OBJECTIVE:
To compare the mRNA level of cell proliferation-related genes Twist1, SIRT1, FGF2 and TGF-β3 in placenta mesenchymal stem cells (PA-MSCs), umbilical cord mensenchymals (UC-MSCs) and dental pulp mesenchymal stem cells (DP-MSCs).
METHODS:
The morphology of various passages of PA-MSCs, UC-MSCs and DP-MSCs were observed by microscopy. Proliferation and promoting ability of the three cell lines were detected with the MTT method. Real-time PCR (RT-PCR) was used to determine the mRNA levels of Twist1, SIRT1, FGF2, TGF-β3.
RESULTS:
The morphology of UC-MSCs and DP-MSCs was different from that of PA-MSCs. Proliferation ability and promoting ability of the PA-MSCs was superior to that of UC-MSCs and DP-MSCs. In PA-MSCs, expression level of Twist1 and TGF-β3 was the highest and FGF2 was the lowest. SIRT1 was highly expressed in UC-MSCs. With the cell subcultured, different expression levels of Twist1, SIRT1, FGF2, TGF-β3 was observed in PA-MSCs, UC-MSCs and DP-MSCs.
CONCLUSION
Up-regulated expression of the Twist1, SIRT1 and TGF-β3 genes can promote proliferation of PA-MSCs, UC-MSCs and DP-MSCs, whilst TGF-β3 may inhibit these. The regulatory effect of Twist1, SIRT1, FGF2 and TGF-β3 genes on PA-MSCs, UC-MSCs and DP-MSCs are different.

Objective To detect the expression levels of cytokines in peripheral blood of patients with diffuse large B-cell lymphoma (DLBCL), and to explore their clinical significances. Methods The levels of interleukin-1β (IL-1β), IL-2R, IL-6, IL-8, tumor necrosis factor (TNF) and IL-10 in peripheral blood of 38 newly diagnosed patients with DLBCL and 20 healthy adults in Anqing Municipal Hospital from April 2013 to January 2017 were detected and analyzed by immunofluorescence method. Results The levels of IL-2R, IL-6, IL-8, TNF and IL-10 in DLBCL group were significantly higher than those in healthy control group (all P< 0.05). The levels of IL-2R and TNF in stage Ⅲ-Ⅳ group [(5985±26) U/ml and (42.2±5.8) pg/ml)] were higher than those in stageⅠ-Ⅱ group [(3672 ±28) U/ml and (30.4 ±2.6) pg/ml] (t values were 34.861 and 28.451; P values were 0.023 and 0.038 respectively). The levels of IL-2R and IL-10 in the high-risk group (IPI 4-5) [(6322±36) U/ml and (77±7) pg/ml] were higher than those in the low-risk group (IPI 0-1) [(2567± 32) U/ml and (50±5) pg/ml)] (t values were 58.327 and 24.264; P values were 0.001 and 0.041 respectively). IL-2R, IL-6 and IL-10 levels in patients with B symptom group [(6234±26) U/ml, (38.1±2.3) pg/ml, (90± 10) pg/ml] were higher than those in patients without B symptom group [(6234 ±26) U/ml, (38.1 ±2.3) pg/ml, (90±10) pg/ml) (t values were 32.263,24.321, 36.529; P values were 0.027, 0.043, 0.020). Conclusion The levels of IL-2R, IL-6, IL-8, TNF and IL-10 in DLBCL patients are higher than those in healthy controls, and the levels of cytokines are correlated with the clinical characteristics of DLBCL patients.

Objective To investigate the adjustment of miRNA-155 on CD4+ CD25+ Treg regulative T cell in peripheral blood in patients with acute cerebral infarction (ACI) and its pathogenesis. Methods Sixty patients with ACI were divided into three groups according to clinical neurological deficit score. Twenty healthy volunteers were enrolled into the control group. The expression levels of plasma miR-155 mRNA and Foxp3 mRNA were detected by real-time quantitative PCR(qRT-PCR). IL-10 levels in plasma were detected by ELISA. Results Expression of miR-155, Treg, Foxp3 mRNA and levels of IL-10 were significantly increased in patients with ACI compared with normal control group, with statistical differences; Expression of miR-155, Treg, Foxp3 mRNA and levels of IL-10 were gradually increased. The values showed significant statistical difference among the mild, moderate and severe ACI groups (P < 0.01). Among the patients, the levels of miR-155, Treg, Foxp3 mRNA and levels of IL-10 in the survival group were obviously lower than those in the non (P<0.05 or P<0.01). There was a positive correlation between miR-155 and Treg, Foxp3 mRNA (P < 0.01). Conclusion This study suggests that miR-155 is involved in the cell proliferation regulation of CD4+ CD25+ Treg cells, and plays some role in the immunological dissonance with ACI.

A new animal model of multiple system organ failure (MSOF) in rabbits is used in this experiment. The purpose of the current study is to observe dynamically the changes in the intestinal flora, the relationship between the changes and bacterial translocation to various organs and endotoxemia. The results show that there is no obvious alteration in population of Bacteroides, but a significant increase in enteric bacilli population in the contents of the ileocecum is found. Changes in the intestinal micro-flora correlate to the positive bacterial culture of the blood, viscera, and the level of endotoxin in the plasma. The results of our work suggest that ecological imbalance in the intestinal flora may promote-the gut-derived septic process and the development of MSOF.
