1.Exploring mechanism of Lycium barbarum polysaccharides in preventing inflam-matory bowel disease in chicks based on network pharmacology

This study aims to explore protective effects of Lycium barbarum polysaccharides(LBP)on intestinal damage caused by lipopolysaccharide(LPS)-induced inflammatory bowel disease(IBD)in chicks.Network pharmacology was initially employed to determine the target proteins of wolfberry in the prevention and treatment of IBD.Following this,protein-protein interaction analy-sis,GO and KEGG pathway enrichment analysis,and molecular docking studies were conducted.Subsequently,an animal study was conducted:a total of 100 one-day-old male Hy-line brown lay-ing hens were randomly divided into five groups:a blank control group(CON),an LPS treatment group(LPS),a low-dose LBP group(LPS+LBP 0.25 g/L,L-LBP),a medium-dose LBP group(LPS+LBP 0.5 g/L,M-LBP),and a high-dose LBP group(LPS+LBP 1 g/L,H-LBP).Upon reac-hing 21 days old,duodenal,jejunal,ileal,and cecal tissues were collected to determine SOD and GSH-Px levels.Furthermore,the mRNA expression levels of TNF-α,AKT1,IL-6,IL-1β and TP53 in the intestinal tissues were measured using quantitative real-time PCR.The results demonstrated that network pharmacology identified 45 active ingredients in wolfberry that target 116 key protein sites,including TNF,AKT1 and IL6.The primary objectives focus on signaling pathways including AGE-RAGE,IL-17,TNF,HIF-1,and NF-κB.Molecular docking showed excellent ligand-receptor docking scores,with stable binding facilitated by hydrogen bonds and hydrophobic interactions.Compared to the LPS group,the 0.5 g/L LBP exhibited notably higher levels of SOD and T-AOC.In comparison with the LPS group,the medium and high-dose LBP experimental groups showed notably decreased the mRNA expressions of TNF-α,AKT1,IL-6,and IL-1β,while TP53 mRNA expression was significantly upregulated(P<0.01).In summary,wolfberry exerts preventive and therapeutic effects on IBD through a multi-component,multi-target,and multi-pathway mecha-nism.
2.Risk factors for heterotopic ossification in patients with proximal humerus fracture after shoulder arthroplasty

Objective:To investigate the risk factors for heterotopic ossification (HO) after shoulder arthroplasty for patients with proximal humerus fracture.Methods:A retrospective study was conducted to analyze the clinical data of 95 patients with proximal humeral fracture who had undergone shoulder arthroplasty at Department of Sports Medicine, Beijing Jishuitan Hospital between January 2002 and December 2020. The cohort included 69 females and 26 males with an age of (66.8±11.8) years. By the Neer classification, 39 patients had three-part fractures, and 56 ones four-part fractures. According to the postoperative occurrence of HO, the patients were divided into an HO group and an HO-free group. Univariate analysis was performed for factors such as age, gender, time from injury to surgery, injury mechanism, involvement of the dominant side, surgical approach, and the Shoulder and Elbow Surgeons (ASES) score, visual analog scale (VAS) for pain, and shoulder range of motion at the final follow-up in the 2 groups. After positive indicators ( P<0.05) were screened through univariate analysis, they were included in a multivariate logistic regression model to analyze the risk factors for HO following shoulder arthroplasty. Results:All patients were followed up for (83.7±39.8) months. At the final follow-up, the incidence of HO was 34.7% (33/95). There were no statistically significant differences between the 2 groups in gender, time from injury to surgery, injury mechanism, surgical approach, or ASES score, VAS pain score, or shoulder range of motion at the final follow-up ( P>0.05). The univariate analysis showed that the age was significantly younger ( P=0.009), and the proportion of the dominant side involvement significantly lower ( P=0.016) in the HO patients. Multivariate logistic regression analysis showed that a younger age ( OR=0.947, 95% CI: 0.910 to 0.985, P=0.007) was an independent risk factor for HO in patients with proximal humerus fracture after shoulder arthroplasty. Conclusion:A younger age is the risk factor for HO after shoulder arthroplasty for patients with proximal humerus fracture.
3.Protective effect of lycium barbarum polysaccharide on testicular injury induced by perfluorooctanoic acid in offspring mice

To examine how Lycium barbarum polysaccharide(LBP)impacts reproductive harm caused by perfluorooctanoic acid(PFOA)in male offspring mice.Sixty expectant female rats were randomly allocated into 6 groups,with each group consisting of 10 rats.From 1 to 17 d of gesta-tion,the blank control group was given 0.2 mL deionized water;The PFOA group was given 3.5 mg/kg PFOA;the LBP intervention group were given different doses(40,80,120 mg/kg)of LBP by gavage based on the PFOA group;the control group was given 80mg/kg.At 21 d of age,the offspring mice were weaned,and the offspring male mice were fed normally until 35 d of age,and testicular tissue was taken.HE staining was used to observe the pathological changes of testic-ular tissue sections.Body mass and testicular index were measured.The levels of SOD,MDA,LDH,SHD,GSH-PX and AR in testicular tissue were determined.Serum levels of LH,FSH,Gn-RH and T were determined by ELISA.Differential genes and differential metabolites of testis were analyzed in combination with transcriptome and metabolome.Real-time fluorescence quantitative PCR was used to detect the expression of key genes in fatty acid metabolic pathway.The results showed that LBP could alleviate PFOA-induced decline in body mass and increase testicular index(P<0.01).LBP could alleviate the necrosis,edema of testicular cells caused by PFOA.LBP can in-crease the contents of SOD,GSH-Px,LDH,SDH and AR in testicular tissue of offspring male mice induced by PFOA,increase the contents of FSH,LH,GnRH and T in serum of offspring male mice,and decrease the content of MDA.Analysis of the transcriptome revealed that 1 388 genes exhibited differential expression between the groups exposed to PFOA and 80 mg/kg LBP,with 696 genes being up-regulated and 692 genes being down-regulated.The differentially ex-pressed genes were predominantly involved in pathways such as fatty acid metabolism,unsaturated fatty acid biosynthesis,cholesterol metabolism,the PPAR signaling pathway,and protein digestion and absorption,with fatty acid metabolism being the most prominent among them.Examination of the metabolome revealed 34 metabolites that were expressed differently between the PFOA group and the 80 mg/kg LBP group.Key metabolic pathways such as fatty acid biosynthesis,unsaturated fatty acid biosynthesis,and steroid hormone biosynthesis were found to be significant.In a joint transcriptome and metabolome analysis,the Differentially expressed genes(DEGs)and Differential expression of metabolites(DEMs)are highly correlated in signaling pathways such as fatty acid bi-osynthesis and unsaturated fatty acid biosynthesis.This study shows that PFOA can cause testicu-lar injury in offspring male mice,and LBP can effectively alleviate the testicular injury caused by PFOA in offspring male mice.Meanwhile,transcriptomic metabolism shows that LBP can effec-tively improve the testicular injury caused by PFOA in offspring male mice through fatty acid bio-synthesis and unsaturated fatty acid biosynthesis signaling pathways.Among them,80 mg/kg LBP has better effect.

7.Effect of soft brace wearing on plantar dynamics in patients with chronic ankle instability

Objective To investigate the effect of daily soft brace wearing on plantar dynamics during walking in patients with chronic ankle instability(CAI). Methods A total of 52 patients with unilateral chronic ankle instability(CAI)in Beijing Rehabilitation Hospital from February,2021 to January,2023 were randomly divided into control group(n=26)and experimental group(n=26).Both groups underwent an eight-week exercise training program.The control group wore placebo brace dur-ing daily activities,while the experimental group wore soft ankle brace.Plantar dynamic parameters were mea-sured using a pressure plate system during walking,including peak plantar pressure and plantar impulse before and after intervention. Results Six participants dropped out in the control group and five in the experimental group,resulting in a final inclu-sion of 41 participants.After intervention,there was no significant difference in peak plantar pressure and im-pulse on the affected side in the control group among different areas(P>0.05).In the experimental group,the peak pressure and impulse in the heel medial,heel lateral and forefoot medial areas increased(|t|>4.192,P<0.001),while the peak pressure and impulse in the midfoot and lateral forefoot areas decreased(t>2.984,P<0.05);the peak pressure and impulse in the heel medial,heel lateral and forefoot medial areas were higher in the experimental group than in the control group(|t|>2.126,P<0.05),and the peak pressure and impulse were low-er in the midfoot and forefoot lateral areas(t>2.133,P<0.05). Conclusion Wearing a soft brace during daily activities may optimize the distribution of peak plantar pressure and plan-tar impulse on the affected side in patients with CAI,which may prevent recurrence of sprains.
8.Supplementing biofeedback training with kinesiotaping better promotes the recovery of knee function after anterior cruciate ligament reconstruction

Objective:To document any effect of combining kinesiotaping (KT) with Monitored Rehab Systems biofeedback training (MRS training) on the recovery of knee function after anterior cruciate ligament reconstruction (ACLR).Methods:Twenty-two patients after ACLR were randomly divided into an MRS group and a combined group, each of 11. In addition to conventional rehabilitation training, the MRS group had placebo taping in place during their MRS training, while the combined group had KT applied. Each training session lasted 45 minutes, with three sessions per week. Before the experiment and after 8 weeks of treatment, the affected foot support phase, impulse of each area of the affected foot′s sole, and lower limb reaction time were measured.Results:Significant improvement in the affected foot′s support phase and lower limb reaction time was documented in both groups, as well as in the average impulse percentage of the affected foot′s plantar forefoot area (F area), mid-foot area (MF area), and heel area (H area). All were significantly better in the combined group than in the MRS group, on average.Conclusion:Supplementing MRS biofeedback training with kinesiotaping can more effectively improve the knee joint functioning and postural control of patients after ACLR surgery. The combination is worthy of clinical promotion and application.
9.Early clinical follow-up after arthroscopy-assisted anatomic coracoclavicular ligament reconstruction using tendon allograft for acute high-grade acromioclavicular joint dislocations

Objective To explore the clinical results after arthroscopy-assisted anatomic coracoclavicu-lar ligament reconstruction(AA-ACCR)using tendon allograft for acute high-grade acromioclavicular joint dislocations.Methods Patients undergoing primary AA-ACCR with tendon allograft for Rockwood type Ⅲ/Ⅴ acromioclavicular joint dislocations between January 2015 and January 2017 were analyzed retrospectively.The coracoclavicular distance(CCD)was obtained on radiographs pre-and post-opera-tively,as well as 6 weeks after the operation and during the final follow-up,and complications were recorded.Moreover,before the operation and during the final follow-up,all patients were evaluated shoulder pain and function using the American Shoulder and Elbow Surgeon score(ASES)and Visual Analogue Scale(VAS).Results In total,21 patients were included for evaluation with a minimum of 6-month follow-up(range,6-18 months).According to preoperative X ray,8 and 13 patients suffered from Rockwood type Ⅲ and V acromioclavicular dislocation,respectively.CCD decreased significantly six weeks postoperatively(8.6±2.6 mm)and at the final follow-up(11.7±3.5 mm)compared with the preoperative value(19.7±4.5 mm,P<0.01).However,CCD at the latter time points was signifi-cantly higher than right after the operation(7.9±2.5 mm,P<0.01).Furthermore,CCD increased sig-nificantly at the final follow-up compared with six weeks postoperatively(P<0.01).At the final follow-up,significant improvement was observed in the average VAS and ASES scores(P<0.01).Moreover,at the final follow-up,10 out of 21 patients(48%)experienced a loss of reduction,including 3 fail-ures in operation and 7 in later restoration.All patients had both medial and lateral clavicular tunnel widening.However,no fracture of the coracoid/clavicle or other complications were recorded.Conclu-sion AA-ACCR with tendon allograft for acute high-grade acromioclavicular joint dislocations contrib-utes to significant improvement in clinical and radiographic outcomes in the early follow-up.Although the loss of reduction rate is relatively high(48%),it is a safe method without obvious complications.
