1.Honokiol inhibits the malignant progression of gastric cancer cells by regulating the ATF4/CHOP/TRIB3 pathway

ObjectiveTo investigate the effect of honokiol on proliferation, apoptosis, migration, and invasion of gastric cancer cells and its underlying mechanistic. MethodsHuman gastric cancer cell lines HGC-27 and AGS were treated with Honokiol at concentrations of 0, 15, and 25 μmol/L. CCK-8 assays were conducted to determine the half maximal inhibitory concentration (IC50) for both cell lines. Cell viability, proliferation, migration, and invasion capabilities were assessed using CCK-8, colony formation, wound healing, Transwell migration and Transwell invasion assays. Apoptosis rates were measured via flow cytometry. Western blot analysis examined proteins related to proliferation, apoptosis, migration, invasion, and the endoplasmic reticulum stress pathway ATF4-CHOP-TRIB3. ResultsCompared with the control group, treatment with 15 and 25 μmol/L Honokiol significantly reduced the proliferation, colony formation, migration, and invasion capabilities of the two gastric cancer cell lines, while significantly increasing the apoptosis rate (P<0.05). Additionally, compared to the control group, the protein expression levels of neural cadherin(N-cadherin), Vimentin, proliferating cell nuclear antigen(PCNA), and B-cell lymphoma/leukemia-2 protein(Bcl-2)decreased in the two gastric cancer cell lines after treatment with 15 and 25 μmol/L Honokiol, while the protein expression levels of epithelial cadherin(E-cadherin), Bcl-2-associated X protein(Bax), activating transcription factor 4(ATF4), endoplasmic reticulum stress-related protein(CHOP), and tribbles homolog 3(TRIB3)increased(P<0.05). ConclusionHonokiol promotes apoptosis and inhibits proliferation, migration, and invasion of gastric cancer HGC-27 and AGS cells by regulating the ERS signaling pathway ATF4/CHOP/TRIB3.
2.Dementia Overdiagnosis in Younger, Higher Educated Individuals Based on MMSE Alone: Analysis Using Deep Learning Technology

Background:
Dementia is a multifaceted disorder that affects cognitive function, necessitating accurate diagnosis for effective management and treatment. Although the Mini-Mental State Examination (MMSE) is widely used to assess cognitive impairment, its standalone efficacy is debated. This study examined the effectiveness of the MMSE alone versus in combination with other cognitive assessments in predicting dementia diagnosis, with the aim of refining the diagnostic accuracy for dementia.
Methods:
A total of 2,863 participants with subjective cognitive complaints who underwent comprehensive neuropsychological assessments were included. We developed two random forest models: one using only the MMSE and another incorporating additional cognitive tests.These models were evaluated based on their accuracy, precision, recall, F1-score, and area under the receiver operating characteristic curve (AUC) on a 70:30 training-to-testing split.
Results:
The MMSE-alone model predicted dementia with an accuracy of 86% and AUC of 0.872. The expanded model demonstrated increased accuracy (88%) and an AUC of 0.934.Notably, 17.46% of the cases were reclassified from dementia to non-dementia category upon including additional tests. Higher educational level and younger age were associated with these shifts.
Conclusion
The findings suggest that although the MMSE is a valuable screening tool, it should not be used in isolation to determine dementia severity. The addition of diverse cognitive assessments can significantly enhance diagnostic precision, particularly in younger and more educated populations. Future diagnostic protocols should integrate multifaceted cognitive evaluations to reflect the complexity of dementia accurately.
3.T cell factor 1 regulates CD8 +T cell immunological and stemness

T cell factor 1(TCF-1)is a downstream transcription factor of the Wnt/β-catenin signaling pathway,and plays an important role in the development,differentiation,and memory formation of T cells. Recent studies have shown that TCF-1 can regulate the formation and maintenance of stem-like memory T cells(Tscm),and has potential application value in evaluating the prognosis of tumor immunotherapy and as a target for tumor immunotherapy. This article reviews the regulatory effects of TCF-1 on the immune memory as well as stemness formation and maintenance of CD8 +T cells,summarizes the transcription network centered on TCF-1,and further elucidates the application and value of TCF-1 in tumor immunotherapy.
4.Clinical and genetic characteristics of 14 children with sodium taurocholate co-transporting polypeptide deficiency

Objective To summarize the clinical and genetic characteristics of children with sodium taurocholate co-transporting polypeptide(NTCP)deficiency.Methods Clinical data of children with NTCP deficiency diagnosed and treated at the First People's Hospital of Yunnan Province from July 2022 to March 2025 were retrospectively analyzed.Results A total of 14 children were included(6 males,8 females),all with normal growth and development.Reasons for initial consultation included elevated serum bile acids in 7 cases,jaundice in 4 cases,cholestatic hepatitis in 1 case,and one case each of pneumonia and cow's milk protein allergy.At the first visit,all patients had elevated serum total bile acids beyond the normal range,with a mean of 152.5 μmol/L.Elevated alanine aminotransferase was observed in 1 case,elevated aspartate aminotransferase in 2 cases,and elevated total bilirubin in 10 cases.Genetic sequencing revealed that all children carried the homozygous SLC10A1 variant c.800C>T(p.Ser267Phe),classified as likely pathogenic.Conclusions NTCP deficiency often lacks obvious clinical symptoms and signs.Some children present with transient hyperbilirubinemia,cholestasis,or other liver function abnormalities.Persistent isolated elevation of serum bile acids warrants suspicion for this disease.Biallelic pathogenic variants in SLC10A1 constitute the basis for definitive diagnosis.There is no specific treatment for this disease,and management is mainly symptomatic.
5.Construction of a predictive model for clinical pregnancy of IVF-ET in patients with secondary infertility

Objective To analyze the influencing factors of the success of clinical pregnancy in secondary infertility patients undergoing in vitro fertilization-embryo transfer(IVF-ET)and construct a nomogram prediction model.Methods A retrospective analysis was conducted on the clinical data of 235 patients with secondary infertility who underwent IVF-ET at Urumqi Maternal and Child Health Hospital from January 2020 to December 2023.They were divided into successful pregnancy group(n=109)and failed pregnancy group(n=1 26)based on whether clinical pregnancy was successful.The general information,ovulation induction data and embryo data of two groups of patients were compared.Multivariate Logistic regression analysis was used to screen out statistically significant indicators,and based on this,a nomogram prediction model was constructed.The receiver operating characteristic curve,calibration curve and decision analysis curve were drawn to verify the discrimination,accuracy and clinical practicability of the model.Results The results of multivariate Logistic regression analysis showed that the female's age,overweight and obesity were all risk factors for clinical pregnancy failure,while anti-Müllerian hormone(AMH)and the total amount of gonadotropins(Gn)were protective factor for clinical pregnancy outcomes.On this basis,a nomogram prediction model was successfully constructed,which has a medium degree of discrimination,good accuracy and clinical practicability.Conclusion For secondary infertility patients undergoing IVF-ET,the female's age,overweight and obesity,AMH,and the total amount of Gn have certain influences on clinical pregnancy.The clinical pregnancy outcome can be predicted through the constructed nomogram prediction model.

7.Investigation of the current situation and cultivation path of science and technology ethics awareness in medical postgraduates

Objective:To understand the current status and influencing factors for science and technology ethics awareness among medical postgraduates, explore scientific cultivation path, and provide empirical evidence for higher medical colleges to carry out science and technology ethics education.Methods:The study adopted a cluster sampling method and designed a questionnaire. A total of 2 095 medical postgraduates from Hebei Medical University participated in the survey from June to December 2023. SPSS 24.0 was used for non-parametric tests, and the ordered logistic regression analysis was used to explore the influencing factors.Results:A total of 1 992 valid questionnaires were collected, with a valid response rate of 95.08% (1 922/2 095). The qualified rate of science and technology ethics awareness among medical postgraduates was 90.76% (1 808/1 992), with 48.24% (961/1 992) of students falling into the good interval and 16.21% (323/1 992) falling into the excellent interval. The regression analysis showed that sex, academic category, students' attention to science and technology ethical issues, and understanding of ethical principles were significant influencing factors for medical postgraduates' science and technology ethics awareness ( P<0.05). Conclusions:The science and technology ethics awareness of medical postgraduates is at a medium level. Higher medical colleges should start from the needs and adhere to the direction of lifelong education to build a multi-subject, all-round, and all-process cultivation path. They can help students cultivate ethical awareness by improving the educational system, building a scientific teaching system, and creating a social education atmosphere. They can also help students internalize ethical norms and externalize them in their behaviors, and ensure that they can maintain ethical sensitivity in scientific and technological activities, thereby cultivating high-quality new medical and health professionals who can anticipate scientific and technological risks and assume ethical responsibilities.

9.Fulminant type 1 diabetic ketoacidosis due to serplulimab

A 57-year-old female patient with small cell lung cancer was treated with serplulimab (100 mg by intravenous infusion on the first day, 21 days as a cycle). After the 10th cycle of treatment, the patient suddenly presented drowsiness, shortness of breath, dry mouth, nausea, vomiting, fatigue and other symptoms. Laboratory tests showed fasting blood glucose 38.9 mmol/L, glycosylated hemoglobin 7.9%, serum C peptide 0.2 μg/L, pH value 7.05, anion gap 31 mmol/L, anti-islet cell antibody 36 kU/L, anti-glutamic acid decarboxylase antibody 131 kU/L, urine ketone body (+++). The patient was diagnosed with fulminant type 1 diabetic ketoacidosis, which was considered to be related to serplulimab. The drug was stopped, and insulin, rehydration, correction of acid-base imbalance, and other treatments were given. After 3 days, the patient′s consciousness returned to normal. After 12 days, her breathing was stable, dry mouth, nausea, vomiting and other symptoms were relieved. Laboratory tests showed random blood glucose 13.6 mmol/L, pH value 7.44, anion gap 6 mmol/L, and urine ketone body negative. After 26 days, her random blood glucose was controlled at 10.0-12.0 mmol/L.
