1.Effects of treadmill exercise on hippocampal autophagy-induced apoptosis in ovariectomized stressed rats

BACKGROUND:Apoptosis and autophagy imbalance in the hippocampal region of perimenopausal depressed rats are closely related to cognitive decline.Whether aerobic exercise can reduce apoptosis by promoting hippocampal autophagy and thus improve the learning and memory abilities of perimenopausal depressed rats is not clear.OBJECTIVE:To investigate the possible mechanism by which 4-week moderate-intensity treadmill exercise improves learning memory ability in ovariectomized stressed rats.METHODS:Forty Sprague-Dawely rats were randomly divided into four groups,namely,sham operation group(n=10),ovariectomized group(n=10),ovariectomized stress group(n=10)and ovariectomized stress exercise group(n=10).Except for the sham operation group,the ovaries were removed in the other three groups to establish a perimenopausal rat model,and then a depressed rat model was established by chronic unpredictable stress in the latter two groups.The rats in the ovariectomized stress exercise group underwent a 4-week moderate-intensity treadmill exercise.Tail suspension test and sucrose preference test were performed to text depression-like behaviors in rats after exercise and stress.The eight-arm maze experiment was used to test the learning and memory behaviors of rats after exercise and stress.Western blot was used to detect the protein levels of AMP-activated protein kinase/UNC-51 like autophagy activating kinase 1/mammalian target of rapamycin(AMPK/mTOR/ULK1),hippocampus apoptotic factor Caspase-3 and the protein expression of autophagy markers LC-3II/Beclin-1 in the hippocampus.RESULTS AND CONCLUSION:(1)Compared with the sham operation group,rats in the ovariectomized and ovariectomized stress groups had prolonged resting time in the tail suspension test and decreased sugar-water intake and sugar-water preference in the sucrose preference test.(2)Ovary removal reduced the learning memory capacity of rats,as evidenced behaviorally by a significant increase in the number of working memory errors,the number of reference memory errors,and the completion time,and an even more pronounced increase in the above measures in the ovariectomized stress group.(3)Compared with the ovariectomized group,there was a significant reduction in the number of working memory errors,the number of reference memory errors,and the completion time in the ovariectomized stress group.(4)Compared with the sham operation group,in the ovariectomized and ovariectomized stress groups,the expression of hippocampal apoptotic factor Caspase 3 protein was significantly elevated,the expression of autophagy-related factors proteins Beclin-1 and LC3II,as well as the protein expression of AMPK and ULK1,was decreased,whereas the expression of mTOR protein was elevated.Changes in the above indicators were more significant in the ovariectomized stress group.(5)Compared with the ovariectomized stress group,in the ovariectomized stress exercise group,the protein expression of Caspase 3 was significantly decreased,the protein expression of Beclin-1 and LC3II was significantly increased,the protein expression of AMPK and ULK1 was significantly increased,and the protein expression of mTOR was significantly reduced.To conclude,4-week moderate-intensity treadmill exercise may promote cellular autophagy and reduce apoptosis through the AMPK/mTOR/ULK1 autophagy signaling pathway,thereby enhancing the learning and memory capacity of rats with ovariectomized depression
2.Survey of genetic diversity of select tick species in Inner Mongolia

The aim of this study was to understand the internal genetic diversity and population history dynamics of ticks in Inner Mongolia,to provide data for designing effective vector control programs and revealing ticks'transmission mechanisms.From 2022 to 2023,the manual collection method was used to collect samples in Inner Mongolia.The 16S rDNA and COI gene sequences of ticks were used to identify Hyalomma marginatum,Haemaphysalis concinna,and Argas persicus,and analyze the sequence characteristics and genetic diversity within the populations.Base composition analysis indicated that the average A+T content of the 16S rDNA gene and CO I gene in the three ticks was significantly higher than that of C+G.Moreover,22 haplotypes of the COI gene and 12 haplotypes of the 16S rDNA sequence were identified in Hyalomma marginatum.Eleven haplotypes were identified according to the COI gene,and nine haplotypes were identified according to the16S rDNA sequence of Haemaphysalis concinna.Two haplotypes were identified on the basis of the COI gene,and six haplotypes were identified on the basis of the 16S rDNA sequence of Ar gas persicus.The minimum 16S rDNA haplotype diversity was 0.264 for Ar gas persicus and 0.579 for the other two species.The nucleotide diversity of the three tick species was less than 0.05.Tajima's val-ue and Fu's Fs value of the neutrality test were negative.Base saturation substitution analysis indicated that neither of the two genes in the three tick species reached saturation.The phylogenetic tree revealed that Hyalomma marginatum,Haema physalis concinna,and Ar gas persicus in Inner Mongolia independently aggregated into branches.In conclusion,the base content of Hyalomma marginatum,Haemaphysalis concinna,and Argas persicus genes in Inner Mongolia was consist-ent with the characteristics of insect mitochondrial DNA content.Furthermore,the three tick populations showed rapid evolu-tionary population expansion,and the phylogeny of three tick species showed independent aggregation into clades,with no pop-ulation isolation.
3.Papillary cystadenoma of the epididymis: a case report

This article reports a case of a 55-year-old patient who was admitted to the hospital due to an 8-year history of a right epididymal mass and intermittent right scrotal heaviness for 1 month. Preoperative ultrasound examination revealed a right epididymal cyst and right testicular hydrocele. Under general anesthesia, a right epididymal mass resection and right testicular tunica vaginalis inversion procedure were performed. Postoperative pathological diagnosis was epididymal papillary cystadenoma. The patient has been followed up for 1 year with no recurrence and malignancy. Epididymal papillary cystadenoma, a rare benign tumor, lacks specific clinical and imaging features, often leading to misdiagnosis as seminal vesicle cysts or malignant tumors. If associated with von Hippel-Lindau syndrome, bilateral occurrence should be considered to prevent missed diagnosis and treatment delay.
4.Evaluation of the clinical effect of TESSYS technique in treating single-segment LSS based on CT imaging and traumatic stress indicators

Objective:To evaluate the clinical effect of transforaminal endoscopic spine system (TESSYS) technique in treating single-level lumbar spinal stenosis (LSS) based on computed tomography (CT) imaging and traumatic stress indicators. Methods:A total of 112 patients with single-level LSS admitted to The Third Hospital of Qinhuangdao from June 2019 to June 2021 were selected,and they were divided into observation group and control group by random number table method,with 56 cases in each group. The observation group was treated with TESSYS technique,and the control group was treated with "broad easy immediate surgery (BEIS)" technique of intervertebral foramen scope. The indicators of CT imaging of spinal canal area and protrusion ratio,as well as traumatic stress indicators included lactate dehydrogenase (LDH),myoglobin (MYO) and cortisol (COR),of two groups were compared before and 1 week after surgery. Oswestry Disability Index (ODI),Visual Analogue Scale (VAS) score and Japanese Orthopaedic Association (JOA) Scores were compared between the two groups. Excellent rates of treatment and postoperative complications of two groups were compared at the 6 th month after surgery. Results:The average hospital stay and operation time of observation group were respectively (4.29±1.08)d and (93.53±22.01)min,which were significantly shorter than (6.61±1.72)d and (112.29±26.68)min of control group,and the intraoperative blood loss of observation group was (30.15±8.26) ml,which was shorter than that (41.35±11.58) ml of control group,and the differences of them between two groups (t=8.548,4.059,5.892,P<0.05),respectively. At the 1st week after surgery,the ODI and VAS scores of observation group were lower than those of control group,and the JOA score was higher than that of control group,and the differences of them between two groups were significant (t=9.443,t=8.969,6.084,P<0.05),respectively. At the 3rd month after surgery,the ODI and VAS scores of observation group were respectively lower than those of control group,and JOA score of observation group was higher than that of control group,and the differences of them were significant (t=9.706,3.753,5.894,P<0.05),respectively. The rate of excellent and good treatment of observation group was 94.64%,which was higher than that (80.36%) of control group,and the differences of them between the two groups were statistically significant (t=5.225,P<0.05). At the 1st week after surgery,the vertebral canal area of observation group was larger than that of control group,and the ratio of protrusion invasion was less than that of control group,and the differences of them between the two groups were statistically significant (t=3.404,5.578,P<0.05),respectively. At the 1st d after surgery,the serum LDH,MYO and COR levels of observation group were lower than those of control group,and the differences of them between two groups were statistically significant (t=7.570,9.687,5.242,P<0.05),respectively. At the 3rd d after surgery,the serum LDH,MYO and COR levels of observation group were respectively than those of control group,and the differences of them between two groups were significant (t=6.856,9.729,2.744,P<0.05). There was no significant difference in complication rate between the two groups (P>0.05). Conclusion:Compared with BEIS technique,TESSYS technique has the advantages of less trauma,faster postoperative recovery and higher excellent and good rate in treating single-segment LSS,which can alleviate stress reaction of early postoperative trauma. Its comprehensively clinical efficacy is ideal.
5.Cultivation techniques and research progress of porcine intestinal organoids

Porcine intestinal organoids are 3D cultures derived from porcine stem cells or porcine intestinal crypts containing stem cells,which can self-renew and differentiate into tissues similar to those in vivo.Due to the high structural and physiological similarity between pigs and humans,porcine intestinal organoids can not only meet the needs of agricultural production and ecological environment research,but also serve as an in vitro model for human drug screening and toxicology studies.Compared with other in vitro models,porcine intestinal organoids have the advantages of being similar to in vivo tissues,stable phenotypes and genetic properties.Porcine intestinal organoids show great potential in simulating host-microbial interactions,drug screening and nutritional development studies,but their construction still lacks a fixed process,which affects experimental repeatability.In this review,we will review culture of porcine intestinal organoids,the exposure method of apical villi,the interactions between microorganisms and hosts,and the current challenges.

8.Finite element analysis of stress distribution on mandibular All-on-4 implant fixed denture with different occlusion

BACKGROUND:The long-term effect of All-on-4 implant fixed denture is closely related to biomechanics.Its occlusion design affects the stress distribution of every part of the restoration.However,there is limited and inconclusive biomechanical research on All-on-4 implant fixed denture both domestically and internationally so far.OBJECTIVE:To analyze the effect of different occlusal contacts on the stress distribution of mandibular All-on-4 implant fixed denture.METHODS:A model of mandibular All-on-4 implant fixed denture was established.By changing the loading force,direction,and position,10 loading conditions were set up to simulate centric occlusion,laterotrusive occlusion,and protrusive occlusion.The stress distribution and maximum stress values of every part of the model in different loading conditions were analyzed and compared among each group of models by loading different tooth positions to simulate different occlusal contact types.RESULTS AND CONCLUSION:(1)The stress distribution cloud diagrams of each group of models were basically similar.The stress of the implant was mainly concentrated at the implant neck.The stress of the surrounding bone tissue was mainly concentrated around the implant neck,and the stress of the cortical bone was significantly higher than that of the cancellous bone.The stress of the abutment was mainly concentrated at the connection between the abutment and the implant and the abutment and the upper restoration.The stress of the titanium bracket in the upper restoration was mainly concentrated at the connection between the abutment and the titanium bracket,and the stress of the resin surface in the upper restoration was mainly concentrated on each loading point.(2)In simulating centric occlusion,the maximum stress value of the implant significantly increased when the cantilever was not loaded.It indicates that the mandibular All-on-4 implant fixed denture can have occlusal contact in the distal cantilever during centric occlusion.(3)In simulating laterotrusive occlusion,the maximum stress values of every part of the model significantly increased when simulating canine-guided occlusion.The maximum stress value of the simulated laterotrusive balanced occlusion was slightly lower than that of the simulated group functional occlusion.It indicates that group functional occlusion can be selected in the permanent prosthesis after osseointegration and laterotrusive balanced occlusion can be selected in the provisional restoration of mandibular All-on-4 implant fixed denture.(4)In simulating protrusive occlusion,the maximum stress values of every part of the model significantly increase when only the front tooth is loaded.In simulating protrusive balanced occlusion,the maximum Von-Mises stress value is relatively small.It indicates that the protrusive occlusion of mandibular All-on-4 implant fixed denture can choose protrusive balanced occlusion.
