1.Preliminary study on remote treatment of relapse in alcohol dependent patients by the alcoholics anonymous

Objective To explore the impact of alcoholics anonymous (AA) online video conferences on relapse among alcohol dependent patients. Method Alcohol dependent patients who were hospitalized and discharged from the Mental Health Center of the First Hospital of Hebei Medical University from March 2020 to March 2022 were randomly divided into the AA group and the non AA group. The AA group underwent a 12-month remote AA treatment,participating in an AA online video conference once a week. The non-AA group did not receive any intervention. At baseline,the general condition of the subjects,such as alcohol consumption and addiction duration,as well as the severity of anxiety and depression symptoms,were evaluated. At 12 months after enrollment,clinicians communicated and assessed the subjects and their families via telephone or WeChat. The assessments included changes in anxiety and depression among the subjects,relapse into drinking,duration of abstinence after discharge,and other relevant factors. Result Eighty subjects were included,including 32 in the AA group and 48 in the non AA group. After intervention,the proportion of the AA group patients who resumed drinking was lower than that of the non AA group (68.8% vs. 93.8%,P<0.01). The median duration of abstinence in the AA group was 7.5 (3.0,12.0) months,while the median duration of abstinence in the non AA group was 1.0 (0.3,5.0) months,with a significant difference (P<0.01). Multivariate logistic regression analysis showed that the risk of relapse in the non AA group was 14.85 times higher than that in the AA group (OR=14.85,95%CI:2.33-94.57). Conclusion Online videos for AA in quitting alcohol are convenient and easy to implement for alcohol dependent patients. This remote treatment can significantly prolong the duration of abstinence among alcohol dependent patients.

3.Expression of hypoxia-inducible factor-1alpha in liver tumors after transcatheter arterial embolization in an animal model.

To examine the effect of transcatheter arterial embolization (TAE) of liver tumors on hypoxia-inducible factor-1alpha (HIF-1alpha) expression in the residual viable tumor, a total of 30 New Zealand White rabbits implanted with VX2 liver tumor were divided into 2 groups. TAE-treated group animals (n=15) were subjected to TAE with 150-250 mum polyvinyl alcohol particles. Control group animals (n=15) underwent sham embolization with distilled water. Six hours, 3 days or 7 days after TAE, the animals were sacrificed, and samples of tumor and adjacent normal liver tissue were harvested. Expression of HIF-1alpha protein was examined immunohistochemically. Real-time PCR was performed to examine the HIF-1alpha mRNA levels. Our results showed that HIF-1alpha protein was expressed in the VX2 tumors but not in the adjacent normal liver tissue. The HIF-1alpha-positive tumor cells were located predominantly at the periphery of necrotic tumor regions. The mean levels of HIF-1alpha protein were significantly higher in TAE-treated tumors than those in control tumors (P=0.002). Among the three sacrificing time points, the difference in increase in HIF-1alpha protein was significant between the two groups at the sacrificing time point of 6 h and 3 days after TAE (P=0.020, P=0.031, respectively), whereas no significant increase was noted 7 days after TAE (P=0.502). In contrast, although HIF-1alpha mRNA was expressed in TAE-treated and control VX2 tumors, there existed no significant difference in the HIF-1alpha mRNA level between the two groups (P=0.372). It is concluded that TAE of liver tumors increases the expression of HIF-1alpha at protein level in the residual viable tumor, which could be attributed to hypoxia generated by the procedure.

5.P120-catenin Isoforms 1A and 3A Differently Affect the Expression of E-cadherin and β-catenin in Lung Cancer Cells

Background and objective Different isoforms of p120-catenin (p120ctn), the members of catenin family, are variably expressed in different tissues and cells. These isoforms may confer different properties with respect to cell adhesion and migration, but this concept needs experimental evidence. Methods To more precisely define their biological roles, we stably transfected isoforms 1A and 3A into the lung cancer cell lines A549 and NCI-H460, which had low expression of p120ctn. Using RT-PCR, Western Blot, and Transwell, we testified their impact on the expression ofE-cadherin, β-catenin, and the invasivenessof lung cancer cells. Results Expression of isoform 1A of p120ctn upregulated E-cadherin and β-catenin, as well as the ability of ceils to invasion. In contrast, overexpression of isoform 3A had a smaller influence on invasion. Conclusion P120ctn isoforms 1A and 3A may differently regulate the invasive, adhesive of hing cancer cells through distinct regulation of the expression of E-cadherin and β-catenin.
