1.Mechanism of treatment of Tibet yak-origin Salmonella infections by Tibetan medicine Terminalia Chebula via Bax/Bcl-2 and PTGS2/SLC3A2 pathway was analyzed based on network pharmacology and experimental validation

In order to understand the potential target and related mechanism of action of Termina-lia Chebula treatment,network pharmacology and molecular docking methods were used in this experiment,and the challenge test of Salmonella from yak was performed.The active ingredients and potential targets of Terminalia Chebula were screened through HERB cluster identification database,TCMSP database and SwissTargetPrediction web page tool,and"gastroenteritis"was searched through OMIM and GeneCards database.Cytoscape and STRING databases were used to construct the Terminalia Chebula PPI network to screen out key targets,the intersection targets between Terminalia Chebula and enteritis were obtained through Venny platform,and gene ontol-ogy(GO)and Kyoto encyclopedia database of genes and genomics(KEGG)were enriched through DAVID database.The core target of screening was verified by molecular docking.After that,the gastrointestinal inflammation model of mice was established,the pathological changes of gastroin-testinal tract were observed,and the effect of Terminalia Chebula on the target protein was veri-fied by Western blot test.The results showed that:after analyzing and sorting out 8 main active in-gredients of Terminalia Chebula,118 targets of Terminalia Chebula were screened,11 161 targets of gastroenteritis and 100 targets of intersection were obtained;the core targets of PTGS2,CASP3,SLC3A2,Bax,Bcl-2 and TP53 of Terminalia Chebula and enteritis were obtained through PPI network.GO and KEGG enrichment analysis collected 337 items and 138 items,respectively,mainly related to chemokine pathway,PI3K-Akt signaling pathway,apoptosis related pathway,i-ron ion transport related pathway,NF-κB signaling pathway,etc.The results of molecular docking showed that chebulidic acid,the first active component of chebulidic acid,can bind to Bax,Bcl-2,PTGS2 and SLC3A2 through hydrogen bonding,hydrophobic action,π-π packing force and other intermolecular forces.The pathological tissue sections showed that Terminalia Chebula could sig-nificantly recover gastrointestinal tissue injury.Western blot test results showed that Terminalia Chebula can regulate the process of apoptosis and iron death through Bax/Bcl-2 and PTGS2/SLC3A2 pathways to achieve the effect of treating intestinal inflammatory damage.The results showed that Terminalia Chebula can regulate the occurrence and development of enteritis by regu-lating apoptosis and iron death through Bax/Bcl-2 and PTGS2/SLC3A2 pathways.Terminalia Chebula has the characteristics of multi-target and multi-pathway in the treatment of enteritis.
2.Effects of SERPINE1 on proliferation,migration,invasion and apoptosis of colon cancer cells

Objective To explore the expression of serine proteinase inhibitor family E member 1(SERPINE1)in colon cancer and its effect on the malignant biological behaviors of colon cancer cells.Methods starBase and TISIDB databases were used to analyze the expression of SERPINE1 in 471 patients with colon cancer and 41 paracancerous tissues,and the relationships of its differential expression with clinical stage and prognostic survival were analyzed.The cancer specimens and paracancerous tissues from 5 patients with colon cancer were collected in No.940 Hospital of Joint Logistic Support Force,and RT-qPCR and Western blotting were employed to detect the expression of SERPINE1 at mRNA and protein levels.After lentiviral vectors of SERPINE1 overexpression and interference were constructed and transfected into colon cancer RKO and LoVo cells.The experimental cells were assessed by RT-qPCR and Western blotting to verify the efficiency of overexpression and interference.Then cell proliferation,migration,invasion and apoptosis were evaluated by CCK-8 assay,clone formation test,Transwell test and Hoechst apoptotic nuclear staining,respectviely.Finally,Western blotting was applied to determine the effect of SERPINE1 on phosphatidyl-inositol 3-kinase(PI3K)/protein kinase B(AKT)signaling pathway.Results Database analysis showed that SERPINE1 was highly expressed,positively correlated with the clinical stage and negatively correlated with the overall survival in colon cancer patients(P<0.05).Transfection of overexpression lentiviral vector resulted in enhanced cell proliferation,invasion and migration abilities while weakened apoptosis in RKO and LoVo cells.On the contrary,SERPINE1 interference reduced the abilities of cell proliferation,invasion and migration but improved cell apoptosis(P<0.05).Western blotting showed that overexpression of SERPINE1 had no effect on the expression of PI3K and AKT,but increased the expression of p-PI3K and p-AKT,and SERPINE1 interference also had no change in PI3K and AKT expression and reduced the levels of their phosphorylation levels.Statistical differences were observed in relative quantitative analysis on the ratios of p-PI3K/PI3K and p-AKT/AKT(P<0.05).Conclusion SERPINE1 is highly expressed,and correlated with clinical stage and prognosis of patients with colon cancer.Its overexpression promotes the proliferation,migration,invasion and inhibits apoptosis of colon cancer RKO and LoVo cells through PI3K/AKT signaling pathway.
3.Effect and mechanism of miR-640 in lumbar disc degeneration

Objectives:To investigate the effect and mechanism of miR-640 in lumbar disc degeneration(LDD).Methods:The nucleus pulposus cells isolated from normal rat nucleus pulposus tissue were allocated into four groups:control group,TNF-α group(50ng/mL TNF-α treated the cells for 48h),TNF-α+miR-640-NC group(50ng/mL TNF-α treated the cells transfected with miR-640-NC for 48h)and TNF-α+miR-640-in-hibitor group(50ng/mL TNF-α treated the cells transfected with miR-640-inhibitor for 48h).The expression of miR-640 in nucleus pulposus cells was detected by RT-qPCR.The cell viability was detected by CCK-8 assay.Cell apoptosis was examined by flow cytometry.The expressions of Bcl-2,Bax,Cleaved caspase-3,Collagen n and Aggrecan were detected by Western blot.The expression of MMP-9 was observed by im-munofluorescence staining.The LDD rat model was established through the intradiscal injection of TNF-α.The 25 rats were randomly allocated into 5 groups(each consisting of 5 rats):sham group,LDD group,Sham+miR-640-NC group,LDD+miR-640-NC group,and LDD+miR-640-inhibitor group.Specifically,one week pri-or to sham surgery or LDD modeling,rats in the Sham+miR-640-NC,LDD+miR-640-NC,and LDD+miR-640-inhibitor groups were administered with the lentivirus vector carrying miR-640-inhibitor or miR-640-NC into the intervertebral disc space.The expression of miR-640 in nucleus pulposus tissue was detected by RT-qPCR.The morphology of lumbar intervertebral discs was observed by MRI and HE staining.The expres-sion of MMP-9 in nucleus pulposus was observed by immunofluorescence.And the expressions of Cleaved caspase-3,Collagen Ⅱ and Aggrecan in nucleus pulposus was observed by immunohistochemistry.Results:The expression of miR-640 was increased in nucleus pulposus tissue of LDD model rats and nucleus pulpo-sus cells treated with TNF-α(P<0.05).Compared with TNF-α+miR-640-NC group,cell viability and the ex-pressions of Bcl-2,Collagen Ⅱ and Aggrecan were all increased in TNF-α+miR-640-inhibitor group(P<0.05),while cell apoptosis,and Bax,Cleaved caspase-3 and MMP-9 expressions were decreased(P<0.05).The LDD+miR-640-NC group showed obvious degenerative changes in lumbar intervertebral disc,the expressions of Collagen Ⅱ and Aggrecan in nucleus pulposus were decreased(P<0.05),and the expressions of Cleaved cas-pase-3 and MMP-9 were increased(P<0.05),however,the above changes were reversed to a certain extent in LDD+miR-640-inhibitor group(P<0.05).Conclusions:In the LDD rat model and in vitro cell model,miR-640 expression is upregulated.Inhibition of miR-640 can alleviate LDD by potentially suppressing nucleus pulpo-sus cell apoptosis,reducing extracellular matrix degradation,and promoting extracellular matrix synthesis.
4.Applications and prospects of electroencephalography technology in neurorehabilitation assessment and treatment.

With the high incidence of neurological diseases such as stroke and mental illness, rehabilitation treatments for neurological disorders have received widespread attention. Electroencephalography (EEG) technology, despite its excellent temporal resolution, has historically been limited in application due to its insufficient spatial resolution, and is mainly confined to preoperative assessment, intraoperative monitoring, and epilepsy detection. However, traditional constraints of EEG technology are being overcome with the popularization of EEG technology with high-density over 64-lead, the application of innovative analysis techniques and the integration of multimodal techniques, which are significantly broadening its applications in clinical settings. These advancements have not only reinforced the irreplaceable role of EEG technology in neurorehabilitation assessment, but also expanded its therapeutic potential through its combined use with technologies such as transcranial magnetic stimulation, transcranial electrical stimulation and brain-computer interfaces. This article reviewed the applications, advancements, and future prospects of EEG technology in neurorehabilitation assessment and treatment. Advancements in technology and interdisciplinary collaboration are expected to drive new applications and innovations in EEG technology within the neurorehabilitation field, providing patients with more precise and personalized rehabilitation strategies.

5. Anatomic morphology analysis of the torcular herophili of the intracranial venous system and its clinical significance to related venous system diseases

Objective: To investigate anatomic and morphological distribution characteristics of the torcular herophili of the intracranial venous system and to provide reference for the treatment and evaluation of common diseases related to the venous system. Method: From January 2016 to July 2018, 2 668 inpatients with cerebrovascular disease related symptoms admitted in Department of Neurology and the other departments, the First Medical Center of the PLA General Hospital were retrospectively recruited. These patients included 1 572 men and 1 096 women. The ages ranged from 11 to 82 years, with an average of (54 ± 14) years old. According to the anatomic morphology and drainage channel of the straight sinus,bilateral or unilateral transverse sinuses,superior sagittal sinus and other blood vessels in the sinus confluence area (occipital protuberance),the anatomic morphology of the torcular herophili from DSA results can be divided into type I (complete torcular herophili type),type II (incomplete torcular herophili type, including II1A,II1B,II2,II3,II4 and II 5 five subtypes), type III (no torcular herophili). Type I refers to venous confluence of the superior sagittal sinus, straight sinus and bilateral transverse sinuses in the occipital protuberance;Type II refers venous confluence of any three of the superior sagittal sinus, straight sinus and bilateral transverse sinuses in the occipital protuberance, and it must contain the superior sagittal sinus and the straight sinus not directly connected with bilateral transverse sinuses; Type III refers to confluence of the straight sinus alone into the one side of the transverse sinus-sigmoid sinus,but without drainage from the contralateral veins and no communication between bilateral transverse sinuses. Results: (1) Among the 2 668 patients admitted to the hospital due to cerebrovascular disease related symptoms and completed DSA examination, the proportion of different anatomic types torcular herophili from high to low was type II 56. 4% (1 504 cases), type I 39. 0% (1 040 cases) and type ID 3.7% (100 cases). (2) Among five subtypes of type II, the proportion of II 1A was 20.4% (544/2 668), II 1B 5. 7% (152/2 668), II2 19. 8% (528/2 668), II3 2. 8% (76/2 668), II4 4. 3% (116/2 668) and II5 3. 3% (88/2 668). (3) The proportion of right dominant drainage was the highest,accounting for 50. 5% (1 348/2 668),followed by bilateral balanced drainage,accounting for 36. 4% (972/2 668). The left dominant drainage was the lowest, accounting for 12. 1% (324/2 668). Conclusions: There are great morphological variations in the sinus confluence of the intracranial venous system. Understanding of the anatomical features of the sinuses and assessment of the drainage characteristics of the venous sinuses could provide references for diagnosis and treatment of venous system diseases.
6.Effect of Acupuncture plus Chinese Herbal Medication on ET-1 and CGRP in Child Patients with Mesenteric Lymphadenitis

Objective To investigate the effect of acupuncture plus Chinese herbal medication on plasma endothelin (ET-1) and calcitonin gene-related peptide (CGRP) in child patients with mesenteric lymphadenitis.Methods One hundred and eighty child patients with mesenteric lymphadenitis were randomly allocated to groups A, B and C, 60 cases each. Group A received acupuncture at Zusanli and pricking Sifeng points plus oral administration of Wudang Babao Zijinding; group B, oral administration of amoxicillin and clavulanate potassium granules; group C, oral administration of Wudang Babao Zijinding alone. ET-1 and CGRP contents were measured in the three groups before and after treatment.Results There were statistically significant pre-/post-treatment differences in ET-1 and CGRP contents in group A (P<0.01). There were statistically significant post-treatment differences in ET-1 and CGRP contents between group A and group B or C (P<0.01).Conclusions Acupuncture plus Chinese herbal medication is an effective way to treat mesenteric lymphadenitis in children. It can regulate ET-1 and CGRP in the patients.
7.Cysteinyl leukotriene and 8-isoprostane in exhaled breath condensate of asthmatic children

Objective To analyze the change of cysteinyl leukotriene ( Cys-LTs) levels and 8-Isopros-tane (8-iso-PG) levels in the exhaled breath condensate (EBC) of asthmatic children from acute exacerbation to clinical remission, and investigate the role of the detection of Cys-LTs and 8-iso-PG in EBC in its severity and pathogenesis , and explore the relationship between the Cys-LTs and 8-iso-PG through measuring Cys-LTs levels and 8-iso-PG levels in the EBC of asthmatic children. Methods The outpatient or inpatient asthmatic children of the pediatrics and a group of healthy children were studied. All subjects′ EBC were collected by the R-Tube produced by American Respiratory Research. The concentration of Cys-LTs and 8-iso-PG in EBC were measured by enzyme-linked immunosorbent assay (ELISA), and compared among children in asthmatic exacerbation, asthmatic remission, and healthy condition. The relevance of their change would be explored at the same time. Results (1) Cys-LTs levels in EBC were higher in asthma exacerbation, compared to healthy controls (P<0. 05), while no significant difference were found between asthmatic remission and asthmatic exacerbation or healthy controls ( P>0. 05 ) . ( 2 ) 8-iso-PG levels was higher in asthmatic exacerbation compared to asth-matic remission ( P<0. 05 ) . Moreover, the 8-iso-PG levels were significantly higher in asthmatic remission than in healthy controls (P<0. 05). (3) Through the relevance analysis of the Cys-LTs and 8-iso-PG levels in EBC among the three groups, Cys-LTs levels in EBC of asthmatic exacerbation significantly were correlated with 8-iso-PG levels (n1 =35, r1 =0. 61, P<0. 05), while there was no significant correlation between 8-iso-PG levels and Cys-LTs levels in asthmatic remission. Conclusion The increase of 8-iso-PG levels in EBC of bronchial asthmatic patients correlates with the disease and its control. Therefore, 8-iso-PG can be an objective indicator for asthmatic diagnosis and healing efficacy. Cy-LTs levels increase in the EBC of bronchial asthmatic according to disease severity. The two levels correlate during asthmatic exacerbation, indicating that a link be-tween airway oxidative stress and inflammation among asthmatics.
8.Preliminary study of the inhibitory effect and mechanism of B16F10-ESAT-6-gpi/IL-21 vaccine on the pulmonary metastasis in mouse models of melanoma.

OBJECTIVETo investigate the effect and mechanism of B16F10-ESAT-6-gpi/IL-21 tumor cell vaccine on pulmonary metastasis in mouse model of melanoma.
METHODSTwelve 8-week old female C57BL/6 mice were used in this study. The mice were injected with wild-type B16F10 cells through tail vein after immunization with B16F10-ESAT-6-gpi/IL-21 tumor cell vaccine, and the pulmonary metastasis was observed. The CD4(+) and CD8(+) T cells were isolated by magnetic activated cell sorting, and then used for the detection of CFSE/7-AAD cytotoxicity by flow cytometry. Serum from the mice immunized with tumor-cell vaccine was used to detect IFN-γ expression by ELISA. The expression of TGF-β2, ZEB1, E-cadherin, and N-cadherin of tumor tissues was detected by RT-PCR and immunofluorescence, respectively.
RESULTSThe mice vaccinated with B16F10-ESAT-6-gpi/IL-21 had significantly fewer nodules in the lung and lower lung weight [(285.8 ± 19.01) mg vs. (406.3 ± 27.12) mg], with lower levels of TGF-β2, ZEB1 and N-cadherin proteins but higher level of E-cadherin protein within the tumor tissue, as compared with the control mice. Meanwhile, the immunized mice had significantly increased CD8(+) T cell killing activity [(42.62 ± 3.465)% vs. (22.29 ± 1.804)%] and IFN-γ expression level [(55.200 ± 7.173) pg/ml vs. (6.435 ± 1.339) pg/ml] over the control mice.
CONCLUSIONSThe B16F10-ESAT-6-gpi/IL-21 vaccine can inhibit the metastasis of melanoma in the lung in vaccinated melanoma-bearing mice. This inhibitory effect is associated with CD8(+) T cell immune response and a higher level of IFN-γ, which may influence on the mesenchymal-epithelial transition of tumor cells.

9.Using FRET to Study The Interaction Domain of TLR4 Binding to MD-2 in Living Cells

TLR4-MD-2 complex plays a key role in LPS recognition and its signal transduction. These steps are the vital elements of the host's defensive reaction. Studying the functional domain of TLR4 and MD-2 is very important to further understand the mechanism of LPS signal transduction. It was studied the interaction domain of TLR4 and MD-2 in living cells based on gene mutation, gene transfection and fluorescence resonance energy tramsfer(FRET) which is considered as one of the best methods used for intracellular protein-protein interaction study. CY-15P which was fused by CFP and YFP through 15 neutral amino acids was used as positive control, while co-expressed CFP and YFP proteins were used as negative control. The results showed that the ability of TLR4 binding to MD-2 decreased dramatically after the deletion of Glu~(24) ~ Met~(41) in N terminal of TLR4. Aggregation of TLR4 to LPS stimulation was observed, however, TLR4 without the Glu~(24)~ Met~(41) mutation did not aggregate. All these results indicated that TLR4 Glu~(24)~ Met~(41) might be the interaction domain of TLR4 binding to MD-2 and participate in the aggregation effect of TLR4 upon LPS stimulation.
10.Problems and countermeasures in cost accounting at the department level in army hospitals

Cost accounting is an important measure in hospital economic management.This article approaches the problems and their causes in cost accounting in army hospitals in terms of the correlation between income and cost,the normality of asset management,the accuracy of internal service cost,the advancedness of cost accounting techniques and so on.It also proposes a some corresponding countermeasures based on our work experience.
