OBJECTIVE To evaluate the efficacy and safety of intranasal corticosteroids (INCS) combined with oral H1- antihistamine (AH) in the treatment of allergic rhinitis. METHODS A comprehensive literature search was conducted in PubMed, Embase, CNKI, and VIP database to collect randomized controlled trial (RCT) that INCS combined with AH (experimental group) versus INCS (control group) in the treatment of allergic rhinitis from the inception to December 31, 2024. After study selection, data extraction, and quality assessment, a meta-analysis was performed by using RevMan 5.3 software. RESULTS A total of 12 RCTs were included in the meta-analysis, with a total of 1 842 patients. Results of meta-analysis showed that, compared with the control group, patients in the experimental group had a greater reduction in sneezing scores, rhinorrhea scores, nasal congestion scores, nasal itching scores, total nasal symptom scores, total ocular symptom scores, and rhinoconjunctivitis quality of life questionnaire scores (P<0.05). The incidence of headache was lower in the experimental group (P<0.05); whereas there were no significant differences between the two groups in the incidence of nosebleed, dryness of the mouth and nose, drowsiness, fatigue, and total adverse events (P>0.05). CONCLUSIONS Compared with INCS monotherapy, the combination of INCS and oral AH provides superior efficacy and safety in the treatment of allergic rhinitis.

ObjectiveTo investigate the change in the expression of magnesium transporter 1 (MagT1) in peripheral blood CD8+ T cells in patients with chronic hepatitis B virus (HBV) infection, as well as the correlation of serum Mg2+ and MagT1 with HBV DNA load. MethodsA total of 102 patients with HBV infection who were admitted to Tangshan Workers’ Hospital from January 2022 to December 2023 were enrolled, and a case-control study was conducted. According to the stage of disease progression, the subjects were divided into chronic hepatitis B group with 40 patients, compensated liver cirrhosis group with 32 patients, and hepatocellular carcinoma group with 30 patients, and 32 healthy volunteers matched by age and sex were enrolled as normal control group. The serum concentration of Mg²⁺ was measured; RT-qPCR was used to measure the mRNA expression level of MagT1 in CD8+ T cells and serum HBV DNA load; flow cytometry was used to measure the expression levels of programmed death-1 (PD-1), T-cell immunoglobulin and mucin-domain containing-3 (Tim-3), and natural killer cell group 2D (NKG2D) on the surface of CD8+ T cells. A one-way analysis of variance was used for comparison of normally distributed continuous data between multiple groups, and the SNK-q test was used for comparison between two groups; the chi-square test was used for comparison of categorical data between groups; a Pearson linear correlation analysis was used to investigate the correlation between the mRNA expression level of MagT1 and other related indicators. ResultsThere were significant differences in the serum level of Mg2+ and the mRNA expression level of MagT1 between the normal control group, the chronic hepatitis B group, the compensated liver cirrhosis group, and the hepatocellular carcinoma group (F=29.014 and 145.578, both P<0.001). The Pearson correlation analysis showed that the serum level of Mg2+ and the mRNA expression level of MagT1 were positively correlated with HBV DNA load (r=0.335 and 0.394, both P<0.05). The hepatocellular carcinoma group had the highest levels of PD-1 and Tim-3, followed by the compensated liver cirrhosis group, the chronic hepatitis B group, and the normal control group; the normal control group had the highest level of NKG2D, followed by the chronic hepatitis B group, the compensated liver cirrhosis group, and the hepatocellular carcinoma group (all P<0.001). The Pearson correlation analysis showed that the mRNA expression level of MagT1 was negatively correlated with PD-1 and Tim-3 (r=-0.643 and -0.640, both P<0.05) and was positively correlated with NKG2D (r=0.655, P<0.05). ConclusionThere is a reduction in the mRNA expression level of MagT1 in peripheral blood CD8+ T cells in patients with HBV infection, which is positively correlated with serum HBV DNA load. The reduction in the expression of MagT1 may contribute to CD8+ T cell exhaustion by impairing Mg²⁺ influx, manifesting as the upregulation of PD-1 and Tim-3 and the downregulation of NKG2D.

【Objective】 To study the concordance of identifying the presence or absence of KIR genes using flow reverse sequence-specific oligonucleotide probe (Flow-rSSO) hybridization and sequencing based typing-PCR (PCR-SBT) methods. 【Methods】 A total number of 131 cases of DNA samples from Han population were subjected to identify the presence or absence of all 16 KIR genes by Flow-rSSO method, and then sequenced at coding sequence for all 14 functional KIR genes using our in-house KIR PCR-SBT assay. The concordance of identifying the presence or absence of all functional KIR genes by Flow-rSSO and PCR-SBT was analyzed. Samples with inconsistent initial results were re-tested using the Flow-rSSO commercial kits with different Lot number, and further tested using the PCR-SSP commercial kit. 【Results】 The presence or absence of 14 functional KIR genes for 129 of 131 samples were completely in accordance via the PCR-SBT and Flow-rSSO methods. Two samples, one with 3DL1 negative, the other with both 2DS3 and 2DS5 negative initially-identified by Flow-rSSO, were actually all positive tested by PCR-SBT. Further retest by Flow-rSSO commercial kits with different Lot number and PCR-SSP commercial kit indicated that the two samples were all positive, which agreed well with PCR-SBT results. 【Conclusion】 In this paper, the initial test results of the presence or absence of KIR genes identified by Flow-rSSO for 2 samples were wrong, which indicated the importance of carrying out the quality control for reagents in KIR gene testing.

Objective:To investigate any anti-aging effect of repeated transcranial magnetic stimulation (rTMS) and explore the relationship between the effect and relief of clinical symptoms in patients with Parkinson′s disease (PD).Methods:A total of 108 PD patients were randomly divided into an rTMS group and a control group, each of 54, while another 54 healthy counterparts were selected to form a normal group. In addition to anti-PD drug therapy, the rTMS group was given daily rTMS treatment, 5 days a week for 4 weeks, while the control group received sham rTMS treatment, with no treatment of the normal group. Before the treatment and after 4 weeks of treatment as well as and 1 month after the ending of the treatment, the subjects′ clinical exercise symptoms were evaluated using the Unified Parkinson′s Disease Rating Scale (UPDRS), a timed exercise test and the 10m re-entry exercise test. Non-exercise symptoms were assessed using the Hamilton Depression Scale (HAMD), the Hamilton Anxiety Scale (HAMA) and the Mini-mental State Examination (MMSE). Fasting venous blood samples were analyzed to quantify the serum levels of tumor necrosis factor (TNF), interleukin-6 (IL-6), interleukin-1β (IL-1β) and matrix metalloproteinase-3 (MMP-3).Results:Four weeks and 1 month after the treatment, the average UPDRS scores, exercise test times and 10m re-entry exercise test results of the rTMS group were significantly better than those before treatment and significantly better than those of the control group at the same time point. The rTMS group′s average HAMA, HAMD and MMSE scores, as well as its average P300 latency and amplitude were also significantly better than those of the control group at the same time point and significantly better than those before treatment. After 4 weeks, the average MMP-3 content in the rTMS group was significantly lower than the control group′s average, and after a month the average levels of TNF, IL-6, IL-1β and MMP-3 of the rTMS group were all significantly different from those before treatment and those of the control group. The TNF, IL-6, IL-1β and MMP-3 levels were all positively correlated with the average UPDRS total score.Conclusion:High-frequency rTMS therapy can change the phenotypes related to cell senescence, and thus has good therapeutic effect on motor and non-motor symptoms of PD.

OBJECTIVE:To investigate the potential mechanism of Salvia miltiorrhiza in the treatment of postoperative abdominal adhesion (PAA). METHODS :Active components and target genes of S. miltiorrhiza were retrieved from TCMSP database,SwissADME database ,Perl database ,UniProt database and other databases. GeneCards ,OMIM and PubMed database were used to retrieve target genes related to PAA. Venn diagram was drawn by using mapping tool of bioinformatic online database so as to screen the intersecting targets of active component-PAA. STRING platform was adopted to establish target network related to active component-PAA and protein-protein interaction (PPI)network of intersecting targets ,etc.,and to screen hub genes. Gene ontology(GO)and Kyoto Encyclopedia of Genes and Genom es(KEGG)pathway enrichment were carried out by using R 3.6.1 software. Using the protein encoded by hub gene as receptor and tanshinone Ⅱ A as ligand ,the molecular docking was carried out with AutoDock 1.5.6 tool. RESULTS :A total of 38 active components of S. miltiorrhiza with high gastrointestinal absorption and their corresponding 72 targets,755 PAA-related target genes were identified. Results of Venn diagram showed that there were 33 intersecting targets of active components of chuqi90@163.com S. miltiorrhiza with PAA. Tanshinone ⅡA,dihydrotanshinolac- tone and other components may be important nodes of the target network related to active component-PAA. FOS,APP,ACHE, CASP3 and PTGS2 may be the hub genes in PPI network of intersecting targets. Results of GO enrichment showed that the intersecting targets were mainly concentrated in adrenergic receptor activity ,catecholamine binding ,G protein-coupled amine receptor activity and so on ;KEGG pathway enrichment analysis showed that the intersecting targets were mainly enriched in neuroactive ligand-receptor interaction ,cGMP-PKG signaling pathway ,endocrine resistance ,EGFR-tyrosine kinase inhibitor resistance and calcium signaling pathway.Molecular docking analysis showed that tanshinone ⅡA could form hydrogen bonds with many amino acid residues such as VAL- 580 of proto oncogenes c-Fos ,amyloid precursor protein ,acetylcholinesterase, caspase 3 and prostaglandin G/H synthase 2. CONCLUSIONS :The active components of S. miltiorrhiza play a role in the treatment of PAA by directly or indirectly acting on neuroactive ligand-receptor interaction ,cGMP-PKG signaling pathway ,endocrine resistance , EGFR-tyrosine kinase inhibitor resistance resistance and calcium signaling pathway.
