1.Correlation Between Intervertebral Disc Degeneration and Paravertebral Muscle Fat Infiltration in Chronic Low Back Pain via Quantitative MRI

Purpose To investigate the correlation between intervertebral disc degeneration and paraspinal muscle fatty infiltration in chronic low back pain(CLBP)using quantitative MRI techniques.Materials and Methods A total of 63 CLBP patients and 42 volunteers in Zigong Fourth People's Hospital from September 2023 to March 2024 were retrospectively collected.Magnetic resonance scans were performed using multi-echo water-fat separation technique and T2 mapping technique.Intervertebral disc degeneration was assessed using the Pfirrmann grading,while paraspinal muscle fatty infiltration was evaluated using the Goutallier grading.Proton density fat fraction(PDFF)of the multifidus and erector spinae muscles,and T2 values of the nucleus pulposus were quantitatively measured.The relationships between these parameters and their associations with age and gender were analyzed.Results Compared with healthy volunteers,CLBP patients showed significantly reduced T2 values of the nucleus pulposusand significantly increased PDFF of the paraspinal muscles(Z=-5.54-4.65,all P<0.001).The Pfirrmann grading of intervertebral discs and the Goutallier grading of paraspinal muscles were significantly correlated with the T2 value of the nucleus pulposus and the PDFF of the paraspinal muscles(|r|>0.4,P<0.05).Age and Pfirrmann grading were independent predictors of paraspinal muscle fat infiltration(β=0.228-5.682,all P<0.001),and gender was also an independent predictor at the L4/5 intervertebral disc level(β=-2.834,-3.352,both P<0.05).Conclusion Intervertebral disc degeneration and paraspinal muscle fat infiltration are closely related in CLBP,age and gender also have an impact.

3.Piperlongumine inhibits the proliferation of triple⁃negative breast cancer cells by regulating STAT3/HIF⁃1α pathway

Objective :
To explore the effects of piperlongumine ( PL) on the proliferation and apoptosis of triplenegative breast cancer MDA⁃MB⁃231 cells in vitro and in vivo as well as its possible molecular mechanism.
Methods:
MDA⁃MB⁃231 cells were treated with different concentrations of PL. MTT assay was performed to detect the level of cell proliferation. The apoptotic level was detected by flow cytometry. Western blot was performed to detect protein expression levels of proliferating cell nuclear antigen(PCNA) , B ⁃cell lymphoma⁃2 (Bcl⁃2) , cyclin⁃dependent kinase inhibitor 1A(CDKN1A/p21) , phosphorylated signal transducer and activator of transcription 3(p⁃STAT3), signal transducer and activator of transcription 3 (STAT3) , hypoxia inducible factor⁃1 α ( HIF⁃1α ) and Survivin. MDA⁃MB⁃231 cells were used to model tumor bearing nude mice and then PL was injected by intraperitoneal ( i.p. ) . The tumor protein expression levels of PCNA , p ⁃STAT3 , STAT3 , HIF⁃1 α and Survivin were detected by Western blot.
Results :
PL inhibited cell proliferation and induced apoptosis in a dose⁃dependent manner. PL down⁃regulated the protein expression levels of PCNA , Bcl⁃2 , p ⁃STAT3 , HIF⁃1 α and Survivin , and up⁃regulated the protein expression level of p21 . Furthermore , PL inhibited tumor growth and down⁃regulated the protein expression levels of PCNA , p ⁃STAT3 , HIF⁃1 α and Survivin in nude mice.
Conclusion
PL inhibits the proliferation of MDA⁃MB⁃231 cells in vitro and in vivo , and the underlying mechanism can be related to the negative regulation of STAT3/HIF⁃1 α pathway.

The suitable exercise plays a particular role in patients receiving maintenance hemodialysis. In recent years, many reports demonstrated the suitable exercise intervention could improve the well being, vitality, fatigue, cardiovascular and mental functions and the efficiency of dialysis. Many kinds of exercise interventions, such as low intensity prolonged aerobic exercise, home-based aerobic exercise, etc., have been applied. It is important to be individualized the intensity, time and frequency of the exercises.
6.Management of complications in endovascular treatment for arteriosclerotic occlusive disease of lower extremities

ObjectiveTo discuss the prevention and treatment of the perioperative complications in endovascular treatment for arteriosclerosis obliterans (ASO) of lower extremities.MethodsEndovascular treatment was performed for 210 patients (248 limbs) with ASO of lower extremities,including 28 iliac lesions,76 femoral-popliteal lesions,56 inferior genicular lesions and 88 multiple segment lesions.The clinical data and complications were reviewed and analyzed.Results The technical success was achieved in 92.74% (230/248).Perioperative complications occurred in 21 patients (21/210,10.00 %),including hematoma (n= 6),pseudoaneurysm (n= 1),heart failure (n= 2),constrast-induced nephropathy (n = 1),artery perforation (n= 3),artery section (n = 3) and restenosis (n= 5).All the complications were relieved after corresponding treatment.ConclusionEndovascular treatment is a safe and effective method for ASO of lower extremities because of its high success rate and few complications.Effective control of systemic diseases,careful operation and regular treatment after operation are all the effective measures to prevent and reduce the occurrenceof complications.
7.ADF/cofilins proteinic family constellation in cytoskeletal protein:How does depolymerization occur through combination of ADF/cofilins with actin?

The actin depolymerizing factor/cofilin (ADF/cofilins) are a family of actin-binding proteins expressed in all eukaryotic cells. The ADF/cofilins appear to have multiple functions, and this is reflected in their very complex association with both monomeric and filamentous actin. Phosphorylation by some kinases and other factors such as LIM kinases 1 and 2, TESK 1 and TESK 2 kinase, Insulin, etc, prevents ADF/cofllins from binding actin. The serial researchs of ADF/cofilins are increasingly becoming study hot spots, especially on the relationship between homo-sapiens disease and mechanism of action of ADF/cofilins.Now in this domain wilderness details are still far from clear, such as the mechanism by which actin filaments are depolymerized by ADF/cofilins has been controversial.
8.Experimental study of water and electrolyte changes following seawater immersion of animals with intestinal rupture

Objective To evaluate the changes in water, electrolyte and acid-base balance after seawater immersion in cases of open abdominal trauma associated with intestinal rupture, and to obtain a theoretical basis for the early treatment of open abdominal injury with intestinal rupture in naval combat. Methods A canine model of open abdominal trauma with intestinal rupture was established in 26 healthy adult dogs, and they were divided randomly into three groups. All animals were subjected to abdominal wall incision and intestinal rupture. Seawater immersion group(n=10) was immersed into artificial seawater after trauma; normal saline solution group(n=6) was immersed into normal saline solution after trauma; control group (n=10) had no immersion. The 3 groups were observed for changes in water, electrolyte and acid-base balance, and the results were analysed and compared. Results Signficant disturbance of water, electrolyte and acid-base imbalance were observed in the seawater immersion groups, but no significant changes of these parameters were seen in the control group and normal saline group. Conclusion Seawater immersion is the main factor leading to the disturbance of body metabolism after open abdominal trauma with intestinal rupture.
9.Early treatment for dogs suffering from open abdominal injury and sea water immersion

Objective To evaluate a protocol of early treatment for dogs suffering from open abdominal injury and seawater immersion. Methods A canine model of open abdominal injury was made with 30 adult health dogs. After 2 hours seawater immersion, the dogs were divided into two groups: treatment group( n =18) and control group(untreated group, n =18). The changes of survival rate, body liquid metabolism and hemodynamics were observed respectively.
