1.Efficiency of proteomics combined with transcriptomics in identifying differentially expressed genes in sepsis-induced lung injury and independent sample validation

Objective To investigate biomarkers for sepsis-induced lung injury based on results of proteomics combined with transcriptomics analyses.Methods A total of 70 patients with sepsis and 70 patients with sepsis-induced lung injury were included as research objects.These patients were di-vided into experimental group and validation group.The experimental group included 10 patients with sepsis and 10 patients with sepsis-induced lung injury.Proteomics was used to analyze differentially expressed proteins in plasma,and Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analyses were performed.The dataset GSE10474 of sepsis-induced lung injury was downloaded from the Gene Expression Omnibus(GEO),and GEO2R online database was used to analyze differential transcriptomics data for sepsis-induced lung injury.A Venn diagram was used online to analyze common differentially expressed genes related to sepsis-induced lung injury in pro-teomics and transcriptomics.The validation group included 60 patients with sepsis(sepsis group)and 60 patients with sepsis-induced lung injury(sepsis-induced lung injury group).Enzyme-linked immunosorbent assay(ELISA)was used to detect and compare the differences in protein expression level in peripheral blood between the sepsis group and the sepsis-induced lung injury group.Receiv-er operating characteristic(ROC)curve was used to analyze the clinical value of differential protein expression level in distinguishing sepsis and sepsis-induced lung injury.Results Proteomics results confirmed the presence of 239 significantly differentially expressed proteins in the plasma of patients with sepsis and sepsis-induced lung injury.Compared with patients with sepsis,there were 96 sig-nificantly upregulated proteins and 143 significantly downregulated proteins in patients with sepsis-induced lung injury.The results of GO enrichment analysis of differentially expressed proteins in-cluded cytoplasm,microtubule binding,adenosine triphosphate(ATP)binding,defense response to virus,and immune response.The results of KEGG enrichment analysis included metabolic path-ways,interleukin-17(IL-17)signaling pathway,and phosphatidylinositol-3-kinase-protein kinase B(PI3K-Akt)signaling pathway.In the GSE10474 dataset,compared with patients with sepsis,there were 77 significantly upregulated genes and 142 significantly downregulated genes in patients with sepsis-induced lung injury.The Venn diagram results showed that there were 6 common differential-ly expressed genes in proteomics and transcriptomics,namely BTNL8,FCGR2B,TAK1,KCNC1,TREM1,and SEC31A.Compared with the sepsis group,the levels of TAK1 and TREM1 proteins in the peripheral blood of the sepsis-induced lung injury group were significantly increased(P<0.01).ROC curve showed that the areas under the curve(AUC)for the expression levels of TAK1 and TREM1 proteins in serum to distinguish sepsis and sepsis-induced lung injury were 0.925 and 0.785 respectively;when the cut-off value for TAK1 was 71.28 pg/mL,the sensitivity and specific-ity were 94.45%and 97.89%respectively;when the cut-off value for TREM1 was 58.22 mg/mL,the sensitivity and specificity were 83.43%and 82.19%respectively.Conclusion Proteomics and transcriptomics results confirm that the activation of TAK1,TREM1 and multiple inflammatory signa-ling pathways may play important roles in the progression of sepsis-induced lung injury.

3.Design and Implementation of the Glasses-free Three Dimensional Laparoscopy System

Minimally invasive surgery (MIS) has the advantages of small trauma, quick recovery and lower risk of the surgery.Compared with the traditional two-dimension laparoscopic technique, three-dimension laparoscopic system uses the double beam imaging system as well as the three-dimensional imaging display device so as to compensate for the lack of depth information in two-dimension laparoscopic imaging technique, reduce the surgeon's difficulty in using surgical instruments and enhance the accuracy of the surgery. For those reasons, the author designed a glassesfree three dimensional wide-field electronic laparoscopic system, which consists of dual CMOS camera module,image processing module and glasses-free display module that can offer depth information. The active shape model is applied to eye tracking, for the purpose of getting the wide-field to help doctor obtain 3D display in more places.Primary experimental results of the simulation using the system show that operators with the aid of proposed system spend less time finishing the task than those using traditional system without depth perception during operation.
4.Protein expression and biological characteristics of colon cancer stem cells:culture and screening

BACKGROUND:EpCAMhighCD44+ colon cancer stem cels have been isolated in recent years, and most of them are related to the study of isolating tumor stem cels in colorectal cancer cel lines. There are less reports on the function of colon cancer stem cels. OBJECTIVE:To isolate, screen and culture colon cancer stem cels from colorectal carcinoma tissues and to explore the protein expression and biological characteristics of colon cancer stem cels. METHODS: The primary cels were isolated from the colon cancer tissues. EpCAMhighCD44+ cels were detected and sorted by flow cytometry. Then, these cels were cultured in serum-free medium. After the cels were replanted subcutaneously into the mice, we observed the daily performance and tumor growth in the mice. When the tumor diameter was above 1 cm, tumor tissues were taken for immunohistochemical testing. The growth of EpCAMhighCD44+ cels cultured in the serum-free medium was observed; expressions of neutral epithelial mucin and CK-20 in EpCAMhighCD44+ cels were detected. RESULTS AND CONCLUSION: EpCAMhighCD44+ colon cancer stem cels were detected in the colon cancer tissues, and the proportion of EpCAMhighCD44+ cels in primary colon cancer cels was 1.7%-38%. After 24 hours of serum-free culture, there were a few of smal suspended cel spheres which were incompact. After 21 days, dense cel spheres with uniform size were found. The formation of transplanted tumor was found in the nude mice. Hematoxylin-eosin staining showed the transplanted tumor had the typical characteristics of colon cancer cels. The expression of neutral epithelial mucin and CK-20 was observed in al the transplanted tumors. Additionaly, the proliferation of EpCAMhighCD44+ cels could be promoted by 5-fluorouracil (10, 1, and 0.1 PPC) with time. These findings indicate that EpCAMhighCD44+ colon cancer stem cels in the colon cancer have a certain ability of proliferation, regeneration, differentiation, tumor formation and chemotherapy resistance.
5.Study on Quality Standard for Compound Jieyu Granules

Objective:To establish the quality control method for compound Jieyu granules. Methods:Liquorice was identified by TLC. saikosaponin a, Saikosaponin d and rutin were determined by HPLC. Results:The spots on TLC plates were clear without any in-terference. The linearity was achieved within the range of 0. 508-16. 200 μg (r=0. 999 8) for saikosaponin a, 0. 503-16. 100 μg(r=0.999 7) for saikosaponin d, and 0.130-4.250 μg(r =0.999 9) for rutin. The average recovery was 99.7%(RSD =2.03%), 99. 8%(RSD=1. 44%) and 102. 6%(RSD=1. 40%), respectively. Conclusion:The method is simple, reliable and accurate, and can be applied as the quality control method for compound Jieyu granules.
6.Effect of sodium hyaluronate on malondialdehyde and superoxide dismutase levels in the synovial fluid of patients with knee osteoarthritis

BACKGROUND:Intra-articular injection of sodium hyaluronate is an effective method for the treatment of knee osteoarthritis, with significant effect and less adverse reactions, but the mechanism is unclear. OBJECTIVE:Through testing the malondialdehyde and superoxide dismutase levels in the synovial fluid of knee osteoarthritis before and after injection of sodium hyaluronate, to evaluate the clinical efficacy of sodium hyaluronate in the treatment of knee osteoarthritis. METHODS:Thirty-seven patients with knee osteoarthritis (40 knees) were enroled and divided into mild (n=10, 10 knees), moderate (n=17, 18 knees), and severe (n=10, 12 knees) groups according to the Japan's knee osteoarthritis indications. Patients were subjected to intra-articular injection of 25 mg sodium hyaluronate, once a week for 5 weeks. The levels of malondialdehyde and superoxide dismutase in the synovial fluid before and 4 weeks after treatment were detected, and then clinical effects were evaluated based on the clinical scores according to the Japan’s knee osteoarthritis indications. RESULTS AND CONCLUSION: The indication rating results of the mild and moderate groups were decreased significantly 4 weeks after injection (P < 0.05), but there were no significant difference in the severe group before and after treatment. The malondialdehyde level in the synovial fluid was decreased obviously in the three groups at 4 weeks after injection (P < 0.05), while the level of superoxide dismutase was increased remarkably (P < 0.05). These findings indicate that sodium hyaluronate can treat knee osteoarthritis by reducing the malondialdehyde level and increasing superoxide dismutase level in the synovial fluid, but this method is more suitable for treatment of mild to moderate knee osteoarthritis.
7.Three-dimensional ultrasound locatization in endoscopic thyroidectomy for multinodular goiter.

OBJECTIVE:
To explore the three-dimensional ultrasound locatization in endoscopic thyroidectomy for multinodular goiter (MG).
METHOD:
Thirty two patients with MG underwent ET via bilateral areolar approach. Preoperative ultrasonography were used to locate lesions and marked in the 3-D map designed by us. Dur ing ET procedure, surgery skills were adopted to expose inferior pole of thyroid, glands in the area of Berry ligament or (and) upper pole were retained according to the preoperative location map.
RESULT:
According to the preoperative ultrasonic orientation, 3 lobectomy were performed in 15 cases of unilateral lesions, 12 subtotal thyroidectomy were performed in 17 cases of bilateral lesions, and 5 patients performed unilateral lobectomy add partial thyroidectomy. All patients had successful ET using bilateral areola approach. No conversions to open surgery, no complication occurred, and no nodules were found missing in the postoperative ultrasonography examinations.
CONCLUSION
Preoperative ultrasonic orientation and intraoperative operating skills are contributing to a successful ET for MG.
