ObjectiveTo clarify whether METTL14 mediates the core role of acupuncture at Neiguan (PC6) in promoting myelination and improving behavior in young autistic rats through gene intervention technology. MethodsThe ASD model was established by intraperitoneal injection of valproic acid (VPA) in pregnant rats. Male offspring were intracerebroventricularly injected with adenovirus-packaged METTL14 shRNA (sh-METTL14) or its control (sh-NC) on postnatal day 1, with a model group set as well. Subsequently, the juvenile rats were divided into model group, acupuncture group, acupuncture+sh-NC group, and acupuncture+sh-METTL14 group. The acupuncture group received acupuncture at Neiguan (PC6) from postnatal day 7, once daily for 21 consecutive days. Neurobehavioral changes were evaluated by behavioral tests; METTL14 knockdown efficiency and the expression of METTL14, METTL3, and PTEN were detected by quantitative real-time PCR (qRT-PCR) and Western blot (WB); PTEN m6A levels were measured by RNA immunoprecipitation-qPCR (RIP-qPCR); myelin ultrastructure, expression of myelin basic protein (MBP) and neurofascin 155 (NF155), and dendritic spine density were observed using transmission electron microscopy (TEM), enzyme-linked immunosorbent assay (ELISA), immunofluorescence, qRT-PCR, and primary neuron culture. ResultsBehaviorally, knockdown of METTL14 significantly counteracted the beneficial effects of acupuncture in improving self-grooming, open field exploration, three-chamber social interaction, and Morris water maze learning and memory (P<0.05, P<0.01). Compared with the acupuncture+sh-NC group, the acupuncture+sh-METTL14 group showed significantly decreased mRNA and protein expression of hippocampal METTL14 (P<0.01), and the upregulating effects of acupuncture on METTL3 and PTEN expression were reversed (P<0.01). Meanwhile, knockdown of METTL14 significantly inhibited the acupuncture-induced increase in PTEN m6A levels (P<0.01). Morphologically, knockdown of METTL14 attenuated the improvement of myelin structure by acupuncture, reversed the downregulation of MBP and upregulation of NF155 induced by acupuncture, and blocked the increase in dendritic spine density (P<0.05, P<0.01). ConclusionMETTL14 is a key molecule mediating the therapeutic effect of acupuncture at Neiguan. Acupuncture at Neiguan upregulates METTL14, thereby enhancing m6A methylation modification of PTEN mRNA to stabilize its expression, ultimately promoting myelin development and improving behavioral symptoms in ASD juvenile rats. This preliminarily reveals the modern biological connotation of “opening Xuanfu and dredging myelin”.

3.Inverse Association Between Alcohol Consumption and Parkinson’s Disease Risk and Identification of RIT2 as a Linked Biomarker

ObjectiveAs a common lifestyle habit, alcohol consumption has a controversial association with the onset of Parkinson’s disease (PD). To demonstrate the correlation between alcohol consumption and PD and to identify associated genes, we integrated findings from clinical surveys, genomics, transcriptomics, and animal experiments. MethodsWe investigated the alcohol consumption rates (including both before and after disease onset) among 244 PD patients in China and 177 PD patients from the U.S. NHANES database. Mendelian randomization (MR) analysis was performed using genome-wide association study (GWAS) data for three alcohol-related traits and seven PD-related datasets from the MRC IEU OpenGWAS database. Transcriptomic data from the substantia nigra of PD patients were obtained from three GEO datasets (GSE7621, GSE20141, and GSE49036) to analyze RIT2 gene transcription. Finally, three groups of animal experiments (water/20% ethanol/20% liquor, with 4 C57BL/6J mice per group) were conducted to examine changes in brain RIT2 gene expression and transcriptomic profiles following alcohol consumption. ResultsThe alcohol consumption rates among PD patients in China and the U.S. (9%-18.87%) were significantly lower than the general population rates of 15%-45% in their respective regions (P<0.001), suggesting a possible negative association between alcohol consumption and PD. Subsequently, in 21 bidirectional MR analyses using 3 alcohol-related GWAS datasets and 7 PD-related GWAS datasets, the forward MR analyses (alcohol intake as exposure, PD as outcome) yielded 12 negative associations (ORIVW<1) and 9 positive associations (ORIVW>1). Among these, only two negative associations reached statistical significance: alcohol intake frequency (ORIVW=0.75, 95% CI: 0.60-0.93, P=0.010) and alcohol consumption (ORIVW=0.20, 95% CI: 0.05-0.83, P=0.026). The forward MR analysis (alcohol intake→PD) identified 235 SNPs, annotated to 316 genes, while the reverse MR analyses (PD→alcohol intake) identified 37 SNPs, annotated to 53 genes. Notably, only the RIT2 gene appeared in both the forward and reverse MR analyses (alcohol intake→PD: rs28597806, rs8083110; PD→alcohol intake: rs4588066). RIT2 is selectively expressed in the human brain (FPKM: 5.259±2.103), with low or no expression in peripheral tissues (FPKM: <1). Analysis of three human substantia nigra transcriptomic datasets revealed a decreasing trend in RIT2 gene expression in PD patients (GSE20141 array signal: 3.49±1.23 vs. 2.33±0.87, P=0.044). Animal experiments demonstrated that administration of 20% ethanol or 20% liquor (approximately 8% ethanol) stimulated a >2-fold upregulation of RIT2 gene expression in the mouse brain. Furthermore, transcriptomic sequencing revealed that the two alcohol-treated groups exhibited 96 (20% ethanol vs. water control) and 4 (20% liquor vs. water control) differentially expressed genes, respectively, indicating that low-dose alcohol consumption can achieve RIT2 upregulation while minimizing impact on other brain genes. In addition to its anti-infective effects, low-dose alcohol consumption primarily influences signaling pathways related to neurodegenerative diseases such as PD and Prion diseases. ConclusionAlcohol consumption is generally considered as a harmful lifestyle habit. However, some studies have also shown a lower risk of mortality among individuals who consume low doses of alcohol (100 g/week of ethanol) or drink occasionally. Currently, one of the research focuses on alcohol consumption is whether the human body can benefit from low-dose alcohol intake. This study provides new evidence supporting a negative association between alcohol consumption and PD, and for the first time, through MR analysis, identifies the RIT2 gene as a potential mediator of the effect of alcohol consumption on PD. RIT2 is selectively expressed in the human brain. Building upon existing evidence indicating downregulated RIT2 gene expression in PD pathogenesis, our experiments confirm that low-dose alcohol consumption can upregulate RIT2 expression in the brain. In brief, alcohol consumption may suppress the pathogenesis of PD by upregulating RIT2 expression in the substantia nigra. China is facing a serious problem of population aging. This study offers important insights for long-term PD prevention and treatment strategies, with the aim of benefiting more potential PD patients through lifestyle modifications, thereby improving the quality of life of the aging population and reducing the economic burden on healthcare.
4.Association between depressive symptoms and risk of stroke and all-cause mortality in young and middle-aged adults

BackgroundCurrent studies on the relationship between depressive symptoms and the risks of stroke and all-cause mortality mostly focus on the elderly population or high-risk groups with chronic diseases. The long-term impact of depressive symptoms in young and middle-aged people on the risks of stroke and all-cause mortality still lacks systematic assessment, and the relevant evidence is insufficient. ObjectiveTo explore the association between depressive symptoms and the risks of stroke and all-cause mortality in young and middle-aged adults, thereby expanding the risk evidence in this age group and providing epidemiological basis for early identification of high-risk individuals and the development of targeted intervention strategies. MethodsBased on data from the National Health and Nutrition Examination Survey (NHANES) database (2009–2018), a total of 14 947 young and middle-aged respondents aged 18 to 64 were included as the research subjects. Logistic regression and restricted cubic spline (RCS) models were employed to examine the association between depressive symptoms and stroke. Cox proportional hazards model, RCS model, and Kaplan-Meier (K-M) survival curves were used to clarify the relationship between depressive symptoms and all-cause mortality risk. To verify the robustness of main findings, sensitivity analysis was conducted stratified by gender, hypertension, diabetes, hyperlipidemia, body mass index (BMI), and age. ResultsLogistic regression analysis revealed that compared with the non-depressed group, the risk of stroke in the mild depression group increased by 53.50% (OR=1.535, 95% CI: 1.147–2.053), while the moderate-to-severe depression group showed the highest risk with a 132.90% increase (OR=2.329, 95% CI: 1.733–3.129). RCS analysis indicated an approximately linear dose-response relationship between PHQ-9 score and the risk of stroke (Pfor non-linearity=0.089), with stroke risk increasing as PHQ-9 scores rose. As the PHQ-9 score increased, the risk of stroke also showed an upward trend. Cox regression analysis demonstrated that compared with the non-depressed group, the risk of all-cause mortality in the mild depression group increased by 48.80% (HR=1.488, 95% CI: 1.164–1.902), and the moderate-to-severe depression group had a 70.60% increased risk (HR=1.706, 95% CI: 1.310–2.221). RCS analysis showed a linear dose-response relationship between PHQ-9 scores and all-cause mortality risk (Pfor non-linearity=0.440). K-M curves indicated that as the severity of depressive symptoms increases, the risk of all-cause mortality gradually rises (P<0.05). Sensitivity analysis confirmed that the associations between depression symptoms and risks of stroke and all-cause mortality were not modified by gender, hypertension, diabetes, hyperlipidemia, BMI, or age, demonstrating robust findings. ConclusionAmong the young and middle-aged population, both mild and moderate-to-severe depression were associated with increased risks of stroke and all-cause mortality, with the moderate-to-severe depression group showed more pronounced risk elevations. A positive dose-response relationship was observed between depressive symptoms and these risks.

The management goals of rheumatoid arthritis are to control pain and disease activity,prevent joint damage,restore function,and improve quality of life.Hand joint pain is a common symptom of rheumatoid arthritis and currently cannot be cured.Systematic symptom management pro-tocols have been established abroad,while management often relies on clinical experience in China.This paper summarized the mechanisms of hand pain in rheumatoid arthritis and non-pharmacological intervention strategies,aiming to provide a scientific basis for patient management.
6.Research Progress on the Role of Immune Cell Dysfunction in the Occurrence and Development of Liver Failure

The liver,as the core metabolic organ of the human body,undertakes multiple critical physiological functions such as protein synthesis,xenobiotic metabolism,and immune regulation.Liver failure represents a major disease process in patients with end-stage liver disease,characterized clinically by coagulopathy,abnormal bilirubin metabolism,and multiple organ dysfunction.Depending on the disease course,liver failure can be classified into four types:acute,subacute,acute-on-chronic,and chronic.The pathogenic mechanisms are complex,involving interactions among various factors such as pathogen infections,dysregulation of the immune microenvironment,and gut microbiota disturbances,which pose significant challenges for clinical diagnosis and treatment.This review summarizes the abnormalities in innate and adaptive immune responses,as well as the molecular mechanisms of immune metabolic dysregulation during the occurrence and progression of liver failure.It further explores the pivotal role of immune cell dysfunction in the disease process,aiming to provide a theoretical basis for targeted immune therapies in liver failure.
7.Immunoregulatory mechanisms,development and application of terpenoid compounds from the Isodon genus

Plants of the Isodon genus are an important source of terpenoids, with their constituents exhibiting rich structural diversity and remarkable biological activities (such as anticancer, antimicrobial, and anti-inflammatory properties), demonstrating significant potential in the field of immunomodulation. This review summarizes recent advances in the immunomodulatory mechanisms, development and application of terpenoid compounds from the Isodon genus. It has been found that these compounds can modulate key inflammatory signaling pathways, such as nuclear factor-kappa B (NF-κB) and mitogen-activated protein kinases (MAPKs), thereby blocking the cascade amplification of inflammatory factors, alleviating chronic inflammatory responses, and correcting immune dysregulation. Additionally, they can influence the polarization direction of macrophages and dynamically regulate the balance among different functional subsets of T cells, restoring immune homeostasis. Their clinical translation faces multiple challenges, including poor druggability, a lack of systematic safety data, the absence of precise pharmacodynamic biomarkers, complexities in clinical trial design, and unclear industrialization pathways.
8.An Electronic Microbial Growth Analyzer-based Method for Rapidly Screening Viable Salmonella in Food

Foodborne illnesses caused by Salmonella pose significant threats to worldwide public health safety.In this study,a rapid method for screening viable Salmonella in oyster sauce and milk was developed by utilizing an electronic microbial growth analyzer(EMGA).Target food samples were diluted 10-fold with RVS broth and loaded into test tubes.Test tubes were positioned in the EMGA to determine the bacterial growth curves and the time required to reach the maximum growth rate(Tmgr).Using Salmonella typhimurium(S.typhimurium)asan model species,there was linear relationship between the logarithmic value of viable bacterial concentration(lgC)and Tmgr over the range of 5×101-5×106 CFU/mL,with a detection limit of 10 CFU/mL.For oyster sauce,the regression equation was Tmgr(min)=-80.775lg[C/(CFU/mL)]+754.96(R2=0.9907),and the recovery rates of S.typhimurium ranged from 95.2%to 119.8%,with relative standard deviations(RSD)ranging from 3.5%to 16.3%.For milk,the regression equation was Tmgr(min)=-71.922 lg[C/(CFU/mL)]+618.65(R2=0.9985),with recovery rates ranging from 98.4%to 110.6%and RSD ranging from 6.4%to 12.8%.The EMGA method required only one portable instrument,and involving only three manual steps,i.e.,dilution,transfer,and insertion.When S.typhimurium contamination reached 106 CFU/mL,the total time consumption,from the unwrapping of samples to the readout of bacterial concentration,was no more than 7 h.When applied to detection of actual oyster sauce and milk samples,the new method demonstrated strong consistency with plate counting results in positive detection rates.This method was superior to the plate counting method,which was generally considered as a gold standard,in terms of accuracy,precision,simplicity and efficiency,representing a promising alternative for the on-site screening and quantification of viable Salmonella in oyster sauce and milk products.
9.An outbreak of neonatal Pseudomonas aeruginosa infection caused by wa-ter contamination

Objective To analyze the causes and intervention measure efficacy of an outbreak of neonatal Pseudo-monas aeruginosa(P.aeruginosa)infection in a medical institution.Methods The epidemiological investigation was conducted on 5 neonates who had P.aeruginosa infection in the neonatal intensive care unit(NICU)of this medical institution from May 26 to June 10,2024.Specimens were taken from potentially contaminated environ-ments to infer the cause of P.aeruginosa transmission in the NICU,infection control measures were also taken,and the efficacy of the control measures was continuously monitored.Results Among the 5 infected neonates,3 had lower respiratory tract infection and 2 had lower respiratory tract combined with bloodstream infection.Seven strains of P.aeruginosa were detected with a consistent antimicrobial resistance spectrum.Infected neonates were in the same ward and had spatial and temporal clustering.Through environmental hygiene monitoring,P.aerugi-nosa with consistent antimicrobial resistance spectrum of neonatal infection was isolated from the NICU handwashing sink and water-related environment(suction connection pipe,bedside isolation gowns).Infection was controlled af-ter improving sink disinfection methods and enhancing contact isolation measures,and no further P.aeruginosa was detected in the environment.Conclusion The outbreak of neonatal HAI in the NICU is inferred to be caused by contamination of ward handwashing sink by P.aeruginosa,then further contaminated the suction connection tube,thereby leading to the transmission of neonatal infection through suction procedures.Healthcare-associated infection surveillance and control professionals need to fully understand the characteristics of outbreak pathogens and take tar-geted measures to curb the transmission of neonatal infection.
10.Value of dual-energy CT parameters in evaluating the pathological grade of pancreatic ductal adenocarcinoma

Objective:To investigate the value of dual-energy CT parameters in the evaluation of pathological grade of pancreatic ductal adenocarcinoma.Methods:80 cases of pancreatic ductal adenocarcinoma confirmed by pathology were retrospectively analyzed and divided into high grade group (36 cases) and low grade group (44 cases) according to their differentiation degree. All 80 patients underwent SOMATOM Force DECT for arterial phase (AP) and pancreatic phase (PP) scanning, and measured dual-energy parameters including dual-phase iodine concentration (IC AP, IC PP) in tumors and normal pancreatic parenchyma, pancreatic phase and arterial phase iodine concentration difference (ICD PP-AP) in tumors, dual-phase iodine uptake ratio (IUR AP, IUR PP) , dual-phase tumor/normal pancreatic parenchyma fat fraction ratio, and dual-phase slope of energy spectrum curve. Differences between two groups were compared by two independent sample t-test or Mann-Whitney U test or chi-square test. The multivariate logistic regression model was used to analyze the influencing factors of pathological grading of PDAC. Results:There were statistically significant differences in gender, age, aspect ratio, positive lymph node, fat fraction ratio in pancreatic phase between the two groups ( P< 0.05) . The multivariate logistic regression analysis showed that fat fraction ratio in pancreatic phase ( OR=1.781, 95% CI 1.127-2.814, P=0.013) , positive lymph node ( OR=4.870, 95% CI 1.488-15.938, P=0.009) , aspect ratio ( OR=0.019, 95% CI 0.001-0.437, P=0.013) were independent factors influencing the pathologic grade of PDAC. Conclusion:Parameters of dual-energy CT are valuable in the evaluation of pathological grading of PDAC.
