1.Indobufen attenuates cerebral ischemia–reperfusion injury by inhibiting the NF-κB/Caspase-1/GSDMD pathway

Indobufen is a new generation of antiplatelet agents and has been shown to have antithrombotic effects in animal models. However, its therapeutic potential and mechanisms against cerebral ischemia/reperfusion (I/R) injury remain unclear. In this study, we evaluated the in vivo neuroprotective effects of indobufen through both pretreatment and posttreatment regimens in a rat model of middle cerebral artery occlusion/reperfusion (MCAO/R). In vitro, human umbilical vein endothelial cells (HUVECs) subjected to oxygen-glucose deprivation/reoxygenation (OGD/R) were employed to investigate the relationship between indobufen and the pyroptosis-associated NF-κB/Caspase-1/GSDMD pathway. The pharmacodynamic tests revealed that indobufen ameliorated I/R injury by decreasing the level of thromboxane B2 (TXB2), infarct size, brain edema and neurological impairment in rats and rescuing cell pyroptosis in HUVECs. The underlying mechanisms were probably related to pyroptosis suppression by regulating the NF-κB/Caspase-1/GSDMD pathway. Overall, these studies indicate that indobufen exerts protective and therapeutic effects against I/R injury by pyroptosis suppression via downregulating NF-κB/Caspase-1/GSDMD pathway.
2.Internal tension relieving technique assisted anterior cruciate ligament reconstruction to promote ligamentization of Achilles tendon grafts in small ear pigs in southern Yunnan province

BACKGROUND:We have successfully established an animal model of small ear pig in southern Yunnan province with internal tension relieving technique combined with autologous Achilles tendon for anterior cruciate ligament reconstruction,and verified the stability and reliability of the model.However,whether internal tension relieving technique can promote the ligamentalization process of autologous Achilles tendon graft has not been studied. OBJECTIVE:To investigate the differences in the process of ligamentalization between conventional reconstruction and internal reduction reconstruction of the anterior cruciate ligament by gross view,histology and electron microscopy. METHODS:Thirty adult female small ear pigs in southern Yunnan province were selected.Anterior cruciate ligament reconstruction was performed on the left knee joint with the ipsilateral knee Achilles tendon(n=30 in the normal group),and anterior cruciate ligament reconstruction was performed on the right knee joint with the ipsilateral knee Achilles tendon combined with the internal relaxation and enhancement system(n=30 in the relaxation group).The autogenous right forelimb was used as the control group;the anterior cruciate ligament was exposed but not severed or surgically treated.At 12,24,and 48 weeks after surgery,10 animals were sacrificed,respectively.The left and right knee joint specimens were taken for gross morphological observation to evaluate the graft morphology.MAS score was used to evaluate the excellent and good rate of the ligament at each time point.Hematoxylin-eosin staining was used to evaluate the degree of ligament graft vascularization.Collagen fibers and nuclear morphology were observed,and nuclear morphology was scored.Ultrastructural remodeling was evaluated by scanning electron microscopy and transmission electron microscopy. RESULTS AND CONCLUSION:(1)The ligament healing shape of the relaxation group was better at various time points after surgery,and the excellent and good rate of MAS score was higher(P<0.05).Moreover,the relaxation group could obtain higher ligament vascularization score(P<0.05).(2)The arrangement of collagen bundles and fiber bundles in the two groups gradually tended to be orderly,and the transverse fiber connections between collagen gradually increased and thickened,suggesting that the strength and shape degree of the grafts were gradually improved,but the ligament remodeling in the relaxation group was always faster than that in the normal group at various time points after surgery.(3)The diameter,distribution density,and arrangement degree of collagen fibers in the relaxation group were better than those in the normal group at all time points,especially in the comparison of collagen fiber diameter between and within the relaxation group(P<0.05).

Objective:This study systematically summarized the construction experience of the immunology platform at the Institute of Basic Medical Sciences, Peking University Third Hospital, aiming to provide theoretical references and practical guidance for research-oriented hospitals in building high-quality research platforms.Methods:This study employed case study analysis to elaborate on the platform development initiatives, integrating literature analysis and in-depth interviews to conduct a horizontal comparison of management models among peer research platforms.Results:Through five years of development, the platform had achieved remarkable outcomes via a model integrating ″Talent cultivation-Technological innovation-Equipment procurement″ Research talents had demonstrated breakthroughs in securing national-level research grants, publishing high-impact papers, and obtaining scientific awards. The technical service system had achieved enhancement in both service scope and professional depth, fostering robust interdisciplinary synergy. The platform had effectively expanded its societal engagement capacity.Conclusions:The sustainable advancement of research-oriented hospital immunology platform necessitates establishing standardized flow cytometry databases and implementing high-dimensional data integration. Building upon multidisciplinary convergence, it is imperative to pioneer innovative operational mechanisms characterized by efficiency, open-access, and shared frameworks.
6.Evaluation of left ventricular function in patients with different degrees of stenosis in the left anterior descending branch by myocardial work combined with stress echocardiography

Objective:To evaluate left ventricular function and myocardial ischemia in patients with different degrees of left anterior descending(LAD)coronary artery stenosis by using post-stress regional myocardial work(MW)and coronary flow reserve(CFR),and to analyze the diagnostic efficacy of each parameter for different degrees of stenosis in the LAD.Methods:A retrospective analysis was conducted on 76 patients with suspected coronary artery disease who underwent coronary angiography and attended the Fourth Affliiated Hospital of Harbin Medical University from January 2023 to June 2024. According to the degree of LAD stenosis,30 cases were categorized into the mild stenosis group(stenosis < 50%),25 cases in the moderate stenosis group(50%≤stenosis < 70%),and 21 cases in the severe stenosis group(70% ≤ stenosis ≤ 90%). Transthoracic Doppler echocardiography was applied to obtain CFR LAD. Longitudinal strain in the left anterior descending region(LS LAD),peak strain dispersion(PSD),left anterior descending region work index(WI LAD)and left anterior descending region work efficiency(WE LAD)were obtained at rest and under stress,respectively. The LAD regional MW indexes were compared between the groups in two pairs. The correlation between CFR LAD and the degree of LAD stenosis was analyzed. ROC curves were plotted to compare the diagnostic efficacy of LAD regional MW parameters and CFR LAD for moderate and severe LAD stenosis. Results:①At rest:compared with the mild stenosis group,the absolute value of LS LAD,WI LAD,and WE LAD were reduced in the severe stenosis group,and the differences were statistically significant(all P < 0.05). Compared with the moderate stenosis group,WE LAD was reduced in the severe stenosis group,and the difference was statistically significant( P < 0.05). Differences in two-by-two comparisons between groups in the resting state for the remaining parameters were not statistically significant(all P > 0.05). Under stress:compared with the mild stenosis group,the absolute values of LS LAD,WI LAD,and WE LAD were reduced in the moderate and severe stenosis groups,and the PSD was increased in the severe stenosis group,and the differences were statistically significant(all P < 0.05). Compared with the moderate stenosis group,the absolute values of LS LAD,WI LAD,and WE LAD were reduced in the severe stenosis group,and the differences were statistically significant(all P < 0.05). Differences between two-by-two comparisons between groups in the remaining parameters at loading state were not statistically significant(all P > 0.05). ②CFR LAD was negatively correlated with the degree of LAD stenosis( r = -0.809, P < 0.05). ③The ROC curves showed that the peak WE LAD of the regional MW parameters predicated the largest area under the ROC curve(AUC)for moderate and severe stenosis in LAD,0.803 and 0.897,respectively,with cutoff values of 92.5% and 91.5%. The cutoffs of CFR LAD ≤ 2.5 and CFR LAD ≤ 2.0 predicted the greatest AUC for moderate and severe stenosis in LAD at 0.743 and 0.901,respectively. Peak WE LAD combined with CFR LAD predicted the greatest AUC for moderate and severe stenosis in LAD at 0.826 and 0.967,respectively. Conclusions:Regional MW parameters and CFR LAD can sensitively detect left ventricular hypo-function in patients with different degrees of stenosis in LAD. Peak WE LAD and CFR LAD are reliable predictors of moderate and severe stenosis in LAD. A combination of the two techniques can significantly improve diagnostic efficacy.
7.Assessment of left ventricular wall shear stress at each segment in non-obstructive hypertrophic cardiomyopathy using vector flow mapping

Objective:To assess the clinical utility of vector flow mapping(VFM)for evaluating left ventricular wall shear stress(WSS)in individuals with non-obstructive hypertrophic cardiomyopathy(nHCM).Methods:Forty patients with non-obstructive HCM(nHCM)and 46 healthy volunteers in Tangdu Hospital of Air Force Medical University from May 2020 to September 2023 were enrolled prospectively in this study. The left ventricular WSS of each of 18 segments was measured and analyzed by VFM during rapid filling phase(T1),slow filling phase(T2),isovolumetric contraction phase(T3)and rapid ejection phase(T4).Results:① WSS decreased gradually from basal to apical segments in control group,while there was no such change pattern in nHCM group. ② Diastolic phase:When compared to the control group,the nHCM group demonstrated significantly increased WSS across all segments of the inferoseptal regions,the basal anteroseptal segment,and the middle and apical inferior segments during the T1 stage. Additionally,at the T2 stage,the WSS was significantly higher in the nHCM group across all segments of the inferoseptal and anteroseptal regions,the basal anterolateral segment,and the apical inferolateral and anterior segments(all P<0.05). Conversely,during the T1 stage,the basal anterolateral segment,basal inferolateral segment,and the middle and apical anteroseptal segments,as well as during the T2 stage,the basal and middle inferior segments,exhibited lower WSS values compared to the control group,with these differences being statistically significant(all P<0.05). ③Systolic Phase:During the T3 stage,the WSS values in all segments of the anteroseptal and anterolateral regions,as well as the apical inferior and anterior segments,were significantly elevated in the nHCM group compared to the control group(all P<0.05).At the T4 stage,WSS in each segment of the inferolateral and inferior regions,as well as the basal and middle anterior segments,was significantly elevated in the nHCM group compared to the control group(all P<0.01). Conversely,during the same stage,the WSS values in the basal inferolateral segment,the basal and middle anterolateral segments,and the basal and middle anteroseptal segments were significantly lower in the nHCM group than in the control group,with all differences reaching statistical significance(all P<0.01).Regarding global WSS,the left ventricular global WSS in the nHCM group was significantly higher than that in the control group at both the T2 and T3 stages(all P<0.001). Furthermore,the global WSS at the T3 stage was negatively correlated with left ventricular end-systolic volume,end-diastolic volume,and left ventricular stroke volume( r=-0.226, P=0.036; r=-0.345, P=0.001; r=-0.303, P=0.005). Conclusions:Significant differences in WSS are observed between patients with nHCM and control groups at various phases. WSS may serve as an early indicator of changes in cardiac function in nHCM patients.
8.Analysis of changes in ocular surface biology and visual quality in diabetic dry eye patients under different influencing factors

Objective To investigate the factors that influence ocular surface biology and visual acuity in individuals with diabetic dry eye(DDE)and analyze how these factors contribute to changes in visual quality.Methods Based on the disease duration,fasting blood glucose(FBG),and glycated hemoglobin(HbA1c)levels of patients with type 2 diabe-tes mellitus(T2DM),the DDE patients were divided into different groups.Logistic regression analysis was used to identify influencing factors related to ocular surface biology and visual quality in each group of DDE patients.Tear film stability was evaluated based on the tear film rupture time(BUT),Schirmer I test(SIt),and ocular surface disease index(OSDI).Lip-iview? Surface interferometers were used to measure tear film lipid layer thickness(LLT),meibomian gland loss rate(MGP),meibomian gland opening number(MGYLS),and meibomian gland secretion score(MGYSS).Wavefront aber-rometry was used to measure corneal wavefront aberration values at 4 mm and 6 mm pupil diameters.Ocular response ana-lyzer(ORA)was adopted to analyze corneal hysteresis(CH)and corneal resistance factor(CRF).Moreover,ELISA ex-periment to evaluate the trend of changes in inflammatory factors in tears.Results Logistic regression analysis revealed that T2DM duration,smoking history,FBG,HbA1c,total cholesterol(TC),triglycerides(TG),OSDI score,LLT,BUT,SIt,MGP,MGYLS,MGYSS,total higher-order aberrations,spherical aberration,coma aberration,trefoil aberration,tumor necrosis factor-α,interleukin-6,matrix metalloproteinase-9,receptor for advanced glycation end products,and insu-lin were all influencing factors for the risk of DDE(all P<0.05).As the T2DM course prolonged and FBG or HbA1 c levels rose,tear film-related indicators(LLT,BUT,and SIt)and meibomian gland-related indicators(MGYLS and MGYSS)inpa-tients gradually decreased,while OSDI scores and MGP gradually increased(all P<0.05).As the T2DM course prolonged and FBG or HbA1c levels rose,the total higher-order aberrations,spherical aberration,coma aberration,and trefoil aber-ration in DDE patients under 4 mm and 6 mm pupil diameters gradually increased;Meanwhile,best corrected visual acuity,corneal hysteresis,and corneal resistance factor gradually decreased;The contents of tumor necrosis factor-α,interleukin-6,matrix metalloproteinase-9,receptor for advanced glycation end products,and insulin in tears all gradually increased,while mucin-5AC gradually decreased(all P<0.05).Conclusion With the prolongation of T2DM duration and the in-crease of FBG or HbA1c,the ocular surface inflammatory response in DDE patients gradually worsens,corneal biological function decreases,and visual quality deteriorates.Timely systemic and local interventions are of great significance for im-proving dry eye symptoms and visual quality in DDE patients.

Body weight abnormalities, including overweight, obesity, and underweight, have become a dual public health challenge in Chinese adults: overweight and obesity lead to a variety of chronic complications, while underweight increases the risks of malnutrition, sarcopenia, and organ dysfunction. To systematically address these issues, multidisciplinary experts in endocrinology, sports science, nutrition, and psychiatry from various regions have held multiple weight management seminars. Based on the latest epidemiological data and clinical evidence, they expanded the guideline to include assessment and intervention strategies for underweight, in addition to the core content of obesity management. This guideline outlines the etiological mechanisms, evaluation methods, and multidimensional management strategies for overweight and obesity, covering key areas such as diagnosis and assessment, medical nutrition therapy, exercise prescription, pharmacological intervention, and psychological support. It is intended to provide a scientific and standardized approach to weight management across the adult population, aiming to curb the rising prevalence of obesity, mitigate complications associated with abnormal body weight, and improve nutritional status and overall quality of life.
10.Ameliorative effects of Lycii Fructus-Chrysanthemi Flos at different ratios on retinal damage in mice.

This study aimed to compare the ameliorative effects of Lycii Fructus and Chrysanthemi Flos at different ratios on retinal damage in mice and to elucidate the underlying mechanisms. A retinal injury model was established by intraperitoneal injection of sodium iodate(NaIO_3) solution. The mice were divided into the following groups: blank group, model group, positive drug(AREDS 2) group, low-and high-dose groups of Lycii Fructus and Chrysanthemi Flos at 1∶1, low-and high-dose groups at 3∶1, and low-and high-dose groups at 1∶3. Administration was carried out 15 days after modeling. The visual acuity of the mice was assessed using the black-and-white box test. The fundus was observed using an optical coherence tomography device, and retinal thickness was measured. HE staining was used to observe the morphology and pathological changes of the retina. The levels of oxidative factors in serum and ocular tissues were measured using assay kits. The levels of inflammatory factors in serum and ocular tissues were detected by enzyme-linked immunosorbent assay(ELISA), and the expression of Nrf2, HO-1, and NF-κB proteins in ocular tissues was analyzed by Western blot. The results showed that after administration of Lycii Fructus and Chrysanthemi Flos at different ratios, the model group showed improved retinal thinning and disordered arrangement of retinal layers, elevated content of SOD and GSH in the serum and ocular tissues, and reduced levels of MDA, TNF-α, IL-1β, and IL-6. Lycii Fructus and Chrysanthemi Flos at 1∶1 and 1∶3 showed better improvement effects. The combination significantly upregulated the expression levels of Nrf2 and HO-1 and downregulated the expression of NF-κB p65. These results indicate that Lycii Fructus and Chrysanthemi Flos at different ratios can improve retinal damage, reduce oxidative stress, and alleviate inflammation in both the body and ocular tissues of mice. The mechanism may be related to the regulation of the Nrf2/HO-1 and NF-κB signaling pathways in ocular tissues. These findings provide a theoretical basis for the clinical application of Lycii Fructus and Chrysanthemi Flos in the treatment of dry age-related macular degeneration.
