Objective To explore the correlation between changes in serum autoantibodies and pregnancy outcome in patients with unexplained recurrent spontaneous abortion(URSA)and to establish a predictive model. Methods The clinical data of 147 patients with URSA were retrospectively collected.Based on pregnancy outcome, patients were divided into the successful pregnancy group(108 cases)and the failed pregnancy group(39 cases).Serum levels of autoantibodies(including antiphospholipid antibodies, antithyroid antibodies, antinuclear antibodies,anti-endometrial antibodies and anti-ovarian antibodies),coagulation function and hormone levels were compared between the two groups.Logistic regression analysis was used to examine the correlation between changes in serum autoantibodies and pregnancy outcome in URSA patients.A nomogram model was constructed to predict the risk of adverse pregnancy outcomes in URSA patients. Results The age, number of previous miscarriages, positive rates of antiphospholipid antibodies, antithyroid antibodies, antinuclear antibodies,anti-endometrial antibodies,anti-ovarian antibodies,as well as the elevated proportions of fibrinogen and D-dimer,and levels of follicle-stimulating hormone(FSH),luteinizing hormone(LH),and prolactin(PRL)were significantly higher in the failed pregnancy group than those in the successful pregnancy group(P<0.05).In contrast, the levels of estradiol(E2),progesterone(P)and testosterone(T)were significantly lower in the failed pregnancy group compared to the successful pregnancy group(P<0.05).Multivariate Logistic regression analysis indicated that age, previous miscarriage count, positive antiphospholipid antibodies, positive anti-endometrial antibodies, elevated FSH levels and reduced E2 levels were independent risk factors for pregnancy failure in URSA patients. The nomogram model constructed based on these indicators demonstrated good predictive ability, with an AUC of 0.795(95%CI:0.697-0.893).Conclusion Serum antiphospholipid antibodies and anti-endometrial antibodies can affect pregnancy outcome in URSA patients, and the constructed prediction model has significant clinical application value.

Aim To detect the mechanism of Wenshen Jianpi recipe(WSJPR)regulating the autophagy by p70S6K signaling pathway on alleviating podocyte inju-ry in diabetic nephropathy(DN)rats. Methods DN model rats induced by streptozotocin were divided into five groups with six rats in each group:model control group, low dose group(7.5 g·kg-1·d-1),medium dose group(15 g·kg-1·d-1),high dose group(30 g·kg-1·d-1),and positive control group(25 mg·kg-1·d-1).In addition, six normal rats were used as negative control group(isotonic NaCl solution 10 mL·kg-1·d-1).All the rats were given continuous ga-vage for eight weeks.Fasting blood glucose, urine al-bumin/creatinine ratio(UACR)and blood viscosity were determined. The changes of podocyte ultrastruc-ture and autophagosome in each group were observed by transmission electron microscopy(TEM).The pro-tein levels of signaling pathway factor p70S6K and au-tophagy factor p62 in renal tissues of rats in each group were detected by Western blot.Besides,p62 expres-sion was observed by immunohistochemistry. Results WSJPR could decrease fasting blood glucose and UACR,and improve the indexes of blood viscosity in rats.TEM indicated that WSJPR could significantly improve the podocyte ultrastructure and autophagy level in DN rats.Western blot showed that the expression level of signaling pathway factor p70S6K and autophagy factor p62 in the kidney of DN rats increased signifi-cantly compared with blank control group(P<0.01).The expression level of p70S6K and p62 in WSJPR groups decreased compared with model control group(P<0.05).Among them, the medium-dose group of WSJPR had the most significant change.Immunohisto-chemical results showed that the level of autophagy fac-tor p62 in kidney tissue of DN rats increased compared with the control group.WSJPR had a certain inhibitory effect on p62 expression in DN rats. Conclusion WSJPR might restore cell homeostasis by inhibiting p70S6K level, reducing the expression of autophagy factor p62 and enhancing autophagy level in renal tis-sue of DN rats.

Foodborne illnesses caused by Salmonella pose significant threats to worldwide public health safety. In this study,a rapid method for screening viable Salmonella in oyster sauce and milk was developed by utilizing an electronic microbial growth analyzer(EMGA).Target food samples were diluted 10-fold with RVS broth and loaded into test tubes.Test tubes were positioned in the EMGA to determine the bacterial growth curves and the time required to reach the maximum growth rate(Tmgr).Using Salmonella typhimurium(S.typhimurium)asan model species, there was linear relationship between the logarithmic value of viable bacterial concentration(lgC)and Tmgr over the range of 5×101-5×106 CFU/mL,with a detection limit of 10 CFU/mL.For oyster sauce, the regression equation was Tmgr(min)=-80.775lg[C/(CFU/mL)]+754.96(R2=0.9907),and the recovery rates of S.typhimurium ranged from 95.2%to 119.8%,with relative standard deviations(RSD)ranging from 3.5%to 16.3%.For milk, the regression equation was Tmgr(min)=-71.922 lg[C/(CFU/mL)]+618.65(R2=0.9985),with recovery rates ranging from 98.4%to 110.6%and RSD ranging from 6.4%to 12.8%.The EMGA method required only one portable instrument, and involving only three manual steps,i.e.,dilution,transfer,and insertion. When S.typhimurium contamination reached 106 CFU/mL,the total time consumption, from the unwrapping of samples to the readout of bacterial concentration, was no more than 7 h.When applied to detection of actual oyster sauce and milk samples, the new method demonstrated strong consistency with plate counting results in positive detection rates. This method was superior to the plate counting method, which was generally considered as a gold standard,in terms of accuracy, precision, simplicity and efficiency, representing a promising alternative for the on-site screening and quantification of viable Salmonella in oyster sauce and milk products.

Objective To investigate the inhibitory effect of β-elemonic acid(β-EA)on the proliferation and inva-sion of colon cancer cells and the underlying mechanisms through PI3K/AKT/mTOR signaling pathway. Methods The effects of β-EA on colon cancer cell proliferation were evaluated using the MTT assay and colony formation as-say.Transwell invasion assay were used to assess the impact of β-EA on invasion.Western blot analysis was con-ducted to detect changes in PI3K/AKT/mTOR pathway proteins after treatment. Results MTT assay showed thatβ-EA effectively inhibited the proliferation of colon cancer HCT8 and HCT116 cells in a dose-dependent manner. The colony formation assay confirmed its inhibitory effect on cell proliferation.Transwell invasion assays demonstra-ted that β-elemonic acid reduced the invasion abilities of the cells.Western blot analysis revealed increased expression of apoptosis-related proteins cleaved-caspase 3,cleaved-caspase 9,and Bax,while Bcl-2 expression was decreased.Invasion-related proteins vimentin,snail,MMP2,and MMP9 were downregulated after treatment.Addi-tionally,β-EAA reduced the levels of p-PI3K,p-Akt,and p-mTOR,and these reductions were more pronounced af-ter the addition of the PI3K inhibitor LY294002.Conclusions β-EA may inhibit proliferation and invasion in colon cancer cell lines HCT8 and HCT116 through PI3K/AKT/mTOR signaling pathway, and potentially be transformed as a novel therapeutic agent for colon cancer.

Objective:To investigate the incidence and high-risk pathogenic factors of cardiovascular disease(CVD)in patients with rheumatic and autoimmune diseases, and to construct and validate a predictive model for the risk of CVD occurrence in these patients. Methods:A retrospective analysis was conducted on 239 patients with rheumatic and autoimmune diseases who underwent treatment and echocardiography at the Affiliated Hospital of Inner Mongolia Medical University between June 2020 and June 2023. General patient data, laboratory test results, and echocardiographic findings were collected. Follow-up was performed via electronic medical records or telephone surveys until December 2024 to determine the incidence of CVD,starting from the date of the first echocardiographic examination. Predictive factors were screened using univariate analysis and Lasso regression, and a Logistic regression model was constructed. Internal validation was performed using the Bootstrap method. The model's accuracy and clinical utility were assessed using the Hosmer-Lemeshow test, calibration curve, and decision curve analysis. Results:Among the 239 patients,111 developed CVD. Logistic regression analysis identified age, diastolic blood pressure, use of immunosuppressants, lymphocyte count(LYM),α-hydroxybutyrate dehydrogenase(α-HBDH)level,serum cystatin C(CysC),and right ventricular fractional area change(RVFAC)as independent predictive factors for CVD in these patients(all P<0.05). The area under the ROC curve(AUC)for the prediction model was 0.895(95% CI = 0.856 - 0.935),and after Bootstrap validation,it was 0.894(95% CI = 0.861-0.925). The Hosmer-Lemeshow test, calibration curve, and decision curve analysis all indicated that the model had good accuracy and clinical utility. Conclusions:Age,diastolic blood pressure, use of immunosuppressants,LYM,α-HBDH,CysC,and RVFAC may serve as independent risk factors for CVD in patients with rheumatic and autoimmune diseases. The prediction model based on echocardiography combined with laboratory indicators can,to some extent, predict the risk of CVD occurrence in these patients.

Pyroptosis is an identified programmed cell death that has been highly linked to endoplasmic reticulum (ER) dynamics. However, the crucial proteins for modulating dynamic ER membrane curvature change that trigger pyroptosis are currently not well understood. In this study, a biotin-labeled chemical probe of potent pyroptosis inducer α-mangostin (α-MG) was synthesized. Through protein microarray analysis, reticulon-4 (RTN4/Nogo), a crucial regulator of ER membrane curvature, was identified as a target of α-MG. We observed that chemically induced proteasome degradation of RTN4 by α-MG through recruiting E3 ligase UBR5 significantly enhances the pyroptosis phenotype in cancer cells. Interestingly, the downregulation of RTN4 expression significantly facilitated a dynamic remodeling of ER membrane curvature through a transition from tubules to sheets, consequently leading to rapid fusion of the ER with the cell plasma membrane. In particular, the ER-to-plasma membrane fusion process is supported by the observed translocation of several crucial ER markers to the "bubble" structures of pyroptotic cells. Furthermore, α-MG-induced RTN4 knockdown leads to pyruvate kinase M2 (PKM2)-dependent conventional caspase-3/gasdermin E (GSDME) cleavages for pyroptosis progression. In vivo, we observed that chemical or genetic RTN4 knockdown significantly inhibited cancer cells growth, which further exhibited an antitumor immune response with anti-programmed death-1 (anti-PD-1). In translational research, RTN4 high expression was closely correlated with the tumor metastasis and death of patients. Taken together, RTN4 plays a fundamental role in inducing pyroptosis through the modulation of ER membrane curvature remodeling, thus representing a prospective druggable target for anticancer immunotherapy.
