1.Recessive loss of pregnant nurses by psychological contract

Objective To explore the content of the psychological contract of pregnant nurses and analyze the main reasons resulting in the recessive loss of pregnant nurses, in an attempt to put forward a management strategy about pregnancy nurses. Methods Totals of 30 pregnant nurses and 15 nurse managers were randomly selected and interviewed to formulate the content of the psychological contract of pregnant nurses. Thereafter, the content was perfected by Delphi method to ensure the main psychological contract factors influencing the recessive loss of the pregnancy nurses. Results The content of the psychological contract of pregnant nurses included 12 factors of management responsibility ( posts of duty, workload, working atmosphere, bonus distribution, social welfare, education and training, opportunity to display the ability, promotion opportunity, respect for dignity, caring about the daily life of pregnant nurses, psychological comfort, pregnancy health care) and 11 factors of pregnant nurses responsibility ( fulfillment of duties, abidance by rules, rigorous thinking, enthusiasm for work, professional knowledge and skills, insist on the job, acceptance to the assignment, report for sick leave, interests of the hospitals, hospital image and development with the hospital) . Conclusions This index models a useful and reliable method to assess the psychological contract of pregnant nurses and can provide a guidance to reduce the recessive loss rate of pregnant nurses.
2.Efficacy of TCM fumigation in patients with active rheumatoid arthritis and its nursing

Objective To explore the clinical efficacy of traditional Chinese medicine ( TCM ) fumigation in patients with active rheumatoid arthritis.Methods A total of 120 patients with rheumatoid arthritis were randomly divided into the control group and the observation group. Patients in the control group ( n=60) received methotrexate and non-steroidal anti-inflammatory drugs, and patients in the observation group ( n=60) received TCM fumigation on the basis of those drugs that used in the control group. The efficacy between two groups was observed and compared after 15 days. Results The effective rate in the observation group was significantly higher than that in the control group (χ2=5.04,P<0.05). Outcome measures (DSA28, Morning stiffness duration, ESR and CRP) between two groups were distinctly improved after treatment (P<0.01), and the improvement in the observation group was better than that in the control group ( P < 0. 05 ) . Conclusions TCM fumigation combined with Western medicine is more effective, safe and worthy of being widely used in treating active rheumatoid arthritis.
3.Expession of Tp0319 recombinant protein from Treponema pallidum and analysis of its immunocompetence

Objective To clone, express Tp0319 gene from Treponemapallidum (T. pallidum), and to assess the immunocompetence of recombinant protein. Methods The immuno-dominant region of Tp0319gene was chosen by computer analysis, amplified from T. pallidum complete genome by PCR, subcloned into the expression vector pQE32 to construct a recombinant plasmid, pQE32/Tp0319, which was then expressed in E. coli M15. The recombinant protein was purified with Ni-NTA affinity chromatography, and identified by using sodium dodecyl sulfate polyacrylamide gel electropheresis (SDS-PAGE) and Western blot. New Zealand rabbits were immunized with the recombinant protein, and the titer of anti-Tp0319 antibodies in sera from immunized rabbits were measured with indirect ELISA. Also, indirect ELISA with the recombinant Tp0319 as coating antigen was performed to detect the anti-Tp0319 antibody in sera from 200 normal human controls and 200 patients with syphilis. Results The prokaryotic expression vector pQE32/Tp0319 was constructed successfully, and the recombinant protein Tp0319 with a molecular weight of about 30 000 was attained. Specific humoral response was elicited by the recombinant protein in New Zealand rabbits and the specific antibody titer was more than 1: 10 240 after immunization for 3 times. Western blot proved that the recombinant protein could specifically react with anti-T. pallidum IgG antibody-positive sera. Indirect ELISA was successfully developed with the recombinant Tp0319, and detected antibodies to T. pallidum in control sera with a sensitivity and specificity of 100% (40/40), respectively. Compared with T. pallidum particle agglutination (TPPA) assay, the sensitivity and specificity of the indirect ELISA were 92.6% and 100%, respectively, in the detection of T. pallidum in sera from patients and controls, and the concordance between the indirect ELISA and TPPA was 96%. Conclusions The prepared recombinant protein shows a satisfactory immunocompetence, which may lay a foundation for its further application in the serodiagnosis of syphilis.
4.Effect of Kechuanning on Adjusting Cytokine in Children with Virus-induced Asthma

Objective:To observe the effect of Kechuanning on Th cytokine expression in pediatric virus-induced asthma.Methods:120 cases of pediatric virus-induced asthma were randomly divided into Kechuanning group(treatment group,n =60,treated with Kechuanning) and Western medicine group(control group,n=60,treated with virazole).The change of Th1 cytokine IL-12 and Th2 cytokine IL-4 in blood serum were detected and analyzed.Results:Kechuanning could increase IL-12 but decrease IL-4 level in the patient's blood,therefore regulate Th cell subset,and the effect was more obvious than in control group.Conclusion:The mechanism of Kechuanning in curing pediatrics asthma induced by virus was related with increasing IL-12 and decreasing IL-4 level.
5.EB virus encoded latent membrane protein 1 modulates the phosphorylation of epidermal growth factor receptor in nasopharyngeal carcinoma cell line.

OBJECTIVETo elucidate the regulation of the phosphorylation of epidermal growth factor receptor (EGFR) by the EB virus encoded latent membrane protein 1 (LMP1) in nasopharyngeal carcinoma cell line.
METHODSThe levels of EGFR expression and phosphorylation in pTet-on LMP1 HNE2 cell, a nasopharyngeal carcinoma (NPC) cell line, in the dynamic expression of LMP1 induced by different concentrations of doxycycline (Dox) were observed. The EGFR dominant negative mutant and LMP1 antisense expression plasmid were transiently transfected into pTet-on LMP1 HNE2 cells by lipofectamine, and the changes in EGFR phosphorylation were observed by immunocoprecitation and Western blot. The changes in EGFR phosphorylation were observed after EGF treatment.
RESULTSIn pTet-on LMP1 HNE2 cells, Dox-induced LMP1 upregulated EGFR expression and phosphorylation in a dose-dependent manner. After EGFR dominant negative mutant was transfected into pTet-on LMP1 HNE2 cells, the increase of EGFR phosphorylation was inhibited completely. When LMP1 antisense expression plasmid was transfected into pTet-on LMP1 HNE2 cells, the levels of EGFR phosphorylation were also inhibited significantly. Meanwhile, after EGF had been added into pTet-on LMP1 HNE2 cells, increase of EGFR phosphorylation was induced, but it was completely blocked by EGFR dominant negative mutant and the introduction of LMP1 antisense.
CONCLUSIONEB virus encoded LMP1 not only induces the dose-dependent expression of EGFR, but also the dose-dependent phosphorylation of EGFR. The phosporylation of EGFR may play a vital role in the development of nasopharyngeal carcinoma.
