1.Protective effect of myrislignan on autoimmune hepatitis in mice

Objective:To investigate the protective effect of myrislignan(MRL)on concanavalin A(Con A)-induced autoimmune hepatitis(AIH).Methods:C57BL/6J mice were divided into the following groups using a random number table,with five mice in each group:control group,MRL group,model group(Con A group),and MRL pretreatment group(MRL+Con A group). MRL was injected intraperitoneally at a dose of 30 μg/g;3 h after pretreatment,Con A(18 μg/g)was administrated by intravenous injection;mouse livers and serum samples were collected 12 h after injection for measuring serum transaminase levels and liver cell apoptosis. The mRNA and protein expression levels of IL-6,IL-12,and TNF-α were measured using qRT-PCR and ELISA. Flow cytometry was performed to detect the proportion and activation status of macrophages in liver tissues. Bone marrow-derived macrophages(BMDMs)were isolated and induced in vitro to analyze the regulatory effect of MRL on macrophages. One-way analysis of variance was used to compare the differences in various indicators among groups. Results:Compared with the Con A group,MRL(30 μg/g)pretreatment significantly reduced alanine aminotransferase( P<0.05)and aspartate transaminase( P<0.01)levels,attenuated liver oxidative stress(increased superoxide dismutase activity,while decreased levels of malondialdehyde and myeloperoxidase;all P<0.05),and suppressed hepatocyte apoptosis( P<0.01). Both in vivo and in vitro experiments confirmed that MRL(30 μg/g)could reduce the proportion of M1 macrophages( in vivo: P<0.05; in vitro:all P<0.001)and inhibit macrophage activation( in vivo: P<0.01; in vitro:all P<0.05). Conclusion:MRL effectively prevents Con A-induced autoimmune hepatitis by inhibiting liver cell apoptosis,attenuating liver oxidative stress,suppressing M1 macrophage polarization,and reducing inflammatory cytokine expression.

3.Evaluation of efficacy of two medical cleaner on the pre-cleaning dental zinc phosphate cement filler

Objective To compare the efficacy of alkaline enzyme-containing versus multi-enzyme cleaner for pre-cleaning dental zinc phosphate cement fillers.Methods One hundred new dental cement fillers were selected and contaminated to moderate-to-heavy levels by simulating clinical contamination,and divided into the alkaline enzyme-containing group and the multi-enzyme group,with 50 cases in each group.The two groups underwent manual brushing followed by automated spray-type washer-disinfector cleaning.The effica-cy of the two groups of pre-cleaning were evaluated by naked-eye visual detection,10×illuminated magnifica-tion and residual protein quantification.Results The alkaline enzyme-containing group demonstrated signifi-cantly higher pass rates than the multi-enzyme group by naked-eye visual detection and 10×illuminated mag-nification(P<0.05),while there was no statistically significant difference in the residual protein quantifica-tion between the two groups(P>0.05).The qualified cleaning rates of the three parts of the instruments in the alkaline enzyme-containing group were all higher than those in the multi-enzyme group,but only the quali-fied cleaning rate of the handle threaded surface was statistically significant(P<0.05).There was no statisti-cally significant difference in the cleaning qualification rates of the instruments three parts of the alkaline en-zyme-containing group(P>0.05),while there was statistically significant difference in the cleaning qualifica-tion rates of the instruments three parts of the multi-enzyme group(P<0.05).Conclusion Compared with the multi-enzyme cleaner,alkaline enzyme-containing cleaner has better effects in the pre-cleaning of zinc phosphate cement fillers.

6.MiR-183-5p Promotes Proliferation, Metastasis and Angiogenesis in Breast Cancer Cells through Negatively Regulating Four and a Half LIM Protein 1

Purpose:
Four and a half LIM protein 1 (FHL1) is involved in breast cancer (BC) development, but the regulatory mechanism involved remain unclear. In the present study, we examined the role of FHL1 in BC development.
Methods:
The expression of FHL1, miR-183-5p, and miR-96-5p in BC tissues was analyzed using StarBase analysis. FHL1 expression in BC tissues, a normal human breast epithelial cell line, and BC cell lines was detected using quantitative reverse transcription polymerase chain reaction (qRT-PCR). The relationship between FHL1 and miR-183-5p/miR-96-5p was analyzed via Pearson's rank correlation, TargetScan, and a dual-luciferase reporter assay. BT549 and MDA-MB-231 cells were transfected with either FHL1 and miR-183-5p mimics, or siFHL1 and a miR-183-5p inhibitor, respectively. The viability, colony number, migration, invasion, and tube length of BT549 and MDA-MB-231 cells were examined using cell counting kit-8, colony formation, wound-healing, Transwell, and tube formation assays, respectively. The levels of FHL1, vascular endothelial growth factor (VEGF), p53, E-cadherin, N-cadherin, and vimentin were quantified using western blotting and qRT-PCR.
Results:
FHL1 expression was downregulated in BC tissues and cells, whereas miR-183-5p and miR-96-5p were upregulated in BC tissues (negative correlation with FHL1 expression).FHL1 overexpression inhibited the viability, colony number, migration, and invasion of BC cells and the expression of VEGF, N-cadherin, and vimentin, and increased the expression of FHL1, p53, and E-cadherin in BT549 cells. Furthermore, a miR-183-5p mimic reversed these effects of FHL1 overexpression, whereas FHL1 silencing caused opposite results to those observed in MDA-MB-231 cells; however, this was reversed by a miR-183-5p inhibitor.
Conclusion
Our study suggests that miR-183-5p promotes cell proliferation, metastasis, and angiogenesis by negatively regulating FHL1 in BC.
8.Efifcacy of Cetuximab in Combination with Chemotherapy in Advanced Non-small Cell Lung Cancer

Background and objective Cetuximab is a monoclonal antibody directed against epidermal growth fac-tor receptor. Emerging evidence showed improved effcacy with the addition of cetuximab to chemotherapy in advanced non-small cell lung cancer (NSCLC), but the data in oriental population are limited. hTe aim of this study is to investigate the eff-cacy of cetuximab in combination with chemotherapy in Chinese patients with advanced NSCLC.Methods NSCLC patients receiving cetuximab in combination with chemotherapy in Cancer Hospital, Chinese Academy of Medical Sciences & Peking Union Medical College were enrolled and retrospectively analyzed. Clinical characteristic, effcacy, outcome and toxicity data were analyzed.Results A total of 40 patients were enrolled into this study in which 29 were male, 36 with adenocarcinoma. In the 23 patients who had received palliative chemotherapy previously (with a median of 2 prior chemotherapy regimens), the median progression-free survival (PFS) atfer the last prior chemotherapy regimen was 2.3 months. For the overall population, 13 (32.5%) patients achieved partial response atfer cetuximab in combination with chemotherapy. Response rate were 52.9% (9/17) and 17.4% (4/23) in chemotherapy-naive patients and chemotherapy-treated patients, respectively (P=0.018). hTe median PFS was 4.8 months for the overall population. In chemotherapy-naive patients and chemotherapy-treated patients, the median PFS was 8.4 months and 4.1 months, respectively (P=0.062). hTe estimated median overall survival was 17.1 months. Toxicities were generally manageable and no treatment-related deaths occurred.Conclusion Cetuximab in addition to che-motherapy appears to be associated with promising effcacy and acceptable toxicity proifle in Chinese patients with advanced NSCLC. Further validation is needed.
9.The significance of genetic expression of IGF-Ⅰ , ERK, and GLUT4 in adipose tissue of patients with metabolic syndrome and colorectal cancer and its significance

[Summary] To explore the mRNA expression of insulin-like growth factor-Ⅰ (IGF-Ⅰ),extracellular signal regulated kinase (ERK),and glucose transporter 4 (GLUT4) in greater omental adipose tissue of patients with metabolic syndrome and colorectal cancer.The mRNA expression of IGF-Ⅰ,ERK,and GLUT4 in greater omental adipose tissue of the subjects was measured by RT-PCR.(1) The mRNA expression level of IGF-Ⅰ and ERK in the metabolic syndrome group was significantly higher than that in the control group (P<0.01),while in colorectal cancer subgroup the expression was significantly higher than that in the non-colorectal cancer subgroup (P< 0.01).The expression of GLUT4 was obviously lowered (P<0.01).(2) The expression of ERK was positively correlated with that of IGF-Ⅰ (r =0.608,P<0.01).The fasting insulin was positively correlated with the expression of ERK and IGF-Ⅰ(r =0.538,0.439,P < 0.01),and negatively with that of GLUT4 (r =-0.457,P < 0.01).There may be relationship between ERK plus IGF-Ⅰ and metabolic syndrome complicated with colorectal cancer.The lowered GLUT4 expression may be related to insulin resistance in metabolic syndrome.
10.Effect of collaborative health education by both doctors and nurses on parturition

Objective To explore the effect of collaborative health education by both doctors and nurses on parturition?Methods One hundred and sixty primiparas at uterogestation undergoing analgesic parturition were assigned equally into the control and observation groups according to their admission sequence? The control group received routine nursing intervention and the observation group the collaborative health education by both doctors and nurses? The two groups were compared in terms of analgesic effect,vaginal delivery and post-partum hemorrhage? Result The rates of analgesic delivery and vaginal delivery in the observation group were significantly higher and the volume of postpartum hemorrhage was significantly smaller than those of the control group(all P < 0?05)?Conclusion The collaborative health education may be effective in alleviating mental stress of the preimiparas,enhancing the analgesic effect,increasing the rate of vaginal delivery and decreasing the volume of postpartum hemorrhage?
