1.Research progress on binge eating disorder's neuropsychological mechanism

Binge eating disorder(BED)is a common eating disorder whose pathogenesis involves both neurobiological and psychological mechanisms.At the neurobiological level,the development of BED is associated with abnormal resting-state brain functional connectivity in the reward circuitry,dysregulation of the endocannabinoid system,and elevated leptin levels.This paper reveals that the neurobiological mechanisms of BED may influence psychological processes,including habitual behavioral imbalances and impaired emotion regulation.Conversely,the dysfunction of behavior in the psychological domain may further modulate neurobiological manifestations.This finding provides insights for future research aimed at systematically integrating neural mechanisms into clinical interventions,ultimately facilitating treatment advancement and prognostic improvement.
2.Research Progress on the Wuwei Qingzhuo Preparation of Mongolian Medicine and Shiliu Jianwei Preparation of Zang Medicine

Based on the results of the National Drug Sampling and Inspection Programme,we summarized the history,the standard collection,the production enterprises and the dosage form specifications,the quality standard study,the pharmacological and pharmacodynamic study,and the clinical application study of Wuwei Qingzhuo preparation of Mongolian medicine and Shiliu Jianwei preparation of Zang medicine,to provide the basis for improved quality standards for both preparations.The development of these two preparations was searched and analyzed through literature.The available information shows that there is very little research on the two preparations and insufficient pharmacological experimental and clinical experimental data.The two preparations are basically the same in prescription and efficacy.However,the quality standards are very different,which are not conducive to the quality control of the two and their related dosage forms.And it is suggested that the Chinese Pharmacopoeia should take the situation of this category into comprehensive consideration,and unify the quality standards of the two preparations.
3.Effect of miR-155-5p/sirt1 signaling pathway on immune function of Candida albicans induced Kawasaki disease model mice

Objective :
To investigate the effect of the miR-155-5p/silent information regulator 1(sirt1) signaling pathway on the immune function ofCandida albicansinduced Kawasaki disease model mice.
Methods :
C56BL/6 mice were separated into control group, Kawasaki disease group, antagonist control group, miR-155-5p antagonist group, miR-155-5p antagonist+si-NC group, and miR-155-5p antagonist+si-sirt1 group, with 12 mice in each group. Except for the control group, mice in all other groups were used to construct a Kawasaki disease model by intraperitoneal injection of water-solubleCandida albicans. After successful modeling, administration was performed once a day for 7 days. QRT-PCR was applied to detect the expression of miR-155-5p in coronary arteries. Western blot was applied to detect sirt1 protein in coronary arteries. HE staining was applied to detect pathological changes in coronary arteries. Mouse thymus index and spleen index were detected. Flow cytometry was applied to detect helper T cells 17(Th17)/regulatory T cells(Treg) in peripheral blood. ELISA was applied to detect the levels of interleukin(IL)-17 and IL-10 in mouse serum. The targeting relationship between sirt1 and miR-155-5p was validated.
Results:
Compared with the control group, there was a large amount of inflammatory cell infiltration in the coronary arteries of mice in the Kawasaki disease group. The miR-155-5p expression, Th17 ratio, Th17/Treg ratio, and IL-17 level increased. The sirt1 protein expression, thymus index, spleen index, Treg ratio, and IL-10 level decreased(P<0.05). Compared with the Kawasaki disease group, the inflammatory cell infiltration in the coronary arteries of mice in the miR-155-5p antagonist group was alleviated. The miR-155-5p expression, Th17 ratio, Th17/Treg ratio, and IL-17 level decreased. The sirt1 protein expression, thymus index, spleen index, Treg ratio, and IL-10 level increased(P<0.05). Si-sirt1 weakened the promoting effect of miR-155-5p inhibition on Th17/Treg balance and the inhibitory effect on vascular inflammation in Kawasaki disease mice, miR-155-5p targeted and regulated sirt1.
Conclusion
The mechanism by which inhibiting miR-155-5p promotes Th17/Treg balance and inhibits vascular inflammation in Kawasaki disease mice may be related to the upregulation of sirt1 expression.
4.Study about the role of long noncoding RNA CCDC18-AS1 in primary hyerparathyroidism

Objective:To explore the role of long noncoding RNA(lncRNA) CCDC18-AS1 in parathyroid carcinoma(PC) diagnosis.Methods:This cross-sectional study included 55 patients with primary hyperparathyroidism treated at Beijing Shijitan Hospital from 2013 to 2024. Of these, 12 patients were diagnosed with PC and 43 with parathyroid adenoma(PA). Tissue samples and clinical data were collected, and lncRNA CCDC18-AS1 expression were measured using real-time quantitative PCR(RT-qPCR).Results:LncRNA CCDC18-AS1 expression was significantly higher in the PC group than that in the PA group( P=0.003). It was identified as an independent risk factor for PC, independent of age, sex, or serum calcium levels. Among patients with hypophosphatemia, no significant differences in lncRNA CCDC18-AS1 expression was observed between the PC and PA groups. However, in patients with normal serum phosphate levels, lncRNA CCDC18-AS1 expression was significantly higher in the PC group( P<0.001) and showed a positive correlation with serum phosphorus concentration( P=0.001). The area under the receiver operating characteristic(ROC) curve(AUC) for lncRNA CCDC18-AS1 in PC diagnosis was 0.758(95% CI 0.620-0.866, P=0.005), comparable to that of lncRNA plasmacytoma variant translocation 1(PVT1). Among patients with normal serum phosphate, the AUC for lncRNA CCDC18-AS1 increased to 0.969(95% CI 0.835-0.999, P<0.001), with 100% sensitivity and 92.31% specificity, suggesting superior diagnostic performance compared to PVT1(0.840, 95% CI 0.653-0.950, P=0.001). Conclusions:LncRNA CCDC18-AS1 may serves as a potential biomarker for PC diagnosis, with greater diagnostic value in patients with normal serum phosphorus levels.
5.The clinical features and advances in psychological treatments of trichotillomania in children and adolescents

Trichotillomania, also known as Hair Pulling Disorder, is a unique obsessive-compulsive spectrum disorder characterized by repeated removal of hair from various parts of the body. Patients attempt to control this behavior but often fail, causing impairment to important functional areas such as social interaction, work, and academics. Trichotillomania typically begins in childhood or adolescence, and is often comorbid with anxiety and depression. The resulting physical damage and changes in appearance further exacerbate the social functional impairment of patients, resulting in most patients being diagnosed only in adulthood, and missing the optimal intervention period. Current pharmacological treatments for Trichotillomania are not satisfactory, while various psychological therapies have shown potential value and prospects. Therefore, this article focuses on Trichotillomania in children and adolescents, providing a comprehensive review from multiple aspects including disease diagnosis, clinical characteristics and typing, functional impairment, neuroimaging mechanisms, and the latest developments in psychological therapy, to provide references for the clinical diagnosis, assessment, and effective intervention of Trichotillomania.
6.The neural and psychological mechanisms of alexithymia in adolescent non-suicidal self-injury

Alexithymia refers to a deficiency of emotional structure, but the neurologic and psychological mechanisms of non-suicidal self-injury (NSSI) in adolescents are still unclear. The neural basis of alexithymia may play a role in adolescents′ NSSI by affecting the function of emotion regulation and emotion expression. At the same time, NSSI is also considered to be a non-adaptive emotional regulation mode for alexithymia individuals, which interacts with personality factors and psychosocial factors. This study explored the neuropsychological mechanism of alexithymia in adolescent NSSI from the perspective of emotional function, and provided theoretical basis for early identification and precise intervention of alexithymia and adolescent NSSI.
7.Investigation and analysis on safety of empirical or experimental medication for adult-onset Still disease before diagnosis

Objective:To investigate and analyze the safety of empirical or experimental medication (EEM) for adult-onset Still disease (AOSD) before diagnosis.Methods:The AOSD inpatients admitted to the Second Hospital of Jilin University from January 1, 2019 to December 31, 2023 were collected through hospital information system, and those who were misdiagnosed on admission were screened out. The main clinical characteristics, laboratory tests, misdiagnosis situation, the use of EEM and their adverse drug reactions (ADR), and the potential drug-drug interactions in the misdiagnosed patients were analyzed by descriptive statistics.Results:During the set time period, a total of 49 patients with AOSD were admitted to the hospital, of which 16 (32.7%) were misdiagnosed with other diseases on admission. Among the 16 patients, 10 were male and 6 were female, with a median age of 53 years. The main clinical manifestations were fever (in 15 patients), arthralgia/arthritis (in 10 patients), lymphadenopathy (in 10 patients), rash (in 9 patients), pleural or pericardial effusion (in 6 patients), pneumonia (in 5 patients), splenomegaly (in 4 patients) and sore throat (in 4 patients). Abnormalities in laboratory tests included white blood cell count elevation (in 13 patients), platelets count elevation (in 8 patients), serum ferritin elevation (>500 μg/L, in 12 patients), and abnormal liver function (in 9 patients). The median time of treatment before admission was 5.5 months (11 days to 27.0 months), and the median time from admission to diagnosis of AOSD was 12 days. Before the diagnosis of AOSD, all patients received a long time of EEM, including antibiotics, traditional Chinese medicine preparations, liver-protection drugs, anti-allergic drugs and antiviral drugs in 15, 12, 11, 3 and 2 patients, respectively. Four patients experienced ADRs related to EEM, all of which were caused by antibiotics. There were potential interactions in the therapeutic drugs in 4 patients.Conclusion:The misdiagnosis rate of AOSD was high. Patients might had accepted multiple EEMs before the definite diagnosis, which posed risks of ADRs and drug interactions.
8.Mechanism of GRK subtypes modulating the unique binding properties of M1 acetylcholine receptor and transducers

Objective·To investigate the mechanisms by which different subtypes of G protein-coupled receptor kinases(GRKs)regulate the biased signaling transduction mediated by the muscarinic acetylcholine receptor 1(M1 receptor),focusing on their molecular effects in modulating the binding of the M1 receptor to the downstream heterotrimeric G protein(Gαq-Gβ1-Gγ2)andβ-arrestin 2(βarr2).Methods·By establishing a highly sensitive protein interaction detection system based on bioluminescence resonance energy transfer(BRET),six M1 receptor agonists/allosteric modulators were selected to measure the dynamic interactions between the M1 receptor and four GRK subtypes(GRK2/3/5/6),βarr2,and the G protein under stimulation.All BRET data were statistically quantified using the area under the curve(AUC)of the time-response curves.First,concentration-effect curves were established by treatment with gradient concentrations of agonists/allosteric modulators and AUC fitting,to comprehensively analyze the differences in efficacy between each agonist/allosteric modulator and the endogenous agonist acetylcholine chloride(ACh)in promoting the interactions of M1 receptor with GRK3/5,βarr2,and the G protein;next,GRKs were divided into two groups based on subtypes:GRK2/3 and GRK5/6.The maximum AUC values for the interaction between the M1 receptor and the two GRK groups under high concentrations were calculated respectively,to further evaluate the regulatory propensity of different types of GRKs on the binding strength of the M1 receptor to βarr2 or the G protein.Results·All six agonists/allosteric modulators effectively induced the association of the M1 receptor with GRK3,while simultaneousey inducing dissociation of the M1 receptor from GRK5.The allosteric modulator BQCA not only activated the M1 receptor alone and triggered its binding to downstream signaling proteins,but also,when co-treated with ACh,caused a significant leftward shift of the concentration-effect curves in the M1-G protein and M1-βarr2 systems,suggesting that its potentiation effect on ACh was mainly achieved by reducing the half-maximal effective concentration.A moderate positive correlation was observed between the maximum AUC values of M1-βarr2 and M1-G protein interactions induced by the seven groups of drug treatments(r=0.722,P=0.067).Further analysis showed that the ratio of the maximum AUC for M1-GRK2/3 interaction to that for M1-GRK5/6 interaction was also positively correlated with the ratio of the maximum AUC for M1-βarr2 interaction to that for M1-G protein interaction(r=0.760,P=0.047).Conclusion·The M1 receptor may be pre-coupled with GRK5/6 under basal conditions,and they dissociate upon receptor activation,suggesting that GRK5/6 may be involved in M1 receptor inactivation or signal reprogramming.The relative efficiency of the M1 receptor's interaction with different GRK subtypes determines its preference for downstream signaling pathways.
9.Quality Analysis and Suggestion of Zukamu Preparation Based on National Drug Sampling and Testing

Objective To examine the quality of Zukamu preparations through the national drug sampling and testing,and further understand their current quality status and existing problems.This work is benefit for improving the quality standard of Zukamu preparations and providing technical support for the drug regulatory authorities.Methods Samples of Zukamu preparations were collected from a total of 29 provinces in China,and were tested for description,identification,other requirements(weight variation,particle size,determination of water,disprsion,and microbial limit items),and assay in accordance with the national pharmaceutical standards.The test data were analyzed to evaluate the quality status of the Zukamu preparations,and exploratory research was carried out to address the problems found in the test.Results A total of 97 batches of Zukamu preparations were sampled,and the passing rate was 100.0%according to the current quality standard.Exploratory study,revealed that Zukamu preparation were subject to 4 testing standards,with uneven test items,missing items,poor operability,and lack of exclusivity in some items.The test based on the existing standards can't comprehensively evaluate the quality of the preparation.Conclusions Based on the national drug sampling and testing,combined with exploratory research on drug safety,authenticity and effectiveness,it is recommended to unify the quality standards of Zukamu preparations by combining with the work of standard improving,revising the identification method of thin-layer chromatography,increasing the content determination,and establishing the quick test method,thereby effectively evaluating and controling the quality of the samples of Zukamu preparations.
10.Application of exercise interventions in the rehabilitation of patients with hemophilic arthropathy: a scoping review

Objective:To conduct a scoping review of studies on the application of exercise interventions in the rehabilitation of patients with hemophilic arthropathy (HA) .Methods:A systematic search was conducted in PubMed, Web of Science, Cochrane Library, Embase, CINAHL, China National Knowledge Infrastructure, Wanfang Data, VIP, and China Biology Medicine disc. The search period was from the database inception to March 1, 2024. Relevant literature was screened, summarized, and analyzed.Results:A total of 31 studies were included. The study population mainly consisted of adult patients with moderate to severe hemophilia suffering from elbow, knee, and ankle arthropathy, who were in a stable bleeding phase without coagulation factor inhibitors. Exercise types included resistance training, aerobic exercise, balance/proprioceptive exercises, and flexibility training. The exercise intensity was predominantly low to moderate. The duration of each session was mostly 30-60 minutes, with a frequency of 2-5 times per week and a total intervention period of 4-12 weeks. Exercises were mostly conducted in medical institutions under face-to-face supervision. Exercise interventions led to varying degrees of improvement in mobility, balance/proprioceptive function, joint function, cardiopulmonary fitness, psychological well-being, and quality of life.Conclusions:Exercise interventions for patients with hemophilic arthropathy are diverse, demonstrating safety, feasibility, and efficacy. Future research should focus on a multidisciplinary, exercise-centered rehabilitation approach, standardizing evaluation criteria, and dynamically monitoring the rehabilitation process to develop standardized exercise rehabilitation protocols.
