Diamond-Blackfan anemia(DBA),also known as congenital pure red cell aplasia,is a rare genetic disorder characterized by bone marrow failure,congenital anomalies,and severe red blood cell abnormalities.The rarity of the condition,and consequently limited patient pool and scarcity of research models,means that the pathogenic mechanisms associated with genetic mutations in DBA remain uncertain,and the clinical treatment options are limited.This review synthesizes the findings from zebrafish,mouse,and human cellular models of DBA mutations.We clarify the pathogenic mechanisms and monitor the progression of drugs into clinical trials,thereby aiding further in-depth explorations into the etiology and therapeutic advancements for DBA.

3.Acute pancreatitis induced by tigecycline in a patient after renal transplantation

A 25-year-old male patient received intravenous infusion of tegacyclin 50 mg once per 12 hours from the 3rd day after renal transplantation because the bacterial culture of the donor kidney preservation solution showed carbapenem resistant Acinetobacter baumannii. On the 4th day after using tegacyclin, the patient developed epigastric pain, which was improved after gastric protection treatment. On the 8th day of medication, the patient developed persistent severe pain in his left lower abdomen. On the 9th day of medication, laboratory tests showed amylase 1 188 U/L and lipase 699 U/L. Pancreatic ultrasonic examination suggested acute edematous pancreatitis. Acute pancreatitis was diagnosed, which was considered to be related to tigecycline. Tigecycline was stopped. An IV infusion of octreotide acetate injection 0.6 mg dissolved in 0.9% sodium chloride injection 50 ml twice daily was given and diet was prohibited at the same time. Five days later, his symptoms of abdominal pain was improved significantly, pancreatic amylase decreased to 69 U/L, and lipase decreased to 646 U/L.
4.Nivolumab-induced severe ulcerative colitis

A 64-year-old male patient with gastric adenocarcinoma received chemotherapy with oxaliplatin (250 mg intravenous infusion on day 1) and tegafur gimeracil oteracil potassium (60 mg orally twice daily on day 1 to day 14, 21 days as a cycle). One month later, he was given nivolumab immunotherapy (240 mg intravenous infusion on day 1, 14 days as a cycle). On the 2nd day of the 4th cycle of nivolumab treatment (the 44th day after the first dose), the patient developed diarrhea with watery stools, 3 to 4 times a day, accompanied by mild abdominal pain; 3 days later, the diarrhea worsened, showing dark red bloody stools, covered with pus moss, up to 10 times a day, accompanied by obvious abdominal pain. According to colonoscopy and histological examination results, ulcerative colitis (initial onset, total colon, active stage, and severe) was diagnosed, which was considered as a grade 3 immune-related colitis induced by nivolumab. After suspension of chemotherapy and immunotherapy, and administration of glucocorticoids and octreotide, the patient′s abdominal pain and diarrhea were gradually improved. On the 45th day of nivolumab withdrawal, the patient received chemotherapy with oxaliplatin and tegafur gimeracil oteracil potassium again, and colitis did not recur.

7.Biomass carbohydrates assimilation and lipid accumulation by Mortierella isabellina.

Biomass carbohydrates assimilation and lipid accumulation by Mortierella isabellina M2 strain were investigated. Corn fiber hydrolysate was specially studied. The results showed M. isabellina M2 strain achieved growth and lipid accumulation while glucose, xylose, mannose and arabinose were introduced as single carbon source, respectively. When M. isabellina M2 strain was cultivated on corn fiber hydrolysate with 6% sugars concentration, the biomass reached 18.2 g/L, the lipid content of dry mycelia was 45.7%, and the lipid yield achieved 8.3 g/L. It provided a promising perspective for microbial oils production with biomass hydrolysates.

8.Effect of culture conditions on cell growth and lipid accumulation of oleaginous microorganism.

We used four strains (Y1, Y2, M1 and M2) to screen out the high lipid production strains. We first adopted cell morphology and cytochemical methods (Sudan III dyeing technique) to observe intracellular characteristics. Observation results indicated that M2 strain had the potential lipid accumulation capacity. To prove this, lipid content of these strains was determined by soxhlet extraction. One strain (M2) was found to produce lipids up to 53.09%. In order to increase the production of oleaginous microorganism, the effects of hydrolysate concentration, nitrogen source, pH, fermentation temperature and time on cell growth and lipid accumulation were studied. The optimal fermentation conditions were obtained as follows: corn starch byproduct hydrolysate concentration at 10(o) Bx as carbon source; NaNO3 as nitrogen source at 0.2%; initial pH of 6.0; temperature at 28 degrees C, cultivated for 6 d. Under these conditions, M2 strain accumulated lipids up to 75.21% on a cellular biomass basis with biomass yield of 30.40 g/L, and the corresponding lipid production reached 22.86 g/L. GC analysis demonstrated that the fatty acid composition of the lipid was similar to that of vegetable oil, which mainly contained 16-and 18-carbon fatty acids. Thus, microbial lipid is a promising material for biodiesel production, and its unsaturated fatty acid content reached around 68%. These unsaturated fatty acids show great potential applications in food, medicine and cosmetics industries.

9.Correlation among expression of CD44, DNA content and apoptosis in female adenocarcinoma of the lung.

BACKGROUNDCluster of differentiation 44 (CD44) is a family of transmembrane glycoproteins. As cell surface hyaluronate receptor, it has been found to be widely expressed in a variety of cells. The aim of this study is to assess the relationship among CD44 expression, DNA content, proliferation index (PI) and apoptosis in female adenocarcinoma of the lung.
METHODSThe expression of CD44, DNA index (DI), PI and apoptotic rate were studied in 61 cases of female adenocarcinoma of the lung, paracancerous tissues and 45 cases of benign lesions by flow cytometry.
RESULTSThe percent of DNA aneuploidy was 75.41% in adenocarcinoma. The expression of CD44, DI and PI in adenocarcinoma were significantly higher than those in paracancerous and benign controls (P < 0.01), however the apoptotic rate was obviously lower in adenocarcinoma than that in paracancerous and benign controls (P < 0.01). There was a positive correlation between CD44 and DI (P < 0.01), and a negative correlation between apoptosis and DI in adenocarcinoma (P < 0.01).
CONCLUSIONSThe expression of CD44, DNA content, proliferation and apoptosis may play important regulating roles in oncogenesis, development and metastasis of female adenocarcinoma of the lung.
