1.Association between mental health status and adverse childhood experiences among sexual minority college students in Guangxi

Objective:
To explore the association between mental health status and adverse childhood experiences (ACEs) among sexual minority college students, so as to provide a scientific basis for mental health education and health promotion in universities.
Methods:
From January to February 2024, convenience and cluster sampling methods were used to select 1 792 college students from 11 colleges in Guangxi. A self reporting method was applied to identify 476 sexual minority individuals. The Symptom Check-List 90 (SCL-90) and the Simplified Chinese Adverse Childhood Experiences International Questionnaire (SC-ACE-IQ) were employed to assess mental health and ACEs. Multivariate Logistic regression analysis was conducted to examine the associations.
Results:
The detection rates of all psychological issues among sexual minority college students in Guangxi were significantly higher than those of non sexual minority college students ( χ 2=56.01-91.39, all P <0.01). Except for physical neglect, bullying, and community violence, sexual minority students exhibited higher reporting rates of other ACEs types compared to nonsexual minority students ( χ 2= 4.52-13.34, all P <0.05). The total ACEs score for college students was 1.00 (1.00, 2.00), while the SCL-90 total score was 96.00 (113.00, 160.00). Spearman correlation analysis revealed a positive correlation between ACEs total scores and SCL-90 total scores ( r=0.29, P <0.05). Additionally, all ACEs subscales, including emotional neglect, physical neglect, emotional abuse, sexual abuse, parental loss, domestic violence, and community violence were positively correlated with corresponding SCL-90 subscale scores ( r =0.05-0.22, all P <0.05). Multivariate Logistic regression analysis showed that family violence increased the risk of mental health issues for sexual minority students ( OR=1.61, 95%CI =1.26-2.09); emotional neglect ( OR= 1.05 , 95%CI =1.00-1.10), physical neglect ( OR=1.20, 95%CI =1.06-1.35), sexual abuse ( OR=1.49, 95%CI =1.15-1.93) increased mental health risks for non sexual minority students (all P <0.05). The cumulative effects of ACEs were all statistically significant in the total sample and both subgroups (all P <0.05).
Conclusion
Mental health status among sexual minority college students in Guangxi is associated with ACEs, and their well being requires active attention

Myocardial fibrosis is a serious cause of heart failure and even sudden cardiac death. However, the mechanisms underlying myocardial ischemia-induced cardiac fibrosis remain unclear. Here, we identified that the expression of sterile alpha and TIR motif containing 1 (SARM1), was increased significantly in the ischemic cardiomyopathy patients, dilated cardiomyopathy patients (GSE116250) and fibrotic heart tissues of mice. Additionally, inhibition or knockdown of SARM1 can improve myocardial fibrosis and cardiac function of myocardial infarction (MI) mice. Moreover, SARM1 fibroblasts-specific knock-in mice had increased deposition of extracellular matrix and impaired cardiac function. Mechanically, elevated expression of SARM1 promotes the deposition of extracellular matrix by directly modulating P4HA1. Notably, by using the Click-iT reaction, we identified that the increased expression of ZDHHC17 promotes the palmitoylation levels of SARM1, thereby accelerating the fibrosis process. Based on the fibrosis-promoting effect of SARM1, we screened several drugs with anti-myocardial fibrosis activity. In conclusion, we have unveiled that palmitoylated SARM1 targeting P4HA1 promotes collagen deposition and myocardial fibrosis. Inhibition of SARM1 is a potential strategy for the treatment of myocardial fibrosis. The sites where SARM1 interacts with P4HA1 and the palmitoylation modification sites of SARM1 may be the active targets for anti-fibrosis drugs.

Osteoporosis is a prevalent skeletal condition characterized by reduced bone mass and strength, leading to increased fragility. Buqi-Tongluo (BQTL) decoction, a traditional Chinese medicine (TCM) prescription, has yet to be fully evaluated for its potential in treating bone diseases such as osteoporosis. To investigate the mechanism by which BQTL decoction inhibits osteoclast differentiation in vitro and validate these findings through in vivo experiments. We employed MTS assays to assess the potential proliferative or toxic effects of BQTL on bone marrow macrophages (BMMs) at various concentrations. TRAcP experiments were conducted to examine BQTL's impact on osteoclast differentiation. RT-PCR and Western blot analyses were utilized to evaluate the relative expression levels of osteoclast-specific genes and proteins under BQTL stimulation. Finally, in vivo experiments were performed using an osteoporosis model to further validate the in vitro findings. This study revealed that BQTL suppressed receptor activator of NF-κB ligand (RANKL)-induced osteoclastogenesis and osteoclast resorption activity in vitro in a dose-dependent manner without observable cytotoxicity. The inhibitory effects of BQTL on osteoclast formation and function were attributed to the downregulation of NFATc1 and c-fos activity, primarily through attenuation of the MAPK, NF-κB, and Calcineurin signaling pathways. BQTL's inhibitory capacity was further examined in vivo using an ovariectomized (OVX) rat model, demonstrating a strong protective effect against bone loss. BQTL may serve as an effective therapeutic TCM for the treatment of postmenopausal osteoporosis and the alleviation of bone loss induced by estrogen deficiency and related conditions.

4.Construction of a high-efficiency risk-prediction model for urinary tract infection after BPH surgery based on machine learning algorithm

Objective To explore the risk factors for urinary tract infection(UTI)after benign prostatic hyperpla-sia(BPH)surgery,and construct a high-efficiency risk-prediction model based on machine learning algorithms(MLA).Methods This retrospective study included 200 cases of BPH surgically treated in Xinyu Hospital of Traditional Chinese Medicine from May 2020 to April 2023.Twenty-three cases were included in the UTI group,and 177 were enrolled into the non-UTI group.The relevant data were collected and analyzed for the statistically significant factors by univariate logistic analysis by using the SPSS software,MLA-based logistic regression,back-propagation(BP)neural networks and decision classification and regression tree(CRT).And the predictive values of the models established via different algorithms using the area under the receiver operating characteristic curve(AUC)were compared.We assessed the prediction accuracy of the models and identified one with the best prediction efficiency based on the mean absolute error(MAE).Results Univariate analysis indicated statisti-cally significant differences between the UTI and non-UTI groups in age,comorbid diabetes mellitus(DM),urinary catheter-in-dwelling time,prostate volume,preoperative catheterization,preoperative IPSS(P<0.05).The independent predictive varia-bles for UTI after BPH surgery were shown to be age,IPSS,comorbid DM and prostate volume by the method of multivariate lo-gistic regression model(P<0.05).Age,urinary catheter-indwelling time,prostate volume and IPSS were assessed as the influ-ence factors by the CRT model(P<0.05),and prostate volume,IPSS,age and urinary catheter-indwelling time were assessed as the influence factors by the BP neural network model(P<0.05).Among the 3 risk-prediction models,the one constructed with the BP neural networks exhibited the best prediction efficiency(AUC:0.992,the optimal truncation value:0.912,corre-sponding sensitivity and specificity:0.957 and 0.955).Conclusion The risk-prediction model constructed by MLA and BP neural networks based on the characteristic factors of age,preoperative urinary retention catheterization,urinary catheter-indwell-ing time,IPSS and comorbid DM has a high predictive value for UTI after BPH surgery which can be applied to the identification and management of such high-risk population.
5.Analysis on effective components and biological activity of volatile oil in anti-URTI formula

Objective:To analyze the extraction, chemical composition, antioxidant, and anti-inflammatory activity of the TCM formula essential oil for the treatment of upper respiratory tract infections (URTI); To provide a scientific basis for its further development.Methods:The formula essential oil was extracted using the steam distillation method and analyzed for chemical composition by gas chromatography-mass spectrometry (GC-MS). The DPPH, ABTS scavenging ability, and hydroxyl radical scavenging ability of volatile oils were measured. The effect of the essential oil on the viability of RAW264.7 cells was assessed using the CCK-8 assay. ELISA and Western blot methods were used to determine the effects of volatile oil on LPS induced inflammatory cytokines IL-6 and TNF-α.Results:The average extraction rate of the formula essential oil was 1.12%, with a density of 0.973 2 g/ml. Twelve main chemical components were identified, with 1,8-cineole (42.9%) and patchoulol (19.9%) being the predominant constituents. The essential oil exhibited DPPH and ABTS radical scavenging capacities of 52% and 59%, respectively, and a hydroxyl radical scavenging capacity exceeding 70%. Essential oil could reduce the levels of IL-6 and TNF-α ( P<0.05). Conclusion:TCM formula essential oil for the treatment of URTI contains multiple bioactive components and demonstrates significant antioxidant and anti-inflammatory effects.
6.Combined nutritiona-immunological-inflammatory indexes for assessing pulmonary tuberculosis severity

Objective :
To explore the value of combined indicators of prognostic nutritional index ( PNI) ,nutri- tional risk screening 2002 scale ( NRS2002 ) ,body mass index ( BMI) ,monocyte / lymphocyte ratio ( MLR) , platelet / lymphocyte ratio ( PLR) ,and neutrophil / lymphocyte ratio ( NLR) in evaluating the severity of pulmonary tuberculosis ( PTB) .
Methods :
A total of 175 patients with pulmonary tuberculosis were selected as the study group,and 175 healthy subjects who underwent physical examination during the same period were selected as the control group.According to the range of lung lesions,the patients were divided into mild to moderate group ( <3 lung fields,n = 110) and severe group ( ≥3 lung fields,n = 65) ,PNI,NRS2002,BMI and peripheral blood MLR , PLR and NLR levels were compared between the two groups.Spearman rank correlation was used to analyze their correlation with the severity of the disease.A multivariate logistic regression model was established.A nomo- gram was drawn,and the efficiency of the model was evaluated by receiver operating characteristic ( ROC) curve, calibration curve,decision curve.
Results:
The PNI,BMI,peripheral blood lymphocyte and albumin levels in the study group were lower than those in the control group ( P<0. 05) ,while the MLR , PLR and NLR levels were sig- nificantly higher than those in the control group ( P<0. 05) .The NRS2002,MLR , PLR and NLR levels in the se- vere group were higher than those in the mild and moderate group ( P<0. 05) ,while the PNI and BMI levels were lower than those in the mild and moderate group ( P<0. 05) .NRS2002 and peripheral blood MLR , PLR and NLR levels in patients with pulmonary tuberculosis were positively correlated with the severity of the disease ( r = 0. 250, 0. 509,0. 431 and 0. 488) .PNI and BMI were negatively correlated with the severity of the disease ( r = -0. 516, - 0. 231) .Multivariate Logistic regression showed that NRS2002 and NLR were independent risk factors for severe disease,while PNI was a protective factor.The areas under the curve ( AUC) of NRS2002,NLR , PNI,and com- bined detection for evaluating the severity of pulmonary tuberculosis were 0. 692,0. 777,0. 786,and 0. 860,re- spectively,the sensitivity of the combined prediction was 81. 54% and the specificity was 76. 36%.The combined detection had better evaluation efficiency for severe pulmonary tuberculosis than the single indicator ( P <0. 05) .
Conclusion
NRS2002 and NLR are independent risk factors for severe disease,while PNI is a protective factor. The combined detection model has a good fit,which can improve the evaluation efficiency and has potential for clin- ical application.
7.Establishment of Fingerprints of Danggui Buxue Granules with Different Drying Processes and Study of the Spectrum-Effect Relationship of Antioxidant Activity

OBJECTIVE To establish HPLC fingerprints of three drying processes(Atmospheric pressure drying,Decompression drying,Freeze-drying)of Danggui Buxue granules,and combine them with antioxidant tests to determine the optimal drying process and main active components of Danggui Buxue granules.METHODS The fingerprints of multiple batches of Danggui Buxue granules were established by HPLC;the"Fingerprint Similarity Evaluation System of Traditional Chinese Medicine Chromatogram"was used to evaluate the similarity;Hierarchical Cluster Analysis(HCA)and Principal Component Analysis(PCA)were used to characterize the different drying processes of Danggui Buxue granules;evaluate the antioxidant activity of Danggui Buxue granules in different drying processes using 1,1-diphenyl-2-picrylhydrazy(DPPH)and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt(ABTS)free radical scavenging method;the spectrum-effect relationship between Danggui Buxue granules fingerprint and antioxi-dant activity was analyzed by grey correlation degree and Pearson,Spearman,Kendall's tau-b correlation analysis methods.RE-SULTS The results of fingerprint showed that there were 16 common peaks in 30 batches of Danggui Buxue granules,and 6 of them were identified by comparison.The results of Cluster Analysis and Principal Component Analysis showed that 30 batches of Danggui buxue granules were divided into 3 categories,and the difference between the groups of atmospheric pressure drying was the least.The results of oxidation test showed that different drying processes of Danggui Buxue granules had good antioxidant activity,and the atmos-pheric pressure drying had the lowest IC50 and the strongest antioxidant activity.Finally,combining the results of gray correlation anal-ysis and correlation analysis,the compounds F2,F10,F13(calycosin),F15(formononetin),F16 might be important characteristic peaks reflecting the antioxidant activity of Danggui Buxue granules.CONCLUSION Compared with other drying processes,atmos-pheric pressure drying has higher batch consistency and stronger antioxidant activity,and can be used as the preferred drying process for Danggui Buxue granules,and components 2,10,13,15 and 16 are the main active ingredients for Danggui Buxue granules to exert antioxidant effects.
8.Medical resource consumption of healthcare-associated infection based on disease diagnosis-related grouping payment model

Objective To analyze the medical resource consumption of healthcare-associated infection(HAI)in patients in different groups of disease diagnosis-related grouping(DRG)based on the DRG payment model,provide reference for optimizing prevention and control of HAI as well as resource management.Methods Medical records and DRG-related indicators of discharged patients from a municipal hospital in Sichuan Province from January 1 to December 31,2024 were analyzed retrospectively.Medical resource consumption of patients in HAI group and non-HAI group was compared.Differences in average length of hospital stay and average expense per hospitalization be-tween two groups of patients were analyzed using stratified analysis.Results In 2024,HAI incidence of discharged patients in DRG management in this hospital was 1.57%.There were statistically significant differences in age,gender,admission and discharge ways between the HAI group and the non-HAI group(all P<0.05).The main HAI sites were lower respiratory tract,surgical site,urinary tract,and blood.The time consumption index(1.63 vs 0.85),average length of hospital stay(21.00 vs 5.00 days),expense consumption index(1.53 vs 0.92),ave-rage expense per hospitalization(44 700 vs 7 300),and multiple expense in HAI group were all higher than those in non-HAI group(all P<0.05).The consumption of medical resources for bloodstream infection was relatively higher.Patients with HAI were mostly concentrated in the groups related to acute leukemia with major complications or co-morbidities(MCC),intracranial or craniotomy surgery with MCC,tracheotomy with mechanical ventilation for 96 hours,as well as gastric,esophageal,and duodenal surgery.The average length of hospital stay and average ex-pense per hospitalization of patients in HAI group were both higher than those in the non-HAI group,differences were statistically significant(both P<0.05).Conclusion HAI significantly increase the consumption of medical resources.Based on DRG analysis,key disease groups for infection prevention and control can be further identified,and the consumption of medical resources can be more accurately and precisely evaluated,thereby optimizing the allocation of medical resources and improving hospital operational efficiency.
