1.Correlation Analysis Between Microbial Community Changes and Medicinal Quality Formation During Processing of Angelicae Dahuricae Radix

ObjectiveTo compare the differences in color, odor, coumarin content and microbial community composition of Angelicae Dahuricae Radix(ADR) during different drying processes, and to explore the correlation between changes in microbial community composition and changes in quality indexes of ADR. MethodsThe fresh ADR was processed at three drying temperatures(50, 70, 100 ℃) by drying and steaming cutting, semi-fresh cutting and drying, fresh cutting and drying, and sulfur fumigation methods. The color values of samples were extracted by Adobe Photoshop 2022 software and subjected to principal component analysis(PCA), electronic nose was used to identify the odor information of medicinal powders and subjected to loadings analysis, PCA, and linear discriminant analysis(LDA), and high performance liquid chromatography(HPLC) was used to determine the contents of five coumarins(bergapten, oxypeucedanin, imperatorin, phellopterin, isoimperatorin). The samples for microbial detection were taken from fresh dried samples, 50 ℃(dried and steamed cut, sulfur fumigated) samples, and 100 ℃(dried and steamed cut) samples when the water content was 50% and 14%, respectively. And the changes of microbial community composition during processing were determined by high-throughput sequencing method. The relationship between the changes of microbial community composition and the changes of odor, color and active component content of ADR during drying process was analyzed by Pearson correlation analysis. ResultsThe color quantification results showed that an increase in drying temperature led to the decrease of brightness value(L), and the increases of red-green value(a) and yellow-blue value(b), and the change of processing method had no obvious effect on the color of medicinal materials. The results of odor quantification showed that W1S, W2S, W5S, W2W and W1W sensor were sensitive to the odor changes of ADR and could be used to distinguish ADR decoction pieces from different processing methods. The results of HPLC showed that the coumarin content of ADR decreased with the increase of drying temperature and the delay of processing time, the optimal processing method was drying and steaming cutting method, and the optimal temperature was 50 ℃. High-throughput sequencing results showed that the dominant bacteria in ADR during processing were Achromobacter, Agrobacterium, Nocardioides, Mycobacterium and Enterobacter, the dominant fungi were Coprinopsis, Meyerozyma and Apiotrichum. The results of correlation analysis showed that the quality indexes of ADR were positively correlated with Agrobacterium, Mycobacterium in bacteria, Candida in fungi, and negatively correlated with Bacillus in bacteria. ConclusionThere are significant differences in the color, odor, coumarin content and microbial community composition of ADR in different drying processes, and the best drying method is drying and steaming cutting at 50 ℃. The relative abundance changes of 9 bacterial genera and 4 fungal genera are closely related to the quality formation of ADR during the drying process.
2.Effects of different dressing methods on wound healing after cosmetic suturing for facial trauma

Objective:To evaluate the effects of two dressing methods on wound healing and patient satisfaction after cosmetic suturing for pediatric facial trauma.Methods:A prospective randomized controlled trial was conducted at Dongying People′s Hospital from October 2022 to October 2023. A total of 180 pediatric patients [105 males, 75 females, aged 3-7 (3.9±1.4) years] with facial trauma requiring cosmetic suturing were enrolled in this study. Participants were randomly divided into the study group ( n=91) and control group ( n=89) using a random number table. During the first postoperative dressing change at 24 hours, the study group received saline cleaning followed by erythromycin ointment coverage, while the control group underwent iodine disinfection with gauze coverage. Pain intensity during the second dressing change was assessed using the Chinese version of the Children′s Hospital of Eastern Ontario pain scale (CHEOPS). Wound healing at 6-7 days post-suturing was evaluated using the Chinese version of the redness, edema, ecchymosis, discharge, and approximation (REEDA) scale. Scar appearance at 14 days was measured via the Stony Brook scar evaluation scale (SBSES). Complication rates (infection, dehiscence) and satisfaction rates were statistically analyzed. Results:During the second dressing change, the CHEOPS score was significantly lower in the study group (6.27±1.32) than that in the control group (6.89±1.21) ( P=0.001). At 6-7 days, the REEDA score in the study group (2.26±1.91) was significantly lower than that in the control group (3.07±2.13) ( P=0.008). At 14 days, the SBSES score was significantly lower in the study group [2.60±1.42) vs (3.89±1.50), P<0.001]. The infection rate was 1.09% (1/91) in the study group and 1.12% (1/89) in the control group, with two cases of epidermal dehiscence observed in the control group. The satisfaction rate in the study group was 93.41% (85/91), which was higher than that in the control group [85.49% (76/89), P=0.020]. Conclusion:Saline cleaning combined with erythromycin ointment coverage reduces pain during wound dressing change, facilitates early wound healing, and improves patient′s satisfaction.

Heart defibrillation monitor is a key device in rescuing patients with cardiac arrest,and its performance directly determines the success rate of rescue.The control circuit of the heart defibrillation monitor is a control module for charging and discharging during the defibrillation and electric shock process.Functionally,it is divided into control module of direct current(DC)inverter,energy storage capacitor charging module,cardiac defibrillation discharge module,and cardiac defibrillation monitoring module.In them,the energy storage capacitor is the core component of the control circuit.During the charging process,the energy storage capacitor stores charges,and it releases charges during the discharging process,which can provide control functions for effective implementing defibrillation and electric shock.The control accuracy of the heart defibrillation monitor is high,and its structural performance is constantly improving.The equipment's users and engineering support personnel need to master its structural composition and working principle,and be familiar with the fault analysis and handling process,and timely and effectively handle faults and restore normal operation of equipment.This study analyzed the structural composition and working principle of the control circuit of the heart defibrillation monitor.According to the control mode of charging and discharging,it analyzed the methods of analyzing and handling common faults of the heart defibrillation monitor,so as to improving the quality of operation management and clinical services of equipment.

6.Expression of ET-1 and PLGF in serum of preeclampsia patients in early pregnancy and its prognostic significance

Objective To investigate the expression of endothelin-1(ET-1)and placental growth factor(PLGF)in patients with preeclampsia in early pregnancy and its prognostic significance.Methods A total of 89 patients with preeclampsia who were admitted to No.904 Hospital of Joint Logistic Support Force of PLA from March 2019 to May 2021 were assigned to observation group.And 81 healthy pregnant women who underwent physical examination during the same period were enrolled as controls.Their clinical data were retrospectively analyzed.Serum ET-1 and PLGF levels were detected after admission.The correlation between serum ET-1 and PLGF levels was analyzed.All the participants were followed up until term pregnancy,and the prognoses of the two groups were compared.The patients in the observation group were subdivided into good prognosis group and poor prognosis group according to the prognoses.Multivariate logistic regression analysis was used to analyze the risk factors of poor prognosis in patients with preeclampsia.The receiver operating characteristic(ROC)curve was prepared,and the area under the curve(AUC)was used to evaluate the predictive value of serum ET-1 and PLGF levels for poor prognosis of preeclampsia patients.Results Serum ET-1 level in the observation group was higher than that in the control group,while serum PLGF level in the observation group was lower than that in the control group(both P<0.05).Pearson correlation analysis showed that there was a negative correlation between serum ET-1 and PLGF in both groups(P<0.05).The incidence of poor prognosis in the observation group was significantly higher than that in the control group(20.22%vs 7.41%,P<0.05).The proportion of severe cases,24-hour urine protein quantitation,uric acid,and serum ET-1 in the poor prognosis group were higher than those in the good prognosis group,while serum PLGF level in the poor prognosis group was lower than those in the good prognosis group(P<0.05).Multivariate logistic regression analysis showed that severe condition,24-hour urine protein quantitation,serum ET-1 and PLGF levels were risk factors for poor prognosis of preeclampsia patients(all P<0.05).The ROC curve showed that the AUC(95%CI)values of serum ET-1 and PLGF in predicting poor prognosis of preeclampsia were 0.825(0.709-0.906)and 0.771(0.651-0.852),respectively.Conclusion Patients with preeclampsia have higher ET-1 and lower PLGF than healthy pregnant women.Both ET-1 and PLGF can be used to predict the prognosis of patients with preeclampsia.
8.Fgf8P2A-3×GFP/+: A New Genetic Mouse Model for Specifically Labeling and Sorting Cochlear Inner Hair Cells.

The cochlear auditory epithelium contains two types of sound receptors, inner hair cells (IHCs) and outer hair cells (OHCs). Mouse models for labelling juvenile and adult IHCs or OHCs exist; however, labelling for embryonic and perinatal IHCs or OHCs are lacking. Here, we generated a new knock-in Fgf8P2A-3×GFP/+ (Fgf8GFP/+) strain, in which the expression of a series of three GFP fragments is controlled by endogenous Fgf8 cis-regulatory elements. After confirming that GFP expression accurately reflects the expression of Fgf8, we successfully obtained both embryonic and neonatal IHCs with high purity, highlighting the power of Fgf8GFP/+. Furthermore, our fate-mapping analysis revealed, unexpectedly, that IHCs are also derived from inner ear progenitors expressing Insm1, which is currently regarded as an OHC marker. Thus, besides serving as a highly favorable tool for sorting early IHCs, Fgf8GFP/+ will facilitate the isolation of pure early OHCs by excluding IHCs from the entire hair cell pool.

9.Exploring the Connotation of Qi based on the Original Holistic Principle of Systematic Traditional Chinese Medicine

This paper systematically analyzed the understanding of qi from the perspectives of matter, energy, information and relationship reality, introduced the original holistic principle of systematic traditional Chinese medicine (TCM), and considered the latest research results of qi (three-layer material theory), trying to optimize the structure framework of the qi theoretical system and exhibit the occurrence and development rules of original qi. It emphasizes the hierarchical order of qi transformation following the original holistic principle, and takes this to guide the clinical understanding of “qi diseases”, helping doctors grasp the basic pathogenesis of the disease, that is abnormal qi movement, and helping them establish the awareness of providing systematic TCM treatment to patients by taking qi regulation as the key. At the same time, it discusses people within the structure of time and space, and points out that the treatment of diseases must comply with the principle of “the harmony of heaven, earth, and human beings”.
10.Long-term efficacy of fibrin sealant treating proximal type Ⅰ endoleak after endovascular aneurysm repair in abdominal aortic aneurysm

Objective To observe the long-term efficacy of human fibrin sealant (FS) in the treatment of proximal type Ⅰ endoleak after endovascular aneurysm repair (EVAR) in abdominal aortic aneurysm (AAA).Methods The clinical data of 104 AAA patients with proximal type Ⅰ endoleak receiving EVAR + FS in Changhai Hospital from 2003 to 2012 was retrospectively analyzed,among those 77 cases were with less than 15 mm proximal neck,21 cases with greater than 60 degrees proximal neck angulation,37 cases with severe calcification or thrombosis in proximal neck.After failure of conventional endoleak therapy FS was injected through AAA catheter and long-term efficacy was evaluated by CTA during the follow-up.Results Intra-sac pressure decreased significantly after FS injection.Three patients (2.9%)died perioperatively.Postoperative 1'-,3' and 5 year survival rate was 91.8%,80.6% and 60.2%respectively.Maximum diameter of AAA decreased from (58.78 ± 13.41) mm to (52.6-± 12.2) mm.There was no FS injection related complications.Conclusion Intra-sac injection of FS is an effective,economical and safe method for treating post-EVAR endoleak,especially for AAA with relatively short and twisted aneurysm neck.
