1.Functional analysis of prolyl oligopeptidase (POP) in foot-and-mouth disease virus replication.

The study aims to investigate the impacts of prolyl oligopeptidase (POP) on the replication of foot-and-mouth disease virus (FMDV) in BHK-21 cells. Firstly, the effects of FMDV replication on POP expression in BHK-21 cells were analyzed by Western blotting and Real-time reverse transcription polymerase chain reaction (RT-qPCR). Secondly, a eukaryotic expression plasmid for POP was constructed, and the effects of POP overexpression on the replication of two different serotypes of FMDV were assessed by Western blotting, RT-qPCR, and virus titer assays. Thirdly, specific small interfering RNAs (siRNAs) targeting POP were synthesized, and their efficiency in interfering with endogenous POP expression was identified by RT-qPCR. The impacts of downregulating endogenous POP expression on FMDV replication were further evaluated by Western blotting, RT-qPCR, and virus titer assays. The results indicated that FMDV infection did not significantly affect POP expression in BHK-21 cells. Overexpression of POP dose-dependently enhanced the replication of both FMDV/O and FMDV/A serotypes. Conversely, siRNA-mediated downregulation of endogenous POP expression markedly suppressed FMDV/O replication. This study is the first to demonstrated that the role of the host POP protein in promoting FMDV replication in BHK-21 cells, thereby providing a critical theoretical foundation and potential molecular targets for developing efficient candidate cell strains for foot-and-mouth disease inactivated vaccines.

2.Correlation between physical fitness condition and depressive symptoms in adolescents

Objective:
To explore the relationship between adolescents physical fitness and depressive symptoms, and to provide reference for the early prevention and intervention of depressive symptoms and improvement of physical fitness in Chinese adolescents.
Methods:
From September to December 2021, a total of 8 102 adolescents were selected by random cluster sampling method in Shanghai, Urumqi, Changsha and Kunming. The Center for Epidemiologic Studies Depression Scale(CES-D) was used to investigate the depressive symptoms, and completed grip strength, standing long jump, 50 m running, modified sitting forward flexion, 20 s repeated traverse, 30 s sit ups, 20 m round trip running (20 m SRT) test. χ 2 test, Goodman Kruskal Gamma and Logistic regression analysis were used to analyze the relationship between physical fitness index(PFI) and depressive symptoms.
Results:
The overall detection rate of depressive symptoms in adolescents with high level PFI was 23.4%, and the detection rate of low level adolescents was 26.3%, with a statistically significant difference ( χ 2=6.73, P =0.01). There was a significant positive correlation between PFI and depressive symptoms in the high school group ( G=0.09, P <0.05) and the boy group ( G=0.12, P < 0.05 ), and there was no significant association between PFI and depressive symptoms in the junior high school group and the girl group ( P >0.05). After adjusting for gender and age in the Logistic regression model, compared with those with high PFI, the risk of depressive symptoms in those with low PFI was 1.18 times (95% CI =1.05-1.33).
Conclusion
There is a correlation between physical fitness and depressive symptoms in adolescents. Adolescents with low PFI are at higher risk of developing depressive symptoms than those with high PFI.
3.Effects of progesterone on the proliferation and expressions of TNF-α and IL-6 genes of human decidual stromal cells in late pregnancy in vitro

Objective:To study the effects of progesterone on the morphology, proliferation, and secretion of cytokines of human decidual stromal cells (DSCs) in late pregnancy, and to explore the mechanism of progesterone in preventing spontaneous preterm birth.Methods:Human decidual stromal cells in late pregnancy were cultured and treated with different concentrations of progesterone (in the experimental groups, 10 -6 mol/L, 10 -5 mol/L and 10 -4 mol/L progesterone was added to the culture medium respectively, and no progesterone was added to the culture medium of control group). The morphology of DSCs was observed under the microscope, the cell length/width ratio was measured, the proliferation was detected by methyl thiazolyl tetrazolium (MTT) method, and the expressions of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) mRNA were detected by RT-qPCR. Results:The length/width ratios of DSCs in progesterone 10 -4 mol/L (5.87±0.19) and 10 -5 mol/L (5.98±0.27) groups were lower than that in control group (6.42±0.19), the differences were statistically significant ( P<0.001, P=0.002). The length/width ratio in the 10 -4 mol/L group was lower than that in the 10 -6 mol/L group (6.28±0.32, P=0.005). The proliferation of DSCs in the 10 -5 mol/L (0.70±0.04) and 10 -4 mol/L (0.78±0.04) groups was higher than that in control group (0.59±0.05; P=0.027, P=0.002), and proliferation of DSCs in 10 -4 mol/L group was higher than that in 10 -6 mol/L group (0.61±0.01, P=0.004). The expression of TNF-α mRNA in each progesterone group was lower than that in control group (all P<0.001) and the expression of TNF-α mRNA in the 10 -5 mol/L and 10 -4 mol/L groups was lower than that in the 10 -6 mol/L group (all P<0.001) . The expressions of IL-6 mRNA decreased gradually in control group, 10 -6 mol/L, 10 -5 mol/L and 10 -4 mol/L groups, the differences were statistically significant (all P<0.001). Conclusion:Progesterone can make the decidual stromal cells wider, promote proliferation, and decrease the expressions of TNF-α and IL-6 mRNA, which may play an important role in the mechanism of progesterone preventing spontaneous preterm birth.

5.Effects of nursing intervention in patients with generalized anxiety disorder based on the theory of anxiety ratification combined with progressive dominant content

Objective:To explore the effect of nursing intervention based on the theory of anxiety ratification combined with progressive dominant content in patients with generalized anxiety disorder (GAD) .Methods:From January 2020 to January 2021, convenience sampling was used to select 103 GAD patients admitted to the First Affiliated Hospital of Wenzhou Medical University as the research object. The patients were divided into the observation group ( n=52) and the control group ( n=51) by random number table method. The control group carried out routine nursing, and the observation group conducted the nursing based on the theory of anxiety ratification combined with progressive dominant content on the basis of the control group. The Medical Coping Modes Questionnaire (MCMQ) and the World Health Organization Quality of Life-BREF (WHOQOL-BREF) were used to evaluate the intervention effect. Results:There were no significant differences in MCMQ and WHOQOL-BREF scores between the two groups before intervention ( P>0.05) . After the intervention, the WHOQOL-BREF score and the confrontation dimension score in the MCMQ of the observation group were higher than those of the control group, and the avoidance and resignation dimension scores in the MCMQ were lower than those of the control group, and the differences were statistically significant ( P<0.05) . Conclusions:Nursing intervention based on the theory of anxiety ratification combined with progressive dominant content can effectively guide GAD patients to adopt a positive coping style to face the disease and improve the quality of life of patients, which is worthy of clinical application.
6.Clinical application value of human papillomavirus typing combined with cervical thin-prep cytology test in cervical cancer screening

Objective To explore the clinical value of the third-generation hybrid capture nucleic acid detection technology (DH3) typing detection of human papillomavirus (HPV) combined with thin-prep cytology test (TCT) in screening cervical cancer. Methods A total of1 582 female patients who received HPV and TCT cervical screening in Liuzhou Workers Hospital, Fourth Affiliated Hospital of Guangxi Medical University, from October 2020 to March 2021, were selected for this retrospective analysis. The cervical histopathological diagnosis was used as the gold standard to evaluate the diagnosis accuracies of HPV and TCT test results. Results Among the 1 582 patients, 334 were positive for HPV and 1 248 were negative; 234 were positive for TCT and 1 348 were negative; 180 were positive for histopathological diagnostic and 1 402 were negative. The sensitivity of HPV detection was 100%, the positive predictive value was 53.9%, the specificity was 89.0%, and the negative predictive value was 100.0%. In the HPV typing test, the positive rate for high-risk types 16/18 was 94.8%, and the positive rate for other 12 high-risk types was 73.8%. There was a little difference between TCT test and pathological test, and the detection consistency rate of high-grade squamous intraepithelial lesions and squamous carcinoma was same. Conclusion HPV testing combined with TCT to screen cervical cancer can reduce the misdiagnosis rate by a single test. The classification test is helpful for the hierarchical management of patients, and has a high clinical value for examination triage and grade screening.
