1.Synthesis, preclinical evaluation and pilot clinical study of a P2Y12 receptor targeting radiotracer 18FQTFT for imaging brain disorders by visualizing anti-inflammatory microglia.

As the brain's resident immune cells, microglia perform crucial functions such as phagocytosis, neuronal network maintenance, and injury restoration by adopting various phenotypes. Dynamic imaging of these phenotypes is essential for accessing brain diseases and therapeutic responses. Although numerous probes are available for imaging pro-inflammatory microglia, no PET tracers have been developed specifically to visualize anti-inflammatory microglia. In this study, we present an 18F-labeled PET tracer (QTFT) that targets the P2Y12, a receptor highly expressed on anti-inflammatory microglia. [18F]QTFT exhibited high binding affinity to the P2Y12 (14.43 nmol/L) and superior blood-brain barrier permeability compared to other candidates. Micro-PET imaging in IL-4-induced neuroinflammation models showed higher [18F]QTFT uptake in lesions compared to the contralateral normal brain tissues. Importantly, this specific uptake could be blocked by QTFT or a P2Y12 antagonist. Furthermore, [18F]QTFT visualized brain lesions in mouse models of epilepsy, glioma, and aging by targeting the aberrantly expressed P2Y12 in anti-inflammatory microglia. In a pilot clinical study, [18F]QTFT successfully located epileptic foci, showing enhanced radioactive signals in a patient with epilepsy. Collectively, these studies suggest that [18F]QTFT could serve as a valuable diagnostic tool for imaging various brain disorders by targeting P2Y12 overexpressed in anti-inflammatory microglia.
2.Clinical effect of tibial periosteal lateral distraction combined with compound Ziyin ointment in the treatment of diabetic foot

Objective:To investigate the clinical effect of tibial periosteal lateral distraction combined with compound Ziyin ointment in the treatment of diabetic foot.Methods:A total of 62 patients with diabetic foot treated in the First People′s Hospital of Wuhu from July 2022 to September 2023 were prospectively selected and divided into routine group and combination group according to random number table method, with 31 cases in each group. The conventional group was treated with tibial periosteal lateral stretch, and the combined group was treated with tibial periosteal lateral distraction combined with compound Ziyin ointment, and the treatment lasted for 2 weeks. Traditional Chinese Medicine (TCM) syndrome scores, serum inflammatory factors, growth factors, foot temperature, toe oxygen saturation, pain and quality of life scores, clinical efficacy and complications were observed before and after treatment in the two groups.Results:After treatment, TCM syndrome score, serum inflammatory factor level and pain score were lower than before treatment, serum growth factor level, life quality score, foot temperature and toe end blood oxygen saturation were higher than before treatment (all P<0.05). After treatment, the pain score, TCM main disease score, TCM secondary disease score and inflammatory factor levels in the combined group were lower than those in the conventional group (all P<0.05), and the serum growth factor level, life quality score, foot temperature and toe end blood oxygen saturation were higher than those in the conventional group (all P<0.05). The total effective rate of combined group was higher than that of conventional group [93.55%(29/31) vs 70.97%(22/31), P<0.05]. There was no significant difference in the total incidence of adverse reactions between the two groups ( P>0.05). Conclusions:Tibial periosteal lateral distraction combined with compound Ziyin ointment can improve the levels of serum inflammatory factors, growth factors, foot temperature and toe oxygen saturation, reduce TCM syndrome score and relieve pain degree, improve patients′ quality of life and clinical efficacy, and have high safety.
3.Multi-source COVID-19 surveillance data in Fujian Province and implications for epidemic prevention and control

To analyze the epidemiological characteristics of COVID-19 in Fujian Province from the 49th week of 2022 to the 5th week of 2023,after further optimization of China's COVID-19 prevention and control measures on December 7,2022(the 49th week of 2022),this study used multi-dimensional surveillance data to dynamically assess population infection levels and their changing trends.The aim of the study was to provide a scientific basis for early warning of epidemic risk,medical resource allocation,and evalu-ation of socio-economic impact.A multi-source data surveillance system was constructed,encompassing surveillance of fever clinics at medical institutions(weekly collection of visits,positive nucleic acid and antigen test results,inpatients,and severe cases in sec-ondary or above hospitals),population nucleic acid test monitoring(weekly person-times and positivity rates of single-tube tests from the provincial system),sentinel hospital monitoring(weekly proportion of influenza-like illness visits at 18 sentinel hospitals and re-lated viral testing data),and monitoring of novel coronavirus variants(weekly systematic collection of genomic sequences of local and imported cases).Line charts were plotted weekly,and time series analysis,molecular epidemiological methods,and an improved SEIAR model were used to simulate epidemic spread.During the study period,the COVID-19 epidemic in Fujian Province exhibited three distinct stages.In the infection peak stage(52nd week of 2022),the provincial fever clinic visits reached 606 893 person-times,and a 49.2%positivity rate in population single-tube nucleic acid tests and 63.8%positivity rate in sentinel hospital monitoring were observed.In the medical load peak stage(2nd week of 2023),274 460 inpatients and 28 487 severe cases were recorded.In the epidemic decline stage(4th to 5th weeks of 2023),fever clinic visits decreased by 96.3%with respect to the peak,the single-tube nucleic acid test positivity rate decreased to 6.3%,and the sentinel hospital COVID-19 nucleic acid test positivity rate was 6.4%.All 508 sequenced local cases were Omicron variants,predominantly BA.5.2 and its sub-lineages(67.4%).Among 56 imported se-quenced cases,BA.5.2 and its sub-lineages accounted for 50.0%,and 16.1%comprised nine variants of interest including XBB and BQ.The model predicted the infection peak in the 52nd week of 2022,whereas the hospitalization peak lagged by approximately 10.6 days.Multi-source data monitoring revealed a three-stage development of the COVID-19 epidemic in Fujian.The BA.5.2 strain was dominant during the epidemic.The combination of multi-source monitoring data and modeling provides important references for epi-demic prevention and control,and highlights the need to improve the monitoring system in follow-up.

The OPT101 photoelectric sensor has the characteristics of good photoelectric response,buih-in transimpedance amplifier and small size,which meet the needs of accurately near-infrared spectral measurement in medical near-infrared spectroscopy (NIRS) range.Our research team has developed a series of portable NIRS instruments for NIRS measuring in intensive care units (ICU).The characteristics and advantages of OPT101 in the development of clinical ICU-specific NIRS instruments were introduced.The research progress of our team on the development of ICU-NIRS instruments using OPT101 was reviewed.The prospect of OPT101 in clinical noninvasive detection was discussed.
7.Extraction and purification of Jo-1 antigen

Objective To improve the purifying method of Jo-1 antigen from rabbit thymus used for detection of anti-Jo-1 antibody by dot-blotting immunoassay(DB).Methods The rabbit thymus glands were cut into pieces,homogenized and extracted by PBS.Total protein was precipitated by acetone to get acetone powder(RTAP).The RTAP was solved in PBS and separated by an by anti-Jo-1 IgG affinity column.Results 5~7 g RTAP was obtained from 100g rabbit thymus glands.There was 19%~24% of protein in RTAP.Jo-1 antigen was enriched around 1900 folds through affinity chromatography,with 2.5% recovery of antigenic activity.In this preparation,there were several bands on SDS-PAGE,but only one band about 50 ku,reacted with anti-Jo-1 antisera on immunoblotting.Dot-blotting also showed that the antigen only reacted with Jo-1 antisera.The purified Jo-1 antigen was not stable for long time,but the antigenic activity could maintain for a long time when there was MgCl2 in the solution.Conclusion Affinity chromatography was a simple and easy method for purifying Jo-1 antigen from rabbit thymus.The antigen purified by affinity chromatography could meet the requirement for detecting Jo-1 antibody bydot-blotting.
8.Quality control of autoantibodies detection with analysis

Objective To make an investigation for autoantibodies detection in China. Method Forty-eight hospitals were included by mail or Email. The items of QC include ANA, anti-ENA antibody, anti-dsDNA antibody, anti-mitochondria antibody and anti-smooth muscle antibody. The distribution of samples and analysis of the testing results was double-blinded. Result The correct rate of ANA, anti-ENA antibody, anti-dsDNA antibody, anti-mitochondria antibody and anti-smooth muscle antibody were 47%, 70%, 46%, 95%and 40% respectively. Conclusion The overall results are not optimistic. The quality of detecting kits is not all good, neither level of technicians. This condition should be changed by selecting better methods and improving understanding.
9.Preparation of Mi-2 antigen and significance of detecting anti-Mi-2 antibody in autoimmune connective tissue diseases

Objective To prepare Mi 2 antigen,detect anti Mi 2 antibody and investigate its significance in autoimmune connective tissue diseases (CTD).Methods Mi 2 antigen was prepared from rabbit thymus acetone powder and purified by DE52 chromatography.Anti Mi 2 antibodies were tested in 40 normal controls and 315 patients with CTD by double immunodiffusion.The distribution of anti Mi 2 antibodies in CTD was analysed.The clinical characteristics of dermatomyositis between anti Mi 2 positive group and negative group were compared.Results The precipitation line developed between the antigen and the standard anti Mi 2 serum.In DE52 chromatography,Mi 2 antigen was found in elute with 0 1 mmol/L and 0 2 mmol/L NaCl.The positive rate of anti Mi 2 antibodies was 26 1%(12/46) in dermatomyositis and 4 5%(2/44) in polymyositis respectively.There were no positive cases in 50 patients with rheumatoid arthritis,30 patients with primary Sjgren syndrome (SS),60 patients with systemic lupus erythematosus,50 patients with systemic sclerosis,35 patients with other CTD and 40 normal controls.The specificity of anti Mi 2 antibody in dermatomyositis was 99 4%.In dermatomyositis,the patients with positive anti Mi 2 antibody usually presented skin lesions at beginning,and most patients had V sign and/or shawl sign during the course of disease.On the other hand,the antibody negative ones manifested muscle involvements initially and easily got fever during the course of disease.There were statistic differences between the two groups in above features ( P
