1.Evaluation of postoperative recurrence and treatment efficacy of glioma using 3.0 T functional magnetic resonance imaging

Objective To investigate the clinical value of 3.0 T functional magnetic resonance imaging in evaluating postoperative recurrence and treatment efficacy of glioma. Methods A retrospective analysis was conducted on the general clinical data of 67 patients who underwent glioma surgery at the Second Affiliated Hospital of Xingtai Medical University. All patients received chemotherapy for more than one month post-surgery. Recurrence of glioma was diagnosed based on secondary surgery or pathological biopsy results as the gold standard. From 3 to 6 months post-surgery, computerized tomography was used to measure cerebral blood volume (CBV), three-dimensional arterial spin labeling was used to measure cerebral blood flow (CBF) and relative CBF (rCBF), and diffusion-weighted imaging with and without contrast enhancement was used to measure apparent diffusion coefficient (ADC). Data were analyzed using SPSS 26.0 statistical software, and the t test or χ2 test was used for inter-group comparisons based on data type. The receiver operating characteristic (ROC) curve was applied to evaluate the value of CBV, rCBF, and ADC in assessing postoperative recurrence and treatment efficacy of glioma. Results Patients with high-grade gliomas showed significantly higher CBV and rCBF and significantly lower ADC compared to those with low-grade gliomas (P < 0.05). The area under the ROC curve (AUC) of CBV, rCBF, and ADC in combination for grading glioma was 0.960, which was higher than those of individual indicators (0.790, 0.955, and 0.795, P < 0.05). The recurrence group had significantly higher CBV and rCBF and lower ADC compared to the non-recurrence group (P < 0.05). The AUC of CBV, rCBF, and ADC in combination for predicting postoperative glioma recurrence was 0.965, which was significantly higher than those of individual indicators (0.729, 0.929, and 0.941, P < 0.05). CBV and rCBF were lower and ADC was higher in the effective treatment group than in the ineffective treatment group (P < 0.05). The AUC of CBV, rCBF, and ADC in combination for evaluating glioma treatment efficacy was 0.985, which was higher than those of individual indicators (0.842, 0.898, and 0.961, P < 0.05). Conclusion The CBV, rCBF, and ADC in combination has shown high diagnostic accuracy and predictive efficacy in the evaluation of postoperative recurrence and treatment efficacy of glioma, which has important clinical application value.
2.Proteomic analysis of NUS1 mutant R290C interaction proteins and their potential roles in Lennox-Gastaut syndrome

Objective:To investigate the changes in interaction proteome of NUS1 mutant R290C and their relations with pathogenicity of Lennox Gastaut syndrome (LGS). Methods:The wild-type and mutant NUS1(R290C) plasmids were constructed and transfected into human embryonic kidney HEK293T cells; 48 h after that, NUS1 protein expression in HEK293T cells was detected by Western blotting. Co-immunoprecipitation, silver nitrate staining, and proteomic analysis were used to analyze the proteins interacted with wild-type or mutant NUS1 and identify the differential interacting proteins. Enrichment analyses of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) were performed to annotate the molecular function and signaling pathways involved in the differential proteins. DisGeNet database was used to analyze the association between differential proteins and human diseases. Protein-protein interaction (PPI) was used to analyze the interaction network of NUS1 with protein folding regulatory proteins (RTN4 and DHDDS) and developmental epileptic encephalopathy related proteins.Results:(1) There was no significant difference in NUS1 protein expression between the wild-type and mutant NUS1 transfected HEK293T cells ( t=0.536, P=0.620). (2) Compared with that with wild-type NUS1 plasmid, number of proteins interacting with mutant NUS1 plasmid was significantly reduced in the transfected cells; 310 differential interacting proteins were screened in the mutant NUS1. (3) GO and KEGG enrichment analyses showed that the differential proteins were mainly involved in protein folding reaction and translation regulation. (4) DisGeNet association analysis showed that the two most relevant proteins in the differential interacting proteins were associated with frontotemporal dementia and developmental epileptic encephalopathy. (5) PPI analysis showed that NUS1 may be involved in occurrence of neurological diseases such as LGS by affecting protein folding signaling pathways. Conclusion:NUS1 mutant R290C alters its interacting protein lineage and mediates the development of LGS and other neurological diseases probably by regulating protein folding-related signaling.

4.The analysis of gene expression profile and related signal pathways in focal cortical dysplasia

Objective To investigate the potential pathogenesis of Focal cortical dysplasia (FCD), we performed cDNA microarray analysis to obtain gene expression profile of FCD. Methods Three FCD samples and three normal controls were enrolled. Total RNA of the brain tissues were extracted. The difference gene expressions between FCD group and control group was detected using Affymetrix gene chip. The up and down-regulated genes were confirmed by Real-time PCR. Further, the related signal pathways involved in the pathogenic mechanisms of FCD were predicted by bioinformatics. Result In FCD, two up-regulated genes C21orF2 and AU152162 and 5 down-regulated genes ENPP2, ANLN, IP6K3, UGT8, and AZGP were found. Compared the FCD samples with the normal controls , there were significantly different in all down-regulated genes (P < 0.05), while the up-regulated genes were not (P > 0.05). Using bioinformatics analysis, the ENPP2 , UGT8 , and AZGP1 protein which located in the cell membrane or secreted into the extracellular matrix may be involved in the formation of the myelin sheath and the development of the nervous system by the lipid metabolism and LPA signaling pathway. Conclusion ENPP2, UGT8 and AZGP1 may be involved in pathogenesis of FCD through the process of myelin sheath formation and LPA signal pathway , which warrants further study to know their roles in the pathogenesis of FCD.
5.The relationship between trace elements and sperm density and motility in male infertility

Objective To investigate the relationship between semen trace elements and sperm density and motility in male infer‐tile patients .Methods A total of 116 cases of male infertility patients recruited in this hospital from September 2013 to September 2014 were collected and divided into the sperm density anomaly group (59 cases)and sperm motility anomaly group(57 cases) .Other 56 cases of fertile healthy men were enrolled into the control group .Sperm density and motility were analyzed and levels of calcium , phosphorus ,magnesium ,copper ,icon and zinc were detected in seminal plasma .Results Seminal level of calcium ,phosphorus ,zinc and iron in sperm density anomaly group were significantly lower than those in the control group ,while the level of copper was higher than that in the control group ,had statistically significant differences (P<0 .05) .Seminal level of copper in sperm motility a‐nomaly group was higher than that in the control group ,while levels of zinc and iron were lower than those in the control group ,had statistically significant differences (P< 0 .05) .Conclusion Sperm motility and density are significantly correlated with trace ele‐ments levels in seminal plasma .Routine determination of trace elements in seminal plasma in infertile men should be recommended , which could be helpful in fertility assessment and treatment .
6.Disability of Children with Autism Spectrum Disorder in China in 2006

Objective To understand the prevalence situation of disabled children with autistic spectrum disorders (ASD) in China. Methods The data of disabled ASD children aged 0~17 years from the Second National Sampling Survey on Disability was selected and analyzed. Results The prevalence rate of disabled ASD was 0.212 among children aged 0~17 years in China, the prevalence rate of disabled ASD among boys (0.282‰ ) was higher than that among girls (0.132‰ ) (χ2=16.119, P=0.000). The prevalence rate of disabled ASD was higher in younger group than that in elder group (P=0.000). There was no significant difference in the prevalence rate of those children between city and countryside (χ2=0.093, P=0.760). There were 73.3% disabled ASD children received rehabilitation training and service, and 26.7% disabled ASD children didn't. Conclusion Disabled ASD children is a big group in China, and their rehabilitation, education, and care after grow-up are badly in need of social attention.

ObjectiveTo study the outcome of 3 kinds of disabled children. Methods269 disabled children found in 2004 Beijing Disabled Children Sampling, which including 237 children with mental retarded disability, 57 with physical disability and 26 with psychological disability, were followed up in 2007. Results52.32% of mental disabled children, 8.77% of physical disabled, and 15.38% of psychological disabled children would not be seen as "disability" any longer. ConclusionThe disability before 6 years old is a kind of developmental disability, which may be recovery as development.
8.Effect of Vocational Rehabilitation and Related Factors in Adult Mental Retarded Persons in Beijing

Objective To investigate the effect of vocational rehabilitation and related factors in adult mental retarded persons in Beijing.Methods 152 mental retarded adult persons were investigated with Scale of Vocational Rehabilitation Effective for Adult with Mental Retarda⁃ tion. Results 59 (38.82%) persons improved their vocational ability after rehabilitation. The related factors of vocational rehabilitation included the causes of mental retardation and maladaptive behavior. Conclusion It is important for the vacational rehabilitation that the consultants deal with the maladaptive behavior appropriately.
9.Expression, purification and activity analyses of three Bcl-2 family proteins.

Bcel-2 family proteins (Bcl-x(L), Bcl-2, Mel-1 etc.) are key regulators of some life processes, including apoptosis and autophagy. They are currently considered as promising targets for developing new anti-tumor therapies. In our study, the human Bcl-2/Bcl-x(L) chimeric gene and the human/mouse Mel-1 chimeric gene were designed and cloned, and the prokaryotic expression vectors for expressing glutathione S-transferase (GST) fusion proteins and histidine tag fusion proteins were constructed respectively. These two proteins as well as the GST-Bcl-x(L) fusion protein were all successfully expressed in E. coli and subsequently purified. In addition, we measured the binding of these Bcl-2 family proteins to the Bid BH3 peptide by fluorescence polarization-based assay. The dissociation constants (Kd) obtained by us were in general agreement with the data reported in literature. The Kd values of all three proteins with or without the GST tag were almost identical. All these results validate the biological functions of these Bcl-2 family proteins obtained by us. These proteins can be used in the experimental screening of small-molecule regulators of Bcl-2 family proteins in vitro.

10.Effect of acupuncture combined with rehabilitation therapy to improve comprehensive function among patients with stroke in communities

Objective To Btudy effect of standardized acupuncture combined with rehabilitation therapy (function training)on improvement of comprehensive function among patients with stroke in communities,Changning District of Shanghai.Methods One hundred and seventy-six stroke patients were divided into rehabilitation therapy group(88 cases)and control group(88 cages)with block randomization.Patients in rehabilitation therapy group were treated with standardized acupuncture combined with function training in addition to regular medical treatment,and patients in control group regular medical treatment only.All the patients were evaluated with functional comprehensive assessment(FCA)scale by the end of 2-and 5-month of treatment,respectively.Results By the end of 5-month of treatment,scores of FCA in both groups increased significantly (P<0.01),as compared to those before treatment,more in rehabilitation therapy group than that in control group(P<0.05).Conclusions Effect of standardized acupuncture combined with rehabilitation therapy Can obviously improve their comprehensive function among stroke patients in communities.
