Objective:To identify potential therapeutic targets for the treatment of hair color and hair shaft abnormalities, thereby offering innovative insights and strategies for the management of the associated conditions.Methods:Using the protein quantitative trait locus(pQTL) data derived from extensive proteomics studies, a two-sample Mendelian randomization was conducted to preliminarily identify potential drug therapeutic targets. Following this, sensitivity analyses were performed to evaluate potential confounding factors, including heterogeneity and horizontal pleiotropy. A leave-one-out sensitivity analysis was also conducted, systematically excluding each single nucleotide polymorphism (SNP) to evaluate their individual impact. Additionally, co-localization analysis was carried out to determine the presence of shared genetic variants between the identified plasma proteins and the relevant traits.Results:The proteomic data analyzed in this investigation encompassed 4 907 pQTLs, while the genetic data pertaining to hair pigmentation and shaft abnormalities included 124 cases and 432 686 controls. Through the Mendelian randomization screening, six candidate protein genes were identified: HLA-DQA2, CTSB, KIR2DS2, SVEP1, HOMER2, and HOMER1. Sensitivity analyses revealed no evidence of heterogeneity or horizontal pleiotropy among these proteins. The leave-one-out sensitivity analysis demonstrated that no single SNP significantly affected the overall findings. Notably, HOMER2 was substantiated by co-localization analysis, which provided robust evidence of its potential role in regulating the genetic mechanisms associated with hair pigmentation and shaft integrity.Conclusion:This study successfully identified six potential therapeutic targets for hair pigmentation and shaft abnormalities, with HOMER2 exhibiting the most compelling evidence. These findings pave the way for novel therapeutic approaches in the treatment of hair color and hair shaft disorders.

Objective:To investigate changes in psychological status before and after sequential therapy in patients with skin circulatory disorders following hyaluronic acid injection.Methods:A retrospective study was conducted on 17 female patients, with a mean age of 28-54 (39.5±1.2) years, admitted to the Fourth Medical Center of PLA General Hospital from December 2021 to July 2022. Patients received ultrasound-guided percutaneous arterial puncture or digital subtraction angiography (DSA) guided catheter intervention for hyaluronidase injection, alongside corticosteroid pulse therapy, diuretic administration, neurotrophic support, wound protection, and laser therapy. Psychological assessments, including the Minnesota multiphasic personality inventory (MMPI) and symptom check list-90 (SCL-90), were performed at admission and 30-45 days post-discharge. Changes in assessment scores and patient satisfaction were analyzed.Results:Two patients failed to complete all assessments, leaving 15 cases for final analysis. Affected anatomical regions included the nose/perinasal area (13 cases), temporal region (3 cases), lips (3 cases), and forehead (7 cases). MMPI scores for hypochondriasis, depression, hysteria, psychopathic deviation, paranoia, psychasthenia, and hypomania showed statistically significant reductions compared to baseline (all P<0.05). SCL-90 scores for somatization, obsessive-compulsive symptoms, interpersonal sensitivity, depression, anxiety, hostility, phobic anxiety, paranoid ideation, psychoticism, and sleep/appetite disturbances also demonstrated significant decreases (all P<0.05). Among the 15 patients, 7 patients reported being very satisfied, 6 patients were satisfied, and 2 patients were moderately satisfied, with no dissatisfaction reported. Conclusion:Sequential therapy is associated with improved psychological outcomes in patients with skin circulatory disorders secondary to hyaluronic acid injection.

Objective:To explore the promotion effect on remineralization of demineralized dentin of urushiol primer applicated in acid-etch-rinse adhesives,and to clarify its impact on the longevity of dentin adhesion.Methods:Ninety-six freshly extracted,caries-free third molars were selected to prepare the dentin specimens.Following acid etching with 37％phosphoric acid gel,the specimens were randomly divided into blank control group,0.3％,0.7％,1.0％,and 1.5％urushiol groups,and positive control group(acetone solvent).The treated samples were placed in modified simulated body fluids for remineralization for 14 and 28 d.Attenuated total reflection Fourier transform infrared spectroscopy(ATR-FTIR)was used to detect the relative mineralization mass of minerals in the dentinal tubules in various groups,and X-ray Diffractometery and Energy Dispersive spectrometer were used to analyze the dentin surface material compositions in various groups.Scanning electron microscope(SEM)was used to observe the surface morphology of the specimens in various groups,Vickers hardness tester was used to measure the microhardness of the dentin surface in various groups,and microtensile strength(μTBS)was used to examine the effect of the bond strengthes in various groups.Results:Compared with blank control group,the conversion rate of adhesive double bonds by primer in positive control group was decreased,but the difference was not significant(P＞0.05);but the conversion rates of adhesive double bonds by primer in 0.3％,0.7％,1.0％,and 1.5％urushiol groups were increased(P＜0.05).The SEM results revealed that at 14 and 28 d,compared with bland control group,a minimal membranous deposit in dentinal tubules was seen in positive control group,minimal mineralization displayed in 0.3％urushiol group,significant deposition of loose mineral particles with blocking the tubule orifices was found in 0.7％urushiol group,noticeable mineral precipitates exhibited in 1.0％urushiol group,and relatively empty dentinal tubules were seen in 1.5％urushiol group.The microhardness results showed that at 14 d after remineralization,compared with blank control group,the microhardness in positive control group showed no significant improvement(P＞0.05),while the differences in 0.3％,0.7％,1.0％and 1.5％urushiol groups were statistically significant(P＜0.05);at 14 d after remineralization compared with positive control group,the microhardness of dertin in 0.7％and 1.0％urushiol groups were increased(P＜0.05);at 28 d after remineralization,compared with blank control and postive control groups,the microhardness in urushiol groups were significantly increased(P＜0.05),espectially in 0.7％to 1.5％urushiol groups(P＜0.05).In the μTBS test,at 14 d after remineralization,compared with postive control group,theμTBS in 0.3％,0.7％,1.0％and 1.5％urushiol groups were increased(P＜0.05);at 28 d after remineralization,the μTBS in blank control group was the lowest;compared with blank control group,there was no significant difference in the μTBS in postive control group(P＞0.05);compared with positive control group,the μTBS in 0.3％,0.7％,1.0％,and 1.5％urushiol groups were increased(P＜0.05),espectially in 0.7％,1.0,and 1.5％urushiol groups(P＜0.05).Conclusion:Natural-derived urushiol,as a novel primer,can pretreat the demineralized dentin substrates,and facilitate collagen cross-linking within the dentin matrix;moreover,it leverages the phenolic hydroxyl groups within its structure to attact calcium and phosphate ions,envelope dentin collagen fibers to promote remineralization,in order to enhance the strength of the resin-dentin bonding interface.

Recent epidemiological studies have increasingly found that pregnant women who are exposed to air pollutants (for example airborne particulate matter, nitrogen oxides, ozone, and sulfur dioxide) increase the risk of various birth defects in their offspring, such as congenital heart disease, neural tube defects, cleft lip and palate, and hypospadias. Hypospadias not only impairs the sexual function of infants but also causes major social and psychological problems during their growth period, therefore, the prevention and treatment of hypospadias infant carry substantial public health importance. However, the association between prenatal exposure to air pollution and hypospadias remains controversial. The study reviews the epidemiological research progress and potential biological mechanisms of prenatal maternal exposure to air pollutants such as particulate matter, nitrogen oxides, ozone, sulfur dioxide, and the risk of hypospadias in offspring. The study also summarizes the limitations of previous research and looks forward to future research directions, to provide scientific evidence for creating a healthy living environment for pregnant women, and reducing the risk of hypospadias in offspring.
