1.Current status and prospects of botulinum toxin type-A combined with platelet-rich plasma injection

In recent years, botulinum toxin type-A (BTX-A) combined with cosmetic injections and radiofrequency lasers have been widely implemented to achieve better tissue repair. Among these therapies, platelet-rich plasma (PRP), rich in growth factors, has demonstrated notable efficacy in promoting tissue regeneration and repair. However, there is some controversy regarding the combined use of these two biomaterials in the treatment of various diseases. This review summarizes the current status of their combined application in peripheral neuropathic pain, seborrheic alopecia, and facial rejuvenation, also analyzes the advantages, interactions, and influencing factors of the combined application. Although the effectiveness and safety of PRP combined with BTX-A injection therapy in many of the above diseases are relatively high, the conclusion should still be carried out in the future multi-center, large-sample clinical studies to improve the credibility and representativeness of the results.
2.Expression and clinical significance of TTK and MDM2 in cervical lesion tissue

Objective To investigate the expression and roles of serine/threonine and tyrosine kinases(TTK)and mouse dual microbody 2(MDM2)in cervical lesion tissue.Methods Patients(n=180)who underwent gynecological surgery at the First Affiliated Hospital of Henan University of Science and Technology from June 2022 to January 2024 were evaluated,including 50 patients in the cervical cancer group,50 patients in the high-grade lesion group,40 patients in the low-grade lesion group,and 40 patients in the non-cervical lesion(control)group.The mRNA expression and protein levels of TTK and MDM2 in each group were detected and compared by real-time quantitative PCR and immunohistochemistry,respectively.The diagnostic value of the mRNA expression levels of TTK and MDM2 in cer-vical cancer was analyzed using receiver operating characteristic curves.Results The mRNA levels of TTK and MDM2 significantly dif-fered among the four groups(P<0.05);with increases in the lesion grade,the mRNA levels of TTK and MDM2 increased gradually and showed significant differences(P<0.05).Immunohistochemical results showed that TTK was mainly located in the cytoplasm,whereas MDM2 was mainly located in the nucleus,with positive expression rates in cervical cancer tissues of 84.00%and 90.00%,respectively.These values were higher than those in other pathological tissues(P<0.05).The mRNA levels of TTK and MDM2 were significantly cor-related with the degree of differentiation and lymph node metastasis(P<0.05).The receiver operating characteristic curve showed that the areas under the curve of TTK and MDM2 mRNA for diagnosing cervical cancer were 0.803 and 0.798,respectively.Additionally,the area under the curve of the combined diagnosis was 0.912,which was higher than that observed for single detection(Z=2.230,2.553,P<0.05).Conclusion The mRNA expression of TTK and MDM2 and their corresponding protein levels were increased in cervical lesions,demonstrating their potential as biological indicators for the clinical diagnosis of cervical cancer.
3.Relationship between postoperative delirium and preoperative frailty in elderly patients undergoing knee or hip arthroplasty

Objective:To evaluate the association between postoperative delirium (POD) and preoperative frailty in elderly patients undergoing knee or hip arthroplasty.Methods:This nested case-control study utilized medical records from elderly patients who underwent knee or hip arthroplasty under combined spinal-epidural anesthesia at Qingdao Municipal Hospital between September 2021 and May 2023. Participants were divided into 2 groups based on clinically diagnosed POD: POD group ( n=53) and non-POD group ( n=256). Univariate analysis was conducted on suspected influencing factors, and logistic regression analysis was utilized to identify the risk factors for POD. Receiver operating characteristic and clinical decision curves were plotted to evaluate the predictive performance of these risk factors for POD. Mediation analysis was performed, and a clinically applicable nomogram was constructed to achieve visual prediction of outcomes. Results:There were statistically significant differences in age, preoperative frailty, body mass index, American Society of Anesthesiologists Physical Status classification, Memorial Delirium Assessment Scale scores, and concentrations of Aβ 42, Aβ 40, phosphorylated tau protein (p-tau protein) and tau protein, Aβ 42/tau ratio and Aβ 42/p-tau ratio in cerebrospinal fluid (CSF) between non-POD group and POD group ( P<0.05). Preoperative frailty was a risk factor for POD ( P<0.05). Mediation analysis revealed that the association between preoperative frailty and POD was mediated by CSF tau protein concentrations. The area under the receiver operating characteristic curve of preoperative frailty and CSF biomarker concentrations in predicting POD was 0.974 ( P<0.05). The clinical decision curve demonstrated that the model combining the preoperative frailty and CSF biomarker concentrations predicted a higher net benefit ( P<0.05). The clinical decision curve showed that the model combining preoperative frailty and CSF biomarker concentrations predicted a higher net benefit. Conclusions:Preoperative frailty is a risk factor for POD in elderly patients undergoing knee or hip arthroplasty, and its combination with CSF biomarker concentrations can effectively predict the occurrence of POD. 7.Labeling of carcinoembryonic antigen-specific CAR-T cells with superparamagnetic iron oxide nanoparticles and in vitro magnetic resonance imaging

Objective To use superparamagnetic iron oxide nanoparticles(SPIONs)to label chimeric antigen receptor(CAR)T cells targeting carcinoembryonic antigen(CEA),and perform magnetic resonance imaging(MRI)to real time trace CEA CAR-T cells in vivo.Methods Appropriate amount of ferumoxytol,heparin sodium and protamine sulfate were mixed at high(ferumoxytol 100 μg/mL,heparin sodium 4 IU/mL,protamine sulfate 120 μg/mL),medium(ferumoxytol 50 μg/mL,heparin sodium 2 IU/mL,protamine sulfate 60 μg/mL),and low(ferumoxytol 25 μg/mL,heparin sodium 1 IU/mL,protamine sulfate 30 μg/mL)concentrations to form a SPIONs complex ferumoxytol/heparin/protamine(FHP),and then co-incubated with CEA CAR-T cells for cell labeling.The biocompatibility of FHP was detected by CCK-8 assay,EdU assay and flow cytometry.The uptake of FHP was detected by Prussian blue staining,and SPIONs content in the cells was quantitatively detected by inductively coupled plasma-mass spectrometry(ICP-MS).Flow cytometry was used to detect the lytic effect of FHP-labeled CEA CAR-T cells on tumor cells,and MRI was employed to scan FHP-labeled CEA CAR-T cells.Results FHP at high,medium,and low concentrations had no significant effect on the activity of CEA CAR-T cells,with cell activity above 100%determined by CCK-8 assay.DNA proliferation was above 94.3%in EdU assays.Prussian blue staining showed that CEA CAR-T cells could take FHP up,with the uptake increased with the increment of FHP concentration.ICP-MS showed that the intracellular Fe content was 440.23±189.36 ng/mL.Tumor cell killing experiment showed that FHP-labeled CEA CAR-T cells had excellent killing capability against tumor cells.MRI scans indicated that T2WI signals of FHP-labeled CEA CAR-T cells were significantly reduced with increasing FHP concentration(P<0.01).Conclusion SPIONs complex FHP shows good biocompatibility and can effectively label CEA CAR-T cells.SPIONs complex FHP can be used as a magnetic marker for CEA CAR-T cells and a feasible MRI tracer for clinical application.
8.Design of Motion Monitoring System Based on Unity3D and MEMS Inertial Sensing

Purpose/Significance To construct a human body model,bind it with actual human motion data,and achieve monitoring of human motion postures.Method/Process The paper designs a motion monitoring system based on Unity3D and MEMS inertial sensors.It utilizes MEMS inertial sensors to obtain human motion posture data,uses Euler angles for posture calculation,and sends the data to the computer system via the Bluetooth wireless transmission protocol to drive the movement of a virtual animated character,thereby achieving real-time 3D posture presentation of human actions.Result/Conclusion After experimental testing,the model can accurately monitor and simulate human motion postures.In the future,it can provide effective data analysis for application areas such as sports training and limb rehabilitation training.
9.Naringenin promotes atherosclerotic palque stability by regulating the expression of TIMP-3 in smooth muscle cells

Aim To investigate the effects of naringenin on atherosclerotic plaque extracellular matrix remodeling and plaque stability.Methods Murine vascular smooth muscle cells were isolated and treated with various doges of naringenin.ApoE-/-mice were fed with high-fat diet and received naringenin by lavage for 16 weeks.Intraplaque nec-rotic core,contents of collagen and fibrous cap thickness were measured by Sirius red-Haematoxylin staining.Elastin was detected by Van Gieson staining.Matrix metalloproteinase(MMP)activity was determined by gelatin zymography and fluorescence-gelatin staining.Results Naringenin(50 μmol/L)increased signal tansducer and activator of transciption 6(STAT6)phosphorylation and promoted tissue inhibitor of metalloproteinase-3(TIMP-3)expression by 3.1-fold(P<0.001).After naringenin(80 mg/kg)treatment,compared with the control group,the area of plaque necrotic core in aor-tic root decreased by 53%(P<0.01),the thickness of fibrous caps increased by nearly 50%(P<0.05),and the degree of elastic fiber degradation decreased.At the same time,naringenin promoted the expression of TIMP-3 in plaques,and corre-spondingly reduced the activity of MMP in plaques.Lentivirus mediated inhibition of TIMP-3 expression in vivo could reduce the protective effect of naringenin on plaque stability.Conclusion Naringin can increase the expression of TIMP-3 in smooth muscle cells,improve the composition of extracellular matrix,and promote the stability of atherosclerotic plaque.
10.Application of pathological indicators based on liver biopsy in the diagnosis of cirrhotic portal hypertension

Hepatic venous pressure gradient (HVPG) is the "gold standard" for diagnosing portal hypertension and determining its severity, but its wide clinical application is limited due to its invasiveness and difficulties in operation. The replacement of HVPG by noninvasive methods has become a research hotspot in recent years; however, the accuracy of the existing serological and imaging methods remains to be discussed, and such methods cannot completely replace HVPG in clinical practice. Liver biopsy has been widely used in clinical practice for many years and is still an indispensable method for the diagnosis of some liver diseases. Recent studies have found that several pathological indicators after liver biopsy, such as collagen area, fibrous septal thickness, nodule size, microvascular density, and density and area of bile ducts and lymphatic vessels, can not only judge the severity of liver fibrosis, but also have a good correlation with portal venous pressure, which provides new ideas for diagnosing cirrhotic portal hypertension and evaluating the severity of portal hypertension.
