Auto-inhibited Ca2+-ATPase (ACA) is one of the Ca2+-ATPase subfamilies that plays an important role in maintaining Ca2+ concentration balance in plant cells. To explore the function and gene expression pattern of the RcACA gene family in castor, bioinformatics analysis was used to identify the members of the RcACA gene family in castor. The basic physical and chemical properties, subcellular location, protein secondary and tertiary structure, conserved domain, conserved motif, gene structure, chromosome location and collinear relationship, as well as the evolutionary characteristics and promoter cis-acting elements were predicted and analyzed. The expression pattern of the RcACA gene under abiotic stress was analyzed by expression (fragments per kilobase of exon model per million mapped fragments, FPKM) in castor transcriptome data. The results showed that 8 RcACA gene family members were identified in castor, acidic proteins located in the plasma membrane. In the secondary structure of all proteins, the α-helix and random coil is more; the RcACA genes were clustered into three categories, and the design of the genes in the same category was similar to the conserved motif. Both of them had four typical domains, RcACA3-RcACA8 had a Ca2+-ATPase N-terminal autoinhibitory domain. The RcACA gene is mostly located on the long arm of the chromosome and has 2 pairs of collinear relationships. There are more light response elements but fewer hormone-induced elements located upstream of the RcACA coding region. Interspecific clustering showed that the evolution of ACA genes among species was conservative. Tissue expression pattern analysis showed that RcACA genes showed apparent tissue expression specificity, and most of the genes showed the highest expression level in male flowers. Expression analysis under abiotic stress showed that RcACA2-RcACA8 were up-regulated under high salt and drought stress, and RcACA1 was up-regulated at 0-24 h under low-temperature stress, indicating that RcACA genes positively responded to abiotic stresses. The above results provide a theoretical basis for exploring the role of the RcACA gene in castor growth, development and stress response.

Neuroscience Bulletin 2022;38(8):953-965
Although posttraumatic stress disorder (PTSD) is on the rise, traumatic events and their consequences are often hidden or minimized by patients for reasons linked to PTSD itself. Traumatic experiences can be broadly classified into mental stress (MS) and traumatic brain injury (TBI), but the cellular mechanisms of MS- or TBI-induced PTSD remain unknown. Recent evidence has shown that the morphological remodeling of astrocytes accompanies and arguably contributes to fearful memories and stress-related disorders. In this review, we summarize the roles of astrocytes in the pathogenesis of MS-PTSD and TBI-PTSD. Astrocytes synthesize and secrete neurotrophic, pro- and anti-inflammatory factors and regulate the microenvironment of the nervous tissue through metabolic pathways, ionostatic control, and homeostatic clearance of neurotransmitters. Stress or trauma-associated impairment of these vital astrocytic functions contribute to the pathophysiological evolution of PTSD and may present therapeutic targets.

The Korean Journal of Internal Medicine 2019;34(4):932-937
BACKGROUND/AIMS:
To evaluate cellular immune function in systemic lupus erythematosus (SLE) patients over 60 years old, the association between antinuclear antibody (ANA) titers and the ratio of CD4âº/CD8⺠was analyzed in this study. The distribution of ANAs and extractable nuclear antibodies (ENAs) in a healthy elderly population was also investigated.
METHODS:
Serum ANA titers were assayed by indirect immunofluorescence (IIF) and the CD4âº/CD8⺠T-cell ratio was determined by flow cytometry in 76 SLE patients and 30 healthy control individuals. IIF of cytoplasm and nuclear and nucleolar staining were performed on samples taken from 286 healthy elderly individuals. ENA levels were determined using a strip enzyme immunoassay among patients that tested positive for ANAs.
RESULTS:
ANA titers were negative in the 30 control individuals, but were positive in the 76 SLE patients. Based on ANA titers, the SLE patients were stratified to low (≤ 1:320), medium (1:640 to 1:1,280), and high (≥ 1:2,560) titer groups. The average CD4âº/CD8⺠ratio of the SLE group was significantly lower than that of the control group. Among the 286 healthy elderly volunteers, 59 (20.63%) tested positive for ANAs. A homogeneous pattern was present in 47.46% of those 59 patients and a granule pattern in the karyoplasm was present in 33.90%. Furthermore, of the 59 patients, ENAs immunoassay was positive in 18 (30.51%); Sjogren syndrome-related antigen A (SSA)/52 kd and Sjogren syndrome antigen B (SSB)/La were the two major antibodies.
CONCLUSIONS
The significantly lower CD4âº/CD8⺠ratio among SLE patients over 60 years old is associated with deregulated immune responses and the development of SLE. A low ANA titer (1:160) is common in healthy elders, emphasizing the importance of considering age when determining if the evaluation of ANA titers is to be included in autoimmune disease diagnosis.

Objective To explore the mechanism of pigment epithelium-derived factor (PEDF) on human liver cells in mitochondrial apoptosis induced by arsenic. Methods Human normal liver cell line (HHL-5) was exposed to NaAsO2 for 24 h, the dose was 0 (control), 2, 5, 8 and 10 μmol/L, respectively. The mitochondrial membrane potential was determined by using flow cytometry, and mRNA levels of PEDF, Bcl-2 and Bax were assayed by Real-time fluorescent quantitative PCR. Results The mitochondrial membrane potential in 10 μmol/L arsenic group (562.33 ± 31.50) was decreased markedly in comparison with that of control group (688.67 ± 47.50, P<0.01), the levels in other groups (5 μmol/L: 634.50 ± 49.03, 8 μmol/L: 655.67 ± 13.65) were not significantly changed (P > 0.05). Compared with control group [(86.69 ± 12.19)%], the mRNA levels of PEDF decreased significantly (P<0.01) when the HHL-5 cells were cultivated with 5 μmol/L [(59.35 ± 11.21)%], 8μmol/L [(58.46 ± 8.69)%] and 10μmol/L [(51.57 ± 7.58)%] arsenic, respectively, and the PEDF mRNA level in 10 μmol/L group was the lowest; but there was no significant change in PEDF mRNA level [(81.52 ± 13.93)%] when HHL-5 cell was cultivated with 2μmol/L arsenic. Similarly, mRNA level of Bcl-2 in 10 μmol/L arsenic group [(53.53 ± 21 . 49 )%] was significantly decreased compared with that of the control group [(96.48 ± 31.14)%, P < 0.05]. Whereas, Bax mRNA levels in arsenic exposure groups [(65.14 ± 32.34)%, (71.81 ± 12.61)%, (72.22 ± 28.40)%] were not significantly changed in comparison with that of control group [(84.47 ± 35.38)%, P > 0.05]. Conclusion PEDF may play a role in mitochondrial apoptosis of human liver cells induced by arsenic through regulating Bcl-2 level.
