1.Advances in Research on Cell Metabolic Interactions in the Tumor Microenvironment

Metabolic reprogramming plays a critical role in tumorigenesis and tumor progression.The metabolism and the proliferation of tumors are regulated by both intrinsic factors within the tumor and the availability of metabolites in the tumor microenvironment(TME).The metabolic niche within the TME is primarily orchestrated at 3 levels:1)the regulation of tumor metabolism by factors intrinsic to the tumors,2)the interaction between tumor cells and T cells,macrophages,and stromal cells,and 3)the metabolic heterogeneity of tumor cells within the tissue space.Herein,we provided a concise overview of the various metabolic regulatory modes observed in tumor cells.Additionally,we extensively analyzed the interaction between tumor cells and other cells within the TME,as well as the metabolic characteristics and functions of different types of cells.Ultimately,this review provides a theoretical basis and novel insights for the precision treatment of tumors.
2.Study on the antibiotic resistance of Staphylococcus aureus isolated from hospitalized children with respiratory tract infections

Objective To investigate the antimicrobial resistance and genotype distribution of staphylococcal cassette chromosome mec (SCCmec) of staphylococcus aureus (S.aureus) isolated from children hospitalized at Pediatric People's Hospital of Zhongjiang County.Methods Seventy-seven strains of S.aureus were collected by nasopharyngeal swabs at the Pediatric Department of People's Hospital of Zhongjiang County from January to December 2015.Methicillin-resistant staphylococcus aureus (MRSA) and methicillin-susceptible staphylococcus aureus (MSSA) were identified by cefoxitin disc diffusion and detection of mecA method.The minimum inhibitory concentrations (MIC) of antibiotics were determined by E-test method.SCCmec typing on MRSA strains was performed by using multiplex PCR.Results MRSA accounted for 54.5% (42 strains) strains of 77 strains.All MRSA strains were resistant to Penicillin,and the rates of antibiotic resistance to Cefuroxime,Ceftriaxone,Erythromycin were 78.6%,95.2% and 97.6%,respectively.The rates of antibiotic resistance of 35 MSSA to Penicillin and Erythromycin were 97.1% and 62.9%,and they were also sensitive to other antibiotics.In 42 strains of MRSA,SCCmec type Ⅳa was the predominant type (27 strains,64.3 %),which was followed by type Ⅳ g and Ⅴ (each 5 strains,11.9%),type Ⅳ c and Ⅳh (each 1strain,2.4%).Non-susceptibility rate of SCCmec Ⅳ to cefuroxime was significantly higher than that of other SCCmec types (P < 0.05).Conclusions All strains from children hospitalized in People's Hospital of Zhongjiang County are often resistant to Penicillin and Erythromycin.The proportion of MRSA isolated from hospitalized children was high.SCCmec type Ⅳa is the main genotype of MRSA.
3.Problems in Huanggang hospital information construction and suggestions for their solution

Put forward in this paper are suggestions for the solution of problems (insufficient investment of funds, shortage of professionals , incapability of interlink and interaction of hospital data) in Huanggang hospital information construction, including quickening medical information professionals training, optimizing their structure, bringing the leading role of government into full play, adding investment of funds, strengthening construction of infrastructures , establishing criteria for data, promoting interlink and interaction of data, enforcing management and maintenance of hospital information system , founding cooperative professional societies .
4.Surrogate biomarkers identification for neuropsychiatric lupus by proteomic fingerprint technology

Objective To identify biomarkers in cerebrospinal fluid (CSF) by proteomic technology and develop a diagnostic model for neuropsychiatric lupus (NPSLE).Methods CSF proteomic spectra of 27 patients with NPSLE before and after treatment,and 27 controls including 17 patients with scoliosis,and 10 SLE patients without neuropsychiatric manifestation (non-NPSLE) were generated by matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) combined with weak cationic exchange (WCX) magnetic beads.Data were analyzed with t test,non-parametric Kruskal-Wallis H test or Wilcoxon sign-rank test.A decision tree model for NPSLE classification was built based on the discriminating peaks.In addition,CSF samples of 12 patients with NPSLE,12 patients with lumbar disc herniation and 9 patients with other neurological conditions were employed as blind test group to verify the accuracy of the model.Results Twelve discriminating mass-to-charge (m/z) peaks were identified between NPSLE and controls.The diagnostic decision tree model,built with a panel of m/z peaks 8595,7170,7661,7740 and 5806,recognized NPSLE with the sensitivity and specificity of 92.6% and 92.6% based on training group samples,91.7% and 85.7% based on blind test group,respectively.Conclusion Potential CSF NPSLE biomarkers are identified by proteomic technology,the novel diagnostic model is sensitive and relatively specific for the diagnosis of NPSLE.
5.Expression of soluble H1N1 haemegglutinin in two distinct insect baculovirus expression systems

Objective To express functional haemegglutinin(HA)protein in two different bacularvirus expression systems.Methods The whole open reading frame of A/Sichuan/1/2009(H1N1)HA was obtained by synthesis,and the HA protein were expressed in insect cells by two different bacularvius expression systems:BaculoGold system and Bac-to-Bac system. Soluble HA protein was identified by Western blot and haemegglutination test. Results The correct full length of HA gene was obtained and cloned into pAcGP67B and pFAST Bacl vectors,respectively.After 3 rounds of virus amplifyjng by re-infection of Sf9 cells,the HA protein was detected in supematant of BaculoGold system and in intracellular of Bac-to-Bac system which is much better than the former.Purified HA protein was positive not only identified by Western blot,but also detected by haemegglutinin test. Conclusion Functional HA protein was successfully expressed in two distinct bacularvirus expression systems,of which the Bac-to-Bac bacularvirus expression system is more suitable for expression of A/Sichuan/1/2009(H1N1)HA protein.
