively. Western blot was used to detect the protein levels of p21, Cyclin D1, C-Caspase-3 and Cyt-C in cytoplasm and mitochondria. Plate cloning assay was used to evaluate cell radiosensitivity. Results The expression of HULC in osteosarcoma cells was significantly down-regulated by shRNA HULC lentivirus infection. Down-regulation of HULC or irradiation inhibited osteosarcoma cell proliferation [(100. 00±9. 65)% vs. (71. 36±5. 27)%, (63. 48± 5. 93)%, t=4. 512, 5. 585, P<0. 05 ] , blocked cell cycle [ ( 50. 15 ± 5. 14 )% vs. ( 62. 35 ± 4. 22 )%, (66. 05±5. 23)%,t=3. 177,3. 756,P<0. 05], induced cell apoptosis [(2. 98±0. 23)% vs. (22. 61± 3. 26)%, (26. 14±2. 81)%,t=8. 898,10. 498,P<0. 05], promoted the expressions of p21 and Cyclin D1 in cells, down-regulated the level of C-Caspase-3 protein, increased the level of Cyt-C protein in cytoplasm, and down-regulated the level of Cyt-C protein in mitochondria. Downregulation of HULC combined with irradiation yield much more effects on cell proliferation inhibition [ ( 71. 36 ± 5. 27 )%, (63.48±5.93)% vs. (49.32±5.76)%, t=4.890, 2.967, P<0.05], cell cycle arrest [(62.35± 4. 22)%, (66. 05±5. 23)% vs. (77. 17±7. 54)%, t=2. 983, 2. 106, P<0. 05], apoptosis induction [(22. 61±3. 26)%, (26. 14±2. 81)% vs. (36. 21±3. 26) %,t=6. 164, 4. 564, P<0. 05] and the expressions of p21, Cyclin D1, C-Caspase-3 and Cyt-C in osteosarcoma cells. The radiosensitization ratio of down-regulation of HULC was 1. 432. Conclusions Down-regulation of HULC enhances radiosensitivity of osteosarcoma cells, which may be related to cell cycle arrest and apoptosis induction.
