1.Constitution of children with idiopathic central precocious puberty and rapidly progressive early development

Objective:Exploratory study on the relationship between TCM syndrome types, TCM constitution and clinical symptoms of idiopathic central precocious puberty children and rapidly progressive early development children in Shanghai, so as to provide a basis for early intervention and syndrome differentiation and treatment.Methods:A cross-sectional study and retrospectively enrolled. Through questionnaire survey, the basic information, clinical symptoms and TCM constitution of children diagnosed with idiopathic central precocious puberty and rapidly progressive early development were collected from the Pediatric Endocrinology Clinic of Affiliated Shuguang Hospital of Shanghai University of Traditional Chinese Medicine from July to August 2022, and the correlation between the distribution, differentiation and clinical symptoms of TCM constitution in children with idiopathic central precocious puberty and children with rapidly progressive precocious puberty in Shanghai were explored through data analysis.Results:① Fast progressive early development children with higher BMI and age than idiopathic central precocious puberty were statistically significant ( P<0.05), and no significant difference in the sex composition ratio between the two groups. ② Among children with idiopathic central precocious puberty and children with rapid progressive early development, the proportion of liver depression fire syndrome was the highest, 59.15% (42/71) and 46.51% (60/129) respectively, and the proportion of Qi depression was the highest, 35.21% (25/71) and 36.43% (47/129) respectively. However, there was no significant difference in the TCM syndrome types and physical distribution of the two groups. ③ The TCM syndrome types of 200 precocious children weres correlated with physical distribution, among which Yin deficiency fire flourishing syndrome was significantly associated with Yin deficiency, liver stagnation fire syndrome was significantly correlated with qi depression, and phlegm dampness obstruction syndrome was significantly correlated with phlegm dampness and Yang deficiency ( P<0.01). ④ A total of 200 children with precocious puberty were more female than male, male BMI was higher than female, and the BMI of children with precocious puberty was higher than other syndrome types, and the differences were statistically significant ( P<0.01). Liver stasis fire syndrome was the highest proportion between boys and girls, but there was no obvious difference in TCM syndrome types between different genders. The highest proportion of breast pain in clinical symptoms was 22.19% (69/311). Conclusion:Children with idiopathic central precocious puberty and rapidly progressive early development children have differences in age and BMI, with no obvious difference in gender composition, TCM syndrome type and physical distribution, but there is a correlation between TCM syndrome type and physical constitution of precocious puberty children.
2.Anti-cancer Effect of Traditional Chinese Medicine in Regulating Cellular Senescence: A Review

In recent years, the incidence and mortality rates of cancer have been increasing, posing a serious threat to human health. Western medicine mainly uses treatments such as surgical resection, chemotherapy, immunotherapy and targeted therapy, but they are prone to complications, drug resistance and adverse reactions. A growing number of studies have shown that traditional Chinese medicine has obvious advantages in the treatment of cancer, reducing the recurrence rate of cancer and improving the quality of survival of patients. Cellular senescence refers to a state of irreversible cell cycle growth arrest when cells cease to proliferate after a limited number of divisions, resulting in a decline in cell proliferation and differentiation capacities and physiological functions, accompanied by morphological changes such as flattening and multinuclear morphology. At the molecular level, it shows increased expression of DNA damage-related genes, reduced expression of cell cycle-related factors and significant secretory activity. The malignant development of cancer is closely related to cellular senescence. With the increasing number of cancer cell proliferation, cancer-related genes undergo continuous mutations, freeing them from cellular senescence and thus achieving unlimited proliferation. Through recent studies, it has been found that induction of tumor cell senescence, possibly through modulation of cellular DNA damage, cell cycle arrest and senescence-associated secretory phenotype (SASP), which converts the suppressive immune tumor microenvironment to an activated immune tumor microenvironment and thus reverses the escape of tumor cell senescence, is a promising strategy for cancer therapy. However, the mechanism of cellular senescence in cancer progression is not fully understood, especially the anti-cancer role played by traditional Chinese medicine in regulating cellular senescence. This article summarized and concluded the specific molecular mechanisms of cellular senescence, the role of cellular senescence in cancer progression, and the mechanism of anti-cancer effects of traditional Chinese medicine based on cellular senescence from the perspective of regulating cellular senescence, with a view to providing ideas and methods for the anti-cancer effects of traditional Chinese medicine and the development of new drugs.
3.Applicability verification of Lowry2 method for protein content determination of intermediate products of component pertussis vaccine

Objective To verify the applicability of Lowry2 method[the second method of Folin-phenol method of Chinese Pharmacopoeia(VolumeⅢ,2020 edition)]for the determination of protein content of intermediate products of component pertussis vaccine.Methods The standard and sample were precipitated and pretreated with deoxycholate and trichloroacetic acid to remove impurities,and then reacted with Folin-phenol,and the absorbance was measured at 750 nm wavelength.The standard curve was drawn with the concentration and absorbance of the standard,and the sample concentration was calculated. The method was verified for the specificity,linearity,accuracy and precision,and used to detect the protein content of intermediate products of three batches of component pertussis vaccine.Results There was no significant difference between the protein content in background buffer of intermediate product detected by this method and that in ultrapure water(t = 0. 277~1. 178,P = 0. 304~0. 795);The linear relationship of standard curve was good at the protein concentration of 0~50 μg/mL,each R~2> 0. 98;The spike recovery of accuracy verification was in the range of 90%~110%;The RSDs of repeatability and intermediate precision verification were both less than 3%. The detection results of protein content in the intermediate products of three batches of component pertussis vaccine all met the requirements of internal manufacturing and verification regulations.Conclusion Lowry2 method has good specificity,accuracy and precision,and can be used to determine the protein content of component pertussis intermediate products,which provides a basis for the applicability of Lowry2 method.
4.Risk factors of early traumatic coagulopathy in elderly patients with thoracic trauma

Objective To explore the risk factors of early traumatic coagulation(TIC)in elderly patients with chest trauma.Methods The data of 113 elderly patients with chest trauma admitted in a hospital from January 2016 to January 2022 were retrospectively analyzed.Patients were divided into TIC group and non-TIC group according to whether they had early TIC.The risk factors of early TIC in elderly patients with thoracic trauma were analyzed by single factor and multi factor methods.Analysis of the predictive value of early TIC in elderly patients with thoracic trauma using multifactor model.Result Of all the patients included in the study,27 were determined as TIC,and the rest 86 were non-TIC.The results of univariate analysis showed that there were statistically significant differences in the data of shock index,platelet count at admission and plasma fibrinogen at admission between patients in TIC group and non-TIC group(P<0.05).Multivariate analysis showed that the shock index at admission,platelet count at admission,and plasma fibrin at admission were all independent influencing factors of early TIC in elderly patients with chest trauma(all P<0.05).The P value of multivariate model and independent variables predict the early TIC of elderly patients with chest trauma.The Yoden index is 36.95%,42.89%,75.58%and 78.85%respectively.Conclusion The shock index,platelet count and plasma fibrin at the time of admission can affect the early occurrence of TIC in elderly patients with chest trauma.
5.Intravenous route to choroidal neovascularization by macrophage-disguised nanocarriers for mTOR modulation.

Retinal pigment epithelial (RPE) is primarily impaired in age-related macular degeneration (AMD), leading to progressive loss of photoreceptors and sometimes choroidal neovascularization (CNV). mTOR has been proposed as a promising therapeutic target, while the usage of its specific inhibitor, rapamycin, was greatly limited. To mediate the mTOR pathway in the retina by a noninvasive approach, we developed novel biomimetic nanocomplexes where rapamycin-loaded nanoparticles were coated with cell membrane derived from macrophages (termed as MRaNPs). Taking advantage of the macrophage-inherited property, intravenous injection of MRaNPs exhibited significantly enhanced accumulation in the CNV lesions, thereby increasing the local concentration of rapamycin. Consequently, MRaNPs effectively downregulated the mTOR pathway and attenuate angiogenesis in the eye. Particularly, MRaNPs also efficiently activated autophagy in the RPE, which was acknowledged to rescue RPE in response to deleterious stimuli. Overall, we design and prepare macrophage-disguised rapamycin nanocarriers and demonstrate the therapeutic advantages of employing biomimetic cell membrane materials for treatment of AMD.
6.Clinical significance of expression of leptin in patients with biliary atresia and hepatic fibrosis

Objective:To study the relationship and the role of leptin in children with biliary atresia and hepatic fibrosis to provide a treatment basis for these patients.Methods:The clinical data of children with biliary atresia or congenital biliary dilatation (CBD) who underwent surgical treatment at the Department of General Surgery of Tianjin Children's Hospital from August 2019 to August 2021 were retrospectively analyzed. Of 31 children included in this study, there were 14 males and 17 females, with age of 60 (30, 63) d. Children with biliary atresia served as the study group ( n=26) and children with CBD served as the control group ( n=5). Leptin protein, α-smooth muscleactin (α-SMA) and phosphorylation of extracellular-regulated protein kinase 1/2 (p-ERK1/2) in liver tissues were detectd by immunohistochemistry (IHC). The expression level of leptin mRNA in liver tissues were detected by quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR). Results:The average optical density values of leptin protein, α-SMA protein and p-ERK1/2 protein in the liver tissues of children in the study group were significantly higher than the control group ( P<0.05). The expression levels of leptin, α-SMA and p-ERK1/2 in liver tissues of children with biliary atresia significantly increased with increase in fibrosis degree ( P<0.05). The expression level of leptin in liver tissues of children with biliary atresia was positively correlated with the liver fibrosis grade ( rs=0.876), α-SMA ( r=0.723) and p-ERK1/2 ( r=0.725) ( P<0.01). The results of qRT-PCR showed that the content of leptin mRNA in liver tissues of children with biliary atresia was significantly higher than that of children with CBD ( P<0.05). Conclusion:Expressions of leptin increased with aggravation of degrees of hepatic fibrosis in biliary atresia. Leptin may be involved in activation of HSCs through the ERK1/2 signaling pathway in the process of hepatic fibrosis due to biliary atresia.

In order to improve the management level of medicine list of medical institutions in China ,and help medical institutions build a medicine list of medical institutions with reasonable drug use structure ,standardized adjustment procedures , convenient operation and application and scientific evaluation methods ,so as to meet the needs of clinical rational drug use to the greatest extent ,with the support of the Pharmaceutical Care Professional Committee of the Chinese Pharmaceutical Association , China-Japan Friendship Hospital and the First Affiliated Hospital of University of Science and Technology of China (Anhui Provincial Hospital )jointly initiate and complete Guideline for the Evaluation of Medicine List in Chinese Medical Institutions jointly with a number of medical institutions. In strict accordance with the methodological requirements of World Health Organization standard guidelines ,based on the Delphi method ,the guideline formulation working group has constructed the quality evaluation index system and quantitative scoring table of medicine list management in medical institutions from the 5 dimensions of organization and management ,structure,adjustment,application and e valuation of the list. It is used to help medical institutions evaluate the quality of their medicine list management ,so asto play a positive role in the fine management of medicine list in medical institutions.
8.Research advances on the relationship between age at Kasai portoenterostomy and prognosis for biliary atresia

Kasai portoenterostomy is the preferred treatment for biliary atresia. Age at Kasai portoenterostomy is an important factor affecting the prognosis of children with biliary atresia and avoiding liver transplantation. Choosing the appropriate surgical age, restoring good bile drainage, improving the native liver survival rate, and avoiding early liver transplantation are the items that clinicians have always been working on. The age at Kasai portoenterostomy was correlated with the jaundice clearance rate, native liver survival rate, and the incidence of postoperative cholangitis. This article systematically reviewed the research advances on the relationship between age at Kasai portoenterostomy and prognosis for biliary atresia, aiming to provide the basis for the ideal surgical age of Kasai.

Parkinson's disease (PD) is the second most common neurodegenerative disease, but none of the current treatments for PD can halt the progress of the disease due to the limited understanding of the pathogenesis. In PD development, the communication between the brain and the gastrointestinal system influenced by gut microbiota is known as microbiota-gut-brain axis. However, the explicit mechanisms of microbiota dysbiosis in PD development have not been well elucidated yet. FLZ, a novel squamosamide derivative, has been proved to be effective in many PD models and is undergoing the phase I clinical trial to treat PD in China. 10.Anatomic study of facial artery

Objective:Facial artery is the main blood supply source of facial soft tissue, which has an important influence on facial plastic surgery and injection procedures. There are many types of facial arteries. They have wide coverage, and travel several layers. In this study, the detailed characteristics of the facial arteries were revealed by cadaver dissection.Methods:In 19 donated fresh cranial (28 sides) specimens, the facial arteries were dissected. And then the type, layer, relationship with nasolabial groove, length, diameter, distance from the oral commissure, branches and submental artery were observed and measured.Results:Facial arteries were found in all cadavers, starting from the external carotid arteries, branching submental arteries below the mandibular margins, ascending to the anterior edge of the masseter muscles, crossing the mandibles and entering the faces. After entering into the faces, the facial arteries branched the submental arteries(100%, 28/28), lower labial arteries (100%, 28/28), upper labial arteries (100%, 28/28), lateral nasal arteries (92.9%, 26/28) and the angular arteries (57.2%, 16/28) throughout the courses. According to the vascular route, the facial artery was classified into four types. In TypesⅠ(8 sides), all branches were included and the angular arteries arose directly from the lower segment of the facial arteries (28.6%). In TypesⅡ(8 sides), all branches were included and the angular arteries were the terminal branches (28.6%). In TypesⅢ(10 sides), angular arteries were absent (35.7%). In Type Ⅳ (2 sides), angular arteries and lateral nasal arteries were absent (7.1%). The lower segment of facial artery coursed under the facial expression muscles, and continued to the middle segment at the point which was (26.0±5.0) mm away from the oral commissure on the lateral side. The layer that the facial artery coursed was varied. It passed through the superficial layer of the facial expression muscles. After passing the horizontal line parallel to the lower margin of nasal sill, the upper segment continued to on the superficial layer of the facial expression muscles. The facial arteries were mostly located medial to the nasolabial fold(24 sides, 85.7%) and occasionally across the nasolabial fold(4 sides, 14.3%). The lengths of the upper, middle and lower segments of facial artery were (26.4±10.9) mm, (29.7±8.4) mm, and(33.5±6.9) mm, respectively. The diameters in starting point of upper, middle and lower sections were(2.45±0.48) mm, (1.85±0.12) mm, and (1.09±0.21) mm, respectively. The facial artery passed lateral to the oral commissure and the distance from the intersection of the horizontal line to the oral commissure was (26.0±5.0) mm. The starting point of the superior labial artery locacted below the horizontal line was (8.0±1.4)mm away from the horizontal line of the oral commissure and(55.0±5.2) mm from the mandibular angle. The starting point of the inferior labial artery located below the horizontal line was (17.1±11.1) mm from the horizontal line of the oral commissure, and the distance from the mandibular angle was (44.2±5.2) mm. The diameter of the submental artery in its starting point was (1.4±0.1) mm. The distance from the starting point of the submental artery to the horizontal line of the oral commissure was (34.9±2.6) mm, and the distance from the mandibular angle was (29.4±5.8) mm. The starting point of the submental artery was (6.4±0.8) mm from the lower margin of the mandible. In the midline of the mandible, the distance between the submental artery and the lower margin of the mandible was (9.0±1.0) mm. In the midsagittal plane, the starting point of the submental artery was(8.4±1.0) mm from the lower margin of the mandible.Conclusions:There were several types of facial arteries and the courses were various, but the way they branched and the layers they coursed had certain rules. Understanding the anatomy of facial arteries is helpful to keep the treatment safe.
