1.Salviae Miltiorrhizae Radix et Rhizoma Extract Regulates Blood Pressure in Rat Model of Metabolic Hypertension Induced by High-sugan and High-fat Diet via TRPC3/6/NOX2/4 Sigraling Pathway

ObjectiveTo explore the mechanisms of Salviae Miltiorrhizae Radix et Rhizoma in treating metabolic hypertension induced by a high-sugar and high-fat diet in rats based on network pharmacology, proteomics, and animal experiments. MethodsThirty male SD rats were randomized into the control, model, positive drug (tetrandrine, Tet, 50 mg·kg-1·d-1), low-dose Salviae Miltiorrhizae Radix et Rhizoma (DS-L, 45×104 mg·kg-1·d-1), and high-dose Salviae Miltiorrhizae Radix et Rhizoma (DS-H, 90×104 mg·kg-1·d-1) groups. Except for the control group, each group was fed a high-fat and high-sugar diet for 8 weeks for the modeling of metabolic hypertension. Following successful modeling, drug interventions were conducted through gavage for 4 weeks. Blood pressure and lipid indicators were monitored, and cardiac function was assessed via echocardiography. Samples from the thoracic aorta and cardiac tissue were collected for histopathological examination. Network pharmacology analysis identified key active components, potential targets, and mechanisms of Salviae Miltiorrhizae Radix et Rhizoma in treating hypertension. Proteomics technology was employed to analyze the differential proteins and major pathway targets to synergistically elucidate the antihypertensive mechanism of Salviae Miltiorrhizae Radix et Rhizoma. Ca2+ concentrations in the thoracic aorta and cardiac tissue were measured. The protein levels of transient receptor potential cation channel (TRPC)3, TRPC6, NADPH oxidase (NOX)2, and NOX4 were quantified via Western blotting. The levels of oxidative stress markers and inflammatory factors, including superoxide dismutase (SOD), malondialdehyde (MDA), tumor necrosis factor-α (TNF-α), and interleukin-1β (IL-1β), were determined by enzyme-linked immunosorbent assay (ELISA). Molecular docking analysis was performed for the main components of Salviae Miltiorrhizae Radix et Rhizoma with TRPC3, TRPC6, NOX2, and NOX4. ResultsThe experimental results indicated that compared with the control group, the model group exhibited abnormally elevated blood pressure and increased lipid levels (P<0.01). Compared with the model group, the DS-L group exhibited reduced systolic blood pressure (SBP), diastolic blood pressure (DBP), and mean arterial pressure (MAP) (P<0.05), decreased serum levels of triglycerides (TG), total cholesterol (TC), and low-density lipoprotein cholesterol (LDL-C) (P<0.05), and increased level of high-density lipoprotein cholesterol (HDL-C) (P<0.01). The DS-H and Tet groups showed more significant effects (P<0.01). Moreover, the interventions attenuated vascular wall thickening, myocardial cell injury, and collagen and lipid deposition. Network pharmacology and proteomics prediction results indicated that the core targets of Salviae Miltiorrhizae Radix et Rhizoma in treating hypertension were TRPC3, TRPC6, NOX2, and NOX4, and the core pathways included calcium signaling, cyclic guanosine monophosphate (cGMP)/cGMP-dependent protein kinase (PKG) signaling, and atherosclerosis-related pathways. The molecular mechanism experiment results indicated that compared with the control group, the model group exhibited significantly elevated tissue Ca2+ concentrations, exacerbated oxidative stress and inflammatory responses, and upregulated protein levels of TRPC3, TRPC6, NOX2, and NOX4 in the thoracic aorta and myocardial tissue (P<0.01). Compared with the model group, DS-L reduced the free Ca2+ concentration, lowered the levels of IL-1β, TNF-α, SOD, and MDA (P<0.05), and downregulated the protein levels of TRPC3, TRPC6, NOX2, and NOX4 in the thoracic aorta and myocardial tissue (P<0.05). DS-H and Tet exhibited more significant effects (P<0.01). Meanwhile, the main components of Salviae Miltiorrhizae Radix et Rhizoma had strong binding affinity with the core targets of hypertension. ConclusionSalviae Miltiorrhizae Radix et Rhizoma may ameliorate oxidative stress and mitigate inflammatory responses by regulating the TRPC3/6/NOX2/4 signaling pathway, thereby alleviating abnormal blood pressure abnormality and myocardial injury in hypertensive rats.
2.Different Effects of Fresh and Dried Dendrobium Huoshanense on Chronic Atrophic Gastritis

ObjectiveTo compare the protective effects of water extracts from fresh and dried Dendrobium huoshanense on gastric mucosa in chronic atrophic gastritis (CAG). MethodsMale SD rats (n=72) were randomly divided into 9 groups, with 8 rats in each group, which were normal group, model group, Yangwei Shu (4 g·kg-1) group, low-, medium-, and high-dose fresh D. huoshanense (3.5, 7, and 14 g·kg-1) groups, and low-, medium-, and high-dose dried D. huoshanense (0.7, 1.4, 2.8 g·kg-1) groups. The CAG rat model was successfully established by inducing with N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and other factors for a total of 11 weeks. Then, the rats were intervened with fresh and dried D. huoshanense for 4 weeks. The serum and gastric tissues of the rats were collected. The changes in gastric juice secretion volume and gastric acid pH value in each group were observed. The gastric mucosal injury was observed by naked eyes and hematoxylin-eosin(HE) staining. The gastric mucus secretion level was determined by Alcian blue and periodic acid-Schiff staining(AB-PAS) staining. The expression levels of tight junction proteins Occludin and ZO-1 in gastric tissues were determined by immunofluorescence. The expression levels of serum pepsinogen Ⅰ (PG Ⅰ), pepsinogen Ⅱ (PG Ⅱ), gastrin 17 (G-17), interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α) were determined by enzyme-linked immunosorbent assay (ELISA). The expression levels of aquaporin 1 (AQP1), aquaporin 3 (AQP3), and aquaporin 4 (AQP4) in gastric tissues were determined by Western blot. ResultsCompared with the normal group, the model group showed an obviously reduced gastric juice secretion volume (P0.05), significantly increased gastric acid pH value (P0.01), gastric mucosa with obvious atrophy, and a significantly reduced gastric mucus secretion volume (P0.01). The expression of Occludin and ZO-1 in the gastric mucosal barrier was significantly decreased (P0.01). The levels of PG Ⅰ and PG Ⅱ in the serum were obviously decreased (P0.05, P0.01), and the levels of G-17, IL-1β, IL-6, and TNF-α were significantly increased (P0.01). The expression level of AQP1 in the gastric tissue was significantly upregulated (P0.01), and the expression levels of AQP3 and AQP4 were significantly downregulated (P0.01). Compared with the model group, each drug administration group could improve the gastric mucosal atrophy of CAG model rats to varying degrees, obviously increase the gastric juice secretion volume of the model rats (P0.05, P0.01), significantly decrease the gastric acid pH value (P0.01), obviously increase the gastric mucus secretion volume (P0.05, P0.01), obviously decrease the expression levels of G-17, IL-6, IL-1β, and TNF-α (P0.05, P0.01), obviously increase the expression levels of Occludin, ZO-1, PG Ⅰ, and PG Ⅱ (P0.05, P0.01), obviously upregulate the expression levels of AQP3 and AQP4 (P0.05, P0.01), and obviously downregulate the expression level of AQP1 (P0.05, P0.01). ConclusionThe water extracts of fresh and dried D. huoshanense can exert therapeutic effects on CAG by improving gastric mucosal injury, reducing inflammation, and regulating water metabolism. Moreover, the dried D. huoshanense has a better effect.

The quality of Chinese materia medica (CMM) is a challenging and focused topic in the modernization of traditional Chinese medicine (TCM). A profound comprehension of the morphology, structure, active constituents, and dynamic changes during the whole process of CMM growth is essential, which needs highly precise contemporary techniques for in-depth elucidation. Magnetic resonance imaging (MRI) is a cutting-edge tool integrating the benefits of both nuclear magnetic resonance (NMR) spectroscopy and imaging technology. With real-time, non-destructive, and in situ detection capabilities, MRI has been previously used for monitoring internal and external structures of plants alongside compounds during physiological processes in vivo. Here, factors involved in the holistic quality evaluation of CMMs were investigated. Given the applications of MRI in various plants, several representative CMMs were used as examples to demonstrate a methodology of quality visualization by MRI, embodying holistically monitoring the real-time macroscopic morphology, mesoscopic structure, and microscopic metabolites non-destructively in situ. Taken together, the review not only presents a pioneering application mode for utilizing MRI for CMM quality visualization but also holds promise for advancing the quality control and evaluation of CMMs.

5.Neuroprotective effects of serum with Tongqiao Huoxue decoction (TQHXD) against glutamate-induced neurotoxicity in PC12 cells.

OBJECTIVETo observe the protective effect of serum with Tongqiao Huoxue decoction (TQHXD) on PC12 cells damaged by glutamate(Glu) and provide clinical proof of the formulae.
METHODSprague Dawley rats underwent intragastric administration of Tongqiao Huoxue decoction twice a day for five days, the administration dose for rats was 4 g x kg(-1). Thus the serum containing TQHXD was prepared. The model of neurocytes damaged by Glu had been established by adding 12.5 mmol x L(-1) Glu to culture medium of PC12 cells. Photic microscope had been used to observe the morphologic changes of cells, the proliferation and activity of PC12 cells had been detected by MTT. Cell membrane permeability had been investigated by the methods of the coloration of trypan blue and AO/EB, the activities of LDH and the contents of NO in the supernatant of PC12 cells had been detected after 5%, 10%, 20% TQHXD being added to the culture medium of PC12 cells damaged by Glu.
RESULTThe morphologic changes in cells were observed under the inverted microscope. Normal PC12 cells in control group were full and bright, but the cells exposed to Glu were shrank; moreover, some cells were disrupted, configuration of the cells in the TQHXD group was closed to that of normal group. Compared with model group, the ratio of living cells in the group being treated by TQHXD after trypan blue staining had a significant increase. After AO/EB staining, cells in TQHXD groups were less being stained by EB. The absorbance (A) values of TQHXD group increased largely. LDH and NO in cell culture supernatant had decreased, it had shown the marked difference.
CONCLUSIONTQHXD showed an apparent protective effect on neurocytes damaged by Glu on proliferation activity and membrane permeability.

6.Studies on anti-platelet activation effect and partial mechanisms of Taohong Siwu decoction.

OBJECTIVETo explore the anti-platelet activation effect and partial mechanisms of Taohong Siwu decoction (TSD).
METHODThe effect to venous thrombosis model and pulmonary thromboembolism model induced by vein injecting ADP and Adr was observed. The platelet adhesion rate was analyzed by using spinning glass bottle, and the platelet aggregation rate induced by ADP, Adr was analyzed by using turbidimetry. The acute blood stasis rat model was established to analyze the content of plasm TXB2 and PGI2 by RIA, and the content of VWF, GMP-14 by ELISA.
RESULTTSD could effectively reduce platelet the adhesion rate of normal rat, inhibit the platelet aggregation of normal rat induced by ADP, Adr. It significantly reduced the plasma TXB2 VWF, and GMP-140 level of blood stasis rats. It also had significant tendency to increase 6-keto-PGF1alpha level.
CONCLUSIONTSD possessed obvious activity of inhibiting platelet activation. The mechanism related with the restraining of platelet adhesion, platelet aggregation and platelet releasion.

AIM:To study the isolation and identification of curdione from Zedoary oil. METHODS: Silica column was adopted to isolate constituents from Zedoary oil;IR,GC/MS and NMR methods were used to identify its structure. RESULTS: Three constituents were isolated from Zedoary oil,including crystal C_1,C_2 and C_3.Crystal C_1 was identified to be curdione and curdione content in Zedoary oil was 11.31%. CONCLUSION: This method is simple and convenient and it can be used to isolate more quantity of curdione for further pharmaceutical study.
9.Effects of Tongqiao Huoxue Decoction on contents of CGRP、 ET、 IL-1?、TNF-? in the brains of rats of cerebral ischemia

AIM: To investigate the protective effects and mechanisms of Tongqiao Huoxue Decoction on the rats of cerebral ischemia. METHODS: Cerebral ischemia madel was established on the basis of cerebral thrombus and at the same time ligaturing bilateral carotid arteries.These models were used to observe the protective effects of Tongqiao Huoxue Decoction on contents of CGRP、ET、IL-1?,TNF? in rats′ cerebral ischemica. RESULTS: The results showed that Tongqiao Huoxue Decoction decreased the contents of IL-1?、TNF-?、 ET,and increased the contents of CGRP. CONCLUSION: Tongqiao Huoxue Decoction had effects on anti-cerebral ischemia.The protective effects of Tongqiao Huoxue Decoction are related to decreasing the contents of IL-1?,TNF-?,ET,and increasing the contents of CGRP.

The effects of TSCP and TSCC on aggregation and adhesion of blood platelets were measured by turbidimetric method and salzman method respectively. TSCP and TSCC strongly increase mouse and rat platelet aggregation and platelet adhesion. These effects are important for blood coagulation. cAMP and TXB2 levels in plasma and platelet were determind by RIA TSCP significantly increased the TXB2 level in plasma and platelet. This effect may be the main mechanism of increasing platelet aggregation by TSCP.
