Objective:To investigate serological characteristics of Rhnull with anti-Rh29 and its rare gene sequence. Methods:Microcolumn gel method was used for ABO and RhCcDEe blood typing, accidental antibody identification and cross matching.RHD/RHCE genotype and sequencing analysis were performed by PCR-SSP method.RHAG gene was sequenced by fluorescence PCR method.Antigen or protein associated with Rh complex was detected by flow cytometry. Results:Blood group of the patient was A,Rh pheno-type was typed as group C-c-D-E-e,presented with symptoms of anemia, proportion of globular and oral erythrocytes in blood smear was 6.0%and 0.8%,respectively,RH genotype was CCDee,and immune anti-Rh29 high-frequency antigen were developed.RHAG gene sequencing revealed exons 8,9 and 10 deletion. Conclusion:Mechanism of"regulator"type Rhnull generation in this study is caused by deletion of exons 8,9 and 10 in RHAG gene.

Journal of Shenyang Medical College 2025;27(5):467-476
Objective:To investigate the expression and biological function of AHNAK nucleoprotein 2(AHNAK2)in thyroid cancer. Methods:The expression of AHNAK2 in thyroid cancer tissues was analyzed using The Cancer Genome Atlas(TCGA),Gene Expression Omnibus(GEO),and Human Protein Atlas(HPA)databases. The correlation between AHNAK2 expression and patients'prognosis, clinicopathological characteristics, immune cell infiltration, and subcellular localization was evaluated.AHNAK2 mRNA expression in thyroid cancer cells was detected by RT-qPCR.A stable AHNAK2 knockdown TPC-1 cell line was established via lentiviral transfection, and the transfection efficiency was validated using RT-qPCR and Western blot.TPC-1 cells were divided into the sh-NC and sh-AHNAK2 groups. The impact of AHNAK2 knockdown on TPC-1 cell proliferation, migration, and invasion was assessed using colony formation assay,CCK-8 assay, wound healing assay, and Transwell assay.Results:AHNAK2 was up-regulated in thyroid cancer tissues and was associated with poor prognosis. It was primarily localized in the cytoplasm and cytoplasmic membrane.GO analysis revealed that AHNAK2 was mainly enriched in biological processes such as organelle disassembly, cellular components such as the spindle, and molecular functions such as microtubule binding.KEGG pathway analysis indicated that AHNAK2 was primarily enriched in the cell cycle pathway.Immune infiltration analysis suggested that AHNAK2 expression was positively correlated with macrophage infiltration and negatively correlated with plasmacytoid dendritic cell infiltration in thyroid cancer.AHNAK2 was overexpressed in multiple thyroid cancer cell lines. In vitro experiments demonstrated that AHNAK2 knockdown inhibited the proliferation, migration, and invasion of TPC-1 cells.Conclusion:AHNAK2 may serve as an important prognostic biomarker and immunotherapy target for thyroid cancer,as it can promote the proliferation, migration and invasion of thyroid cancer cells.

Bacteroides uniformis(B.uniformis)is an obligate anaerobic Gram-negative bacteria,it belongs to Bacieroides spp..It is an important component of the normal gut microbiota in human body and rarely causes op-portunistic infection in clinical practice.Currently, there is no consensus on the clinical characteristics, diagnosis, treatment, and prognosis of infection caused by B.uniformis, and the resistance of anaerobic bacteria is difficult to be detected. This article reports a case of a lung tumor patient who developed diarrhea and bloodstream infection due to B.uniformis after chemotherapy. After empirical anti-infective treatment with cefoperazone/sulbactam,the pa-tient recovered and was discharged from hospital. This paper aims to improve clinical understanding on infection caused by opportunistic pathogens in the gut,and provide reference for timely diagnosis and effective treatment.

Objective To investigate the key targets and pathways of Juhongtai formula granules in improving acute lung injury(ALI).Methods Juhongtai formula granules and their drug-containing serum components were analyzed by UPLC-QTOF-MS/MS,and the active ingredients were screened based on the"Lipinski Rule of Five".The action targets of the above active ingredients were obtained through the TCMSP,Swiss Target Prediction and SuperPred databases, and the ALI-related targets were obtained from the GeneCards,OMIM,PharmGKB,CTD databases and GEO chip. The target protein-protein interaction network was constructed using the String database and Cytoscape to analyze the key targets. The DAVID database was used for GO functional annotation and KEGG pathway enrichment analysis, and the CB-dock2 platform was used for molecular docking verification. After one week of adaptive feeding, the experimental SD rats were randomly divided into the blank group, the model group, the positive control group, the negative control group and the Juhongtai formula granules group, with 6 rats in each group. The rat model of ALI was replicated by tracheal infusion of lipopolysaccharide. The pathological morphology of lung tissues in each group of rats was observed by HE staining, and the mRNA expressions of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-1 β(IL-1β)and nitric oxide were detected by reverse transcription quantitative polymerase chain reaction(RT-qPCR).Results In total,44 components of the Juhongtai formula granules were identified, including 26 drug-containing serum components, and 21 key compounds were screened.A total of 22 intersection targets were obtained. The pathway enrichment results indicated the action of these targets on the advanced glycation end product receptor(AGE-RAGE)and TNF signaling pathways. The docking results showed that limonin, kaempferol and naringenin could be matched with prostaglandin peroxidase 2,nitric oxide synthase 2 and TNF.Animal experiments confirmed that the Juhongtai formula granules can alleviate inflammatory infiltration in lung tissue and reduce the expressions of TNF-α,IL6,IL-1β mRNA and nitric oxide. Conclusion Juhongtai formula granules can inhibit ALI-related inflammatory targets through the synergistic effects of multiple components such as limonin and kaempferol, regulate signaling pathways such as AGE-RAGE and TNF,reduce the production of inflammatory factors and nitric oxide, and improve ALI.

Objective:To explore the mediating role of self-perceived aging in the relationship between perceived social support and self-efficacy for exercise in aged patients undergoing surgery for lung cancer and provide a basis for formulating relevant clinical intervention strategies. Methods:Totally 220 aged lung cancer patients who underwent surgery and were hospitalized at the Cancer Hospital, Chinese Academy of Medical Sciences and Peking Union Medical College, from April to June 2024 were recruited by convenience sampling. Patients completed a demographic questionnaire, the Self-Efficacy for Exercise (SEE) , the Ageing Perceptions Questionnaire (B-APQ) , and the Perceived Social Support Scale (PSSS) . The mediation effect of self-perceived aging between perceived social support and self-efficacy for exercise was analyzed using Model 4 in the SPSS Process macro. Results:A total of 220 questionnaires were distributed, and 207 valid questionnaires were returned, with an effective recovery rate of 94.09%. The total score for B-APQ was (46.93±11.35) , the SEE score was (44.12±12.83) , and the PSSS score was (53.18±13.53) . Perceived social support had a direct effect on self-efficacy for exercise (effect value: 0.278; P<0.01) . Self-perceived aging played a partial mediating role between perceived social support and self-efficacy for exercise, with a mediation effect value of 0.138 ( P<0.01) , accounting for 33.17% of the total effect (0.138/0.416) . Conclusions:Perceived social support directly influences self-efficacy for exercise in aged patients undergoing surgery for lung cancer and also exerts an indirect effect through self-perceived aging. It is recommended that healthcare providers guide these patients to establish a positive perception of aging while focusing on enhancing their social support levels to improve their self-efficacy for exercise and promote recovery.
