1.Effects of imperatorin on malignant biological behavior of gastric cancer cells by regulating ThPOK expression

OBJECTIVE To investigate the effects of imperatorin (IMP-SD) on malignant biological behavior of gastric cancer (GC) cells by regulating zinc finger and BTB domain 7B (ThPOK). METHODS Human GC cells MKN-7 were used as the research object and then divided into control group (no treatment), IMP-SD low-, medium- and high-concentration groups (40, 80 and 160 μmol/L IMP-SD), si-ThPOK and si-NC group [treated with 160 μmol/L IMP-SD and then transfected with ThPOK small interfering RNA (si-ThPOK) or its negative control (si-NC)]. After treatment, cell clone formation, migration and invasion abilities and apoptosis of MKN-7 cells were detected; the killing activity of NK cells, T cells classification, the protein expressions of ThPOK, programmed death-1 (PD-1) and programmed death-ligand 1 (PD-L1) were all determined. RESULTS Compared with the control group, the number of cell clones, migration number, invasion number, and the protein expressions of PD-1 and PD-L1 were decreased or down-regulated significantly in IMP-SD groups, while the cell apoptotic rate, NK cell killing activity, CD4+ T proportion, the ratio of CD4+ T proportion and CD8+ T proportion (CD4+ T/CD8+ T), and the protein expression of ThPOK were increased or up-regulated significantly, in a concentration-dependent manner (P<0.05). Compared with IMP-SD high-concentration group and si-NC group, the number of cell clones, migration number, invasion number, and the protein expressions of PD-1 and PD-L1 were increased or up-regulated significantly in si-ThPOK group, while the cell apoptotic rate, NK cell killing activity, CD4+ T proportion, CD4+ T/CD8+ T, and the protein expression of ThPOK were decreased or down-regulated significantly (P<0.05). CONCLUSIONS IMP-SD may reduce the clonal formation, migration and invasion abilities of GC cells, promote their apoptosis and inhibit their immune escape by promoting ThPOK expression.

Nuclear factor-kappa B (NF-κB) is an important intracellular transcription factor widely involved in the processes such as immune response, inflammatory response, cell proliferation, and apoptosis. The abnormal activation of the NF-κB signaling pathway plays a pivotal role in various liver diseases including chronic hepatitis, liver fibrosis, liver cirrhosis, and hepatocellular carcinoma. Extensive studies have shown that inhibiting NF-κB activity may effectively reduce inflammation and fibrosis and improve metabolic disorders. Several natural compounds, such as matrine and salvianolic acid B, have shown the potential in suppressing NF-κB activity, thereby exerting anti-inflammatory, anti-fibrotic, and anti-tumor effects. This article systematically reviews the critical role of the NF-κB signaling pathway in liver diseases and its potential as a therapeutic target, in order to highlight its potential as a therapeutic target for liver diseases and provide new directions for the treatment of liver diseases.

Chronic pelvic inflammatory disease(CPID)is a common chronic inflammatory disease in women,and has a long course and is easy to relapse.Danggui Shaoyao Powder is from Jin Gui Yao Lve(Synopsis of the Golden Chamber),which was a commonly-used formula for the treatment of women's abdominal pain in ancient medical records.It is now often used in the treatment of CPID and has achieved satisfactory therapeutic effect.The article summarizes and analyzes the achievements in the clinical research and experimental study of Danggui Shaoyao Powder in the treatment of CPID over the past 10 years,and invesigates the clinical efficacy of Danggui Shaoyao Powder in the treatment of CPID and its therapeutic mechanism.In the field of clinical studies,Danggui Shaoyao Powder for the treatment of CPID was used by modification,or alone,or in combination with antibiotics and Chinese medicine external treatment,and its combined use was effective on significantly improving the indicators of inflammatory response and immune function,alleviating the clinical signs and symptoms such as pain,and did not increase the incidence of adverse reactions compared with the application of western medicines alone.In the field of experimental studies,Danggui Shaoyao Powder played the therapeutic role in CPID by decreasing the adhesion of endothelial cells,regulating the degradation of extracellular matrix,improving the level of inflammatory factors,and down-regulating the expression of proteins related to the nuclear factor κB(NF-κB)pathway.
7.Effects of Electroacupuncture on Skeletal Muscle Mitochondrial Dynamics in SAMP8 Mice

Objective To observe the effects of electroacupuncture on the motor function and mitochondrial dynamics of skeletal muscle of SAMP8 mice;To explore the mechanism of electroacupuncture in improving the motor dysfunction of Alzheimer disease(AD)from the perspective of mitochondrial dynamics.Methods Totally 18 SAMP8 mice were divided into model group and electroacupuncture group,with 9 mice in each group,and the SAMR1 mice with the same age were set as control group."Baihui","Dazhui"and"Shenshu"were selected in the electroacupuncture group,and electroacupuncture was performed daily for 20 min,8 d as a course of treatment.Each course of treatment was separated by 2 d,for a total of 3 courses of treatment.The model group and the control group were not intervened.The motor function of mice was tested by grip strength test,suspension test,hind limb extension test and Morris water maze experiment.The morphology and structure of gastrocnemius were observed by HE staining,ATP content in gastrocnemius was determined by colorimetry,the mRNA expression of optic atrophy 1(OPA1),mitofusin 2(MFN2)and dynamin-related protein 1(DRP1)in gastrocnemius were detected by real-time quantitative PCR,the expressions of OPA1,MFN2 and DRP1 in gastrocnemius were detected by Western blot.Results Compared with the control group,the grip strength,the score in suspension test,and the average speed and maximum speed of Morris water maze experiment of mice in model group significantly decreased(P<0.01);the arrangement of fibers in the gastrocnemius muscle tissue was disordered,the gaps become wider,and the distribution of nuclei was uneven;the ATP content in the gastrocnemius muscle tissue was significantly decreased(P<0.01),the mRNA and protein expressions of OPA1 and MFN2 were significantly decreased(P<0.01),and the expression of DRP1 mRNA and protein significantly increased(P<0.01).Compared with the model group,the grip strength,the score in suspension test,and the average speed and maximum speed of Morris water maze experiment in electroacupuncture group significantly increased(P<0.01);the arrangement of gastrocnemius muscle tissue was relatively neat,the gaps become narrower,and the distribution of nuclei was more uniform;the ATP content in gastrocnemius muscle tissue significantly increased(P<0.01),while the mRNA and protein expressions of OPA1 and MFN2 significantly increased(P<0.05,P<0.01),the expression of DRP1 mRNA and protein significantly decreased(P<0.01).Conclusion Electroacupuncture can improve the skeletal muscle morphological structure and motor dysfunction of SAMP8 mice,and the mechanism may be related to the correction of skeletal muscle mitochondrial dynamic imbalance and the increase of skeletal muscle ATP content.
8.Exploring the Effect of Yuye Decoction on the Prevention and Treatment of Diabetic Nephropathy Based on Intestinal Flora and Intestinal Mucosal Barrier

Objective To investigate the effects of Yuye Decoction(YYD)on diabetic nephropathy(DN)rats with Qi and Yin deficiency based on intestinal flora and intestinal mucosal barrier.Methods The content of the active ingredients puerarin,mangiferin and calycosin 7-O-β-D-glucopyranoside in YYD freeze dried powder was tested by HPLC to control the quality of YYD freeze dried powder.Forty-five Wistar rats were randomly divided into normal group(NC group),Qi-Yin deficiency DN group(M group)and YYD administration group(YYD group;5.03 g·kg-1)according to body weight.Intraperitoneal injections of streptozotocin(STZ)at a dose of 25 mg·kg-1 were administered,accompanied by oral gavage of adix Aconiti Lateralis,Citri Reticulatae Pericarpium Viride and Aurantii Fructus for 8 weeks to establish a Qi-Yin deficiency DN model in rats.During this period,rats in the YYD group were also treated with YYD extract.Weekly body weight and blood glucose levels were monitored for each group,along with observations of physical signs,food and water consumption,swimming time,and urine output in the rats.After 8 weeks,serum levels of cholesterol(TC),triglycerides(TG),low-density lipoprotein(LDL-C),high-density lipoprotein(HDL-C),serum creatinine(SCr)and urinary nitrogen(BUN)were analyzed in rats using automatic biochemical analysis.Hematoxylin-eosin staining(HE)was used to observe the pathological changes in the kidney and colon tissues.Enzyme-linked immunosorbent assay(ELISA)was used to measure urine protein concentration and serum levels of IgG,IgM,cAMP and cGMP.16SrDNA sequencing was used to sequence faecal flora and some flora were validated by quantitative real-time fluorescence PCR(qRT-PCR).Western blot method was used to measure the levels of intestinal barrier proteins and inflammatory factor proteins.Results The yield of YYD freeze-dried powder was 27.36%and the content of the active ingredients in YYD was 0.3496 mg·g-1 for puerarin,0.1851 mg·g-1 for mangiferin and 0.0429 mg·g-1 for calycosin 7-O-β-D-glucopyranoside.The results of animal experiments indicated that compared with Qi and Yin deficiency DN rats,YYD significantly improved the symptoms of lethargy and fatigue,increased swimming time,reduced water intake and increased food intake;increased the abundance of Bacteroidetes and decreased the abundance of Firmicutes in feces.Furthermore,according to measurements of blood lipids,blood glucose,renal function,IgG and IgM,as well as cAMP and cGMP,which are the energy homeostasis factors of the body,YYD could significantly improve the levels of these indicators in the model rats.HE staining showed that compared with model rats,YYD remarkably alleviated kidney and colon damage;inhibited the expression of inflammatory factors TNF-α and IL-6 in the kidney and colon,and markedly raised the expression levels of renal and colonic kinetic proteins CHRM3 and 5-HT3A as well as mucosal barrier proteins Occludin and ZO-1.Conclusion YYD play a role in preventing and treating in rats with Qi-Yin deficiency DN,and this effect may be related to YYD correcting intestinal flora dysbiosis,delaying intestinal mucosal damage and improving renal and colonic inflammation in the model rats.

10.Impact of early and timely treatment and initial antiviral treatment regimen on antiviral treatment mortality and attrition among HIV-infected patients in Liuzhou, Guangxi

Objective To understand the impact of early and timely treatment and initial antiviral treatment regimen on mortality and attrition of antiretroviral therapy. Methods A retrospective cohort study was conducted using download data on antiretroviral therapy for HIV-infected patients in Liuzhou City, Guangxi Province, from the database of the Basic Information System for AIDS Control and Prevention (BISAC) from 2010 to 2020. The Cox proportional risk regression model was used to analyze the influencing factors of mortality and attrition. Results A total of 15 713 infected patients were included, including 53.4% aged 18-<50 years, 69.4% male, 61.0% farmer, 75.1% CD4 count <350 cells /μL before initial antiviral treatment, the overall mortality rate was 4.30/100 person-years, and the overall attrition was 2.42/100 person-years. The results of Cox regression analysis showed that the influencing factors of mortality were pretreatment CD4 counts of 350-<500 cells/μL(AHR=0.72, 95%CI: 0.63-0.81) and ≥500 cells/μL (AHR= 0.64, 95%CI: 0.55-0.76); duration from diagnosis to initial antiviral treatment 91-180 days (AHR=1.25, 95%CI: 1.08-1.45), 181-365 days (AHR=1.26, 95%CI: 1.08-1.47), and ≥365 days (AHR=1.26, 95%CI: 1.11-1.44); initial antiviral treatment regimens of D4T+3TC+EFV/NVP (AHR=1.47, 95%CI: 1.32-1.63) and AZT/D4T/TDF+3TC+LPV/r (AHR=1.73, 95%CI: 1.50-1.99). Factors affecting attrition were pretreatment CD4 counts of 350-499 cells/μL (AHR=1.32, 95%CI: 1.16-1.50) and ≥500 cells/μL (AHR=1.28, 95%CI: 1.10-1.50); interval from HIV positivity confirmation to initial dosing ≥365 days (AHR=1.21, 95%CI: 1.04-1.40), initial antiviral treatment regimens of TDF+3TC+NVP (AHR=1.32, 95%CI: 1.13-1.55), AZT+3TC+EFV/NVP (AHR=1.43, 95%CI: 1.26-1.62) and AZT/D4T/TDF+3TC+LPV/r (AHR=1.33, 95CI%: 1.06-1.67). Conclusions Early and timely treatment and the initial antiviral treatment regimen of TDF+3TC+EFV have good efficacy, but attention should be paid to the high risk of attrition of HIV-infected people with high CD4 count before treatment.
